The objective of the present work was to determine the corresponding uptake and assimilation of ${NO_3}^-$ in roots and shoots of leafy radish by applying of mixed amino acids (MAA). The amino acids used in this experiment were alanine (Ala), ${\beta}-alanine\;({\beta}-Ala)$, aspartic acid (Asp), asparagines (Asn), glutamic acid (Glu), glutamine (Gln), and glycine (Gly). Leafy radish was grown by conventional fertilization with macro- and micronutrients under controlled conditions. The 15-day-old seedlings were treated 0, 0.3 and 3.0 mM of MAA containing 5 mM ${NO_3}^-$ in growth medium. Nitrate uptake was determined by following ${NO_3}^-$ depletion from the uptake solution. The activity of the enzymes related to the process of ${NO_3}^-$ reduction (NR: nitrate reductase; NiR: nitrite reductase; GS: glutamine synthetase) and the content of ${NO_2}^-\;and\;{ND_3}^-$ were analyzed in shoots and roots. The results of this study showed that ${NO_3}^-$ uptake was inhibited 38% with treatment of 0.3 mM of MAA. However, there was more than three times increase of N03- uptake in 3.0 mM MAA. In addition, the enzymatic activities were positively affected by the high MAA rate. Finally, the ${NO_3}^-$ content was increased slightly both in shoots and roots of leafy radish by MAA treatments.
To develope a technique for efficiently managing fertilizer for cucumber, a quick test method to quantify nitrate content in soil solution and leaf petiole juice using a simple instrument was investigated. Among the nitrate analyzing instruments such as compact ion meter, nitrate ion meter, and test strip with reflectometer, the paper test-strip used in conjunction with a hand-held reflectometer was most closely correlated with ion chromatography method in nitrate content, and then it would be suggested with a tool that a farmer can use rapidly, conveniently and accurately for nitrate analysis in a field. Nitrate content in soil solution collected by porous cup was very variable on the lapsed time after drip irrigation and the sampling positions such as soil depth and the distance from dripper. As a result, a significant correlation between nitrate contents of soil solutions and 2M KCl soil extract was not found. However, nitrate content in soil solution extracted with a volume basis (soil:water=1:2) showed the highly significant correlation with that in 2M KCl extract. Nitrate contents of cucumber leaf petiole juices was greatly different between upper and lower leaves. Eleven to sixteen positioned-leaf would be a proper sampling position to determine nitrate content in leaf petiole for evaluating nutrient state by plant tissue analysis. From the secondary regression equations between nitrate contents of soil and petiole juice and the yield of cucumber, nitrate levels for real time diagnosis were estimated as $400mg\;l^{-1}$ soil solution by porous cup. $300mg\;l^{-1}$ in a soil volume extraction, and $1400mg\;l^{-1}$ in petiole juice from spring to summer season. In addition, the maximum yield of cucumber fruit in pot test was obtained in nitrate $1500mg\;l^{-1}$ level of petiole juice, which was similar to nitrate $1400mg\;l^{-1}$ in greenhouse trial.

This study was designed to investigate the content of inorganic nitrogen($NH_4$, $NO_3$) and its change in the 7 different sites; the permanent meadows of alfalfa, orchardgrass, and reed canarygrass, the fields of rye. corn, rice and a mountain region. the results from this study were summarized as follows; 1. The contents of ammonium and nitrate in meadows were 15.89-16.46 and 1.32-1.86ppm, respectively, showing the lower level than in other sites and no significant diffirences among three meadows. 2. In the rice field, the average content of ammonium(7.19ppm) was the highest among all sites, but the content of nitrate(1 l.l7ppm) was the lowest. In addition, almost all of nitrate were spreaded around the surface. 3. In the mountain region, the content of $NH_4$(5.79ppm) was slightly high, but the content of nitrate ( 15.18ppm) was very low. 4. In the rye field. the average content of $NO_3$ was 35.46ppm and show the highest (92.63ppm) at the deep part of soil in the specific season, but it decreased with increasing temperature. 5. In the corn field, which kept bare after harvesting the corn silage, the content of $NO_3$ was high, regardless of seasons and depths, and increased up to maximum of 103ppm. 6. The ground water concentrations of $NO_3$ were not more than 50ppm in the pasture. In the vegetable house, they were less than 30 ppm during the winter and spring and were 80-100ppm during the summer and fall. 7. The concentrations of $NO_3$ in Han river water were lower during the winter and spring(20-30ppm) and higher dxring the summer and fall (50-90ppm).
High rates of nitrogen fertilization dangerously increase the nitrate content of vegetable crops, and the accumulation of nitrate in edible crops is undesirable because of potential risks to human health. Micronutrient solution containing Cu, Mn, Mo, Zn was tested for the suppression of nitrate accumulation in lettuce grown in pots treated with Mg fertilizer under a greenhouse condition. The micronutrient solution was sprayed on leaves at 3 and 4 weeks after transplanting of 20-day old seedlings. Plants were harvested after 5-week growth, and yield, contents of chlorophyll, sugar, micronutrient and nitrate, and also nitrate reductase activity were measured. Fresh weight of lettuce was significantly increased by the application of Mg and micronutrients, and the effect was the most significant in the Mg+micronutrient treatment. Also contents of chlorophyll and micronutrients were higher in the plants of micronutrient treatments. Contents of nitrate were reduced by about 14-18% in lettuce with Mg and/or micronutrient applications. Compared to the plants of control treatment, nitrate reductase activity was also higher in those plants treated with micronutrients, and in the treatment of Mg+micronutrients the enzyme activity was six times as high as that of control treatment. Although the effect of mineral nutrients on the suppression of nitrate accumulation in lettuce was relatively small in this study, an appropriate supply of mineral nutrients could be one of the solutions for the nitrate accumulation in vegetables.
Suppression of nitrate accumulation in spinach and lettuce through foliar application of chitosan formula containing micronutrients is related with the increase of the nitrate reductase (NR) activity. If NR in spinach were highly expressed to increase the assimilatory activity, nitrate content could be reduced. For this, NR cDNA was cloned from the isolated mRNAs of spinach using reverse transcriptase-PCR. Nucleotide sequence of cloned spinach NR cDNA showed highly deduced amino acid sequence identity ($71{\sim}82%$) with other known plant NR genes. Only two nucleotide-base differences were observed in the cloned NR cDNA compared with that of the published spinach NR cDNA.
Studies on the effect of soil moisture, different seasons, harvesting periods and nitrogen application on the nitrate content of radish(Raphanus sativus L. var. niger(Mill.) S. Kerner) were carried out in pot and green house. The results are as follows; The low level of soil moisture leads to an increased nitrate accumulation in radish root. The nitrate content was found to be higher in spring crops and lower in summer ones. It was decreased during harvesting period. The application of nitrogen fertilizer increased the nitrate content in radish root. We found the highest content in petioles with lesser content in roots and leaf blades in that order. The multiple regression analysis and the nutritional value of nitrate content in radish were discussed.
The amount of nitrate in the tobacco leaf has been shown to be correlated with the levels of alkaloids and nitrosamines. Also the nitrate content of the tobacco correlated closely with the smoke delivery of nitric oxide and tobacco-specific nitrosamines (TSNAs). These are related with the effect of the reconstituted tobacco leaf(Recon) using the tobacco stems. Adsorption process is gaining interest as one of the effective processes of advanced liquid treatment for liquid containing unnecessary materials. This study is focused on the evaluation of four anion exchangers, a cation exchanger and an activated carbon, as adsorbents for reduction of nitrate anion from Recon extract. In order to analyze the nitrate anion, the IC method used in this work was carried out with a Dionex ICS-2000 system. The effects of dosages of adsorbents and concentration of extract on the removal of nitrate anion were examined. Experimental results showed that for nitrate-anion exchanger, nitrate-cation exchanger and nitrate-activated carbon adsorption system, approximately 70 %, 10 %, and 4 % removal efficiencies were achieved at the Brix 10 and the 20 % addition. Although the activated carbon was little efficient for removal of nitrate ion, the removal of nicotine was very efficient at given conditions.
This study was conducted to determine the physiological differences betweenhealthy and wilted plants with respect to nitrate assimilation and ascorbic acid content. Wilting was artificially induced in spinach plants by treating the seeds with nutrient solution containing high nitrogen and low potassium. The plants were cultured in different plots 4 types of media: 1N-1P-1K (control), 6N-1P-0K (0K), 6N-1P-0.5K (0.5K), and 6N-1P-2K (2K). The rate of wilting among the plants was as follows: control, 0%; 2K, 10%; 0.5K, 40%; and 0K, 70%. This shows that under high nitrogen conditions, the lower the amount of potassium provided, higher was the rate of wilting. There were no differences in plant growth among the plants treated with different levels of potassium under high nitrogen conditions. The nitrate content in both the leaves and the roots was higher in plants grown under high nitrogen media than those in the control. Furthermore, the nitrate level decreased with increasing potassium concentration. The ascorbic acid content of spinach under high nitrogen conditions was lower than those of the control.

The aim of this study was to investigate dry matter productivity and nitrate nitrogen content in the growth stages of barnyard millet (Echinochloa esculenta) cultivated for feed, which was treated with different nitrogen fertilization levels. An early variety of barnyard millet (cv. Shirohie) was used for the test, and the different treatments with nitrogen fertilizer were as follows: 50% (N-40 kg/ha, T1), 100% (N-80 kg/ha, reference amount, T2), 150% (N-120 kg/ha, T3), 200% (N- 160 kg/ha, T4), 250% (N-200 kg/ha, T5), and 300% (N-240 kg/ha, T6). Sowing was done on May 13, 2021 and plants were harvested for four stage; vegetative stage, elongation stage, heading stage, and milk stage. The length of the millet increased significantly as the amount of nitrogen fertilization increased during the harvest period (p<0.05), but the difference was insignificant during the milk stage (p>0.05). Moreover, barnyard millet dry matter yield increased significantly as the levels of nitrogen fertilization increased (p<0.05), but there was no significant difference in dry matter yield among nitrogen fertilization levels during the heading stage (p>0.05). Chlorophyll also was significantly higher in T5 (250%) at all harvesting times, whereas nitrate nitrogen content was highest at the vegetative stage, gradually decreased as growth progressed, and lowest at the milk stage. Finally, as the nitrogen fertilization levels increased, the nitrate nitrogen content was significantly higher in all treatment groups (p<0.05). Therefore, our results suggest that the most appropriate nitrogen fertilizer levels is between 150%-200%, considering the dry matter yield, feed ingredients and nitrate nitrogen content in barnyard millet for feed.
Methyl methacrylate-butyl methacrylate copolymer (MMBM)-povidone (PVP) films were investigated as a potential topical drug delivery system for the controlled release of econazole nitrate as a model drug. The effect of changes in film composition, drug concentration, film thickness, pH and temperature of release medium on the in vitro release of econazole nitrate were studied. The release rate constant was found to be increased with increasing povidone content in dry films. Drug release followed zero-order kinetics in the initial stage and then release rate increased gradually with time, espicially in the films having larger proportions of PVP. The release rate was found to be dependent on drug content, film thickness, the pH and temperature of release medium. Antimicrobial test showed that microbial growth was inhibited markedly with increasing proportions of PVP in films. Also drug content and film thickness affected the antimicrobial activity.
