일장 및 온도조건에 따르는 잎담배 품종간 반응 (Varietal response in tobacco under photoperiodic and temperature Conditions)

  • 1) 본 실험은 Y.S.A. Gold leaf, oxford 1, Awha Yungzung, Samsun, Maruha, Hatano, 등 8개 품종을 온도와 일장 반응으로 본 것이다. 2) 온도는 저온을 야외(10~$20^{\circ}$) 고온을 온실($20^{\circ}C$~$35^{\circ}C$)에서 맞추었으며 일장은 8.10.12.14로 처리했다. 3) 이를 요약하면 1) 단일 조건에서는 고온보다 저온에서 장일조건에서는 저온보다 고온에서 개화가 촉진되었다. ii) 대체로 고온 장일에서 보다는 저온 단일에서 다소 개화가 촉진 되었다. iii) 그러나 위와 가튼 경향은 품종간에 많은 차이를 볼 수 있다.

감마선에 대한 고구마 Peroxidase 형질전환 담배식물체의 반응 (Responses of Transgenic Tobacco Plants Expressing Sweet Potato Peroxidases to Gamma Radiation)

  • 고구마 산성 peroxidase (POD) (swpal) 또는 중성 POD(swpnl)을 도입한 형질전환 담배에 다양한 선량의 감마선을 조사한 후 30일에 항산화효소 활성과 식물생장에 미치는 영향을 조사하였다. 감마선은 조사선량에 비례하여 형질전환 식물체와 정상식물에 관계없이 모든 식물체의 생장을 크게 억제시켰다. 50-70 Gy 처리에서 식물체의 신장이 크게 억제되었고 새로운 잎이 발생되지 못하였다. 10-50 Gy의 조사선량에서는 POD, superoxide dismutase, catalase 활성에 큰 변화를 주지 않았다. 이러한 결과는 고구마 POD는 감마선 조사에 의해 유도되는 산화적인 스트레스에 대한 보호효과에 관여하지 않음이 시사된다.

담배 성분에 관한 연구 -니코틴 함량 변화에 미치는 K, Ca 및 Mg의 조성 비율- (Studies on the Tobacco Alkaloids -Influence of K, Ca and Mg on Nicotine Contents-)

  • Tobacco, which has nicotine for its main component, has been in medical use for a long time and in great demand for smoking throughout the world. The purpose of this study is to control nicotine amount and to increase yield more efficiently by the method of systematic variations. Nutrient solutions for tobacco culture were designed and prepared in 10 kinds with mineral ions: $NO_{3^-},\;SO_{4^{--},\;PO_4{^{-3},\;K,{^+}\;Ca,{^{++}\;Mg,{^{++}$, Mn, B, Cu, Zn, Mo, and Fe. Nicotiana tabacum L. Yellow Special A, grown for 50days from sowing, was replanted in plastic pot and cultured for 65 days supplying with prepared nutrient solutions. After harvest, their nicotine amount was determined by means of acid-base titrimetry and gas chromatography. The tobacco plants in KCa 4 and KMg 9 groups demonstrated the highest yield in total leaves weight and KMg 7 group in average nicotine amount. They have shown the increase of nicotine amount from lower leaf to 16th leaf, and thereafter decreased gradually. The author also pursued the optimum ionic proportions for the absolute nicotine amount in tobacco by means of systematic variations method.

Partial Biological and Molecular Characterization of Tomato yellow fruit ring virus Isolates from Potato

  • Eight potato-producing provinces of Iran were surveyed during the growing seasons of 2004-2006 to detect the presence of Tomato yellow fruit ring virus (TYFRV), a tentative species in the genus Tospovirus. A total of 1,957 potato leaf samples were collected from plants with tospovirus-like symptoms of chlorotic or necrotic spots, chlorosis and necrosis. The samples were tested by enzyme-linked immunosorbent assay using TYFRV-specific antibodies. Among those tested, 498 samples (25.4%) were found to be infected with the virus. The virus was detected in 72.4% of the potato fields in all provinces surveyed. Thirteen potato isolates of TYFRV were selected for further biological and molecular studies. Based on their reactions on Nicotiana tabacum plants, the isolates were separated into two groups, namely L (local infection) and N (systemic infection). The nucleotide sequences of the nucleoprotein (N) genes of the isolates were determined and compared with the homologous sequences in Genbank. No recombination evidence was found in the isolates using different recombination-detecting programs. In the phylogenetic tree, the potato isolates fell into two major groups: IRN-1 and IRN-2 corresponding to the two biologically separated groups. This study shows for the first time the biological and phylogenetic relationships of geographically distant TYFRV isolates from potatoes in the mid-Eurasian country of Iran.

순무 모자이크 바이러스(TuMV)의 새로운 기주식물 탐색 (New Host Plants of Turnip Mosaic Potyvirus in Korea)

  • Turnip mosaic potyviruses (TuMV) were isolated from Rorippa indica and Armoracia lapathifolia showing mosaic symptoms in field. Identification of the TuMVs were carried out by host reactions of indicator plants, electron micrograph, serological properties and reverse transcription-poly-merase chain reaction (RT-PCR). Both viruses systemically infected Chenopodium quinoa, Nicotiana clevelandii, Brassica rapa, B. campestris subsp. pekinensis, B. juncea and Raphanus sativus, and developed local infection on inoculated leaves of C. quinoa, C. amaranticola, C. album, N. tabacum cv. Xanthi nc and Gomphrena grobosa. However, the viruses did not infect on N. glutinosa, Cucumis sativus and Vigna unguiculata. The filamentous particles, about 720 nm in length, and inclusion bodies were observed from the infected leaf tissues by dipping on electron microscopy. Crude sap of leaf infected with the viruses was reacted positively with an antiserum of TuMV in agar gel double diffusion. For detection of the viruses, RT-PCR was carried out with TuMV--specfic oligonucleotide primer. The RT-PCR products, a 1,092 bp DNA fragment, were obtained from naturally infected leaves of R. indica and A. lapathifolia. In inoculation test to seven cruciferous weeds with TuMV, infection occurred in Arabis glabra, Barbarea orthoceras, Capsella bursa-pastoris, Draba nomorosa var. hebecarpa, Rorippa cantoniensis and Thlaspi arvense.

담배, 토마토 및 고추에서 분리한 TMV 계통의 추가적인 생물학적 특성 (Additional Biological Characteristics of TMV Strains Isolated from Tobacco, Tomato and Pepper Plants)

  • Tobacco (Nicotiana tabacum) cultivars including NC 82 and KF 114, and Datura stramonium, Physalis floridana, Gomphrena globosa, and Chenopodium spp. were added to the previous host plants tested for the further examination on the biological characteristics of tobacco mosaic virus (TMV) strains isolated from tobacco (TMV-Common), tomato (TMV-Tomato), and pepper (TMV-Pepper), In TMV-Tomato and TMV-Pepper, different symptoms were noted in P. floridana (no symptom development), and NC 82 (local lesion production on the inoculated leaves) from TMV-Common with which systemic mosaic symptoms were developed. Local lesions were developed in KF 114, D. stramonium, G. globosa, and Chenopodium spp by TMV-Common and TVM-Tomato, while no symptom was observed in KF114 and G. globosa. Also the number and size of local lesions were smaller in KF 114 than Xanthi-nc tobacco (local lesion host) infected with TMV-Tomato. Systemic necrosis was induced in Xanthi-nc and KF 114 when infected with TMV-C at high temperature, but not with the other strains.

Characterization of Cucumber mosaic virus Isolated from Water Chickweed(Stellaria aquatica)

  • A strain of Cucumber mosaic virus (CMV) was isolated from a weed, water chickweed (Stellaria aquatica), growing in the pepper field in Chunchon, Korea. This isolate, CMV-Sa, was differentiated from other CMVs based on biological properties and nucleotide sequence analysis of the coat protein (CP) gene. CMV-Sa showed different reactions to all the tested plants, except Capsicum annuum and Cucumis sativus, when compar-ed with those of CMV-Mf (subgroup I) and CMV-PaFM (subgroup II). Remarkably, in Nicotiana tabacum cvs. Samsun, Xanthi-nc and Ky-57, CMV-Sa induced local necrotic ring spots on the inoculated leaves and venal wave pattern and mosaic on the upper leaves. RNA analysis, serology, and RT-PCR of CP gene showed that CMV-Sa belonged to subgroup I of CMV. However, restriction enzyme analysis of the cDNA using AluI, HhaI, HincII, HindIII, HinfI and MspI showed that CMV-Sa was distinct from that of CMV-Mf. Based on comparison of the nucleotide of CP gene and deduced amino acid sequences between other CMV strains, CMV-Sa was closely related to CMV-Mf with 93.7% and 97.2 % identity, respectively.

버어리종 잎담배의 건조과정중 암모니아 함량 변화

  • This study was carried out to investigate the effects of curing methods on the concentration of ammonia during curing in burley tobacco leaves. The air-cured tobacco(KB108; Nicotiana tabacum L.) was grown at Chonju Tobacco Experiment Station in 1998 and the tenth leaves from the top on the stalk were harvested. Half of the harvested leaves were cured in normal air curing facility and the other leaves were cured in excessive curing facility. Stalk cut tobaccos were cured in horizontal curing facility. The leaves were sampled every five days from harvesting time to the end of curing(25 days). Ammonia concentration of leaves increased during curing period with a remarkable increase at yellowing stage. The concentration of ammonia was high in the primed cured leaves, while that of the excessive cured leaves was low. It is considered that the lower increase of ammonia in stalk cured leaves may be caused by the translocation from the leaves to the stalk during curing, while that of excessive cured leaves may be caused by the poor decomposition of protein and amino acid during curing by excessive moisture loss and high temperature condition.

Effects of Storage Temperature and Humidity on Germinability and Longevity of Primed Tobacco Seeds

  • Tobacco seeds (Nicotiana tabacum L. cv KF109) were primed in the polyethylene glycol 6000(PEG) solution and then stored at 5 and $25^{\circ}C$ under 40, 60 and 80% relative humidity (RH) conditions for six months. The effect of storage temperature and humidity on mean germination time ($T_{50}$), longevity and germination of the primed tobacco seeds were compared. Untreated seeds (control) stored at $5^{\circ}C$ showed high germinability throughout the entire storage period and humidity, and a decline in germinability showed after 6 months at 60% RH and after 3 months at 80% RH when stored at $25^{\circ}C$, Primed seeds retained high germinability until 6 months at 60% RH and 3 months at 80% RH when stored at $5^{\circ}C$ but showed a significant decline in germinability after 3 months at 40% RH, and 1 months at 60% and 80% RH, respectively when stored at $25^{\circ}C$, Primed seeds were completely lost viability when stored at $25^{\circ}C$ under 60% RH for 6 months and under 80% RH for 3 months.

담배잎의 노화과정에 따른 단백질의 생화학적 변화 (Biochemical Changes of Protein during the Senescence of Tobacco Leaf)

  • 담배 생육단계별 RNA, protease 활성도와 단백질 패턴의 변화를 파악하여 노화가 진행되는 과정에서 생리 ㆍ생화학적인 변화의 기초자료를 얻고자 본 실험을 수행하였으며 결과는 다음과 같다. 가용성 단백질 함량은 출엽 후 15일까지 증가하여 출엽 후 35일까지 일정하게 유지하였다. 총 RNA 함량은 출엽 후 15일에 가장 높았으며 출엽후 30일까지 급격한 감소를 보였다. Protease 활성 변화는 중성 protease (pH 7.8)가 활성이 높았으며 노화말기인 출엽 후 50일부터 갑자기 증가하였다. 전기영동 패턴은 큰 변화가 없었으나 61.0kd의 polypeptide은 출엽 후 35일부터 생성되어 노화말기까지 증가하였다.
