The Effect of Distinctiveness of stimulus and Partial Retrieval on Memory (자극의 구별성과 부분 인출이 기억에 미치는 영향)

  • The present study is designed to investigate the effect of perceptive, emotional and semantic distinctiveness on retrieval-induced forgetting(RIF). Experiment 1 was designed to construct a category and category list for RIF experimental paradigm and to investigate the effects of perceptual distinctness on retrieval-induced forgetting. It was used for the list consisting of the six categories and six words in each category list. In controlled conditions, all the stimuli were presented in black and Gothic. In contrast, perceptual distinctiveness conditions, half of the category list were presented in red and Gungseoche. RIF was observed in all conditions. Experiment 2 was designed to investigate the effects of semantic and emotional distinctiveness on retrieval-induced forgetting. In neutral conditions, adjectives related to items were added. In the emotional distinctiveness condition, half of the items in the category were manipulated in such a way as to add the negative adjectives. In the semantic distinctiveness condition, half of the items in the category were manipulated in such a way as to add the inappropriate adjective. As a result, RIF occurred in the neutral condition, but RIF did not occur in both the emotional discrimination condition and the semantic discrimination. These results suggest the possibility that the RIF will not occur when the distinctiveness occurs within a categorical relationship.

Inhibition of Tyrosinase by Metabolites Originating from Thrichoderma atroviride (Thrichoderma atroviride 배양액의 tyrosinase 억제제에 관한 연구)

  • In today's society, functional whitening cosmetics are important to beauty. Fungi are known to produce a variety of whitening-related metabolites. In this study, we searched for tyrosinase inhibitors with metabolic products derived from Trichoderma atroviride supernatant in order to apply a material for whitening functional cosmetics. In addition, the inhibitory effect was compared to arbutin, which has already been approved as a whitening raw material by the Korea Ministry of Food and Drug Safety (KMFDS). The metabolites from the T. atroviride supernatant showed higher tyrosinase inhibitory activity than that of arbutin. Some of the tyrosinase inhibitors were stable to heat, whereas some were unstable. The heat unstable material was exhibited in the case of samples treated with little amounts, such as 0.02~0.2%. They were very unstable in acidic and alkali pHs, especially under acidic conditions. However, it was found that a weakly-acidic to neutral pH range was the optimal working pH, especially neutral pH. Since the activity of the inhibitory substances in the T. atroviride supernatant was maintained regardless of proteinase K treatment, it was assumed that the metabolites, but not the bioactive peptides, were involved in the activity. In summary, we propose that the metabolites derived from T. atroviride supernatant have strong potential as whitening raw material.

Antimicrobial Activity of Pectin hydrolysate and its Preservative Effect (펙틴분해물의 항균특성과 식품보존효과)

  • Pectin hydrolysate prepared from citrus pectin by enzymatic hydrolysis has antimicrobial activity. The antimicrobial activity was increased by its hydrolyzing rate and it was rapidly increased after 70% hydrolysis of the pectin. The antibacterial activity of pectin hydrolysate against Escherichia coli ATCC 11229 was the strongest at pH 4.9~5.5, but it diminished slightly at neutral pH values. The antibacterial activity of pectin hydrolysate was stronger than those of against molds and yeasts. The growth of bacteria submitted to this test except lactics was completely inhibited for 48 hrs at $35^{\circ}C$ by adding 2.0~3.0% pectin hydrolysate. While the growth of Lactococcus lactis ATCC 19435 and Lactobacillus bulgaricus and Penicillium funiculosum ATCC 11797 were reached about 60~70% compared with those of the controls in the same condition. But there was no significant effect on the growth of the yeasts. The antibacterial effect of pectin hydrolysate was significantly stimulated by addition of glycine, ethanol, sodium ascorbate, sodium chloride and sodium acetate. The shelf life of Kimchi containing 1.0% pectin hydrolysate was prolonged above 15 days at $4^{\circ}C$ than that of its control. In case of whitish bean jam viable cell counts were inhibited about 2 log cycles by 10 days at $25^{\circ}C$. According to these results, author can sincerely suggest that pectin hydrolysate will be used as a natural food preservative for inhibition of common bacterial growth without inhibition of lactics and yeasts.

Studies on the Cytotoxicity and Antitumor Activity of Perilla frutescens (소엽의 세포독성 및 항암작용에 관한 연구)

  • The cytotoxic and antitumor activity of Perilla frutescens extract on cultured 3T3 fibroblast and skin melanoma cells were evaluated by tetrazolium MTT (MTT) and neutral red (NR) colorimetric assay methods. Lactate dehydrogenase activity was also measured. The light microscopic study was carried out to observe morphological changes of cultured mouse fibroblast and skin melanoma cells. The results were as follows: 1. Water and ether extracts showed a significant cytotoxicity in 3T3 fibroblast and all extracts exhibited a significant anti-tumor activity in skin melanoma cells. Methanol, ethyl acetate and ethanol extracts showed low cytotoxic effects, but exhibited a high anti-tumor activity. 2. The MTT absorbance in 3T3 fibroblast was significantly decreased by treatment with ether, water, chloroform and ethanol extracts and skin melanoma cells was significantly decreased by treatment with all extracts. The difference in MTT absorbance in two cell types was most remarkable when treated with methanol and ethanol extracts. 3. Methanol and ethyl acetate extracts showed the strongest effect in growth inhibition of melanoma cells. These results indicated that methanol extract possessed a low cytotoxicity and a strong anti-tumor activity.

Possible Anticancer Activity of Rosuvastatine, Doxazosin, Repaglinide and Oxcarbazepin

  • Background: Rosuvastatine, doxazosin, repaglinide and oxcarbazepin are therapeutic drugs available in the market for the treatment of different diseases. Potential to display antitumor activities has also been suggested. The aim of the current study was to evaluate their in vitro effects on some human transformed cell lines. Materials and Methods: Cytotoxicity of the four drugs was tested in MCF-7, HeLa and HepG2 cells by the neutral red assay method and also the effect of rosuvastatine and doxazosin against Ehrlich Ascities Carcinoma Cells (EACC) by trypan blue assay. Results: Rosuvastatine exerted the greatest cytotoxic effect against HepG2 cells with an $IC_{50}$ value of $58.7{\pm}69.3$; in contrast doxazosin showed least activity with $IC_{50}=104.4{\pm}115.7$. Repaglinide inhibited the growth of both HepG2 and HeLa cells with $IC_{50}$ values of $87.6{\pm}117.5$ and $89.3{\pm}119.5$, respectively. Oxcarbazepine showed a potent cytotoxicity against both HeLa ($IC_{50}=19.4{\pm}43.9$) and MCF7 cancer cells (($IC_{50}=22{\pm}35.7$).On the other hand the growth of EACC was completely inhibited by doxazosine (100% inhibition) while rosuvastatine had weak inhibitory activity (11.6%). Conclusions: The four tested drugs may have cytotoxic effects against hepatic, breast and cervical carcinoma cells; also doxazosine may inhibit the growth of endometrial cancer cells. Further investigations in animals are needed to confirm these results.

Antiplasmodial and Cytotoxic Activities of Toad Venoms from Southern Amazon, Brazil

  • The drug-resistance of malaria parasites is the main problem in the disease control. The huge Brazilian biodiversity promotes the search for new compounds, where the animal kingdom is proving to be a promising source of bioactive compounds. The main objective of this study was to evaluate the antiplasmodial and cytotoxic activity of the compounds obtained from the toad venoms of Brazilian Amazon. Toad venoms were collected from the secretion of Rhinella marina and Rhaebo guttatus in Mato Grosso State, Brazil. The powder was extracted at room temperature, yielding 2 extracts (RG and RM) and a substance ('1') identified as a bufadienolide, named telocinobufagin. Growth inhibition, intraerythrocytic development, and parasite morphology were evaluated in culture by microscopic observations of Giemsa-stained thin blood films. Cytotoxicity was determined against HepG2 and BGM cells by MTT and neutral red assays. The 2 extracts and the pure substance ('1') tested were active against chloroquine-resistant Plasmodium falciparum strain, demonstrating lower $IC_{50}$ values. In cytotoxic tests, the 2 extracts and substance '1' showed pronounced lethal effects on chloroquine-resistant P. faciparum strain and low cytotoxic effect, highlighting toad parotoid gland secretions as a promising source of novel lead antiplasmodial compounds.

  • In this study, tin phosphate glass system($SnO_2-(1-x)P_2O_5-xB_2O_3$) that occur during the melting of the metal oxide inhibition of the oxidation reaction, and to reduce oxides of high melting temperature in the following three methods were melting. The first is the general way in the atmosphere, and the second by injecting $N_2$ gas under a neutral atmosphere, and finally in the air were melted by the addition of a reducing agent Melt in the atmosphere when the oxidation of the metal oxide is inhibited by low temperatures were melting. In addition, the deposition of crystals within glassy or inhibit devitrification phenomenon is also improved over 80% transmittance. This phenomenon, when the melting of glass, many of $Sn^{4+}$ ions are reduced to the $Sn^{2+}$ was forming oxides SnO, because it acts as a modifier oxide.

Purification and Characterization of Fibrinolytic Enzyme from Tricholoma saponaceum (II) (할미송이버섯으로부터 혈전용해효소의 정제 및 특성 연구 (II))

  • Fibrinolytic enzyme (FE-2) was purified from the fruiting bodies of Tricholoma saponaceum using DEAE-Cellulose chromatography and Mono-S column chromatography, The enzyme has a molecular weight of 18.23 kDa and include Zn$^{2+}$ ion as found by ICP/MS. The N-terminal amino acid sequence of the enzyme was A-L-Y-V-G-X-S-P-X-Q-Q-S-L-L-V It has a pH optimum at pH 7.5, suggested that FE-2 was a neutral pretense. The activity of FE-2 was highly inhibited by EDTA and 1,10-phenanthroline, indicating that the enzyme is a metalloprotease. The activity of FE-2 was increased by $Mg^{2+}$, Zn$^{2+}$, Fe$^{2+}$, and Co$^{2+}$, but the enzyme activity was totally inhibited by Hg$^{2+}$. No inhibition was found with PMSF, E-64, pepstatin and 2-mercaptoethanol. The enzyme hydrolyzed both $A\alpha$ and B$\beta$ chains of human fibrinogen. The $\gamma$ chain was resistant to hydrolysis by FE-2.

It shows the maximum fibrinolytic activity at $55^{\circ}C$, is completely inactivated above at $65^{\circ}C$.

Effect of silver nanoparticles on the performance of riverbank filtration: Column study (강변여과에서의 은나노입자의 영향 : 실험실규모 컬럼 실험)

  • Soil column experiments were evaluated effects of silver nanoparticles (i.e., 0, 2.5, 5, and 10 mg/L) on the microbial viability which is strongly associated with the degradation of organic matter, pharmaceutically active compounds(PhACs) and biological oxidation of nitrogenous compounds during river bank filtration. The addition of silver nanoparticles resulted in almost no change in the aqueous matrix. However, the intact cell concentration decreased with addition of silver nanoparticles from 2.5 to 10 mg/L, which accounted for 76% to 82% reduction compared to that of control (silver nanoparticles free surface water). The decrease in adenosine triphosphate was more pronounced; thus, the number and active cells in aqueous phase were concurrently decreased with added silver nanoparticles. Based on the florescence excitation-emission matrix and liquid chromatograph - organic carbon detection analyses, it shows that the removal of protein-like substances was relatively higher than that of humic-like substances, and polysaccharide was substantially reduced. But the extent of those substances removed during soil passage was decreased with the increasing concentration of silver nanoparticles. The attenuation of ionic PhACs ranged from 55% to 80%, depending on the concentration of silver nanoparticles. The attenuation of neutral PhACs ranged between 72% and 77%, which was relatively lower than that observed for the ionic PhACs. The microbial viability was affected by silver nanoparticles, which also resulted in inhibition of nitrifiers.
