한국산 접목 선인장에서 분리한 괴사 반점 원인 바이러스의 생물리화학적 특성 (Purification and Biophysicochemical Properties of the Virus Causing Necrotic Spot Symptom on Korean Cactus)

  • A virus was purified from Korean cactus (Chamaeaereus silverstrii f. Variegata Hort) showing necrotic spots and its biophysicochemical properties were examined. The typical necrotic spots were observed on healthy looking cactus inoculated with the purified virus and the various symptoms from necrotic spots to systemic were observed on 17 species of test plats used, which suggests that the purified virus has the normal infectivity and wide host range. Ultraviolet absorption scanning analysis revealed that the viral preparation had the typical curve with shoulder at 290 nm and its ratio of absorbances at 260 nm to 280 nm and extinction coefficient was 1.156 and 2.86, respectively. The virus represented one distinct and homogeneous band and molecular weight of viral coat protein was determined to 17, 000 Dalton by SDS-PAGE. The virus revealed rod-shaped virion (325$\times$20 nm) by electron microscopy. Based on these results, it has been thought that the virus purified in this study is the pathogenic agents causing necrotic spot symptom on Korean cacuts and one of the member of tobamovirus family.

Ultrastructural Aspects of Mixed Infections with Turnip mosaic virus (TuMV- ACl8 and -C5) and Ribgrass mosaic virus (RMV-CA1) in Oriental Cabbage

  • Mixed infections of two economically important viruses, Turnip mosaic virus(TuMV) in the family Potyviridae and Ribgrass mosaic virus(RMV) in the genus Tobamo-virus, were studied ultrastructurally on oriental cabbage. TuMV-ACl8 (alpine isolate in Korea) induced chlorotic spots on inoculated leaves of both ‘SSD63’ inbred line known as susceptible to TuMV, and ‘Tambok’ commercial cultivar, known as resistant to the virus, in the early stages of infection. TuMV-C5 (Taiwan isolate) caused severe mosaic and malformation on the upper leaves of ‘SSD63’, and necrotic spots in both inoculated and upper leaves of ‘Tambok’. RMV-CA1 (oriental cabbage isolate from alpine in Korea) induced vein chlorosis, leaf malformation, and midrib necrotic streak in the upper leaves of both ‘SSD63’ and ‘Tambok’. Both oriental cabbages infected with a combination of TuMV-ACl8 and RMV-CA1 showed synergistic symptoms of severe yellowing, severe mosaic, and necrotic spot or vein necrosis on their leaves. A combination of TuMV-C5 and RMV-CA1 produced synergistic symptoms only in ‘SSD63’. In ‘Tambok’ infected with the combination of TuMV-C5 and RMV-CA1, the number of necrotic spots on the inoculated leaves was one half lesser than that on singly infected with TuMV-C5. A few necrotic spots progressed systemically. In cells infected with a combination of TuMV-ACl8 and RMV-CA1 or TuMV-C5 and RMV-CA1, the particles of the two viruses made nonagon-like rings(NLR); one TuMV particle was surrounded loosely by nine RMV particles. Two unrelated viruses of TuMV and RMV were compacted in the central part of the spiral aggregates(SA) that was induced strikingly in cells by the mixed infections. The SA showed NLR in its center of the cross-sectioned side. Many particles of RMV of Tobamovirus were closely associated with Potyvirus-characteristic cylindrical inclusions. The SAs in the mixed infections were formed easily by the Potyvirus of TuMV-ACl8 or -C5 isolates.

닭의 Leucocytozoon병(病)의 발생(發生) (An Occurrence of Leucocytozoonosis in Chicken)

  • This investigation was performed to study of chickens naturally infected with leucocytozoon. The results obtained were summarized as follows: 1. The characteristic macroscopical changes observed were hemorrhages and elevated greyish white spots on the oviduct, the liver, the pancreas, the kidney and the pectoral muscles. 2. Microscopically, megaloschizonts were located in groups in the elevated greyish white spots. Hemorrhage and degenerative necrotic changes were observed around the megaloschizonts.

멜론괴저반점바이러스-Me의 몇 가지 특성과 멜론 품종의 저항성 선발 (Some Characteristics of Melon necrotic spot virus-Me and Resistance Screen to the Virus in Melon Cultivars)

  • Melon necrotic spot virus (MNSV)는 국내에서 재배되고 있는 멜론(Cucumis melo)에 심각한 피해를 주고 있다. 2009년 전남 나주 멜론재배단지에서 괴저반점을 나타내는 멜론 잎에서 MNSV를 분리 동정하였다. 지표식물 반응, immuno captured(IC) RT-PCR 및 염기서열 상동성 비교 등 몇몇 분석으로 이 분리주를 MNSV-Me로 명명하였다. 멜론 시판 35품종에 대한 MNSV-Me의 저항성 선발을 위하여 유묘의 자엽에 인위적 즙액 접종 방법을 이용하였다. 접종된 모든 품종의 자엽에서는 괴저반점을 형성하였다. 이들 중 25품종은 상엽으로 괴저 증상의 이행 또는 일부 개체에서는 접종 약 40일 후 줄기에 3 cm 이상의 괴저를 나타내는 감수성 반응을 나타냈다. 5품종은 상엽으로 잘 이행이 되지 않지만 줄기에 3 cm 미만의 괴저증상을 형성하는 중간 저항성을 발현하였다. 접종한 상엽으로는 바이러스 증상이 발현되지 않고, 단지 줄기에 희미한 괴저가 형성되는 얼스타이탄, 얼스럭서리, 베타리치 하계, 베타리치 및 원더플하계1호가 저항성을 나타냈다.

Characterization of Melon necrotic spot virus Isolated from Muskmelon

  • A severe disease of muskmelon (Cucumis melo cv. Alsnight) grown on rockwool in a plastic house was characterized by leaf and stem necrosis followed by death of the plants. In 2001, an isolate of Melon necrotic spot virus-MN (MNSV-MN) of the genus Camovirus was identified as the causal agent of the disease on the basis of biological reactions and nucleotide sequence analyses of coat protein (CP) gene. MNSV-MN induced necrotic local lesions on mechanically inoculated leaves and systemic necrotic spots on the upper leaves of melon cvs. Alsnight, Rui III, Party, Imperial, and Seolhang. However, the inoculated leaves of watermelon and cucumber showed only necrotic lesions. DsRNAs extracted from the melon infected with MNSV-MN were separated into three components. Molecular sizes of the dsRNAs were estimated at approximately 4.5, 1.8, and 1.6 kbp. The amplified cDNA products of CP gene for MNSV-MN by RT-PCR showed approximately 1.2 kbp. The amplified DNA was digested to three fragments by MspI treatment. The cDNA of the genomic RNA of MNSV-MN was cloned and the region deduced to encode the CP was sequenced. The CP coding region, located near 3' end of the genome, consisted of 1,170 nucleotides and had the potential to encode a 390 amino acid protein. The nucleotide and amino acid sequences of MNSV-MN CP gene were 84.0-94.6% and 90.8-94.9% identical with other MNSV isolates found in the GeneBank database, respectively. This is the first report on the occurrence of MNSV in Korea.

채소(가지, 알타리무, 슈가로프)에 발생한 토마토반점위조바이러스 (Tomato spotted wilt virus) 발생과 병징 특성 (Occurrence and Symptoms of Tomato spotted wilt virus on Egg Plant, Whole Radish and Sugar Loaf in Korea)

  • Tomato spotted wilt virus(TSWV)의 상습 발생지인 안양지역에서 가지, 알타리무, 슈가로프에서 TSWV가 발생하였다. 알타리무에서는 잎에 괴저반점과 뿌리에 괴저 병징이 나타났다. 가지에서는 잎에 전형적인 다중 원형반점을 나타냈으며 열매에 심한 괴저를 나타냈다. 슈가로프에서는 잎에 전형적인 원형반점과 심한 위축 병징이 나타났다. 가지, 알타리 무, 슈가로프에서 분리한 TSWV의 생물적 특성은 흰 명아주, 붉은 명아주, 담배(N. devney)에서는 국부 감염이었으며, 담배(N. glutinosa, N. benthamiana)와 독말풀(D. stramonium)에서는 전심감염이었다. 가지와 슈가로프에서 분리한 TSWV는 병원성이 유사하였으나, 알타리 무에서 분리한 TSWV는 N. tabacum 'Xanthi NC' 등 5종의 담배에서 국부 감염되어 병원성이 매우 상이하였다.

Clavibacter michiganensis subsp. michiganensis에 의한 고추 궤양병 (Occurrence and Distribution of Bacterial Canker of Red Pepper Caused by Clavibacter michiganensis subsp. michiganensis)

  • Bacterial leaf spot by Xanthomonas campestris pv. vesicatoria has been known to cause serious problem in red pepper in Korea. However recent survey showed that most smptoms in the leaves were mixed with two different symptoms one was leaf spot and the other was canker. bacteria isolated from canker were identified as Clavibacter michiganensis subsp. michiganensis on the basis of biochemical and physiological characteristics. The causal bacteria were non-motile rod-shaped and Gram-positive. The lesions on pepper leaves appeared at first as small blisters or pimple-like white spots which enlarged in size at a later stage. The centers of some of the spots became necrotic and brown and were surrounded by a white halo. Pathogenicity tests were performed on pepper cv. Alchan seedling by spraying of bacterial suspension. During 1997 and 1998 total 17% of 527 fields surveyed were infected by C. michiganensis subsp. michiganensis. The canker of red pepper caused by C. michiganensis subsp. michiganensis was first identified in this study in Korea, and new name "gueyangbyung" was tentatively given to the disease.

Entomosporium mespili에 의한 채진목 점무늬병 (Leaf spot of Amelanchier asiatica Caused by Entomosporium mespili)

  • Leaf spot of Amelanchier asiatica was observed in Korea.. The major symptoms of the disease were small necrotic spots and severe early defoliation. On the basis of morphological characteristics of the fungus observed from naturally infected leaves, the causal organism was identified as Entomosporium mespili (DC. : Duby) Sacc. The conidial suspension prepared from sporulating lesions was sprayed on healthy leaves with or without wounding to prove pathogenicity of the fungus. Small spots were noticed tow days after inoculation, and acervuli containing numerous conidia were observed 7 and 10 days after inoculation from wounded and unwounded leaves, respectively. This is the first record of the disease on A. asiatica in Korea.

  • Anthracnose of statice (Limonium sp. var. Misty Blue and L. sinuatum Mill. cv. Early Blue) occurred up to 20% in vinyl-houses at Buan, Imsil and Namwon from July to November, 1993. All isolates from the anthracnose symptoms were identified as Glomerella cingulata (Stonem.) Spauld. & Schrenk and its anamorph was Colletotrichum gloeosporioides (Pens.) Sacc. Pathogenicity of the isolates was ascertained on the statice host by artificial inoculation. The symptoms on leaves appeared as circular brown spots in early stage and developed to zonate spots. The spot aggregated and diseased leaves were blighted. The symptom also developed on the stems of severely infected plants and the infected branches were blighted. A lots of acervuli developed on the lesions of stems. Acervuli on necrotic lesions were setose, rounded and elongated. Setae variable in length, 1~4 septate, brown, slightly swollen at the base and tapered to the apex on which conidia were occasionally borned. Statice was revealed as the new host of G. cingulata.

First Report of Leaf Rust Caused by Puccinia caricis in Farfugium japonicum in Korea

  • Farfugium japonicum is used in traditional medicine and as an edible herb in China and Korea. In July 2013, leaf spots were observed in F. japonicum seedlings at Ulleung Island, Gyeongsangbuk Province, Korea. Early symptoms on the leaf adaxial surface included roughly circular yellow spots that later developed brown, necrotic centers. The aecia were hypophyllous, cupulate, yellowish, $180{\sim}430{\mu}m$ in diameter, clustered, and erumpent with a peridium with a recurved margin. The aeciospores were globoid, $14{\sim}17{\times}13{\sim}16{\mu}m$, light yellow or colorless, and densely verrucose. The 28S rDNA sequence of the isolate was identical to each other and shared 99% identity with Puccinia caricis. This is the first report of rust caused by P. caricis in F. japonicum in Korea or elsewhere in the world.
