Plant biomass, or lignocellulose, is one of the most abundant natural resources on earth. Lignocellulosic biomass, such as agricultural and forestry residue, serves as a renewable feedstock for microbial cell factories due to its low price and abundant availability. However, the recalcitrance of lignocellulosic biomass requires a pretreatment process prior to microbial fermentation, from which fermentable sugars including xylose and glucose are generated along with various inhibitory compounds. The presence of furan derivatives, such as 5-hydroxymethyl-2-furaldehyde and 2-furaldehyde (furfural), hampers the microbial conversion of lignocellulosic biomass into value-added commodities. In this study, furfural tolerance was improved by investigating the detoxification mechanism in non-model yeast. The genes encoding aldehyde dehydrogenases were overexpressed to enhance furfural tolerance and resulted in improving cell growth and lipid production that can be converted into biofuel. Taken together, this approach contributes to the understanding of the reducing toxicity mechanism of furfural by the aldehyde dehydrogenases and provides a promising strategy that the use of microorganism as an industrial workhorse to treat efficiently lignocellulosic biomass as sustainable plant derivatives.

Objectives : As a substitute for high-price ginseng, this study attempted to examine a possibility of the ferment extract of Panax ginseng sprout whether leaves and roots can be used together as a cosmetic ingredient with anti-oxidative and wrinkle-care effects. Methods : In terms of a test method, antioxidant activities were confirmed through total polyphenol contents, total flavonoid contents, DPPH radical scavenging activity and ABTS radical scavenging activity using the Panax ginseng sprout. In addition, to assess wrinkle-care effectiveness, the cytotoxicity of the extract was analyzed through MTT assay, and inhibition of collagenase activity in the cells was tested using the Panax ginseng sprout fermented by Saccharomyces cerevisiae. Resuits : The content of polyphenols and flavonoids in natural plants was highest in Panax Ginseng Sprout Extract at 100℃, which also demonstrated high DPPH, ABTS radical scavenging activity. MTT assay demonstrated that the Panax Ginseng Sprout Ferment Extract did not have a cytotoxic effect in CCD-986SK cell. Also, Panax Ginseng Sprout Ferment Extract was found to inhibit MMP-1 expression by 51.85±6.09% at a concentration of 10%. Conclusions : Therefore, this study has confirmed a possibility of Panax ginseng sprout ferment extract as a cosmetic ingredient with MMP-1-inhibitory effects.

This study was conducted to determine effects of cellulolytic microbes inoculation to sawdust-based spent mushroom substrate(SMS) during deepstacking on fermentation parameters, total microbial counts and cellulolytic enzyme activity and to on SMS nutrients utilization by sheep. For sheep metabolism trials, six sheep(ram, average 54.8kg) were fed a Control diet(70% concentrates, 15% rice straw and 15% SMS with no microbial treatment on a dry basis) and a Treatment diet(the same diet including SMS with a microbial treatment) for 2 trials. Spent mushroom substrates with or without a microbial(4 strains including 1 strain of Enterobacter ludwigii, 1 strain of Bacillus cereus and 2 strains of Bacillus subtillis) treatment (1% of SMS on wet basis) were deepstacked for 7 days. The internal temperatures in 1.2 M/T of SMS deepstacks reached to 50±5℃ within 7 days of storage. Total microbial counts remarkably decreased (P<0.05) with a deepstacking process and were not affected(P>0.05) by the microbial treatment. For fibrolytic enzyme activity, CMCase and xylanase activities were decreased(P<0.05) by a deepstacking process. After deepstacking, the microbial treatment showed about 2.5-times higher(P<0.05) for CMCase activity and about 4-times higher(P<0.05) for xylanase activity than those of the Control. Activities of ligninolytic enzymes such as laccase and MnP were not affected by the microbial treatment. The sheep fed the microbially treated SMS diet had a tendency of greater total tract digestibilities of ash(P=0.051), NFE (P=0.071), hemicellulose(P=0.087) and NDF(P=0.096) than those fed the untreated SMS diet. Nitrogen balance of sheep was not affected(P>0.05) by feeding of microbially treated SMS. Accordingly, these results indicate that cellulolytic microbes inoculation during deepstacking of SMS may improve the bio- utilization of SMS by sheep.

This study was conducted to evaluate the feasibility of food waste as a feed resources by fermentation and fermented food waste as a substitute of rat feed on the performance with measuring the liveweight gain, feed consumption, feed conversion and digestibility Sixty-two grams Sprague-Dawley line 36 rats were allocated three treatments 12 rats of each(3replication ${\times}$ 4 rats). The substitution level of fermented food waste to commercial broiler feed were control, 0: 100: treatment I, 10: 90: treatment II, 20:80. The chemical composition of fermented food waste was appeared to follows : dry matter, 88.47% : crude ash. 12.95: crude protein, 20.82%; crude fiber, 13.62; ether extract, 9.15%. The body weight of treatment I and II at 1 weeks was significantly lower than those of control(p<.05) and weekly weight gain of control at 0-1 weeks was significantly higher than those of treatment I and II(p<.05). Those were higher in treatment I than those of rest groups at 1-2 weeks(p<.05). Total weight gain of treatment II was significantly lower than those of control and treatment I(p<.05) Total feed consumption of treatment II was significantly higher than those of control (p<.05) and weekly feed consumption of control and treatment II at 3-4 weeks was significantly higher than those of treatment II(p<.05). but those were higher in treatment I and II than those of control at 2-3 weeks(p<.05). Commutative feed conversion of treatment II was significantly higher than those of control(p<.05) and weekly feed conversion of treatment II and III at 0-1 weeks was significantly higher than those of control(p<.05) Dry matter digestibility of control and treatment I was significantly higher than those of treatment II(p<.05) and organic matter digestibility was higher in control than those of treatment II(p<.05).

Meju and nuruk (respectively soybean and malt) are traditional Korean fermentation starters that are vulnerable to contamination by harmful microorganisms such as aflatoxigenic fungi and their associated aflatoxins (AFs). In this study, Aspergillus spp. were isolated and identified from a total of 57 meju and 18 nuruk samples collected from Korean markets. Their potential aflatoxigenicity was investigated by examining the presence of three aflatoxin biosynthetic genes (aflO, aflP, and aflR) using multiplex polymerase chain reaction (mPCR) assays. Thereafter, aflatoxin production of isolates and the natural occurrence of AFs in meju and nuruk samples were analyzed by high-performance liquid chromatography (HPLC). A total of 177 Aspergillus isolates were identified and 130 isolates were obtained from meju samples. Of these, 25 isolates (19.2%) contained all three aflatoxin biosynthetic genes, and five (20%) of these isolates produced aflatoxins. Forty-seven of the Aspergillus isolates were obtained from nuruk samples, five of which (10.6%) expressed all three AF biosynthetic genes; however, none of these strains produced AFs. HPLC analysis showed that 88% (51/58) of the meju samples and 39% (7/18) of nuruk samples were not contaminated with AFs (below limit of detection). Among the isolates isolated from meju and nuruk, there were aflatoxigenic strains containing all three aflatoxin biosynthetic genes or producing aflatoxin in medium, but the frequency of aflatoxin contamination was low in the meju and nuruk samples.

Four lactating Holstein Friesian crossbred cows, with an average initial weight of 450 kg, 48${\pm}$12 days in milk and initial milk yield of 18 kg/h/d, were randomly arranged according to a 2${\times}$2 factorial arrangement in a 4${\times}$4 in Latin square design with 21-d period to investigate the effects of type of total mixed ration (TMR) and type of whole cottonseed (WCS) on intake, digestibility and milk production. The dietary treatments were i) TMR and WCS supplementation at 0.5 kg/h/d, ii) TMR and cracked WCS (cWCS) supplementation at 0.5 kg/h/d, iii) fermented TMR (FTMR) and WCS supplementation at 0.5 kg/h/d, and iv) FTMR and cWCS supplementation at 0.5 kg/h/d. Voluntary feed intake was 15.9, 15.2, 15.4 and 15.6 kg DM/d in dietary treatment 1, 2, 3 and 4, respectively. Digestibility of DM, OM, CP, EE, NDF and ADF were not significantly different among dietary treatments. Ruminal pH, $NH_{3}-N$ and volatile fatty acids in the rumen were also not significantly different among type of TMR or type of WCS. Blood urea-N concentration was not significantly different among dietary treatments. Ruminal bacteria population tended to increase but ruminal protozoa population tended to decrease with supplementation of cWCS, but they were not affected by FTMR. Milk yield and 3.5% FCM were not statistically different among treatments (16.6, 16.2, 17.0, 16.3 kg/d and 18.0, 18.6, 19.9 and 19.0 kg/d, respectively). Milk composition was not significantly different among dietary treatments. However, unsaturated fatty acids in milk fat in cows fed FTMR were lower (p<0.05) than in cows fed TMR. In conclusion, fermentation is a conceivable method to improve the quality of TMR for long-time storage and the cracking method is suitable to release the fat from cottonseed for enhancing fatty acid deposition in milk. Thus, the combination of FTMR and cWCS supplementation would be an alternative strategy to improve performance of lactating cows.

The farm house structure, ventilation system and manure treatment of two-storey buildings and sawdust pig houses were surveyed and analyzed. Based on the data for ten selected farms in five provinces during eight months, the goal is to eventually establish a standard two-storey pig house. Manure treatments were composting, slurry and activated sludge in two-storey pig houses, while fermentation method was done in sawdust pig house. The depth of sawdust as a litter material were 10 to 60cm, with a duration of 1/2, 1, 3 and 6 months, respectively. The ventilation systems were the mechanical type in two-storey pig houses and natural system in the sawdust pig house. Side wall in the two-storey pig house was enclosed with insulation materials such as block, colored metal sheet and sandwich panels. The minimum ceiling height in the first floor of the two-storey pig house was 2.0m and the maximum was 3.0m. On the second floor, ceiling height ranged from 2.0 to 2.7m. The construction cost in the two-storey systems were $700\~140$, and sidewall curtain systems were $30\~40$ thousand Won/pyung.
Made using a natural mixed starter of lactic acid bacteria and yeast, Koumiss is a slightly alcoholic fermented mare's milk beverage, and a traditional drink of the nomadic populations of Central Asia. Cichorium intybus L. (chicory) is a sedative with potential cardioactive properties, and its oligosaccharides are beneficial in maintaining healthy gastrointestinal flora. Hence, in this study, we have generated a functional Koumiss containing two different concentrations of chicory. After fermentation of the Koumiss premix, the TA increased to 0.85~0.88%, and the pH decreased to ~4.3. The addition of either concentration of chicory had no significant effect on pH and TA. However, the taste, flavor, color, texture, and overall acceptability decreased in proportion to the added amount of chicory. This study has provided the first data on Koumiss supplemented with chicory. The results could be useful in developing high-quality Koumiss with functional activity using chicory, and allowing large-scale industrial production. Further studies are needed to determine if chicory root extract is beneficial for lifestyle-related diseases.
This study was performed to confirm the antioxidative and hair-growth-promotion effects of a fermented compound composed of five materials, namely Allium cepa, Cucurbita spp., Glycine max Merr., Pinus sylvestris leaf, and Sesamum indicum. An additional compound composed of two materials was composed of Allium cepa and Cucurbita spp. Antioxidative effects were measured based on DPPH radical-scavenging activity and total flavonoid content. Fermented 5-type natural compounds (5NC) showed higher DPPH radical-scavenging activity and flavonoid content than those of fermented 2-type natural compounds (2NC). Hair-growth promotion was demonstrated in vivo. Alkaline phosphatase (ALP), oid contentontenteptidase phospactivity, and macroscopic observation were measured. Experimental animals were divided into four groups based on the samples they were administered: water, pansildil (P-CON), 5NC, and 2NC. Samples were administered orally once a day at a fixed time for four weeks. Both ALP and ${\gamma}eeksactivity$ increased in the 5NC group compared with the 2NC group. In macroscopic observation, the P-CON group grew the most and were most similar to the state before shaving. The 5NC group grew hair similar to the P-CON group. In all the results, the 5NC group showed greater antioxidative and hair-growth-promotion effects than the 2NC group. Consequently, 5NC could be used as an ingredient for hair-growth promotion.
The study performed to identify the type of ESBL against strains which are producing extendedspectrum ${\beta}$-lactamases and isolated from sewage in Suyeong sewage disposal plant, Busan Environmental Corporation. By the standard activated sludge method, Suyeong sewage disposal plant purify living and lavatory sewage gathering from the northeast Busan and the facility purify total 550,000 tons of living sewage disposal a day. 14 strains were isolated by double disk synergy test and the third generation cepha-antibiotics test. Indole, methyl-red, Voges-Proskauer, Simmon's citrate, decarboxylasedihydrolase and sugar-fermentation tests identified as Klebsiella pneumoniae (n=4) and Escherichia coli (n=10). Plasmid-mediated transmission test against isolated 14 strains proved 11 strains transmitted resistance to recipient E. coli J53 (sodium $azide^R$, $ceftazidime^S$). 9 strains of conjugant were expressed ESBL genes transferred from parental strain but 2 conjugants did not expressed. The type of ESBL from each strain was determined by isoelectric focusing points, DNA and amino acids sequencing. The results indicated that the types of ESBL transmitted to recipient E. coli J53 were TEM-1, the parental TEM type and SHV-12 type.
