Optimization of Submerged Culture Conditions for Exo-biopolymer Production by Paecilomyces japonica

  • Optimization of submerged culture conditions for the production of exo-biopolymer from Paecilomyces japonica was studied. Maltose, yeast extract and potassium phosphate were the most suitable sources of carbon, nitrogen, and inorganic salt, respectively, for both production of the exo-biopolymer and mycelial growth. The optimal culture conditions in flask culture were pH 5.0, $25^{\circ}C$ and 150 rpm in a meidum containing of 30 g maltose, 6 g yeast extract, 2 g polypeptone, 0.5 g $K_2HPO_4$, 0.2 g $KH_2PO_4$, 0.2 g $MnSo_4\;{\cdot}\;5H_2O$, 0.2 g $MgSO_4\;{\cdot}\;7H_2O$ in 1-L distilled water. Exo-biopolymer production and mycelial growth in the suggested medium were significantly increased in a 2.5-L jar fermentor, where the maximum biopolymer concentration was 8 g/1.

홍화, 황련, 세신 배지첨가제가 팽이버섯 생육에 미치는 영향 (Effects of Addition of Cathamus tinctorius, Coptis japonica and Asarum sieboldii to Sawdust substrate on the Growth and Development of Winter Mushroom, Flammulina velutipes)

  • Cathamus tinctorius, Coptis japonica and Asarum sieboldii were tested as substrate for the production of Flammulina velutipes. Among the C. tinctoris , C. japonica and A. sieboldii , C. tinctoris was the best substrate for the production of fruitbody. The effects of addition of C. tinctoris to sawdust substrate resulted in the increased mycelial growth on inoculum culture, 3.1% in ratio of fully culture and shorted one day in culture period. C. tinctoris was decreased 6.1% in ratio of fully culture, 11.0% in ratio of fruitbody productive culm. The addition of C.tinctoris, C.japonica to sawdust substrate increased 134.6%, 114.1% on the yield of the mushroom fruitbody respectively . But A. sieboldii decreased the mycelial growth and pineheading ratio delayed the production of fruitbody.

Effects of Culture Conditions on Mycelial Growth and Polysaccharide Production of Tricholoma matsutake in Bioreactor

  • 본 연구에서는 담자균류의 일종인 Tricholoma matsutake의 균사체 생육과 polysaccharide 생산을 최적화하기 위하여 5L jar fermentor에서 교반속도와 통기량의 영향을 검토하였다. 교반속도는 150 rpm에서 가장 좋은 결과를 보였으며, 균체량은 배양 10일째 21.87 g/L, polysaccharide는 교반속도는 300 rpm에서 배양 8일째 8.86 g/L 로 가장 높은 값을 나타내다. 통기량은 11일째 0.5 vvm에서 가장 좋은 균체량 20.85 g/L, polysaccharide는 1.5vvm에서 배양 11일째 8.83 g/L로 가장 높은 값을 나타내었다.

Hypoglycemic Effect of Exo- and Endo-biopolymers Produced by Submerged Mycelial Culture of Ganoderma lucidum in Streptozotocin-Induced Diabetic Rats

  • The hypoglycemic effect of an exo-biopolymer (EXO) and endo-biopolymer (ENDO) produced from submerged mycelial culture of Ganoderma lucidum was investigated in streptozotocin (STZ)-induced diabetic rats. Both the EXO and ENDO showed hypoglycemic potential, however, the former proved to be more potent than the latter. The administration of the EXO at the dose of 100 mg/kg body weight (BW) significantly reduced the plasma glucose level (23.5%) and increased the plasma insulin level (2.2 fold) in the diabetic animals. The EXO also lowered the plasma total cholesterol, triglyceride, low-density lipoprotein (LDL) cholesterol, and athrogenic index by 14.7, 31.4, 24.1, and 45.4%, respectively, and reduced the liver total cholesterol and triglyceride levels by 6.7 and 25.8%, respectively. It increased the plasma high-density lipoprotein (HDL) cholesterol (37.7%), compared to the control group. Furthermore, the alanine transaminase (ALT) and aspartate transaminase (AST) showed lower activities in the EXO administered groups than the other experimental groups. Taken together, these results suggest that the exo-biopolymer may alleviate the blood glucose level by increased insulin secretion.

Utilization of Robinia pseudoacacia as Sawdust Medium for Cultivation of Edible and Medicinal Mushrooms

  • This study was undertaken to examine the feasibility of black locust (Robinia pseudoacacia) as substrates for several edible mushrooms. For the cultivation of several edible and/or medicinal fungi on black locust, optimum bulk densities, synthetic or semisynthetic additives, natural additives and pretreatment methods were investigated. Fruit body yields of the fungi on various sawdust media composed of different wood species were also analyzed for testing the capability of black locust as a substrate for mushroom production. Mycelial growths decreased proportional when the bulk density increased. The most suitable carbon and nitrogen sources as additives to promote the mycelial growth were sucrose (2%, w/w) and ammonium phosphate (0.2%, w/w) respectively. When corn-powder and beer-waste as natural additives were added to sawdust of black locust showed the significant growth of mycelia. And the optimum mixing ratio was 10:2:1 (sawdust: corn-powder: beer-waste, w/w). Black locust after cold water treatment showed the outstanding mycelial growths. Any significant changes of pH, moisture content (%) and dry-weight losses (%) could not be found among culture substrates (sawdust of black locust, oak and poplar wood) examined before and after harvesting of fruit bodies. Yield of fruit bodies on black locust culture media were comparable with those culture media composed with oak and poplar wood. The present work indicated strongly the potentiality of black locust as raw materials for edible and medicinal mushrooms.

느타리버섯 배지에 담배가루의 처리가 버섯균과 푸른 곰팡이병원균의 균사생장과 수량에 미치는 영향 (Additive effect of tobacco waste in substrates on yield and mycelial growth of oyster mushroom, Pleurotus spp. and Trichoderma disease)

  • 담배가루를 첨가하므로서 버섯균의 균사생장을 촉진하고, 푸른곰팡이병원균의 발생 감소의 가능성을 검토하기 위하여 시험을 실시한 결과. 볏짚배지의 담배가루 표면처리에서는 느타리버섯은 0.1g의 처리구까지는 무처리구보다 약간의 균사생장이 빠르며, 혼합처리에서는 1.0g처리구까지 무처리구보다 균사생장이 빠르고 2.0g에서는 생장이 감소되었다. 푸른곰팡이병원균 처리구는 담배가루의 모든 처리구가 무처리구에 비하여 균사생장이 억제되었다. 느타리버섯 품종별 담배가루 처리에 따른 균사생장 조사에서는 표면처리에서는 ASI 2072 담배가루 처리량 처리방법에 상관없이 억제되었으며, ASI 2180 균주의 0.1, 0.2g처리에서 약간 억제되었고, 혼합처리구에서는 ASI 2072균주는 전체적으로 억제되었으며 그 이외의 균주들은 2.0 g처리에서만이 억제되었다. 포장시험 중 볏짚재배 시험에서는 3%처리시 수량이 높았으며, 폐면재배에서는 6% 처리구가 수량이 높았으며 담배가루 처리에 따른 푸른곰팡이병의 방제효과는 인정되지 않았다.

톱밥재배용 표고 품종의 단핵균사체와 2핵균사체의 한천과 톱밥배지에서의 균사생장 (Mycelial Growth of Monokaryotic and Dikaryotic Strains of Lentinula edodes Cultivars for Sawdust Cultivation on the Agar and Sawdust Culture media)

  • 표고 육종 소재를 개발하기 위하여 톱밥봉지재배용 품종의 2핵균사체와 단핵균사체를 한천배지와 톱밥배지에서 배양적 특성을 조사하였다. 품종은 산조 701호, 산조 705호, 참아람, 추제 2호를 사용하였다. 공시 표고 품종의 균사생장은 산조 705호를 제외한 품종 모두 PDA에서 가장 양호하였으나 산조 705호는 MCM에서의 균사 생장이 가장 양호하였다. 품종에 관계없이 표고 균사는 MEA에서 가장 안정적인 균사생장을 보였다. 톱밥배지에서 공시균주의 균사생장은 품종 간의 큰 차이는 보이지 않았으나 산조 701호와 705호보다 약 10%정도 빠르게 생장하는 것으로 나타났다. 산조701호와 참아람의 단핵균사체는 2핵 균사체보다 균사생장이 저조하였다. 2핵균사체는 참아람이 산조 701호보다 균사생장이 양호하였으나 단핵균사체는 전체적으로는 산조 701호가 참아람보다 균사생장이 양호하였다. 선발된 단핵 균사체는 균사생장이 매우 다양하고, 형성된 균총의 형태도 매우 다양하여 유전적으로 매우 다양한 단핵 균사체가 선발된 것으로 추정할 수 있고, 이들 단핵 균사체는 교배육종을 위한 좋은 육종 소재로 기대된다.

노린재동충하초의 배양 최적화 및 NO 생성 저해 효과 (Optimization for Mycelial Growth and Inhibitory Effect on Nitric Oxide Production of Cordyceps nutans Pat.)

  • 동충하초는 예로부터 아시아권에서 한방약재로 사용되어 온 곤충병원성진균이다. 이 중 노린재동충하초(Cordyceps nutans)는 자연 상태에서 비교적 많이 발견되나 이에 대한 연구가 미미한 편이다. 따라서 본 실험에서는 C. nutans 균사체의 최적 배양 조건을 확립하고 배양액 처치 시대식세포의 NO (nitric oxide) 생성 억제 효능을 조사하였다. 균사 생육 적정 온도는 $25^{\circ}C$이었으며 pH는 7.0~8.0 사이의 중성범위로 조사되었다. MCM (mushroom complete medium), V8A (V8 juice agar), YMD (yeast malt dextrose) 배지에서는 균사 생육이 우수하였으나 MMM (mushroom minimal medium) 배지의 경우 균사 생육이 이루어지지 않아 영양원이 필수적이었다. 영양원 선발에 있어 탄소원은 dextrose와 sucrose가 적합하였고 질소원은 ammonium citrate가 균사 생장에 적합하였다. RAW 264.7 세포에 대한 C. nutans 배양액의 세포 독성은 나타나지 않았으며 LPS (lipopolysaccharide)를 처리 한 세포의 NO 생성량은 농도 의존적으로 줄어들었다. 따라서 본 실험 결과는 C. nutans 배양 시 다량의 균사체를 확보할 수 있는 최적 조건을 제공할 뿐 아니라 C. nutans의 항염 관련 우수한 생리 활성이 있음을 보여준다.

Cultural Characteristics of Ectomycorrhizal Mushrooms

  • Ectomycorrhizal (ECM) mushrooms play a major role in plant growth promotion through symbiotic association with roots of forest trees. They also provide an economically important food resource to us and therefore they have been studied for their artificial cultivation for decades in Korea. We have secured bio-resources of ECM mushrooms from Korean forests and performed their physiological studies. To investigate the cultural characteristics, the fungi were cultured under different conditions (medium, temperature, pH of the medium, inorganic nitrogen source). More than 90% of total 160 strains grew on three solid media (potato dextrose agar, PDA; sabouraud dextrose agar, SDA; modified Melin-Norkrans medium, MMN). The rate of mycelial growth on malt extract agar (MEA) was lower than those of three media (PDA, SDA, MMN). None of the Tricholomataceae strains grew on MEA. Many strains of ECM mushrooms were able to grow at the temperature range of $15{\sim}25^{\circ}C$ on PDA, while they showed poor growth at $10^{\circ}C$ or $30^{\circ}C$. In particular, the growth rates of both Gomphaceae and Tricholomataceae were significantly lower at $10^{\circ}C$ than at $30^{\circ}C$. The optimal pH of many strains was pH 5.0 when they cultured in potato dextrose broth (PDB). Fifty-seven percent of tested strains grew well on medium containing ammonium source than nitrate source. Many strains of Tricholomataceae showed a notable growth on ammonium medium than nitrate medium. Twenty-three percent of strains preferred nitrate source than ammonium source for their mycelial growth. The production and activity of two enzymes (cellulase and laccase) by ECM fungi were also assayed on the enzyme screening media containing CMC or ABTS. Each strains exhibited different levels of enzymatic activities as well as enzyme production. The number of laccase-producing strains was less than that of cellulase-producing strains. We found that 77% of tested strains produced both cellulase and laccase, whereas 2% of strains did not produce any enzymes. The morphological characteristics of mycelial colony were also examined on four different solid media. Yellow was a dominant color in mycelial colony and followed by white and brown on all culture media. ECM mushrooms formed mycelial colonies with a single or multiple colors within a culture medium depending on the strains and culture media. The most common shape of mycelial colony was a circular form on all media tested. Other families except for Amanitaceae formed an irregular colony on MMN than PDA. All strains of Tricholomataceae did not form a filamentous colony on all media. The pigmentation of culture media by mycelial colonies was observed in more than 50% of strains tested on both PDA and SDA. The degree of pigmentation on PDA or SDA was higher than MMN and brown color was dominant than yellow color. The production of exudates from mycelial colony was higher on PDA than MMN. Brown exudates were mainly produced by many strains on PDA or SDA, whereas transparent exudates were mainly produced by strains on MMN. We observed the mycelial colonies with a single or multiple textures in just one culture plate. Wrinkled or uneven colony surfaces were remarkably observed in many strains on PDA or SDA, while an even colony surface was observed in many strains on MMN. Sixty percent of Tricholomaceae strains formed wrinkled surface on PDA. However, they did not form any wrinkle on MMN plate. Cottony texture was observed in mycelia colonies of many strains. Velvety texture was often observed in the mycelial colonies on SDA than PDA and accounted for 60% of Suillaceae strains on SDA.

Artificial cultivation of Medicinal Mushroom, Phellinus linteus using Mulberry logs

  • The optimal conditions for mycelial growth of P. linteus ASI 26011 were 25-30$^{\circ}C$ and pH 6.0, respectively. The mycelial growth of P. linteus was excellent on MCM medium. In case of carbon sources, the mycelial growth of P. linteus was best on the culture media that were contained with sucrose, mannose and glucose. Potassium nitrate and sodium nitrate were good for the mycelial growth of P. linteus as a nitrogen source. For comparison of the mycelial colonization of P. linteus on logs, several techniques of inoculation were tested; the sterilized short log inoculation, drilling inoculation and log-end sandwich inoculation. The mycelial colonization of P. linteus on logs was good in the treatment of sterilized short log inoculation, but poor in the traditional methods such as drilling inoculation and log-end sandwich. The initial mycelial growth and the full mycelial colonization of P. linteus were the best on 20 cm logs under the condition of 42% of moisture content in log. Also the initial mycelial growth of P. linteus was accelerated over 12 hours of sterilization. Burying method of logs after 5-6 months of incubation was the best for formation of basidiocarp of P. linteus. The formation of fruiting body of P. linteus was quite good in the cultivation house at the 31-35$^{\circ}C$ and over 96% of relative humidity.
