Purification and Characterization of 2,4-Dichlorophenol Oxidizing Peroxidase from Streptomyces sp. AD001

  • Streptomyces sp. AD001 is a Gram-positive soil actinomycetes secreting an uncharacterized 2,4-dichlorophenol (DCP) oxidizing enzyme, whose activity is similar to the previously known Actinomycetes lignin-peroxidase (ALiP). This extracellular peroxidase was purified from Streptomyces sp. AD001 as a single protein band on an SDS-PACE by ammonium sulfate fractionation, Q-sepharose, concanavalin A, and Bio-Gel HTP column chromatographies. The molecular mass of the purified peroxidase was determined by SDS-PAGE to be 45.2 kDa, and 49.7 kDa with MALDI-TOF-MS, respectively. The highest level of peroxidase activity was observed at pH 7.5 and $30^{\circ}C$. The amino terminal sequence of the purified peroxidase (G-E-P-E-E-G-N-V-D-G-T-L) showed no significant homologies to my known proteins, suggesting that Streptomyces sp. AD001 may secrete a novel kind of bacterial peroxidase Initial rate kinetic data of the 2,4-DCP oxidation were best modeled with a random-binding bireactant system.

Drinking behaviors by stress level in Korean university students

  • The purposes of this study are to estimate the stress level of university students, and to verify the relationships between stress level and drinking behavior. A questionnaire survey was administered to 430 university students in the Gangwon area in Korea from November 5 to November 28, 2008, and data from 391 students were used for the final statistical analysis. The most stressful factor was "Worry about academic achievements" (2.86 by Likert-type 4 point scale). The subjects were divided into two groups, a low stress group (${\leq}65.0$) and a high stress group (${\geq}66.0$), by the mean value (65.1) and median value (66.0) of the stress levels. The drinking frequency was not different between the two stress groups, but the amount of alcohol consumption was significantly different ($P$ < 0.05). The portion of students reporting drinking "7 glasses or over" was higher in the lower stress group than in the higher stress group. In addition, factor 6, "Lack of learning ability", was negatively correlated with drinking frequency and the amount of alcohol consumption ($P$ < 0.05), and factor 3, "Worry about academic achievements", was negatively correlated with the amount of drinking ($P$ < 0.05). The major motive for drinking was "When overjoyed or there is something to celebrate" (2.62), and the main expected effect of drinking was "Drinking enables me to get together with people and shape my sociability" (2.73). The higher stress group showed significantly higher scores on several items in the categories of motives ($P$ < 0.01), negative experience ($P$ < 0.05), and expected effects ($P$ < 0.05) of drinking than the lower stress group. Our results imply that university students at the lower stress level may drink more from social motives in positive drinking environments, while those at the higher stress level may have more problematic-drinking despite their smaller amount of alcohol consumption.

  • This study was conducted to explore and classify the types of the subjectivity on the perception of the spouse using Q methodology. In this study, 42 Q statements were selected as the Q sample through interview. 42 people who married person with their spouse as P samples were forced to distribute 42 Q samples on 9 point scale. The collected data was analyzed using PC QUANL program. The results of the analysis are defined as two distinctive types. First, Type I(n=36) was defined as 'Love-Ties Type' and Type II(n=6) as 'Skein of thread-Person in charge Type'. Type I perceived their spouse as fate and ties emphasizing love for their spouse. And Type II recognized their spouse as someone requires effort like unravelling a thread and a person to be responsible for. In the result of this study, structural differences between the two types were found. And it will be used as a basic resource to support a healthy and happy marriage.

A Study on the Current Trend about Research of School Facilities (학교건축계획 연구의 동향에 관한 고찰)

  • It has been almost 20 years since Korea has started the research of school facilities, but there has not been any form of organized information or studies done about such a trend of the research. In this respect, this research places its focus on categorizing 44 policy studies initiated and proceeded by different organizations for last twenty years(1983-2004) and 186 volumes of the research about education facility plans and projects released by journal of Architectural Institute of Korea and journal of Korean Institute of Educational Facilities in 17 subjects of this study. It also concentrates on organizing data based on different levels of schools and periods of time as well as understanding the trend. The result of this research is followings. Considering every volume of the paper and the subjects that have previously been treated in those papers, the trend of this study is divided into the first stage of research period(1983-1993), the second stage of research period(1994-2001), and the third stage of research period(2002-present). Of all the papers, there has been the most number of papers objecting elementary schools(110 volumes, 56.1%), and most of them focused on space formation, size, and the present situation of utilization of each classroom in elementary schools. In the meantime, the studies about educational information, application of the media, space formation and size under teachers'consent, two-dimension types, a design of outdoor space(including gardens), the present situation of utilization, remodeling of school facilities, an evaluation of space and recognition-awareness analysis have been mainly done since 1995. Such a result mentioned above provides a sense of understanding about a level of studies about education facilities in our country, and as a basic source for future guidelines of studies and contents, in my personal opinion, it contains a high level of application.

A Case Study of Developing XML-based Web Contents Supporting PC and PDA Browser (PC 및 PDA 브라우저 지원을 위한 XML 기반의 웹 컨텐츠 개발 사례 연구)

  • Due to rapid advance of wireless communication technology and popularization of wireless devices, demand on wireless internet contents is gradually increasing. Therefore, there are many researches and solution developments to provide good qualified contents quickly for wireless internet. For example, researches into converting wired web contents into wireless web contents or using integrated markup language like XHTML basic to make contents. In this paper, I propose a method to develop XML-based web contents which uses PHP language for data fetch from MySQL database. This method use open source software for a cost saving. Due to use of PHP extension as a XSLT engine, this method is very easy to apply. For a example of this method, a web content of movie information is implemented for PC and PDA browser. Developing XML-based web contents is useful not only for supporting devices of multiple type, but also for rapid changes of user interface design and for exchange of contents between web sites.

A Study on the Consciousness of University-Students Concerning the Environmental Protection (環境保全에 관한 대학생의 認識調査 연구)

  • The purpose of this study is to provide the basic data in solving the environmental problem and in environmental education to University-students by searching the consciousness of Universitystudents' attitude toward and general views on the environmental protection. Out of total of 34 universities located in Seoul I randomly selected 11 universities and I again selected 700 university-students to answer my questionnaires. After that the questionnaires were added up by using computer. The major findings of this study are as follows 1. Regarding the overall conditions of the environmental pollution in our country, 91.1% of respondents marked "serious". 2. In suffering from Air pollution, 38.9% of respondents frequently suffered and 36.1% of them always suffered. Only 25.0% of total respondents represented their dislike about so-called "pollution-giving industries", which they do not wish to introduce for the sake of environmental protection. 3. In the source of information concerning the environment, 48.3% of them from the Television, 34.9% them from the newspaper. 4. In the attitude toward the participation for environmental protection, 28.0% of respondents expressed spontaneous attitude. 79.1% of total respondents have the basic desire to establish anti-pollution facilities and only 9.1% of total respondents have replied the ultimate responsibilities of enviironmental pollution. 5. Finally, only 13.3% of total respondents expressed their opinion that hte government officials willingly comply with students' request for better environmental protection and reflect them on their basic policies, and 32.1% of them said that the concerned government officials' attitude were positove and friendly toward the civilians, and 30.6% of them expressed their opinion that those concerned government officals ate holding professional and technicals know-which are necessaty and enough to the concerned field.

  • In accordance with the analysis results on the questionnaires about the awareness of employees according to positions and educational backgrounds in a public health organization, this thesis suggests the basic data on the purpose of the efficient operation and improvement of working environment in a public health organization. Two answers of the questions that "there is a possibility for promotion in the organization" and "the unit section in organization which I work for has a well-designed structure of organization" showed respectively high points in the questionnaire. On the other hand, three questions like there is a resonable way to bind the organization up, completed mission and rewards are connected, and my salary is fit for the current mission showed high points among the questions about the awareness on the organization according to the positions.

Adjunctive buccal and palatal corticotomy for adult maxillary expansion in an animal model

  • Objective: This study aimed to explore the usefulness of adjunctive buccal and palatal corticotomy for adult maxillary expansion in an animal model using cone-beam computed tomography (CBCT). Methods: Twelve adult sheep were randomly divided into two groups (each n = 6): a control group, where no treatment was administered, and a treatment group, where buccal and palatal corticotomy-assisted maxillary expansion was performed. CBCT scans were taken before (T1) and after (T2) treatment. Differences in all transverse dental and alveolar dimensions, alveolar width at crest level, hard palate level, horizontal bone loss, interdental cusp width and inter-root apex were assessed using Wilcoxon signed-rank and Mann-Whitney U-tests. Kruskal-Wallis tests and pairwise comparisons were used to detect the significance of differences among the inter-premolar and inter-molar widths. Results: CBCT data revealed significant changes in all transverse dental and alveolar dimensions. The mean interpremolar alveolar width showed an increase of 2.29 to 3.62 mm at the hard palate level, 3.89 to 4.38 mm at the alveolar crest level, and 9.17 to 10.42 mm at the buccal cusp level. Dental changes in the vertical dimension were not significant. Conclusions: Our findings based on an adult animal model suggest that adjunctive buccal and palatal corticotomy can allow for both skeletal and dental expansion, with the amount of dental expansion exceeding that of skeletal expansion at alveolar crest and hard palate levels by two and three folds, respectively. Therefore, this treatment modality is potential to enhance the outcomes of maxillary expansion in adults.

A Study on the Performances of Pullets and Subsequent Egg Production with the Substitution of Ground Sunflower Seeds (해바라기씨앗으로 사육된 병아리의 성장 및 산란계로서의 생산능력에 관한 연구)

  • Ten-week-old layer type pullets were used in this study, housed 10 birds per cage(61$\times$41cm) and were replicated 15 times After raising them on a 19% protein diet up to ten weeks of age they were fed a 12% protein corn-soy control diet or one with half or all of the soybean mea replaced with ground full-fat sunflower seeds. At 19 weeks of age, pulltes were transferred to a layer house and their respective diet was gradually changed to a 13.5% layer diet containing 635% oats. The 19% protein SFS diet in treatment 2 did not show my, adverse effect on body weight or subsequent egg productio. The higher level of SES(38%) appeared to delay sexual maturity somewhat as was indicated by body weight and egg production data. However, the higher replacement of soybean with SFS(38%) in the pullet ration may not be critical for sexual maturity, but beneficial in a full laying cycle. There were no adverse effects on later stages of production.
