Inhibitory Effect of Mugwort Extracts on Tyrosinase Activity (쑥 추출물의 Tyrosinase 효소활성에 미치는 억제효과)

  • To determine the inhibition of mushroom tyrosinase activity, fresh and dried mugwort, Artemisia princeps was extracted initially with water and ethyl alcohol, and subsequently with hexane, chloroform and ethyl acetate in that order. The highest yield was obtained from the ethyl acetate (15.2%) and hexane (15.5%) fraction of the ethanolic extract of fresh and dried mugwort, respectively. For all fractions tested, the inhibition of tyrosinase activity by fresh mugwort was higher than that of dried mugwort, and the inhibition ratio of tyrosinase activity was 98.9% in the chloroform fraction and 96.7% in the hexane fraction.

  • Eight compounds were isolated from the roots of Glycyrrhiza glabra by the tyrosinase inhibitory activity guided fractionation, and their structures were identified as liquiritigenin (1), isoliquiritigenin (2), isoliquiritigenin-2'-O-methyl ether (3), liquiritin (4), isoliquiritin (5), ononin (6), glycycoumarin (7), glycyrol (8) by analysis of spectral data. Compound 3 exhibited the most potent inhibitory effect on mushroom tyrosinase activity ($IC_{50}$, 47 M).

  • There are a lot of efforts to develop new compounds having skin whitening effect from natural products. Sparassis crispa is a medicinal mushroom containing more than 40% β-glucan, which exhibits anticancer and immunostimulating effects. The aim of this study was to assess the availability of β-glucan extracted from cauliflower mushroom S. crispa as a skin whitener through the evaluation of inhibitory effects of melanin synthesis and tyrosinase activity and their mechanisms. B16F1 cells were treated with S. crispa β-glucan (10, 100, and 1,000 ㎍/ml, respectively) and α-melanocyte stimulating hormone (α-MSH), simultaneously. Content of melanin synthesis and tyrosinase activity were determined. The expressions levels of tyrosinase, tyrosinase related protein-1 (TRP-1), TRP-2 and microphthalmia-associated transcription factor (MITF) were also measured by western blotting. Treatment with 10, 100 and 1,000 ㎍/ml S. crispa β-glucan and 200 nM α-MSH significantly decreased melanin synthesis by 13.9%, 18.7% and 39.5%, respectively, and tyrosinase activity by 15.6%, 26.9% and 43.2%, respectively, compared to the α-MSH alone group. In addition, S. crispa β-glucan inhibited expressions of tyrosinase, TRP-1, TRP-2 and MITF induced by α-MSH. These results indicated that S. crispa β-glucan inhibited MITF expression, thereby reducing tyrosinase expression and inhibiting melanin production in B16F1 melanoma cells. Therefore, S. crispa β-glucan might be available as a skin whitener.

Inhibitory Effect of Methanol Extract and Its Fractions from Grape Seeds on Mushroom Tyrosinase (포도씨 추출물과 분획물의 Tyrosinase 저해활성)

  • The objectives of this study were to determine inhibitory effect on tyrosinase activity and antioxidant activity of the methanolic extract from grape seeds and to investigate relationships between tyrosinase inhibitory activity and antioxidant activity of the extract. The 80% methanol extracts of grape seeds were fractionated subsequently with hexane, chloroform, ethyl acetate and water. The methanolic extract and fractions from grape seeds inhibited tyrosinase activity in a concentration dependent manner. The methanolic extracts showed the highest inhibitory effects on tyrosinase activity. The inhibitory effects of the ethyl acetate fraction from the methanolic extract on tyrosinase activity was higher than other fractions. The ethyl acetate fraction from methanolic extracts showed higher antioxidant activity and contained higher polyphenolic and flavonoid contents compared to other fractions. The correlation coefficients among the polyphenoilc content of methanolic extracts, ABTS radical cation scavenging activity and inhibitory effect of tyrosinase were relatively high. These results suggest that grape seeds may have potential as a depigmentation agent for cosmetics and functional food products.

Antimelanogenesis Effects of Fungal Exopolysaccharides Prepared from Submerged Culture of Fomitopsis castanea Mycelia

  • Fungal exopolysaccharides are important natural products having diverse biological functions. In this study, exopolysaccharides from Fomitopsis castanea mycelia (FEPS) were prepared, and the highest mushroom tyrosinase inhibitory activity was found. FEPS were prepared from cultivation broth by ethanol precipitation method. The extraction yield and protein concentration of FEPS were 213.1 mg/l and 0.03%, respectively. FEPS inhibited mushroom tyrosinase with the half maximal inhibitory concentration ($IC_{50}$) of 16.5 mg/ml and dose-dependently inhibited cellular tyrosinase activity (63.9% at $50{\mu}g/ml$, and 83.3% at $100{\mu}g/ml$) in the cell-free extract of SK-MEL-5 human melanoma cell and ${\alpha}$-melanocyte-stimulating hormone (${\alpha}-MSH$)-stimulated melanin formation in intact SK-MEL-5 human melanoma cell. The $IC_{50}$ of FEPS against NO production from RAW264.7 macrophage cells was $42.8{\pm}0.64{\mu}g/ml$. By in vivo study using a zebrafish model, exposure of FEPS at $400{\mu}g/ml$ to dechorionated zebrafish embryos for 18 h decreased the pigment density, compared to that without FEPS-treated control.

Screening for Antioxidizing and Tyrosinase-inhibitory Activities of the Extracts of Marine Algae from Busan Coastal Area (부산 연안 해조류 추출물의 항산화 활성 및 Tyrosinase 저해 활성 스크리닝)

  • The crude extracts of marine algae were screened for tyrosinase-inhibitory and radical scavenging effects. Among the samples tested, Symphyocladia latiuscula and Gloiopeltis furcata were found to be the most effective in DPPH radical scavenging test while Gigatina tenella, Sargassum thunbergii, and Sargassum sp. were moderately active. For the inhibition against mushroom tyrosinase, Symphyocladia latiuscula and Sargassum confusum showed the strongest inhibition. Codium adhaerens, Corallina pilulifera, Carpopeltis cornea, Halymenia acuminata, Hizikia fuziformis, Porphyra suborbiculata, and Enteromorpha linza exhibited mild inhibitory potency.

Isolation of Antioxidant and Antibrowning Substance from Chionanthus retusa Leaves (이팝나무 잎으로부터 항산화 및 항갈변물질의 분리)

  • This study was performed to examine in vitro antioxidative activities such as DPPH radical scavenging activity, reducing power and tyrosinase inhibitory effect of various solvent fractions from Chionanthus retusa leaves. Ethyl acetate fraction showed potent antioxidative activity and tyrosinase inhibitory effect. The active compound was isolated from the butanol fraction by silica gel column chromatography and MPLC. The isolated compound was luteolin-4'-O-glucoside determined by $^1H$, $^{13}C$-NMR and 2D NMR. Compared with several antioxidant compounds, luteolin-4'-O-glucoside exhibited effective DPPH radical scavenging activity and reducing power in a concentration dependent manner. Bioassay with pure luteolin-4'-O-glucoside showed a dose-independent inhibitory effect on L-DOPA oxidation by mushroom tyrosinase and its $IC_{50}$ values were established as 23.2 ${\mu}g/mL$. Therefore, we may suggest that luteolin-4'-O-glucoside can be used as a food additive possessing the potent antioxidative activity and skin-whitening cosmetic material.

  • Tyrosinase is one of the important enzymes in the mammalian melanin synthesis. In the process of melanin synthesis, tyrosine is oxidized to DOPA (3,4-dihydroxyphenylalanine), and DOPA is further oxidized to dopaquinone. Tyrosinase is an enzyme catalyzing this oxidation of tyrosine, so chemicals that inhibit the activity of tyrosinase can be used as skin whitening agents. In this study, we isolated five constituents from the 80% MeOH extract of Morus bombycis cortex by bioactivity-guided fractionation. We performed mushroom tyrosinase inhibition assay. As a result, 7,2',4'-trihydroxyflavanone (1), 2',4',2,4,-tetrahydroxychalcone (2), and oxyresveratrol (3) showed the more potent inhibitory effect compared to kojic acid, a well-known skin whitening agent with antityrosinase effect. Moracinoside M (4) and moracin M-3'-O-${\beta}$-D-glucopyranoside (5) also showed the moderate tyrosinase inhibitory activities.

Isoliquiritigenin : A Competitive Tyrosinase Inhibitor from the Heartwood of Dalbergia odorifera

  • Effect of isoliquiritigenin isolated from the heartwood of Dalbergia odorifera T. Chen (Leguminosae) on mushroom tyrosinase activity was investigated in vitro using L-tyrosine and L-3, 4-dihydroxyphenylalanine (L-DOPA) as the substrates. When L-tyrosine was used as a substrate, both isoliquiritigenin and kojic acid, a positive control, inhibited tyrosinase activity in a concentration-dependent manner. IC$_{50}$ values of isoliquiritigenin and kojic acid were 61.4 and 52.2 ${\muM}$, respectively. However, isoliquiritigenin showed week inhibitory effect on the oxidation of L-DOPA by tyrosinase with inhibition ratio of 9.1 ${\pm}$ 7.1% at 100 ${\muM}$. It is also suggested that 3-unsubstituted and 4-hydroxyl phenyl group in isoliquiritigenin plays an important role on the inhibition of tyrosinase activity when L-tyrosine was used as a substrate. Analysis of Lineweaver-Burk plot showed that isoliquiritigenin acts as a competitive inhibitor in case of L-tyrosine as a substrate.

  • Objective : The aim of this study is to assess the effect of Schizandrae Fructus on melanin synthesis inhibition and whitening effect. Methods : We assessed inhibitory effects of Schizandrae Fructus on melanin-release from B16F10, on melanin production in B16F10, on mushroom tyrosinase activity in vitro, on tyrosinase activity in B16F10 and effect of Schizandrae Fructus on the expression tyrosinase, TRP-1, TRP-2, PKA, ERK-1 ERK-2, AKT-1, MITF in B16F10. Results and Conclusion : 1. Schizandrae Fructus inhibited melanin-release, melanin production in B16F10. 2. Schizandrae Fructus inhibited tyrosinase activity in vitro and in B16F10. 3. Schizandrae Fructus suppressed the expression of tyrosinase, TRP-1, TRP-2, PKA, ERK-2 in B16F10.
