Effect of Tree Species, Inoculation Amount and Inoculation Methods on Mycelium Growth and Sclerotia Formatino of Poria cocos Wolf (樹種, 接種量 및 接種方法이 茯笭 菌絲生長과 結笭에 미치는 영향)

  • When medicinal mushroom, Poria cocos, is cultured , inoculation method of spawn is cross slice inoculation of which the both sides of pine tree were peeled and spawn of P.cocos was inoculated. However, this method required lots of inoculation amount. This study was carried out to improve the culturing method of P. cocos. A good growth of P.cocos was observed in MCM(mushroom complete medium), showing proper mycelia growth and density. In inoculation amount, conventional method(cross slice inoculation) requires 20 bottles of spawn. In contrast, short log method required 8 bottles of spawn and drilling inoculation method 2~3 bottles, which could save by 60% and 85-90% respectively. In the selectrion of tree species, pine and larch had better condition for spawn culture and sclerotia formation condition.In terms of yield , pine was 33.7kg/3.3$m^2$. In the yield of pine, conventional method was 23.4kg/3.3$m^2$, drilling inoculation 29.4kg/3.3$m^2$, short log inoculation 31.7kg/3.3$m^2$, therefore drilling inoculation could increase by 25% and short log inoculation 35%, In addition, management cost was also saved.

Promotion of Tricholoma matsutake mycelium growth by Penicillium citreonigrum

  • Tricholoma matsutake has been the most valuable ectomycorrhizal fungi in Asia because of its unique flavor and taste. However, due to the difficulty of artificial cultivation, the cultivation of T. matsutake has relied on natural growth in forests. To cultivate the T. matsutake artificially, microorganisms in fairy rings were introduced. In this study, we isolated 30 fungal species of microfungi from the soil of fairy rings. Among them, one single fungal strain showed a promoting effect on the growth of T. matsutake. The growth effect was confirmed by measuring the growth area of T. matsutake and enzyme activities including a-amylase, cellulase, and b-glucosidase. In comparison with control, microfungal metabolite increased the growth area of T. matsutake by 213% and the enzyme activity of T. matsutake by 110-200%. The isolated fungal strain was identified as Penicillium citreonigrum by BLAST on the NCBI database. The Discovery of this microfungal strain is expected to contribute to artificial cultivation of T. matsutake.

Comparison of Antibacterial and Antioxidant Activities of Mushroom Mycelium Culture Extracts Cultivated in the Citrus Extracts (감귤농축액 첨가배지에서 배양한 버섯균사체 추출물의 항균활성 및 항산화활성 비교)

  • This study was carried out to investigate the antimicrobial and antioxidative effects of mycelium cultural extract from mushroom. Mushroom mycelium was grown in a defined synthetic liquid medium and citrus extracts, and the culture extracts were examined for antioxidant and antibacterial activity. Myceliums of Phellinus linteus, Cordyceps militaris, Coriolus versicolor, Sparassic crispa, Agaricus blazei, lnonotus obliquus, Lentinus edodes, Hericium erinacium, Gonoderma lucidium in 10% citrus extract supplemented medium and synthesis medium were incubated in a shaking incubator (120 rpm, $24{\sim}30^{\circ}C$ ) for $7{\sim}15$ days. The antimicrobial activity of the culture fluid of mushroom mycelium grown in submerged liquid culture was tested against 12 microorganisms which were fish pathogens and common bacterial species. The culture extracts showed high activity against Vibrio sp. and had poor effect on Streptocouus sp., S. parauberis, S. iniae. The culture extracts obtained from the synthetic medium showed $30{\sim}93%$ of the 1,1-diphenyl-2-picrylhydrazyl radical scavenger activity, the culture extracts obtained from the citrus extracts medium exhibited antioxidant activity up to 55%.

Development of Effective Screening Method for Resistance of oyster mushroom to Trichoderma disease in vitro (느타리버섯 푸른곰팡이병 저항성 실내검정 방법의 개발)

  • Trichoderma disease of oyster mushroom has not been effectively detected in the field for testing its resistance against the disease with its varieties. In this study, we investigated the methods to detect its resistance in the laboratory by using media, which enables us to understand the relevant characteristics (e.g., lysis, toxin enzyme, mycelial growth rate). In coculturing with strains of Trichoderma and oyster mushroom, it is possible to observe the difference in the resistance of oyster mushroom against Trichoderma with the phenomena of barrage reaction, overgrowth and lysis. We also observed the inhibition of mycelial growth of oyster mushroom using the dilution method with 48-well plate, but could not observed the inhibition of mycelial growth using the filter paper method of cultural supernatant. In simultaneously culturing both Trichoderma and oyster mushroom, it was possible to detect the inhibition of the mycelial growth of oyster mushroom, but Trichoderma mycelium did not overgrow against oyster mushroom. We found that the pathogenicity was efficient in using solid medium with the phenomena of overgrowth and lysis by inoculating Trichoderma on top of mycelia of oyster mushroom. In conclusion, the methods (e.g., coculture method, dilution method with 48-well plate, post-inoculation method) are recommended to detect the resistance of oyster mushroom against Trichoderma disease.

Varietal characteristics of new white button mushroom 'Seolwon' in Agaricus bisporus

  • Commonly known as the button mushroom, Agaricus bisporus is one of the most widely cultivated mushroom species of edible fungi. In the breeding of new button mushroom, Seolwon was developed by crossing two homokaryons. Because of the predominantly pseudohomothallic life cycle, only a small percentage of homokaryotic meiospores are produced, which do not fruit. Homokaryotic cultures derived from these types of single spores produce a vegetative mycelium that contain a variable number of genetically identical nuclei per cell. After crossing two homokaryons, hybrids were cultivated on a small scale and on a commercial scale at a farm. The spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of Seolwon on CDA was better at $25^{\circ}C$ when it was compared with that of Seolgang. The mature cap shape of new strain Seolwon is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 39.7 mm on average. In comparison with white strain Seolgang, the strain had a yield that was 11% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 9.7% firmer with a good shelf life. Days of fruiting body were 1-2 days later than those of Seolgang. The physical characteristics such as springiness, chewiness, adhesiveness, gumminess were better than that of Seolgang.

  • This experiment was carried out to investigate proper conditions for protoplast isolation and reversion from Ganoderma lucidum and Gctnoderma sp.. In G. lucidum, 10 mg. $ml^{-1}$ Novozyme 234 with 0.6 M sucrose was proper for protoplast isolation. The optimal reaction time of mycelium with lytic enzyme was five hrs. Protoplast isolation from four-day-old mycelium was the most effective. Protoplast isolation from four-day-old mycelium in G. sp. was optimum in the combination of N ovozyme 234 and ${\beta}-glucuronidase$ with 0.6 M sucrose. MCM was suitable for reversion in G. lucidum while SCM was good for G. sp.. The most effective osmoticum stabilizer for protoplast reversion in G. lucidum and G. sp. was 0.6 M sucrose.

Anti-neuroinflammatory effects of cultivated red ginseng with fermented complex mushroom-cereal mycelium on lipopolysaccharide activated BV2 microglial cells (LPS로 인해 활성화된 BV2 Microglia에서 발효 복합버섯-곡물 숙성균주 배양 홍삼(紅蔘)의 뇌신경염증 보호효과)

  • Objectives : Neuroinflammation is a common pathological mechanism of neurodegenerative diseases, and the development of therapeutic agents is urgently needed. Red ginseng has been known to be good for the immune stimulation in Eastern Asia. Although the immuno-stimulatory activity of red ginseng are already known, the neuro-protective effects of cultivated red ginseng with fermented complex mushroom-cereal mycelium (RGFM) have not been conducted. Thus, in this study, we tried to investigate the anti-neuroinflammatory effect of RGFM water extract on lipopolysaccharide (LPS) stimulated BV2 cells. Methods : BV2 cells were pretreated with RGFM 1 h prior to LPS exposure. To determine the neuro-protective effects of RGFM water extract, we measured the expression of inflammatory mediators including inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2 and nitric oxide (NO) and pro-inflammatory cytokines such as interleukin (IL)-1𝛽, IL-6 and tumor necrosis factor (TNF)-𝛼 in LPS-stimulated BV2 cells. In addition, to find out the regulatory mechanism of RGFM water extract, we assessed the protein levels of mitogen-activated protein kinases (MAPKs) and inhibitory 𝜅B𝛼 (I𝜅B𝛼) by western blotting. Results : In our study, treatment of RGFM reduced the mRNA expression of iNOS and COX-2 and suppressed NO production in LPS-stimulated BV2 cells. Additionally, the secretion of IL-1𝛽 and TNF-𝛼 but not IL-6 was significantly inhibited by RGFM. Furthermore, RGFM water extract inhibited the phosphorylation of c-Jun N-terminal kinase (JNK). Conclusions : Taken together, these findings suggest that RGFM water extract has a protective effect on neuroinflammation through inhibition of JNK.

  • The effects of mycelium cultural extract supplemented diet, hematology and disease resistance against Vibrio anguillarum in juvenile flounder, Paralichthys olivaceus were evaluated. Fish were fed the Phellinus linteus with Coriolus militaris versicolor mixed mycelium cultural extract supplemented diet, Phellinus linteus mycelium cultural extract supplemented diet and Coriolus militaris tmycelium cultural extract supplemented diet a commercial diet for 12 week. The body weight and length gain from the fish fed on daily the phellinus with coriolus versicolor mixed mycelium cultural extract supplemented diet, phellinus linteus mycelium cultural extract supplemented diet and Coriolus versicolor mycelium cultural extract supplemented diet of each mycelium cultural extract were higher than the control and the Glutamic oxalacetic transaminase (GPT) were of lower than the control. The relative precent survival rate (RPS) after an artificial challenge with $7{\times}10^5\;CFU$ of Vibrio anguillarum per fish was higher than the control.

  • Interspecific fusion products were obtained from fusion of Ganoderma applanatum and Ganoderma luridum. Frequency of fusion was 0.77-1.38%. Fusion products were selected by the comparision of morphology and color of colony. Fusion Products had chracteristics of two parental strains and generally grew faster than the parents. Some fusants were segregated on GCM. Fusion products were confirmed by mycelial morphology and electrophoretics pattern of esterase isozyme from mycelium. Most of fusion products were lack in clamp connection but those fusion products, that were segregated produced mycelium with and without clamp connection. Some of the fusion products produced fruit body on sawdust medium.

The effect of different culture conditions of liquid spawn on the quality characteristics of shiitake mushroom (Lentinula edodes) (표고의 액체종균 배양 조건 및 품질 특성)

  • To improve the productivity of shiitake mushroom (Lentinula edodes), seven different types of media for liquid spawn (denoted as "A" to "G") were prepared with 0.3% soybean meal and varying sugar and glucose concentrations. During 14 days of incubation, the pH of the liquid culture gradually acidified with increasing incubation period. Additionally, there was a significant, but not prominent, difference in the degree of acidification depending on the sugar to glucose ratio. Liquid spawn culture "G," which had the highest sugar content was the most acidic on the last day of incubation. Mycelium dry weight increased significantly with increasing incubation period, and there was no significant difference in mycelium dry weight irrespective of the sugar to glucose ratio even after 14 days of culture. The inoculation of liquid spawn in sawdust medium with an inoculation volume ${\geq}45mL$ and incubation period of 15 to 18 days were the optimal culture conditions. Productivity of fruit bodies in sawdust medium and mushrooms treated with liquid spawn was significantly higher compared to solid spawn treatment. The mushrooms treated with liquid spawn had better chewiness, and the free amino acid content, which is associated with savory taste, was higher in these mushrooms compared to those treated with solid spawn.
