Comparison with various mulberry leaves' and fruit's extract in lipid accumulation inhibitory effect at adipocyte model

  • In relation to the global increase trend of obesity population, there is a demand for the development of foods having high functional activity by mass-extracting anti-obesity active substances using mulberry product such as leaf and fruit. Therefore, we evaluated the antiobesity efficacy according to varieties by using the mulberry leaves and fruit extracts. At the same time, high active varieties were selected. For this purpose, the effects of the extracts of the mulberry leaf and fruit on 3T3-L1 adipocyte differentiation were examined. As a result, in the case of mulberry leaves, the lipid accumulation inhibitory rate of 'Cheongolppong' was higher than that of the control at $500{\mu}g/ml$ treatment. And in the case of the extract of mulberry fruit, 'Daesim' showed the highest lipid accumulation inhibitory rate compared with the control at 50 times of diluted extract.

오디, 뽕잎 및 누에의 혼합비율에 따른 Streptozotocin 유발 당뇨쥐에서의 항산화 효과 및 지질대사개선 효과 (Antioxidant Effects and Improvement of Lipid Metabolism of Mulberry fruit, Mulberry Leaves and Silkworm Powder with Different Mixing Ratios in Streptozotocin-Induced Diabetic Rats)

  • This study was conducted to investigate the effects of mulberry fruit, mulberry leaves and silkworm powder with different mixing ratios on hepatic antioxidative system and lipid metabolism in streptozotocin-induced diabetic rats. Sprague-Dawley male rats weighing $100{\pm}10g$ were induced diabetic by 50 mg/kg bw streptozotocin and randomly assigned to following experimental groups; normal diet group (DM), 0.3% and 0.6% mulberry fruit diet groups (F and 2F), 0.3% mulberry leaves diet group (M), 0.3% silkworm powder diet group (S), 0.15% mulberry fruit+0.15% mulberry leaves diet group (FM), 0.15% mulberry fruit+0.15% silkworm powder diet group (FS), 0.1 % mulberry fruit+0.1 % mulberry leaves+0.1% silkworm powder diet group (FMS). The experimental diets were fed for 4 weeks. Hepatic SOD activity was not changed significantly by any of single or combined supplementations of mulberry fruit, leaves and silkworm powder but GSH-px and catalase activities were increased by the groups supplemented with two or three of the test ingredients (FM, FS, FMS) as compared with the DM group. Hepatic TBARS value was not reduced significantly by any of the supplementations but lipofuscin contents were significantly reduced in the FM, FS and FMS groups as compared with the DM group. Hepatic mitochondria and microsomal carbonyl values were reduced by the single and combined supplementations of the test ingredients. Hepatic HMG-CoA reductase activities were increased in the all supplementation groups as compared with the DM group. Hepatic total lipid and triglyceride contents were increased but cholesterol contents reduced in the supplemented groups. The effects on the enzyme activities, peroxide or its products and lipid contents were most remarkable in the FMS group. In conclusion, mulberry fruit, mulberry leaves and silkworm powder have the favorable effects on antioxidative system and lipid metabolism in the diabetic liver and the mulberry fruit, leaves and silkworm powder with equal ratio exert the synergistic effect expectedly to prevent diabetic complications.

Flavonoids analysis in leaves and fruits of Korean mulberry cultivar, Baekokwang having white fruits

  • Morus alba has white and/or purple fruits with a very sweet taste and low acidity. Most Korean mulberry trees have purple fruits. However, Baekokwang is a unique mulberry genetic resource in Korea with white fruits. In this study, flavonoids contents of Baekokwang mulberry leaf and fruit were analyzed using ultrahigh performance liquid chromatography coupled with diode array detection and quadrupole time-of-flight mass spectrometry (UPLC-DAD-QTOF/MS) technique. UPLC-DAD-QTOF/MS chromatogram showed that 15 flavonoids and 9 flavonoids were isolated and identified from the mulberry leaf and fruit. Total flavonoids contents of Baekokwang leaves and fruits were 812.7 mg and 35.0 mg, respectively. Baekokwang leaves had 4 major flavonoids including quercetin 3-O-(6"-O-malonyl) glucoside, 235.3 ppm, kaempferol 3-O-(6"-O-malonyl) glucoside, 132.3 ppm, kaempferol 3-O-rutinoside (nicotiflorin), 108.1 ppm, and quercetin 3-O-rutinoside (rutin), 103.8 ppm. Baekokwang fruits had 3 major flavonoids including quercetin 3-O-(6"-O-malonyl) glucoside, 13.0 ppm, quercetin 3-O-rutinoside (rutin), 7.8 ppm, and kaempferol 3-O-rutinoside (nicotiflorin), 5.7 ppm. From the above results, mulberry leaves have rich flavonoids compared to its fruits.

Quali-Quantitative Analysis of Flavonoids for Mulberry Leaf and Fruit of 'Suhyang'

  • BACKGROUND: Globally, mulberry (Morus sp.) is exploited for feeding leaf to silkworms in order to obtain silk fiber or for animal feedstock production. Also, mulberry fruit is known to a by-product that was produced from mulberry tree after harvesting leaves for silkworm rearing, as a yield and consumption of mulberry fruit was increased, it has been fixing to a newincome crop. Mulberry leaves and fruits are used for the health benefits of human beings. Mulberry contains various bioactive components, such as alkaloids and flavonoids. Mulberry flavonoids are an important part of the diet because of their effects on human nutrition. The flavonoids in mulberry leaf and fruit of 'Suhyang'(Morus alba L.) were determined. METHODS AND RESULTS: Flavonoids for mulberry leaf and fruit of 'Suhyang' were analysed using ultrahigh performance liquid chromatography coupled with diode array detection and quadrupole time-of-flight mass spectrometry (UPLC-DAD-QTOF/MS)technique. An UPLC-DAD-QTOF/MS system was used, and identification of mulberry leaves constituents was carried out on the basis of the complementary information obtained from LC spectra, MS ions, and MS/MS fragments. The mulberry leaf (16 flavonoids) and fruit (9 flavonoids) were isolated and analyzed from Suhyang using UPLC-DAD-QTOF/MS chromatogram. To the best of our knowledge, Quercetin 3-O-(6"-O-malonyl) glucoside and quercetin 3-O-rutinoside (rutin) was detected on the highest content in leaf and fruit, respectively and further research will be devoted to evaluate their biological activity. CONCLUSION: Obtaining information about the concentration of functional materials in mulberry leaves could contribute to the development and promotion of processed, functional products and offer possible industrial use of 'Suhyang', holding promises to enhance the overall profitability of sericulture.

오디 재배양식 및 지역에 따른 균핵병 방제 효과와 농약 잔류 특성 (Pest Control and Analysis of Residual Pesticides of Mulberry Fruit and Leaf against Popcorn Disease by Cultivated Type and Region)

  • Background: As the Positive List System (PLS) is implemented in broad application to agricultural products, attention to the correct use of pesticides is also needed in the production of mulberry leaves and fruit. In this regard, three types of pesticides against mulberry popcorn disease were applied 2 - 3 times both in the field and greenhouses to prepare safety standards. Residual pesticide analysis was conducted on mulberry fruits and leaves. Methods and results: Three pesticieds, thiophanate-methyl, thiophanate-methyl·triflumizole and fluopyram registered as PLS pesticides for mulberry popcorn disease, were sprayed in the Wanju and in Buan regions, after which residual pesticide analysis was conducted using high-performance liquid chromatography (HPLC). As three pesticieds were either undetected or below the permissible level in mulberry fruit, demonstrating that they were suitable for safe spraying. However, 5.6 mg/kg of thiophanate-methyl was detected in the greenhouse after three application, which was slightly above maximum residue limit (MRL). Furthermore the level of thiophanate-methyl·triflumizole was higher than 0.2 mg/kg ("Gwasang No. 2" variety, spraying twice) or similar to 0.09 mg/kg ("Daesim" variety, spraying thrice) the permissible level (0.1T mg/kg) as the thiophanate-methyl was detected in mulberry leaves in the greenhouse. Conclusions: The spraying frequency for controlling mulberry popcorn disease in greenhouses should be limited to two times or less, especially when mulberry leaves are treated with thiophanate-methyl·triflumizole careful consideration is required if the leaves are to be used as food materials.

대사증후군 개선을 위한 뽕잎, 오디, 누에 분말의 혼합 비율 최적화 (Optimization of Mixing Ratio of Mulberry Leaf, Mulberry Fruit, and Silkworm for Amelioration of Metabolic Syndrome)

  • Objectives: The aim of this study was optimized mixing ratio of mulberry leaf, mulberry fruit, and silkworm for amelioration of the metabolic syndrome by using response surface method (RSM). Methods: Antioxidant, antidiabetic and antihypertensive activities of fifteen mixed powder of mulberry leaf, mulberry fruit, and silkworm by RSM were measured as indicators of metabolic syndrome. Results: The optimal mixing ratio of mulberry leaves, mulberry fruits, and silkworm with the greatest antioxidant, antidiabetic and antihypertensive activities was as follows: 2.5890 of mulberry leaf (A), 0.1222 of mulberry fruit (B), 2.9999 of silkworm (C). At this time, it was predicted that the total polyphenol content was estimated to be 185.51 tannic acid equivalent mg/g, 1, 1-diphenyl-2-picrylhy drazyl radical scavenging activity 84.77%, 1-deoxynojirimycin content 415.66 mg/100 g, ${\alpha}-glucosidase$ inhibitory activity 64.31%, ${\gamma}-aminobutyric$ acid content 267.77 mg/100 g, potassium content 1,899.11 mg/100 g, and angiotensin-converting-enzyme inhibitory activity was estimated to be 73.78%. Conclusions: It was concluded that the significant effect of the mulberry leaf, mulberry fruit, and silkworm on the metabolic syndrome-related biological activity indices (antioxidant activity, antidiabetic activity and antihypertensive activity) was as follows: 2.5890 of mulberry leaf (A), 0.1222 of mulberry fruit (B), 2.9999 of silkworm (C).

뽕잎오디쌀이 흰쥐의 혈당농도와 체지방 및 혈청지질 분획에 미치는 영향 (Effects of Rice Added with Mulberry Leaves and Fruit on Blood Glucose, Body Fat and Serum Lipid Levels in Rats)

  • 재성형 뽕잎오디혼합쌀 개발과 관련하여 당뇨유발쥐와 고지방식이를 급여한 흰쥐에게 뽕잎오디쌀이 혈당농도와 체지방 및 혈청 지질 농도 등에 미치는 영향을 조사하였다. 혈당실험에서 뽕잎오디쌀조성물은 곡류와 잡곡류 등 85%, 뽕잎과 오디즙 15%로 구성하였고, AIN-diet와 50:50 비율로 혼합하여 4주간 급여한 결과 뽕잎오디쌀 급여군은 당뇨대조군보다 실험종료 시 체중은 28% 증가한 반면 공복혈당치는 12.4%, 혈청 중성지방 농도는 34% 감소하였다(p<0.05). 지질실험의 경우 4주 동안 고지방식이(50%)와 뽕잎오디쌀조성물(50%)의 혼합식이와 고지방식이(50%)와 기본식이(50%)의 혼합식이를 급여한 결과 뽕잎오디쌀 급여군은 고지방대조군에 비해 체중과 체지방(EFP)이 각각 8%씩 낮았고, 혈청 중성지방 농도는 약 19% 감소하였다(p<0.05). 이상에서 뽕잎오디쌀식이는 혈당저하 및 중성지방을 감소시키는 것으로 나타났고, 유의성은 없었으나 다소간 체중과 체지방 감소 효과를 확인할 수 있었다.

오디 생산용 뽕나무 재배 현황 조사 (Survey and analysis of mulberry tree for mulberry production)

  • 지금까지 오디는 누에 사육을 위한 뽕나무 재배를 하고 남은 나무에서 생산되는 부산물로 여겨져 왔으나, 오디가 생산량과 소비가 크게 늘어나면서 새로운 소득작목으로 정착되어 가고 있다. 그러나 기상재해에 의한 피해로 오디 안정생산이 문제가 되고 있어 농업재해보험 도입이 필요하다. 농업재해 보험 도입을 위한 기초 연구로서 오디 생산용 뽕나무 재배양식, 수형, 품종별 결실수, 오디 균핵병 발생양상, 생산량 및 판매가격 등을 조사하였으며, 그 결과는 아래와 같다. 10a당 뽕나무 식재주수는 낮추만들기에 적합한 품종을 재배하는 부안과 양평지역이 많았으며, 크게 재배하는 고창지역은 적었다. 최장 결과모지당 오디 결실수는 과상 2호, 수원뽕, 익수뽕, 청일뽕 순으로 많았으며, 오디 균핵병 발병율은 과상2호, 익수뽕, 청일뽕, 수원뽕 순으로 높았다. 10a당 오디 수확량은 청일뽕, 과상2호, 수원뽕, 익수뽕 순으로 많았으며, 수령별 수확량은 나무수령이 오래될수록 많았으나, 과상 2호는 어린 나무가 많았다. 오디 판매가격은 경기도 양평지역이 가장 높았으며, 고창지역이 가장 낮았다.

몇 가지 뽕품종에 따른 오디의 형태 및 화학적 성분의 특성 (Morphological and Chemical Characteristics of Mulberry(Morus) Fruit with Varietes)

  • The morphological and chemical characteristics of leaves and fruits were observed in the three mulberry varieties, including Daeryugppong(Morus Lhou(Ser.) koidz), Kugsang 20(Morus Lhou(Ser.) koidz) and Cataneo(Morus alba L.). The leaf development in spring was earliest in Cataneo and latest in Kugsang. Content of total nitrogen and Mg in leaf was the highest in Cataneo while that of P2O5, K and Ca in Daeryukppong. Flowers started to bloom from May 8 and in full bloom around May 15. Mature fruits began to set from June 10 to 15 and lasted by July 10 in Cataneo. Average fruit weight was heaviest in Kugsang 20(3.52 g/fruit), while lowest in Daeryukppong(1.61 g/fruit). In fruits, glucose and fructose were the major sugars. Citric acid was the most abundant organic acid in three varieties with its average content from 0.8 to 0.14%. The major pigment in fruit was anthocyanin and its content varied among varieties. The stability of anthocyanin was evaluated under various pH, temperature, and sugar concentrations. Rutin was the major flavonol glycoside present in fruits and its content varied from 0.92 to 3.36 mg/gDW. Other flavonol glycosides such as isoquercitrin and quercitrin were also detected in fruit.

강원도 일대에서 채취한 야생뽕의 시기별 뽕잎과 오디의 항산화능 분석 (Antioxidant Capacity Analysis of Water-Soluble Substances according to Maturity Stages in Yield-type Mulberry Leaves and Fruits Collected from Kang-Won Province)

  • 야생 뽕나무의 자원화 및 이용기술을 개발하기 위해 강원도 원주, 인제, 양양 3개 지역에서 성숙 단계별로 채취한 야생 뽕나무를 대상으로 뽕잎과 오디의 항산화능을 비교분석 하였다. 항산화능 측정장치(munilum L-100, ABCD GmbH) 및 ARAW-KIT(anti-radical ability of water-soluble substance)를 사용하였으며, ascorbic acid를 표준물질로 사용하여 calibration curve를 작성하였다. 각각의 지역에서 채취한 야생뽕의 형태학적 특성을 조사한 결과, 원주 지역 채집 개체는 백상형(Morus alba L.), 인제 지역 채집 개체는 산상형(Morus bombycis Koidz) 및 양양 지역 채집 개체는 노상형(Morus Lhou (Ser.) Koidz)의 뽕나무로 분류되었다. 강원도 원주, 인제 및 양양 지역에서 자생하고 있는 야생 뽕나무의 뽕잎에 대한 전체적인 항산화능을 분석한 결과 춘기 5개엽기의 어린 뽕잎의 항산화능이 가장 높았으며, 시기가 경과함에 따라 뽕잎의 항산화능은 점차 감소하는 경향이었다. 그러나 개체마다 각각의 특성을 나타냈다. 오디가 결실된 야생 뽕나무 중 오디의 수량성 및 크기 등 착과상태를 고려하여 이용가치가 있을 것으로 예상되는 개체는 No 7, 8, 10이었다. 산상형의 특징을 갖는 No. 7 개체 뽕나무는 뽕잎의 항산화능이 높을 뿐 만 아니라 뽕나무의 생육 상태 및 오디의 결실 상태도 양호했으므로 기능성 및 오디생산용 육종 유전자원으로 이용할 가치가 있는 것으로 판단되었으며, No. 8 및 No. 10 개체 뽕나무는 오디 생산을 위한 유전자원으로 이용할 수 있을 것으로 보인다.
