Seasonal Resveratrol Contents of Wild-type Mulberry Leaves Collected from Gangwon Province in Korea (강원도에서 채취한 야생 뽕잎의 생육 시기별 resveratrol 함량)

  • Resveratrol is naturally occurring phytoalexin compounds produced by grape berries, peanuts, pine tree, mulberry fruits and their products in response to stress such as fungal infection, heavy metal ions or UV irradiation. The objective of this study was to determine resveratrol contents in wild-type mulberry leaves (Morus alba L., M. bombycis Koidz, and M. Lhou (Ser.) Koidz) collected from Won-ju, In-je and Yang-yang regions, Gangwon province, Korea. Resveratrol contents in wild-type mulberry leaves were higher in Autumn's than in Spring's, and higher than 16 cultivar's. Among the samples, 'No. 3', collected from Won-ju region and 'No. 7' collected from In-je showed high resveratrol contents of 539 and $491{\mu}g$/100 g DW (dry weight; DW), respectively, through the collection time. Therefore, two mulberry trees might be promising as potent resources for mulberry breeding and functional material development.

  • In this study, we tried to find the subject to inhibit H. pylori from Cheongmoknosang mulberry leaves extracts and to purify and identify them. Total phenolic compounds of hot water and 80% ethanol extracts are 17.6 and 16.1 mg/g. The activity of H. pylori inhibition at 80% ethanol extracts was determined as 15mm clear zone. The purification of inhibitory compounds were carried on $C_{18}$ column and MCI-gel CHP-20 column chromatography which were used a gradient procedure as increasing ethanol in $H_2O$. The chemical structure of purified inhibitory compounds on H. pylori were identified chlorogenic acid, caffeic acid, and rosmarinic acid by FAB-MS, $^1H-NMR$, $^{13}C-NMR$ and IR spectrum.

Antifungal Activity of Methylene Chloride Fraction of Pimpinella brachycarpa Against Aspergillus niger (참나물 Methylene Chloride 분획의 Aspergillus niger에 대한 항진균 활성)

  • In order to develop safe and economic novel antifungal agents, we prepared 73 methanol extracts from medicinal and edible herbs and examined their 365 solvent fractions using n-hexane, methylene chloride, ethylacetate, butanol and water residue based on the sequential organic solvent fraction method. When using the various fractions in the screening step for antifungal activity, we discovered ethylacetate fraction of Morus alba L., methylene chloride fraction of Pimpinella brachycarpa (MCPB), and n-hexane fraction of Salvia miltiorrhiza Bunge, which all have activities in methanol extracts, as potential sources of antifungal agents. Amongst these, the antifungal activity of P. brachycarpa has not to date been reported on. In addition, the mycelial growth inhibition and spore germination inhibition activities of MCPB against A. niger were confirmed by disc-diffusion assay in a 10 day culture. The MIC and MFC of MCPB were determined as 0.25 and 0.5 mg/ml, respectively. The MCPB has no hemolytic activity against human RBC at 0.5 mg/ml and glycoside-flavonoids are theorized to be active constituents. These results suggest that MCPB has a prominent antifungal activity and that the application of sequential organic solvent fractions, instead of simple natural product extracts, is useful in the screening process of novel bioactive substances.

  • Nuclear factor-kappaB (NF-kappaB) is a critical transcription factor for maximal expression of many of the cytokines that are involved in the pathogenesis of inflammatory diseases. In this study, we found that 12 plant extracts among 200 plants, namely, Forsythia koreana, Capsicum annuum L, Mentha arvenis, Duchesnea chrysantha, Morus alba, Saururus Chinenis (Lour) Baill, Pine needle, Zingiber mioga (Thunb.), Roscoe, Houttuynia, Prunus yedoenis, Sasa quelpaertenis, significantly inhibited LPS- induced NF-kappaB activation in a concentration-dependent manner. Additionally, 12 plant extracts were found to have antioxidant activities in DPPH assay Therefore, we have attempted to determine whether 12 herbal extracts could inhibit the expression of cytokines possessing NF-kappaB promoter in their promoter regions. Consistently 12 herbal extracts inhibited LPS-induced production of TNF alpha and interleukin-8 (IL-8). These results show that 12 herbal extracts suppresses the production of pro-inflammatory mediators through the inhibition of the NF-kappaB signaling pathway, we suggest that 12 herbal extracts can be used as a anti-inflammatory and soothing agent.

Functional quality characteristics of extracts by sugar-leaching and lactic acid fermentation of mulberry leaves (Morus alba L.) (뽕잎의 당침 및 유산발효에 의한 추출물의 기능성 품질 특성)

  • This study was carried out to investigate functional quality characteristics of extract obtained after sugar-leaching for 12 weeks (SLE) and extract obtained after lactic acid fermentation for 8 weeks (LFE) of mulberry leaves. The yield, sugar content, pH, and total acidity of SLE were 27%, 43 $^{\circ}Brix$, 4.6, and 0.45%. The yield, sugar content, pH, and total acidity of LFE were 166%, 33 $^{\circ}Brix$, 3.6, and 1.17% respectively. The lactic acid bacteria viable numbers ($1.2{\times}10^{10}$ CFU/ml) of LFE were more than those of SLE ($2.8{\times}10^2$ CFU/ml). The LFE expressed activities of hydrolytic enzymes (amylase, cellulase, pectinase, protease), but SLE did not express. The contents of acetic acid, citric acid, and malic acid of SLE were higher than those of LFE, but lactic acid content of LFE was higher than that of SLE. The main free sugars of SLE were glucose (200.93 mg/g), fructose (236.32 mg/g), and sucrose (18.41 mg/g), but LFE did not detect all free sugars. The contents of polyphenol, anthocyanin, and piperidine alkaloid of LFE were higher than those of SLE. ${\alpha}$-Glycosidase activities were inhibited 3.4% and 16.2% by SLE and LFE. These results suggest that lactic acid fermentation extraction is an effective method to increase the yield and contents of functional quality of mulberry leaves extract.

Effects of Host Tree Species, Temperature and Humidity on ex vitro Seed Germination in Endangered Species of Loranthus tanakae (기주목, 온도 및 습도가 멸종위기 종 꼬리겨우살이 종자의 기외발아에 미치는 영향)

  • This study was conducted to investigate the effects of host tree species, temperature and humidity on ex vitro for seed germination in endangered species of Loranthus tanakae. In addition, we compared seed shapes between Loranthus tanakae and Viscum album that we could have easily shown in ex vitro condition. Seeds were germinated after one week inoculation and followed to develop radicles. Seed germination rates of Loranthus tanakae were 80~95% in most of the experimental conditions. The highest rate of holdfast penetrated to host plants was 72% in Populus alba var. pyramidalis among 13 different species tested at $20^{\circ}C$. Also the rates of their penetration were 57% in Morus bombycis, 55% in Acer palmatum and 42% in Castanea crenata at $20^{\circ}C$. Seeds were germinated under condition without irrigation and followed to withered in 12 weeks later. Stages of seed germination of the Loranthus tanakae were followed by radicle induction, holdfast development, haustorium formation and penetration in order in total period of 14 weeks.

Effect of Dietary Medicinal Herb Extract Mix and Antibiotics (Albac G150) on the Oxidative Stability of Chicken Meat (금은화, 황련, 상엽 추출물 혼합물과 항생제(Albac G150) 급여가 계육의 산화 안전성에 미치는 영향)

  • This study was evaluated the oxidative stability of the breast and leg meat from chicken fed extract of medicinal herb mix (Lonicera japonica Thunberg : Morus alba L. : Coptis chinensis = 48.5 : 48.5 : 3.0%) and antibiotics (Albac G150). Ross chickens were divided into three treatments: basal diet (control), basal diet with antibiotics (0.05%) and basal diet with extract of medicinal herb mix (0.3%). They were fed the experimental diets for 35 days and slaughtered. After that, the breast and leg meat samples were stored at $4^{\circ}C$ for 10 days. Total phenol content of breast meat in Ross chicken fed extract of medicinal herb mix was higher than that of antibiotics treatment and control during storage. DPPH radical scavenging activity was the highest in leg meat of Ross chicken fed extract of medicinal herb mix at 0 day. $ABTS^+$ reduction activity was higher in chicken meat fed extract of medicinal herb mix than antibiotics treatment and control. This results showed that the diet with extract of mixed medicinal herb was more effective in oxidative stability of chicken meat than basal diet and diet with antibiotics.

오디추출물의 기능성 물질 탐색에 관한 연구

  • 오디는 옛부터 보혈자음(補血滋陰), 생진윤조(生津閏操), 현훈이명(眩最耳鳴), 심계실면(心悸失眠), 수발조백(鬚髮早白) 등을 치료하는 효능을 가진다고 알려져 있다. 상심자(桑 子)라 하여 뽕나무 열매로서 한방에서 상심(桑 ), 상실(桑實), 오심(烏 ), 흑심(黑 ) 등으로 지칭되며, 뽕나무과(Moraceae)에 속하는 뽕나무(Morus alba L)의 성숙한 과실로 취화과(聚花果)에 속하며 작은 수과(瘦果)가 많이 모여 이루어진 장원형으로 길이 1 내지 2 cm, 지름 0.5 내지 0.8 cm이며 황갈색, 갈홍색 또는 암자색을 띠고 짧은 줄기가 있다. 작은 수과(瘦果)는 난원형으로 조금 납작한 편이며, 길이는 약 2 mm, 너비는 약 1 mm 이고 육질의 화편(花片) 4개가 둘러싸고 있다. 그러나 아직까지는 오디에 대하여 자연과학적인 연구방법을 이용하여 구체적으로 연구된 바가 많지 않다. 또한 이의 생리활성 성분에 대해서도 그 작용과 연관지어 보고된 바가 많지 않다. 뇌졸중 가운데 뇌조직으로 가는 혈액 공급의 감소 혹은 차단으로 발생되는 허혈성 뇌졸중(ischemic stroke)은 전체 뇌졸중 환자의 약 80%정도를 차지하고 있으나 아직까지 뇌신경세포 손상기전의 복잡성 등으로 뇌졸중으로 발생하는 뇌신경세포의 손상을 보호해 줄 수 있는 물질이 개발되어 있지 못한 실정이다. 한편, 천연물로부터 뇌허혈 보호작용을 가지는 물질의 도출은 주로 한방에서 처방을 중심으로 이루어지고 있으며, 따라서 처방으로부터 신경보호작용을 가지는 물질의 도출은 그 처방에 함유되어 있는 각종 생약이 갖는 다양한 활성으로 인해 어려운 점이 있으며, 비록 효과가 있다고 하더라도 과학적인 입증자료가 매우 미비한 실정이다. 이에 본 연구에서는 위에 언급된 자료를 토대로 오디추출물이 뇌허혈에 효과가 있을 것이라는 가정하에 in vitro system을 이용하여 오디추출물의 신경보호작용기전을 검색하고자 DPPH radical 의 생성억제효과, PC12 cell line을 이용한 산화적 스트레스에 의한 오디추출물의 방어효과, LSP에 의하여 활성화된 BV-2 cell에 미치는 영향 등을 검색하였다. 오디의 DPPH 소거활성은 단일화랍물인 bacicalein과 유사한 효과를 나타냈으며 PC12 cell line을 이용한 산화적 스트레스에 의한 오디의 방어 효과는 LDH activity를 행한 결과 농도 의존적으로 LDH 유리 량을 감소시켰다. 뇌손상시에 오디추출물의 염증방어 효과를 관찰하기 위하여 microglial cell line인 BV2 세포주를 선택하여 배양한 후 LPS로 자극을 준 후 일차적인 염증지표인 NO양을 측정한 결과 BV2 cell에 LPS 100 ng/ml을 처리하는 경우 nitrite량이 유의적으로 증가하였는데 이때 오디추출물을 1, 5, 10 ug/ml의 용량으로 처리하는 경우 역시 저농도에서 NO생성량을 감소시켰다. 정리해보면 뽕나무과 식물인 오디는 신경보호효과가 있는 것으로 사료되는데 이러한 효과는 아마도 오디의 항산화효과에 기인한 것으로 추정된다.

Effects of Ehwa Makgeolli Containing Oriental Herbs on Skin Whitening and Wrinkles (한방 이화주의 미백 및 피부 주름 개선 효과)

  • Extrinsic skin aging is characterized by the loss of skin tone and resilience, irregular pigmentation, and deep wrinkles. The aim of this study was to investigate the effects of Ehwa Makgeolli containing oriental herbs (Glycyrrhiza uralensis Fisch., Lycium chinense MILL., Morus alba L., and Saururus chinensis Baill) on skin whitening and wrinkling in human skin cells. We prepared Makgeolli extracts (HEE) with 70% ethanol. HEE significantly inhibited in vitro mushroom tyrosinase activity and reduced the cellular and secreted melanin content of mouse melanoma melanocytes (B16F1 cells). HEE down-regulated the protein expression of tyrosinase related protein (TRP)-1/-2, a key player in melanogenesis. Treatment with HEE in human keratinoctyes (HaCaT cells) inhibited the proteolytic activities of matrix metalloproteinase (MMP)-2/-9 in a dose-dependent manner and dramatically reduced the expression of MMP-2/-9. In addition, HEE attenuated lipopolysaccharide (LPS)-induced nitric oxide production in murine macrophages (RAW264.7 cells). These results indicate that HEE may be a great cosmeceutical ingredient for its whitening, anti-wrinkle, and anti-inflammatory effects.

Growth Performance and Caecal Fermentation in Growing Rabbits Fed on Diets Containing Graded Levels of Mulberry (Morus alba) Leaves

  • Growth performance, nutrient digestibility and changes in caecal fermentation pattern was studied on four groups of 8 grower rabbits (soviet chinchilla) each, fed on diets containing 17 to 18% CP and $10.4-11.00MJ\;DE\;kg^{-1}$ feed. The complete diets contained mulberry leaves along with other feed ingredients at 0 (LH), 15 (ML15), 30 (ML30) and 45% (ML45) levels. In LH diet ground lucerne hay was added at 15% of the diet as roughage source. The diets were uniform in other nutritional parameters except the higher amount of hemicellulose (18.7 and 16.5%) in LH and ML15 diets. The performance of grower rabbits was better (p<0.05) on LH and ML15 diets in terms of twelve weeks weight (kg), total gain (g), average daily gain (ADG) and feed efficiency (%). Different levels of mulberry leaves in the diet did not influence the dry matter intake. Digestibility of nutrients for DM, OM, CP, NDF and energy was uniform in all the diets but the digestibility of CF, ADF and cellulose was higher (p<0.01) on ML15, ML30 and ML 45 diets. The hemicellulose digestibility was significantly higher (p<0.01) on LH (44.52%) and ML15 (48.00%) compared to ML30 (33.54%) and ML 45 (39.17%) diets. The nitrogen retention (% of intake) was higher (p<0.05) in LH and ML15 diets than ML30 and ML45 diets. The caecum weight as percent of intestine weight consistently increased (p<0.05) with increasing content of mulberry leaves in diets. Total nitrogen and $NH_3$-N was higher on LH and ML15 diets. It is concluded that incorporation of mulberry leaves can replace Lucerne hay in complete rabbit feed (15% in diet). Even at higher level i.e. 30 and 45% of the diet mulberry leaves based complete feed gave promising results.
