Molecular Characterization of Filenchus cylindricus (Thorne & Malek, 1968) Niblack & Bernard, 1985 (Tylenchida: Tylenchidae) from Korea, with Comments on Its Morphology

  • Filenchus cylindricus (Thorne & Malek, 1968) Niblack & Bernard, 1985 was reported from the sandy rhizospheric soils of Poa pratensis and for the first time in Korea. Females and males are molecularly characterized and morphological and morphometric data supplied. Identification was made using an integrative approach considering morphological characteristics and inferences drawn from the analyses of the D2-D3 expansion segment of 28S rRNA and ITS1-5.8S-ITS2 of rRNA partial sequences. Females and males from Korea conform to the type descriptions and also to subsequent species descriptions from Iowa and Colorado USA, Sudan and Pakistan. Despite the close morphological and morphometric similarities with F. thornei (Andrássy, 1954) Andrássy, 1963, the two species can be adequately differentiated based on molecular data inference.

  • Gulgastrura reticulosa, first described by Yosii (1966) as a monotypic new genus from a Korean limestone cave, was reviewed for its systematic position by c1adistic analysis of morphological characteristics, investigation of its intermaulting period and reproductive cycle as well as allozyme and 18S rDNA analysis. The great extent of divergence was strongly suggested by its combined lack of sensory organs (third antennal organ, postantennal organ, eyes, pseudocelli) with simultaneous development of an 'apical organ'at the tip of the antennae. The obvious divergence from any existing Collembola families was additionally supported by the extremely prolonged intermoulting period as well as by the low strap value it showed with Onychiuridae as obtained by 18S DNA sequence analysis. All these were considered Justifying the creation of a new family, Gulgastruridae, but still revealing more allied to Onychiuvidae rather than to Hypogastruridae.

Three Dimensional Study of the Tidal Flat Geomorphology in the $K\breve{u}$m River Estuary by Multidate Image Analysis

  • The geomorophology of the tidal near $K\breve{u}$m river estuary on the west coast of Korea was studied using satellite images. There Landsat image of different tidal moment were geometrically corrected to be overlapped one another. Then topographic contours were extracted for the analysis with concept of 3-D. According to the morphological analysis, the tidal flats around $K\breve{u}$m river estuary area can be classified in three morphologically different groups: one is developped in a belt type along the shore; another is a delta type in the estuary; the other is developped between the shore line and islands. The morphological characteristics of the tidal flats in the study area seems to be mainly influenced by the tidal dynamics which are very storg on the weatern coastal zone of Korea. However the waves and human activities seem also to be important in some part of the area.

  • This study attempted to comparative analysis about the physical and morphological damage degrees of hair according to the bleaching methods by $H_2O_2$ concentration. As a result of the changes in the physical characteristics of the hair according to the deceleration operation methods by 3-6-9% and 9% $H_2O_2$ concentration, the intensity of tension considerably decreased but the elongation was increased. And the more the damage of hair, the larger the degree of change. Regarding the method of deceleration operation, the 3-6-9% deceleration operation method was more damaging than the 9% single deceleration operation method. In the change of the characteristics of the form of the hair, the control group of the 5-level deceleration hair generally showed a smooth external appearance and regular and closely overlapped epidermis and undamaged cuticle were observed. But in the 3-6-9% deceleration operation method and 9% single deceleration operation method, damages such as the irregular form of the edge of the epidermis and the unclear boundary between the epidermis occurred.

Molecular Identification and Morphological Characteristics of the Longjaw Bonefish, Albula argentea Collected in Korea (한국산 여울멸의 분자동정과 형태적 특징)

  • One specimen of Albula argentea (296.3 mm in standard length), belonging to the family Albulidae, was collected in a set net in the coastal waters of Jejudo Island, Korea. As the adult A. argentea has not been caught from Korean waters, we described its morphological characters based on the collected specimen and compared them with those of the previous reports on A. argentea and A. koreaus. As a result, the present specimen was characterized by having 68~73 pored lateral line scales (vs. 77~78 for A. koreana), 72~73 vertebrae pored lateral line scales (vs. 77~80) and none streak on the check (vs. yellowish streak). Molecular analysis using the mitochondrial cytochrome b DNA sequences was also conducted to confirm the correctness of species identification and taxonomic status of the sample.

A new record of epiphytic red alga Madagascaria erythrocladioides (Erythropeltidales, Rhodophyta) in Korea

  • The Erythropeltidales are a common group of small, mostly epiphytic, marine red algae. However, they are little known in Korea. Many of the described species of Erythropeltidales differ subtly in morphology, and often the morphological differences are due to the substrate or environmental changes. Integration of molecular data with standardized culture conditions has been recommended to account for these algae. A Madagascaria species was first collected from the western coast of Korea and was identified as Madagascaria erythrocladioides based on the morphological and molecular characteristics. Morphological characteristics conformed well with its original description, and the phylogenetic analysis based on rbcL sequence showed Korean M. erythrocladioides nests in the same clade with the original species described in Japan with a genetic distance of 0.0-0.1%. This species was isolated from a red alga, Pterocladiella capillacea, in laboratory culture. The thallus ontogeny and host preference were examined by a co-culture with 13 different species of algae. Results showed a relatively broad host preference in mono-spore attachment and epiphyte development of Madagascaria erythrocladioides. Mono-spores of M. erythrocladioides attached to most of the red algal hosts' surfaces but no crustose thalli developed on some of the algal hosts even after one month of co-culture.

Morphological, Molecular and Pathogenic Characteristics of Botrytis elliptica and Botrytis cinerea Associated with Leaf Blight of Lily (백합 잎마름병에 관여하는 Botrytis elliptica와 Botrytis cinerea의 형태적, 분자적 특성과 병원성)

  • The seventy nine isolates of Botrytis spp. were obtained from leaf blight lesions of lily in Korea, Japan and Netherlands. Morphological and cultural characteristics of them were investigated and molecular characteristics of them were determined using sequence analysis of RNA polymerase II (RPB2) and heat-shock protein 60 (HSP60) gene. A selection of Botrytis isolates were evaluated for their pathogenicity to lily. Based on morphological and cultural characteristics, the Botrytis isolates were divided into two groups, and identified as B. elliptica (n = 54) and B. cinerea (n = 25). Based on analysis of RPB2 and HSP60 sequences, the Botrytis isolates were also divided into two groups and well supported morphological groupings. Spore suspensions of B, elliptica showed significant pathogenicity on lily leaves and flowers, however those of B. cinerea showed pathogenicity only on flowers but not on leaves. The latter showed pathogenicity on lily leaves only when spore suspensions amended with PDB were used as inocula.

Phylogenetic Analysis, Morphology and Pathogenicity of Penicillium spp. associated with Blue Mold of Apple in Korea (사과푸른곰팡이병에 관여하는 Penicillium의 계통분석, 형태 및 병원성)

  • Blue mold is the most important postharvest disease of apples in Korea. Apple fruits with blue mold symptoms were collected from storages in different locations in Korea and were investigated for their association with Penicillium species. A total of sixty five isolates of Penicillium were sampled from the collected apples. Based on DNA sequence analysis of ${\beta}$-tublin gene and ITS and lsu rDNA (ID region) and morphological characteristics, they were identified as P. crustosum, P. expansum, P. italicum, P. solitum and P. sp.. P. sp. which is closely related to P. hirsutum is a new species, not reported before. P. expansum (35%) was predominant species followed by P. crustosum. The phylogenetic tree inferred from combined ${\beta}$-tublin and ID region sequence showed good correlation with species that are defined by morphological characteristics. In pathogenicity test, apples were wound-inoculated with conidial suspension and incubated at $20-22^{\circ}C$. The most severe and destructive species was P. expansum. The species caused a decayed area 42-50mm in diameter after 8-10days. Decayed area caused by P. crustosum and P. sp. was 26-32mm and 20-26mm, respectively. This is the first record of P. crustosum, P. italicum and P. sp. from apple in Korea.

Confirmation of Chestnut Powdery Mildew, Erysiphe castaneigena, in Korea with Morphological Characteristics and Molecular Analyses (형태적 특징 및 분자적 분석에 의한 밤나무 흰가루병균 Erysiphe castaneigena의 확인)

  • The powdery mildew on Castanea crenata in South Korea was first recorded as Microsphaera alni in 1958. On the basis of its morphological characteristics and host range, the mildew was determined as Microsphaera sinensis in 1988. According to the rules of The International Code of Botanical Nomenclature, M. sinensis was renamed Erysiphe castaneigena in 2006. Nevertheless, taxonomic re-consideration of the morphological and molecular characteristics for the Castanea powdery mildew in Korea has not been performed. In the present study, we studied 34 powdery mildew samples collected from C. crenata in the Korea University Herbarium. On the basis of microscopic examinations of the holomorph and phylogenetic analysis of the internal transcribed spacer regions, we confirmed the powdery mildew fungus as E. castaneigena. In addition, sequence comparison between E. castaneigena and E. alphitoides ex Castanea sp. suggested a close phylogenetic affinity.

A Study on Preparation and Characteristics of SPE-Composites for Water Electrolysis (물 전기분해용 SPE-composites의 제조 및 특성 고찰)

  • SPE-composites which was prepared with impregnation-reduction method for hydroen production were investigated with morphological and electrochemical analysis. As Pt reagent's concentration increased, Pt loadings increased. However, reductant's concentration had a little effect on Pt loadings. By the morphological analysis, it was found that Pt was deposited inside of SPE. Furthermore, with electrochemical analysis, we found that Pt loading, electricity, roughness factor and active surface area were $2.05mg/cm^2$, 14.20 mC, 21.55, $10.51cm^2/mg$ respectively at reducing agent concentration 0.05 mol/L. Therefore, we found the deposited Pt inside of SPE would give an effect on electrical characteristics of SPE-composites.
