Morphological and molecular investigation of Melosira octogona Schmidt (Melosirales, Bacillariophyta) isolated from the Jeju Island

  • Melosira octogona Schmidt has been documented primarily through illustrations and light microscopy photographs, resulting in incomplete morphological descriptions. In this study, the morphological characteristics of M. octogona were investigated using scanning electron microscopy, and the small subunit rDNA of the strain isolated from the coast of Jeju Island, South Korea, was analyzed for the first time. The cells were organized in short chains, with the frustules taking on an octagonal shape in the girdle view because of the straight, obliquely oriented valve margin. The cells were joined together in specific pairs by a cingulum. Imaging analysis via SEM revealed the presence of a hemispherical valve with a well-developed corona on the valve face, along with stellate thickening and rimoportulae. The epicingulum was composed of six ligulate bands punctuated by rows of circular to elongated pores. Genetic analysis of the 18S rDNA sequence suggested a close relationship between M. octogona and Melosira sp. (KJ961704), demonstrating a high degree of similarity. Meanwhile, it exhibited comparatively low similarity (99.0 %) to Melosira cf. octogona (AY485518). The results of our analysis can enhance the precise recognition of M. octogona and enrich our understanding of its phylogenetic position within the order Melosirales.

Particle Size Control by the Addition of PVA and HNO3 in γ-Al2O3 Synthesis Using by Sol-Gel Method (졸-겔법을 이용한 γ-Al2O3 합성 시 PVA와 HNO3 첨가에 따른 입자크기 제어)

  • Alumina(Al2O3) is a ceramic material used in industry with a range of particle sizes and characteristics. In this study, a boehmite sol was prepared by a hydrolysis and peptizing process using the Sol-Gel method from aluminum isopropoxide (AIP). γ-Al2O3 was prepared by drying and calcining. To prevent particle agglomeration during the manufacturing process, four kinds of polyvinyl alcohol (PVA) with molecular weights of 9,000~10,000, 31,000~50,000, 89,000~98,000, and 130,000 were added and three concentrations of HNO3 (0.1, 0.3, 0.5 molar ratio) were added to determine their effects on the particles. The crystal structure, composition, particle size and shape of the prepared γ-Al2O3 were confirmed through x-ray diffraction (XRD), x-ray fluorescence analyzer (XRF), particle size analyzer (PSA), and field emission scanning electron microscopy (FE-SEM). As a result, γ-Al2O3 with a purity of approximately 98.2% was synthesized, and the particle size decreased and the uniformity increased with increasing ratio of HNO3 addition and PVA molecular weight. From these results, the particle size can be controlled during the manufacturing process of γ-Al2O3 by controlling the addition ratio of PVA and HNO3.

  • Antheraea yamamai vitellin was purified from matured eggs by polyacrylamide gel electrophoresis for characterization of its biochemical properties : molecular weight, sugar and lipid composition, amino acid composition and electron microscopic morphology, etc. 1. A yamamai vitellin was composed of two subunits, large and small, showing different mobility in SDS-polyacrylamide gel electrophoresis. 2. The molecular weight of the vitellin was estimated to be approximately 450,000 dalton and the large and small subunits were 174,000 dalton and 44,000 dalton, respectively. 3. The vitellin seemed to be a glycolipoprotein since it showed a positive reaction to coomassie brilliant blue, sudan black B and PAS staining. Both subunits were similiar in this aspect. 4. Lipid of the witellin reveraled several different types including saturated lipids. 5. When the vitellin was incubated at 7$0^{\circ}C$ for 60 minites its apoprotein still cross-reacted to the specific antiserum to the native vitellin. Its sugar components were also detected by PAS staining, but its lipid portion was not detected by sudan black B staining. 6. Its amino acid composition was similar to that of other insects, but its glycine content was peculiarly very high. 7. The vitellin molecule was spherical in shape with a diameter of 14$\pm$0.8nm by negatively.

A Study on the Properties of Condensed Ethyl Silicate Synthesized according to Water Contents in Ethanol (에탄올의 수분함량에 따라 합성된 Condensed Ethyl Silicate의 물성에 관한 연구)

  • Condensed ethyl silicate(CES) solutions were prepared from $SiCl_4$ and ethanol which is different in water content(2.5, 5.0 and 7.0wt.% ). To investigate the sol-gel transition behaviors of CES and PHT(partially hydrolyzed TEOS) solutions, the measurement of volume change, density, $SiO_2$ content, intrinsic viscosity and number average molecular weight were conducted with elapsed time. The polymer shapes of CES were discussed from the relation between the intrinsic viscosity[$\eta$] and the number average molecular weight($\bar{Mn}$). CES-3 prepared from ethanol containing 7.0wt.% $H_2O$ had different sol-gel transition behaviors from CES-1 (2.5wt.% $H_2O$) and CES-2(5.Owt.% $H_2O$), but similar behaviors to those of PHT which is used for controlling the hydrolysis rata of TEOS when composite ceramic materials were manufactured from silicon alkoxides. On the basis of Mark-Hawink relation, $[{\eta}]=K\bar{Mn}^{{\alpha}}$, the polymer shape of CES solutions were determined linear siloxane polymers because all solutions had ${\alpha}$ values in the range from 0.53 to 0.84. Especially, CES-3 showed to be a favorable raw materials for composite ceramics due to its similar properties to those of PHT.

  • A suspension copolymerization of styrene and butyl methacrylate (BMA) in the aqueous phase was conducted at a selected temperature between 65 and $95^{\circ}C$. Hydrophobic silica was selected as a stabilizer and azobisisobutyronitrile (AIBN) as an initiator. Optimum dispersion of silica in water was obtained at pH 10 while polymerization reaction was run at pH 7. TGA and EDS measurements revealed that 90% of silica functioned as a stabilizer and 10% were incorporated into polymeric particles. Average particle diameter decreased with increasing amounts of stabilizer. Molecular weights displayed an increase when the stabilizer concentration reached 1.67 wt%. An increase in the initiator concentration and/or reaction temperature raised the reaction rate but decreased molecular weights. Particle diameter was nearly independent of the initiator concentration and reaction temperature. An increase in the BMA proportion decreased the glass transition temperature and increased the particle diameter with irregularity in shape. Incorporation of carbon black into the particles composed of styrene and BMA prolonged the reaction time before reaching completion. We have confirmed that a suspension copolymerization of styrene and BMA with hydrophobic silica as a stabilizer can produce spherical composite particles with $1-30{\mu}m$ in diameter containing carbon black.

Genetic comparison between Spirometra erinacei and S. mansonoides using PCR-RFLP analysis (만손열두조충과 북미열두조충의 중합효소연쇄반응-마디길이여러꼴 분석법을 이용한 유전 형질 비교)

  • The only observed morphological difference between Spirometra erinqsei and S. mcnsonoides is the uterine shape of the mature proglottid. Two species of worms are thought to be evolutionarily closely related. Biomolecular colnparison of the ho worms by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis was conducted to observe the genetic distance. The 285 rDNA, mitochondrial cytochrome c oxidase subunit I (mCOI), and ribosomal internal transcribed spacer 1 (ITSI) fragments were obtained from the worms by PCR. The PCR products were cleaved by 5 four-base pair restriction enzyme combinations (Msp I, Hae III, Alu I, Cfo I, Rsa I) , electrophoresed and analyzed with PAUP 3.1.1. The fragment Patterns or 285 rDNA and Lni demonstrated that two worms were in identical systematic tree with bootstrap number 94 and 100, respectively As for mCOI, bootstrap number was 74 in a different tree. Above results are indicative of recent common ancestry between S. etinocei and S. mansonoides.

  • Linear and branched copolymers consisting of poly(ethylene glycol) (PEG) and $Poly({\varepsilon}-caprolactone)$ (PCL) were prepared to compare the characterization of star-shaped copolymers with various molecular architecture. Linear and branched PEG-PCL (1-arm, 2-arm, 4-arm, and 8-arm) copolymers were synthesized by the ring-opening polymerization of ${\varepsilon}-caprolactone$ in the presence of HCl $Et_2O$ as a monomer activator at room temperature. The synthesized copolymers were characterized with $^1H-NMR$, GPC, DSC, and XRD. As a result of the DSC and XRD, each copolymers showed different thermal properties and crystallinity according to the number of ms. The micellar characterization of linear and branched copolymers in an aqueous phase was carried out by using NMR, dynamic light scattering, AM, and fluorescence techniques. The critical micelle concentration (CMC) and diameters of micelles depended on the number of arms. Most micelles exhibited a spherical shape in AFM. In this study, we characterized star-shaped PEG-PCL copolymers and investigated their molecular architecture effect on the various properties. Furthermore, we confirmed that the micelles termed with linear and branched PEG-PCL copolymers have possibility as a potential hydrophobic drug delivery vehicle.

The study for the mouse cerebellum developments irradiated with γ-ray during embryogenesis (감마선 조사에 의한 발생과정 중인 마우스의 소뇌발달에 관한 연구)

  • In order to study about the lobule and layer formation and cell migration of the mouse cerebellum from at the birth to 15th day effected by 2.5, 5 and 10 Gy r-raddiation at the 19th pregnancy. The routine tissue preparation and staining procedure, Immunohistochemical staining method by the several antibody and western brotting method were utilized from the birth to the15th day. The results were as followings. 1. The body and cerebellum weights were more slowly increase of the the 2.5 Gy, 5 Gy and 10 Gy irradiation group compare to the control group, and the health condition of the 2.5 Gy group was a little bad. but the 10 Gy group was more severe and begun to die from the 12th day after birth. 2. The thickness, proliferation and migration of the 2.5, 5 and 10 Gy irradiated external granular cells from the maginal zone to the medullary area forming the molecular layer from the 6th day to the 15th day after birth were thinner, weaker and more slower according to the radiated dosages than the control group in the cresyl violet staining. 3. The proliteration, migration and lobulation of the 5 Gy radiated groups from the first day to the 15th day after birth were more weak, incomplete and irregular shape in the immunostaining with Dab, Cdk5, P35, calbindin and Zebrin antibody. 4. In the western blotting analysis using the Reelin, Dab, Cdk5 and P35 antibody. The Bands were in the 60 KD, 80 KD, 33 KD and 35 KD, and there were no differences between the control and irradiated groups in the molecular band except the Reelin. 5. As a results, the proliferation and migration of the outer granular and purkinje cells, and lobulation of the cerebellum by the several dosaege of the ${\gamma}$-ray radiation were proportionally incomplete according to dosage.

Preparation of Fragrant Microcapsule for Reducing Stress (긴장완화를 위한 향기나는 마이크로캡슐의 제조)

  • A fragrant microcapsule was prepared for use by students to reduce the stress of taking examinations. Rosmarinic acid was used as a fragrant oil which had the effect of relaxing stress, polycaprolactone (PCL) was used as a capsule wall material, and poly(vinyl alcohol) (PVA) as a stabilizer. The solvent evaporation method was used to form the microcapsule. The microcapsules were prepared by changing the stirring rate, the concentration of the stabilizer, and the molecular weight of PCL. The shape of the microcapsule was characterized by scanning electron microscopy (SEM). The size of the microcapsule was reduced by increasing the stirring speed. The release rate of rosmarinic acid was decreased when the higher molecular weight PCL was used. When the prepared microcapsule was tested in an aromatherapy class, the microencapsulated fragrant oil had a longer release time than the original fragrant oil. The study data showed that this fragrant oil was effective for increasing concentration ability, reducing stress, increasing digestive power, and increasing memory for the students.

Characteristics of Agronomy Traits to Transgenic Rice Selected by Molecular Breeding Method (분자육종기법에 의해 선발된 형질전환 벼 계통의 작물학적 특성)

  • This study was carried out to develop new cultivars using the $T_5$ generation of transformed rice by PCR analysis with DNA marker in each generation $(from\;T_3\;to\;T_5)$. In the previous study, we successfully developed the transgenic rice plants over-expressing the Arabidopsis $H^+/Ca^{2+}$ antiporter CAX 1 (accession no. U57411) gene. The calcium concentration in brown rice of transgenic plants was higher than that of donor plants, Iipum, and was selected 3 lines out of 25 lines at cultured GMO field. The major agronomic traits such as culm length, panicle length and panicle number of 3 lines at transgenic plants $(T_5)$ were similar to wild type. Also these lines appeared to have disease resistance to rice blast, cold resistance as compared with donor types. The grain shape was similar to donor plant, however, the 1000 grain weight of brown rice was different from transgenic plants. These finding would be used for basic data of new variety registration.
