진핵 세포에서 히스톤의 변형은 크로마틴 구조를 조절하는 데에 있어서 중요한 메커니즘이다. Set1 복합체에 의한 히스톤 H3의 네 번째 라이신 잔기(H3K4)에 발생하는 메틸화는 다양하게 잘 알려져 있는 히스톤 변형 중 하나이다. Set1 complex는 H2B의 유비퀴틴화에 의존적으로 발생하는 H3K4 메틸화에 중요하다고 알려진 Swd2를 포함하여 7개의 소단위 단백질을 가지고 있다. Swd2는 Set1의 RNA recognition motif (RRM) 도메인 근처에 결합하여 Set1의 활성을 조절하고, 또 RNA의 3' 말단 형성에 관여하는 CPF (Cleavage and Polyadenylation Factors) 복합체의 구성성분이라고 보고되었다. 최근 보고들에 따르면, 이런 Swd2의 이중적인 기능이 서로 독립적으로 작용하며, Swd2 결실돌연변이 균주가 살지 못하는 이유가 CPF 복합체의 구성성분으로써의 기능 때문이라고 알려져 있다. 본 연구에서 우리는 Swd2가 Set1의 RRM 도메인에 결합하여 Set1의 활성을 조절할 수 있을 뿐만 아니라, Set1의 안정성에도 영향을 줄 수 있음을 발견하였다. 또 우리는 Swd2가 결합할 수 없는 truncated-Set1을 가지고 있는 ${\Delta}swd2$ 돌연변이가 사멸하지 않고 정상적으로 자라는 것을 관찰하였다. 이런 결과들은 Saccharomyces cerevisiae에서 H3K4 메틸화와 RNA 3' 말단 형성과정에서의 Swd2의 이중적인 기능이 서로 독립적인 것이 아님을 제안하다.
진핵생물에서 THO/TREX 복합체는 전사 신장, pre-mRNA 가공 및 mRNA의 핵에서 방출에 중요한 역할을 담당한다. 이 복합체는 진화적으로 잘 보존되어 있지만, 생명체에 따라 구성성분과 기능에 차이가 존재한다. 이 논문에서는 출아효모보다는 고등생물과 더 유사한 분열효모 Schizosaccharomyces pombe에서 THO/TREX 복합체의 한 구성요소인 spTex1가 생장과 mRNA의 방출에 필수적이지 않다는 것을 보였다. spTex1 유전자의 결실과 과발현 어느 것도 생장의 결함과 $poly(A)^+$ RNA가 핵 안에 축적되는 현상을 거의 보이지 않았다. 또한 spTex1-GFP 단백질은 주로 핵 안에 위치하였다. Yeast two-hybrid와 Co-immunoprecipitation 분석에서 S. pombe Tex1은 THO/TREX 복합체의 주요 구성인자인 spHpr1 (THOC1), spTho2 (THOC2)과 상호작용을 하였다. 이와 같은 결과들은 S. pombe의 Tex1도 THO/TREX 복합체의 구성인자이지만, 생장과 mRNA 방출에는 중요한 역할을 하지 않음을 의미한다.

We aimed to explore whether changes in circulating levels of miRNAs according to type 2 diabetes mellitus (T2DM) or prediabetes status could be used as biomarkers to evaluate the risk of developing the disease. The study included 462 patients without T2DM at baseline from the CORDIOPREV trial. After a median follow-up of 60 months, 107 of the subjects developed T2DM, 30 developed prediabetes, 223 maintained prediabetes and 78 remained disease-free. Plasma levels of four miRNAs related to insulin signaling and beta-cell function were measured by RT-PCR. We analyzed the relationship between miRNAs levels and insulin signaling and release indexes at baseline and after the follow-up period. The risk of developing disease based on tertiles (T1-T2-T3) of baseline miRNAs levels was evaluated by COX analysis. Thus, we observed higher miR-150 and miR-30a-5p and lower miR-15a and miR-375 baseline levels in subjects with T2DM than in disease-free subjects. Patients with high miR-150 and miR-30a-5p baseline levels had lower disposition index (p = 0.047 and p = 0.007, respectively). The higher risk of disease was associated with high levels (T3) of miR-150 and miR-30a-5p ($HR_{T3-T1}=4.218$ and $HR_{T3-T1}=2.527$, respectively) and low levels (T1) of miR-15a and miR-375 ($HR_{T1-T3}=3.269$ and $HR_{T1-T3}=1.604$, respectively). In conclusion, our study showed that deregulated plasma levels of miR-150, miR-30a-5p, miR-15a, and miR-375 were observed years before the onset of T2DM and pre-DM and could be used to evaluate the risk of developing the disease, which may improve prediction and prevention among individuals at high risk for T2DM.
Glucagon-like peptide-1 (GLP-1) has a broad spectrum of biological activity by regulating metabolic processes via both the direct activation of the class B family of G protein-coupled receptors and indirect nonreceptor-mediated pathways. GLP-1 receptor (GLP-1R) agonists have significant therapeutic effects on non-alcoholic fatty liver disease (NAFLD) and steatohepatitis (NASH) in animal models. However, clinical studies indicated that GLP-1 treatment had little effect on hepatic steatosis in some NAFLD patients, suggesting that GLP-1 resistance may occur in these patients. It is well-known that the gut metabolite sodium butyrate (NaB) could promote GLP-1 secretion from intestinal L cells. However, it is unclear whether NaB improves hepatic GLP-1 responsiveness in NAFLD. In the current study, we showed that the serum GLP-1 levels of NAFLD patients were similar to those of normal controls, but hepatic GLP-1R expression was significantly downregulated in NAFLD patients. Similarly, in the NAFLD mouse model, mice fed with a high-fat diet showed reduced hepatic GLP-1R expression, which was reversed by NaB treatment and accompanied by markedly alleviated liver steatosis. In addition, NaB treatment also upregulated the hepatic p-AMPK/p-ACC and insulin receptor/insulin receptor substrate-1 expression levels. Furthermore, NaB-enhanced GLP-1R expression in HepG2 cells by inhibiting histone deacetylase-2 independent of GPR43/GPR109a. These results indicate that NaB is able to prevent the progression of NAFL to NASH via promoting hepatic GLP-1R expression. NaB is a GLP-1 sensitizer and represents a potential therapeutic adjuvant to prevent NAFL progression to NASH.
This study evaluated the change in anti-allergy activity of ${\beta}-glucan$ by electron beam irradiation. ${\beta}-Glucan$ was irradiated at dose of 50 kGy and then orally pre-treated with electron beam irradiated and non irradiated ${\beta}-Glucan$ for 7 days. After pre-treatment, allergy was induced by injection of ovalbumin (OVA). Serum total immunoglobulin E (IgE) and OVA-specific IgE levels in the allergic mice was significantly increased but the mice pre-treated 50 kGy electron beam irradiated ${\beta}-glucan$ was significantly decreased the levels of total IgE and OVA-specific IgE, respectively. Moreover, cytokine production (interleukin-4) was also decreased in the 50 kGy electron beam irradiated ${\beta}-Glucan$ pre-treated mice. These results indicate that pre-treatment of 50 kGy electron beam irradiated ${\beta}-glucan$ may elevate the anti-allergy activity. Therefore, electron beam-irradiated ${\beta}-glucan$ could be used for nutraceutical foods in food industry.

Background: Panax ginseng (i.e., ginseng) root is extensively used in traditional oriental medicine. It is a modern pharmaceutical reagent for preventing various human diseases such as cancer. Ginsenosidesd-the major active components of ginsengd-exhibit immunomodulatory effects. However, the mechanism and function underlying such effects are not fully elucidated, especially in human monocytes and dendritic cells (DCs). Methods: We investigated the immunomodulatory effect of ginsenosides from Panax ginseng root on $CD14^+$ monocytes purified from human adult peripheral blood mononuclear cells (PBMCs) and on their differentiation into DCs that affect $CD4^+$ T cell activity. Results: After treatment with ginsenoside fractions, monocyte levels of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6, and IL-10 increased through phosphorylation of extracellular signal-regulated kinase (ERK)1/2 and c-Jun N-terminal kinase (JNK), but not p38 mitogen-activated protein kinase (MAPK). After treatment with ginsenoside fractions, TNF-${\alpha}$ production and phosphorylation of ERK1/2 and JNK decreased in lipopolysaccharide (LPS)-sensitized monocytes.We confirmed that DCs derived from $CD14^+$ monocytes in the presence of ginsenoside fractions (Gin-DCs) contained decreased levels of the costimulatory molecules CD80 and CD86. The expression of these costimulatory molecules decreased in LPS-treated DCs exposed to ginsenoside fractions, compared to their expression in LPS-treated DCs in the absence of ginsenoside fractions. Furthermore, LPS-treated Gin-DCs could not induce proliferation and interferon gamma (IFN-${\gamma}$) production by $CD4^+$ T cells with the coculture of Gin-DCs with $CD4^+$ T cells. Conclusion: These results suggest that ginsenoside fractions from the ginseng root suppress cytokine production and maturation of LPS-treated DCs and downregulate $CD4^+$ T cells.
버섯은 예전부터 단백질, 당질, 비타민, 무기질 등 일반 영양소들이 다량 함유되어 있을 뿐 아니라 다양한 약리작용을 가지고 있기 때문에 예로부터 민식용 및 민간요법으로도 많이 이용되어 왔다(17). 또한 현재에는 건강식으로 다양하게 이용되고 있다. 본 연구는 여러 가지 버섯 중, 팽이버섯의 추출물을 이용하여 이의 항암 활성을 실시하였다. 팽이버섯(Flammulina velutipes)의 항암활성을 검증하기 위하여 in vitro 실험으로 wound healing 활성 실험을 하였으며, in vivo 실험으로 마우스를 이용한 항암모델 실험을 실시하였다. Wound healing 활성 실험 결과 팽이버섯 추출물의 농도를 0-100 ug/mL로 처리함에 따라 B16 세포의 운동성이 감소하는 것을 알 수 있었다. 이를 바탕으로 마우스를 이용하여 항암 모델에 적용한 결과 B16 세포로 인해 형성되는 colony가 팽이버섯 추출물을 처리한 군에서 약 60% 내외로 현저히 감소하는 것을 알 수 있었다. 본 실험 결과는 팽이버섯 추출물의 항암 활성을 확인 한 것으로 향후 이의 추출물을 이용한 세포신호전달과정 등에 관한 연구와 건강기능성식품의 소재로의 제품 개발에 유용하게 사용될 수 있음을 나타내는 주요한 자료로 판단된다.
유기성 폐기물의 재활용은 떠오르는 연구분야로서, 슬러지와 같은 재활용 가능한 유기성 폐자원을 찾기 위한 노력이 한창이다. 그러나, 완숙기법은 시간 및 공간적인 문제와 연관된 경제성문제로 인하여 실용성이 없는 것으로 여겨지고 있어서, 속성부숙기법이나 미숙된 슬러지의 활용방법이 준비되어야만 할 필요가 있다. 본 연구는 임업에서의 슬러지의 활용기술 개발을 위한 기초연구로서, 단풍나무 묘포장에 슬러지를 살포할 때 적합한 양과 시기를 찾아보고자 실시한 것이다. 본 연구를 위하여 제지공장의 슬러지를 사용하였으며, 일부는 질산암모늄을섞어 탄질률을 40이하로 낮추었다. 최대 살포량은 1 ha당 년간 7톤에 달하였으며, 살포시기는 봄과 늦여름이었다. 슬러지 살포는 1년동안 수목의 생장을 촉진하는 효과가 없었다. 슬러지 처리가 토양의 화학성에 특기할만한 변화를 일으키지 못하였으므로 처리한 슬러지의 양은 수목의 생장을 촉진하기에는 너무 적은 양이었던 것으로 생각되었다. 하지만, 토양수는 늦여름에 슬러지를 처리한 조사구에서 구리의 양이 늘어나는 것으로 나타났다. 즉, 지하수 오염의 위험성을 고려한다면 슬러지의 처리시기를 신중하게 정하여야 할 필요가 있다. 결론적으로, 우리가 단기적인 결과를 얻기 보다는 산림생산성과 같은 장기적인 개념으로 목표를 설정하고 슬러지를 이용하고자 한다면, 소량의 미숙한 슬러지를 생육기에 임업묘포나 산림지역에 살포하는 방안도 고려해 볼만하다.
Purpose: To assess the effect of combination probiotic Saccharomyces boulardii CNCM-I 3799 and Bacillus subtilis CU-1 in outpatient management of acute watery diarrhea in children. Methods: A randomized double-blind placebo-controlled study was conducted in 180 participants aged six months to five years with acute mild to moderate diarrhea. All were enrolled from six centers across India and centrally randomized to receive S. boulardii CNCM-I 3799 and B. subtilis CU-1 or a placebo along with oral rehydration salts and zinc supplementation. Each participant was followed up for three months to assess recurrence of diarrhea. Results: The mean duration of diarrhea in the probiotic and placebo groups were 54.16 hours and 59.48 hours, respectively. The difference in the duration of diarrhea in those administered with probiotic or placebo within 24 hours of diarrhea onset was 25.21 hours. Furthermore, the difference in duration of diarrhea was 13.84 hours (p<0.05) for participants who were administered with probiotics within 48 hours. There were no significant differences in the stool frequencies between the two arms. After three months, 15% in the probiotic group and 18.5% in the placebo group reported episodes of diarrhea. The mean duration of diarrhea was considerably lower in the probiotic group, 31.02 hours versus 48 hours in placebo (p=0.017). Conclusion: S. boulardii CNCM-I 3799 and B. subtilis CU-1 combination was effective in reducing the duration of diarrhea when administered within 48 hours of diarrhea onset. Similarly, it reduced recurrence of diarrhea and its intensity in the subsequent three months.
Background: Colistin and carbapenem-resistant bacteria have emerged and become a serious public health concern, but their epidemiological data is still limited. Objectives: This study examined colistin and carbapenem resistance in Escherichia coli and Salmonella from pigs, pig carcasses, and pork in Thailand, Lao PDR, and Cambodia border provinces. Methods: The phenotypic and genotypic resistance to colistin and meropenem was determined in E. coli and Salmonella obtained from pigs, pig carcasses, and pork (n = 1,619). A conjugative experiment was performed in all isolates carrying the mcr gene (s) (n = 68). The plasmid replicon type was determined in the isolates carrying a conjugative plasmid with mcr by PCR-based replicon typing (n = 7). The genetic relatedness of mcr-positive Salmonella (n = 11) was investigated by multi-locus sequence typing. Results: Colistin resistance was more common in E. coli (8%) than Salmonella (1%). The highest resistance rate was found in E. coli (17.8%) and Salmonella (1.7%) from Cambodia. Colistin-resistance genes, mcr-1, mcr-3, and mcr-5, were identified, of which mcr-1 and mcr-3 were predominant in E. coli (5.8%) and Salmonella (1.7%), respectively. The mcr-5 gene was observed in E. coli from pork in Cambodia. Two colistin-susceptible pig isolates from Thailand carried both mcr-1 and mcr-3. Seven E. coli and Salmonella isolates contained mcr-1 or mcr-3 associated with the IncF and IncI plasmids. The mcr-positive Salmonella from Thailand and Cambodia were categorized into two clusters with 94%-97% similarity. None of these clusters was meropenem resistant. Conclusions: Colistin-resistant E. coli and Salmonella were distributed in pigs, pig carcasses, and pork in the border areas. Undivided-One Health collaboration is needed to address the issue.

제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
