국내 일부 오염 토양 및 퇴적물 내 토착 미생물에 의한 중금속의 지구화학적 거동 연구 (Study on Geochemical Behavior of Heavy Metals by Indigenous Bacteria in Contaminated Soil and Sediment)

  • 지하 심부 토양 및 퇴적물 내 토착 미생물의 활성화에 따른 중금속 거동을 이해하기 위하여, 독성 중금속으로 오염된 국내 일부 지역의 시료를 대상으로 혐기적 환경에서 유산염(lactate)을 탄소원으로 투입한 후 약 25일간에 걸친 비소 및 중금속(카드뮴, 구리, 납, 아연) 함량 변화를 관찰하였다. 실험 결과, 미생물이 투입된 시료의 경우 미생물이 투입되지 않은 비교시료에 비하여 용존 카드뮴, 납, 아연이 효과적으로 제거되었으며, 이는 토착 황산염 환원(sulfate-reducing) 박테리아의 활성화로 인해 생성된 환원상태의 황이 이들 중금속과 황화물을 형성하며 침전시켰기 때문으로 여겨진다. 비소의 경우, 미생물을 투입한 시료 중 황산염의 함량이 높은 덕음 토양에서는 제거율이 높은 반면 황산염의 함량이 상대적으로 낮은 화북 토양, 동두천 퇴적물에서는 지속적으로 그 함량이 증가하였다. 적절한 탄소원 및 황산염을 투입해 독성 중금속으로 오염된 지질 매체 내의 토착 미생물을 활성화시킨다면 이들 중금속의 원위치(in situ) 고정화를 통한 이동도 감소의 효과를 볼 수 있을 것으로 여겨지나, 비소로 동시에 오염된 경우 발생할 수 있는 비소 용출 가능성에 대한 고려가 필요하다.

리기다소나무 잎 에탄올 추출물의 항균 및 생리특성 (Antimicrobial & Physiological Characteristics of Ethanol Extract from Pinus rigida Miller Leaves)

  • 리기다소나무 잎추출물의 항균력을 조사한 결과, $100{\sim}250{\mu}g/mL$ 이상의 농도에서 뚜렷한 항균활성을 보였고, 광역의 병원성 및 부패성 미생물들에 대하여 항균활성을 나타내었다. 특히, 리기다소나무 알코올 추출물에서는 $100{\mu}g/mL$의 낮은 처리농도에서도 뚜렷한 항균력을 나타내었다. 또한 추출물의 농도가 증가함에 따라 미생물의 생육저해환의 직경도 커졌으며, 알코올추출물에서 좀 더 뚜렷한 항균력을 관찰할 수 있었다. 또한, 리기다소나무 잎추출물은 넓은 범위의 열 및 pH에서 안정성을 나타냈다. 따라서 리기다소나무 잎추출물의 항균물질은 항균활성이 높고, 항균 spectrum이 광범위 할뿐 아니라, 넓은 범위의 온도 및 pH에 안정하여 이상적인 천연 항균제로서의 개발가능성을 제시하였다. 아울러, 리기다소나무 잎추출물의 주요 항균작용기작은 미생물 세포막의 perturbation 기능에 기인한 것으로 사료되었다. 또한, fomalin 투여후 PRLE처리로 혈청내 formaldehyde의 분해가 촉진되어 제거비율이 증진되는 효과를 보였으며, formalin 투여로 인한 생체내 hemoglobin 생성비율도 짧은 시간내에 회복됨을 예측할 수 있었다.

젓갈의 숙성 및 저온 저장이 미생물 균수 및 균총에 미치는 영향 (The effects of low temperature storage and aging of Jeot-kal on the microbial counts and microflora)

  • 젓갈 미생물 계수용 배지의 소금농도를 결정하기 위한 실험에서는 NaCl을 5% 첨가한 배지에서 가장 많은 수의 미생물이 계수되었고, 시료를 보존하기 위한 동결보호제는 glycerol 15%를 사용하였을 때 $-170^{\circ}C$에서 30일 보존 후 총균수는 75.5%, 젖산균수는 61.3%로 가장 높은 생존율을 나타내었다. 조개젓의 숙성과정 중 pH는 초기 6.8에서 5.0으로 낮아졌고, 숙성 후 단백질 함량은 조개젓은 10%, 멸치액젓은 $6{\sim}7%$이었다. 조개젓의 미생물수는 멸치액젓의 미생물수보다 많았고, 조개젓의 경우는 숙성기간에 따라 미생물수가 증가했다가 감소하는 경향을 보여주었으나 멸치액젓은 서서히 감소하는 경향을 보여주었다. 조개젓의 경우 숙성 4주까지 그램 양성, 카탈라아제 음성 세균들의 비율이 크게 증가하는 전형적인 젖산 발효 형태를 보여주었다. 젓갈을 $-170^{\circ}C$의 액체질소 탱크에서 2년간 보존 후 미생물수는 전체적으로 90% 이상 사멸하였고, 균총 변화를 보면 조개젓의 경우에는 그램 음성 세균과 간균의 생존율이 더 높았으나 거제 멸치액젓의 경우는 간균의 비율이 구균에 비해 감소하였다. 두 가지 젓갈에서 모두 효모의 비율은 크게 떨어졌다.

국내 담수환경에서 분리된 국내 미기록 효모 6종 보고 (Six Yeast Strains Isolated from Freshwaters for the First Record in Korea)

  • 본 연구는 광주광역시와 전라남도 여수시의 담수환경으로부터 야생효모를 분리하여 특성 분석을 하였다. 분리된 효모의 동정을 위하여 26S rDNA의 D1/D2부위염기서열을이용하였다. 결과적으로 Apiotrichum chiropterorum (NNIBRFG36995), A. domesticum (NNIBRFG32938), A. dulcitum (NNIBRFG33144), A. laibachii(NNIBRFG36991), S. quercus (NNIBRFG33183), Tausonia pullulans (NNIBRFG33181)등 6종의 야생효모들을 동정하였다. 이들 6종의 효모는 국내에 기록된 바가 없으며, 본 논문이 최초보고이다. 미기록 효모 모두 PD, YM 배지에서 잘 자랐으며, 15-30℃ 온도 범위에서 생육하였다. A. domesticum (NNIBRFG32938), A. laibachii (NNIBRFG36991), Tausonia pullulans (NNIBRFG33181) 균주들은 20% glucose를 함유한 YPD 배지에서 생육하는 내당성을 보였으며A. domesticum (NNIBRFG32938)과 A. laibachii (NNIBRFG36991)는 5% NaCl을 함유한 YPD 배지에서도 생육하여 내염성을 가졌다.

Expression of Toll-like Receptors 2 and 4 and Immunoglobulins in Children wih Recurrent Otitis Media with Effusion

  • Background: Toll-like receptors (TLRs) detect microbial infection and can directly induce innate host defense responses, which are thought to play critical roles in protecting the tubotympanum from infection. However, little is known about the relationship between TLRs, which are related to innate immunity, and immunoglobulins, which are related to adaptive immunity, in recurrent otitis media with effusion (OME). We therefore investigated the expression of TLR2 and TLR4 and immunoglobulin in children with OME. Methods: The study population consisted of 72 children with OME, 31 with more than 4 episodes in 12 months or more than 3 episodes in 6 months (otitis-prone group), and 41 with fewer than 3 episodes in 12 months (non-otitis prone group). The expression in middle ear effusion of TLR2 and TLR4 mRNA, as determined by Real time- -polymerase chain reaction (RT-PCR), and the concentrations of IgG, IgA, and IgM, as determined by Enzyme-linked immunosorbent assay(ELISA), were compared between the two groups. Results: Expression of TLR2 and TLR4 mRNA was lower in the otitis prone than in the non-otitis prone group, but the difference was not statistically significant (p>0.05). Between group differences in the concentrations of IgG, IgA and IgM in effusion fluid were not significant (p>0.05), and there were no correlations between immunoglobulin concentration and the expression of TLR2 and TLR4. Conclusion: Although there was a trend toward lower expression of TLR2 and TLR4 in the otitis-prone group, the differences, and those in immunoglobulin concentration, did not differ significantly between the otitis-prone and non-prone groups.

Artificial induction and isolation of cadmium-tolerant soil bacteria

  • Environmental pollution caused by various heavy metals is a serious global problem. To solve this problem, microbial bioremediation of contaminated metals has developed rapidly as an effective strategy when physical and chemical techniques are not suitable. In this study, cadmium (Cd)-tolerant soil bacteria were isolated via artificial induction in laboratory conditions instead of screening bacteria naturally adapted to metal-contaminated soils. Wild-type (WT) bacteria grown in uncontaminated soils were artificially and sequentially adapted to gradually increasing Cd concentrations of up to 15 mM. The resultant cells, named Soil-CdR15, survived at a Cd concentration of 10 mM, whereas WT cells failed to survive with 4 mM Cd on solid media for 2 d. In liquid media containing Cd, the SoilCdR15 cells grew with 15 mM Cd for 7 d, whereas the WT cells could not grow with 5 mM Cd. Both Soil-CdR15 and WT cells removed approximately 35% of Cd at the same capacity from liquid media containing either 0.5 or 1.0 mM Cd over 2 d. In addition to Cd, the Soil-CdR15 cells showed increased resistance to nickel, zinc, and arsenic compared to WT cells. The Soil-CdR cells were identified as Burkholderia sp. by partial sequencing of 16S rRNA. The data presented in this study demonstrate that isolation of heavy metal-tolerant microorganisms via artificial induction in laboratory conditions is possible and may be useful for the application of the microorganisms for the bioremediation of heavy metals.

Biodegradation of Ethylene in an Activated Carbon Biofilter

  • The objective of this study was to investigate the biodegradation of ethylene in an activated carbon biofilter inoculated with immobilized microbial consortium. The biofilter performance was monitored in terms of ethylene removal efficiency and carbon dioxide production. The biofilter was capable of achieving ethylene removal efficiency as much as 100% at a residence time of 14 min and an inlet concentration of 290 ppm. Under the same conditions, carbon dioxide with a concentration of up to 546 ppm was produced. Its was found that carbon dioxide was produced at a rate of 87 mg day$\^$-1/, which corresponded to a volume of 0.05 L day$\^$-1/. During operation with an inlet ethylene of 290 ppm, the maximum elimination capacity of the biofilter was 34 g of C$_2$H$_4$m$\^$-3/ day$\^$-1/. The biofilter could provide an attractive treatment technology for removing ethylene, an extremely volatile and slowly adsorbed compound.

Leuconostoc mesenteroides CBI-110에 의한 Alternan의 생산 (Production of Alternan by Leuconostoc mesenteroides CBI-110)

  • Alternan known as a kind of dextran is a linear and cyclic polysaccharide which is synthesized with sucrose by alternaansucrase (EC2.4.1.140) of Leuconostoc mesenteroides. But this polysaccharide is different from dextran in its structure and other properties. For the purpose of applying alternan for the food, cosmetic, and pharmaceutical industries, we isolated an excellent alternan-porducing strain, Leu. mesenteroides CBI-110. The optimum culture conditions were studied to improve the yield, and the activity of alternansucrase was increased up to 33U/ml in the optimum culture media. The concentrations of $Mn^{2+}$ and $Cu^{2+}$ ions were 0.01% and 0.03%, respectively, for the effective production of alternan. Finally, we obtained 25.6g/L of alternan.

Monasucs anka의 적색조 생산 특성 (Characterization of Red Pigment Production by Monascus anka)

  • Optimal media and cultural conditions for the production of red pigment were established using Monasurs anka KFCC 4478. The optimal temperature and initial pH for the production of red pigment were 30$^{\circ}C$ and 7.0, respectively. Glucose turned out to be most suitable carbon source for red pigment production. Optimal glucose concentration was 3.0%. Addition combined of nitrogen sources of peptone and NaNo3 induced good red pigment production. Thiamine-HCI and nicotinic acid were increased the production of red pigment. Under optimal conditions, maximum red pigment production and cell growth were observed after 5 days of incubation.
