Lactobacillus plantarum is a lactic acid bacterium that promotes animal intestinal health as a probiotic and is found in a wide variety of habitats. Here, we investigated the genomic features of different clusters of L. plantarum strains via pan-genomic analysis. We compared the genomes of 108 L. plantarum strains that were available from the NCBI GenBank database. These genomes were 2.9-3.7 Mbp in size and 44-45% in G+C content. A total of 8,847 orthologs were collected, and 1,709 genes were identified to be shared as core genes by all the strains analyzed. On the basis of SNPs from the core genes, 108 strains were clustered into five major groups (G1-G5) that are different from previous reports and are not clearly associated with habitats. Analysis of group-specific enriched or depleted genes revealed that G1 and G2 were rich in genes for carbohydrate utilization (${\text\tiny{L}}-arabinose$, ${\text\tiny{L}}-rhamnose$, and fructooligosaccharides) and that G3, G4, and G5 possessed more genes for the restriction-modification system and MazEF toxin-antitoxin. These results indicate that there are critical differences in gene content and survival strategies among genetically clustered L. plantarum strains, regardless of habitats.
A composite microbial system (XDC-2) was used to pretreat and hydrolyze corn stalk to enhance anaerobic digestion. The results of pretreatment indicated that sCOD concentrations of hydrolysate were highest (8,233 mg/l) at the fifth day. XDC-2 efficiently degraded the corn stalk by nearly 45%, decreasing the cellulose content by 22.7% and the hemicellulose content by 74.1%. Total levels of volatile products peaked on the fifth day. The six major compounds present were ethanol (0.29 g/l), acetic acid (0.55 g/l), 1,2-ethanediol (0.49 g/l), propionic acid (0.15 g/l), butyric acid (0.22 g/l), and glycerine (2.48 g/l). The results of anaerobic digestion showed that corn stalks treated by XDC-2 produced 68.3% more total biogas and 87.9% more total methane than untreated controls. The technical digestion time for the treated corn stalks was 35.7% shorter than without treatment. The composite microbial system pretreatment could be a cost-effective and environmentally friendly microbial method for efficient biological conversion of corn stalk into bioenergy.
Objective: The objective of this study was to investigate how cellulase or/and lactic acid bacteria (LAB) affected the fermentation characteristic and microbial community in wet brewer's grains (WBG) and corn stover (CS) mixed silage. Methods: The WBG was mixed thoroughly with the CS at 7:3 (w/w). Four treatment groups were studied: i) CON, no additives; ii) CEL, added cellulase (120 U/g fresh matter [FM]), iii) LAB, added LAB (2×106 cfu/g FM), and iv) CLA, added cellulase (120 U/g FM) and LAB (2×106 cfu/g FM). Results: All additive-treated groups showed higher fermentation quality over the 30 d ensiling period. As these groups exhibited higher (p<0.05) LAB counts and lactic acid (LA) content, along with lower pH value and ammonia-nitrogen (NH3-N) content than the control. Specifically, cellulase-treated groups (CEL and CLA) showed lower (p<0.05) neutral detergent fiber and acid detergent fiber contents than other groups. All additives increased the abundance of beneficial bacteria (Firmicutes, Lactiplantibacillus, and Limosilactobacillus) while they decreased abundance of Proteobacteria and microbial diversity as well. Conclusion: The combined application of cellulase and LAB could effectively improve the fermentation quality and microbial community of the WBG and CS mixed silage.

The study was conducted to evaluate differences of growth characteristics of rice cultivated in two different regions (Cheonan and Jeonju). It focused on nutritional composition and anti-nutritional factors of rice straw produced from 21 rice varieties including GM rice (Iksan 483). The range of general nutrition ingredient is that crude was 0.97 ~ 3.2 %, carbohydrate was 67.45 ~ 80.01 %, crude protein was 1.46 ~ 4.81 %, crude ash was 6.52 ~ 18.96 %, crude fiber was 25.77 ~ 40.02 %, NDF was 51.84 ~ 67.77 %, ADF was 27.11 ~ 40.44 %, calcium was 0.49 ~ 5.18 mg/g and phosphorous was 0.26 ~ 2.77 mg/g. The general nutritional contents of GM rice were included above range. The range of phytic acid of rice straws cultivated in Cheonan and Jeonju was 0 ~ 0.056 mg/ml and 0 ~ 0.059 mg/ml, respectively. The phytic acid content of GM was 0.033 mg/ml, which was in the range of the content of rice straw in Cheonan and Jeonju. The range of trypsin inhibitor of rice straws cultivated in Cheonan and Jeonju was 0.061 ~ 0.461 TIU/mg and 0 ~ 1.278 TIU/mg, respectively. The trypsin acid content of GM was 0.461 TIU/mg, which was in the range of the content of rice straw in Cheonan and Jeonju. In addition, we investigated microbial community from each soil sample by using metagenomics sequencing based on rRNA microbial diversity in order to inspect indirect changes of soil environment with cultivation of GM rice. Metagenomics analysis was carried out using soil samples cultivated with GM and non-GM rice for before transplanting, young panicle differentiation stage, heading stage, and ripening stage. Beta diversity of microbial community in both soil environments were calculated by using Bray-Curtis distance method and showed low value with an average of 0.24 (dissimilarity = 1). As a result, it was confirmed that the cultivation of GM does not give a significant effect on the change of microbial composition in soil. Therefore, Our study demonstrates that there is no difference in the composition of soil microorganism due to GM and non-GM rice.
To increase the collagen recovery rate, bromelain (PB) and a microbial enzyme (PM) were used to treat to pork skin with single agent or combinations. The quality of collagen from the pork skin was evaluated by enzymatic treatments. The highest results for the solid contents and pork skin recovery rate obtained with the microbial-enzyme-bromelain mixtue (PMB) were 13.60% and 18.05% respectively. The result also showed that the color was affected by different types of enzyme treatments. Although PM treatment showed the highest result in the protein content of 251.30 mg/100 g, PMB treatment was the highest in the test of collagen content of 37.73 g/100 g among the treatments. However bands of the pork skin were detected widely at 130 kDa and 170 kDa ranges in SDS-PAGE. The band of PB treatment showed at the range of below 17 kDa, changed into a smaller molecular weight. The collagen content test of the pork skin by the treatments, collagen contents with combination treatment of pork skin with PMB (0.5%) resulted the highest in 43.76 g/100 g. Also the fat content at the above treatment was reduced to 11.12% compared to the other treatments. With these results of this experiment, we conclude that the enzymatic treatments were effective for the processing property of pork skin like enhancing the yield of collagen.

Capsaicinoids in red pepper powder are known to show anti-bacterial effects; however, their effects during kimchi fermentation are not known. This study aimed to investigate the effects of various concentrations of capsaicinoids on kimchi fermentation. Five sets of kimchi samples were prepared using 0 mg/kg (control), $98.34{\pm}5.34mg/kg$ (mild), $243.47{\pm}3.71mg/kg$ (medium), $428.63{\pm}30.78mg/kg$ (hot), and $1,320.49{\pm}28.27mg/kg$ (extreme) capsaicinoid. The characteristics of each kimchi sample, including pH, acidity, organic acid, sugars, sugar alcohol, capsaicinoid content, and microbial community were periodically investigated during fermentation. Kimchi with red pepper powder shows significantly higher acidity than control kimchi, whereas pH values were the same. Organic acid in kimchi with red pepper powder was higher than in control kimchi, probably caused by higher lactic acid bacteria (LAB) counts in kimchi samples with red pepper powder. Our results show that addition of red pepper powder decreased Leuconostoc spp. counts in the bacterial community. In particular, Lactobacillus sakei and Leuconostoc gelidum counts increased and decreased, respectively, with increasing capsaicinoid content of red pepper powder added to kimchi. Overall, the results of this study indicate that physicochemical properties and LAB such as L. sakei and L. gelidum are influenced by capsaicinoid content. However, further studies are necessary to investigate the effects of the percentage of red pepper powder in kimchi on fermentation to provide practical guidelines for producing standardized kimchi.
This study examined the growth, fatty acid (FA) content, and carotenoids of a newly isolated freshwater microalga, Mychonastes sp. 246, in various culture media. The appropriate temperature and light intensity for culturing Mychonastes sp. 246 were determined as 18℃-22℃ and 200-250 µmol/m2/s using a high throughput photobioreactor. The microalgal cells were cultivated in 0.5 L bubble column photobioreactors using BG11, Bold's Basal media, and f/2 media. According to the growth results of the microalgae, BG11, among the tested media, showed the highest biomass concentrations (3.5 ± 0.1 g/L in 10 d). To enhance the biomass growth of the microalgae, the N:P ratio in BG11 was manipulated from 45:1 to 7:1 based on the stoichiometric cell composition. The biomass concentrations of Mychonastes sp. 246 grown on the manipulated BG11 (MBG) increased to 38% (4.6 ± 0.3 g/L in d) compared with the original BG11 (3.3 g/L). The FA content of the microalgae grown on the MBG was lower (8.4%) than that of the original BG11 (10.1%) while the FA compositions did not exhibit any significant differences. Furthermore, three kinds of carotenoids were identified in Mychonastes sp. 246, zeaxanthin, lutein, and β-carotene. These results suggest an effective strategy for increasing biomass concentrations, FA content, and carotenoids of microalgae by performing a simple N:P adjustment in the culture media.
The aim of the present study was to determine the effects of soil microbial agent with red ginseng marc on growth of watermelon during 5 months. The three treatments were distributed in a completely randomized design with four replicates per plot. After 1 week in planting dates, the growth of watermelon (full length, stem thichness, leaf length and lead width) showed no significant difference in all treatments. During elongation stage (20 days), soil microbial agent with red ginseng marc was increased by 5% in leaf thickness (May 23) and 7~14% in leaf length (May 16 and 23) when compared to other treatments. For changes in fruit bearing thickness, there were no differences among treatments. Characteristics of watermelon in harvest season have an effect on harvest and length, stalk length, naval length, weight, sugar content and yield, except for harvest and width. In particular, yields increased with treatments with two soil microbial agent (7~12%), indicating that soil microbial agent with red ginseng marc showed higher yield than the other treatments. In conclusion, red ginseng marc-treated soil microbial agents have a positive effect on the harvest season of watermelon and can provide useful information for the selection of the functional microbial properties and the registration of microbial fertilizer.
Changes of sodium chloride content in Chinese cabbage were investigated at different conditions. The diffusion rate of sodium chloride into the cabbage increased with increasing the temperature of brine solution. Sodium chloride content of Chinese cabbage at the lower portion of tank was higher than that at the upper position. The more washing and dewatering, the lower sodium chloride content of the cabbage was found. Microstructure pattern of salted cabbage tissue depended upon height of tank. The changed epidermis cell was recovered after several times of washing.
Pig slurry is a good soil amendment not only because of its high organic matter content, but also because of its ability to provide various nutrients. The objective of this study was to estimate the influence of raw and processed pig slurry application on pot soil over chemical fertilizer and non-amended control soil. Change in the chemical parameters (pH, organic matter (OM), organic carbon (OC), macro and micronutrients) and microbial mass of the treated soils were monitored over 30 to 90 days. Pot soil was treated with the recommended dose of pig slurry and chemical fertilizer, and was sampled after 30, 60 and 90 days of incubation. The least significanct difference (p<0.05) was observed on Fe, Cu, Zn, available P and K between treatments. All treatments increased N, P and K content and microbial mass of soil over control soil. Interestingly, no significant effects were detected on OM, OC, total bacteria, actinomycetes and fungi mass in soil irrespective of treatments given. However fungal and bacterial counts, as well as available nutrients, were found to be higher in processed slurry (PS)-treated soil compared to other soils. In general a significant correlation existed between the fungal count and OM, OC, Zn, T Kjeldahl N (TKN), available P and K of soil. A strong negative correlation was observed between pH and Fe in soil. This study clearly demonstrated that the use of processed manure as a fertilizer could be a key for sustainable livestock agriculture.
