Monasucs anka의 적색조 생산 특성 (Characterization of Red Pigment Production by Monascus anka)

  • Optimal media and cultural conditions for the production of red pigment were established using Monasurs anka KFCC 4478. The optimal temperature and initial pH for the production of red pigment were 30$^{\circ}C$ and 7.0, respectively. Glucose turned out to be most suitable carbon source for red pigment production. Optimal glucose concentration was 3.0%. Addition combined of nitrogen sources of peptone and NaNo3 induced good red pigment production. Thiamine-HCI and nicotinic acid were increased the production of red pigment. Under optimal conditions, maximum red pigment production and cell growth were observed after 5 days of incubation.

  • The production of extracellular polysaccharide, EPS WN9, from Paenibacillus sp. and its suitability as a viscosity modifying admixture for cement mortar mixing were investigated. After 48h culture in an optimized medium, cell growth and EPS production were 1.2g/L and 4.0g/L, respectively. By adding EPS WN9 to mortar, it was possible to prepare a homogeneous mortar without material segregation and excess air entrapment. The optimal amount of EPS addition to mortar found to be 0.02 to 0.05%(w/w) of the cement used. Increasing the dosage of EPS WN9 from 0 to 0.05%(w/w) resulted in a setting retardation of 0.14h to 0.8h and an increase in the compressive strength of mortar of 10 to 20%.

  • The effect of dissolved oxygen concentration on the metabolism of glucose in Pseudomonas putida BM014 was investigated. Glucose was completely converted to 2-ketogluconate via extracellular oxidative pathway and then taken up for cell growth under the condition of sufficient dissolved oxygen concentration. On the other hand, oxygen limitation below dissolved oxygen tension (DOT) value of 20% of air saturation caused the shift of glucose metabolism from the extracellular oxidative pathway to the intracellular phosphorylative pathway. Specific activities of hexokinase and gluconate kinase in intracellular phosphorylation pathway decreased as the DOT increased, while 2-ketogluconokinase activity in extracellular oxidative pathway increased under the same condition. This result can be usefully applied to microbial transformation of glucose to 2-ketogluconate, the synthetic precursor for iso-vitamine C, with almost 100% yield via extracellular oxidation by simple DOT control.

Growth performance, carcass traits and gut health of broiler chickens fed diets incorporated with single cell protein

  • Objective: This study was conducted to evaluate single cell protein (SCP), produced from Methylococcus species, as a protein source on the growth performance, carcass traits and gut health of broiler chickens. Methods: Ten iso-nitrogenous and iso-caloric diets containing 0 (Control), 2.5%, 5%, 7.5%, and 10% SCP replacing either soybean meal (T1 to T5) or fish meal (T6 to T10) were formulated. Each diet prepared for starter (0 to 14 days), grower (15 to 21 days), and finisher (22 to 42 days) phases was offered to four replicates of 10 chicks each (n = 400). Growth performance at different phases and carcass characteristics and intestinal morphology on 42nd day of trial were measured. Results: Body weight gain in groups fed 2.5% and 5% SCP diets were comparable to control during different phases and cumulatively, however lower (p<0.01) in 7.5% and 10% SCP diets. Feed conversion ratio was better (p<0.01) in 2.5% and 5% SCP diets. Dressing percentage, abdominal fat percentage and meat:bone ratio were not affected (p>0.05) by SCP inclusion in the diets. However, breast percentage was higher (p<0.01) in 2.5% and 5% SCP groups and thigh percentage higher in 7.5% and 10% SCP groups. Total microbial count in duodenum, jejunum and ileum were not affected (p>0.05) by SCP inclusion up to 10% in diets. Duodenal villi length and crypt depth were highest (p<0.01) in group fed 5% SCP diets and lowest in group fed 10% SCP diets. Jejunal villi length and crypt depth as well as ileal villi length were lowest (p<0.01) in group fed 10% SCP diets. Body weight gain, feed consumption, feed conversion ratio and gut health were better (p<0.01) in broilers fed fish meal based diets compared to soybean meal based diets. Conclusion: It was concluded that inclusion of SCP up to 5% replacing soybean meal in broiler diets is beneficial in improving growth rate, breast yield and gut health status.

  • Most medium formulations for improving culture of entomopathogenic nematodes (EPN) based on protein sources have used enriched media like animal feed such as dried egg yolk, lactalbumin, and liver extract, among other ingredients. Most results, however, showed unstable yields and longer production time. Many of the results do not show the detailed parameters of fermentation. Soy flour, cotton seed flour, corn gluten meal, casein powder, soytone, peptone, casein hydrolysates, and lactalbumin hydrolysate as protein sources were tested to determine the source to support optimal symbiotic bacteria and nematode growth. The protein hydrolysates selected did not improve bacterial cell mass compared with the yeast extract control, but soy flour was the best, showing 75.1% recovery and producing more bacterial cell number ($1.4{\times}10^9$/ml) than all other sources. The highest yield ($1.85{\times}10^5$ IJs/ml), yield coefficient ($1.67{\times}10^6$ IJs/g medium), and productivity ($1.32{\times}10^7$ IJs/l/day) were also achieved at enriched medium with soybean protein.

Features and Functions of Purple Pigment Compound in Halophytic Plant Suaeda japonica : Antioxidant/Anticancer Activities and Osmolyte Function in Halotolerance

  • Suaeda japonica is a halophytic plant that lives in mudflat at intertidal zone of western and southern coastal areas of Korea. The seawater-living plants showed a purple color during their whole life. In contrast, freshwater-living plants displayed a green color in leaves. When seawater-living plants were transferred to potting soil, the purple color was gradually changed to green in the leaves. The extracted purple pigment compound exhibited typical characteristics of betacyanin that were represented by water solubility, pH- and temperature-dependent color changes, sensitivity to light, UV-Vis spectra, and gel electrophoretic migration pattern. The LC-MS analysis of the extracted pigment compound showed the presence of two major protonated molecular ions ($[M+H]^+$) at m/z 651.1 and m/z 827.1. Antioxidant activity of the pigment compound was determined using stable free radical DPPH assay. It was found to have an antioxidant activity that is linearly increased in proportion to the reaction time for up to 30 min, and the activity was comparable to that of control BHA at 9.0 mg/ml. The anticancer activity against several tumor cell lines was also examined following the MTT assay. The significant growth inhibitory effect was observed on two tumor cell lines, SW-156 (human kidney carcinoma) and HEC-1B (human endometrial adenocarcinoma). Probably, the pigment compound may function as an osmolyte to uphold halotolerant physiological processes in saline environment.

강낭콩 잎에서 정제한 키틴분해효소의 항균활성 (Antifungal activity of a chitinase purified from bean leaves)

  • 강낭콩 잎에서 에틸랜에 의하여 유도되는 분자량 30KD인 염기성 키틴분해효소를 정제하고 그 항균활성을 연구하였다. 이 단백질은 chitinase 활성과 lysozyme 활성을 가졌으며, Aspergillus fumigatus, Botrytis cinerea, Fusarium oxysporum, Rhizoctonia solani의 균사 생장을 억제하였다. 그러나 함께 실험한 2종류의 미생물 chitinase, 달걀 lysozyme, 파파야 protease는 이들에 대한 항균작용을 갖지 않았다. 이상의 결과는 lysozyme 활성을 가진 식물 chitinase가 병원균의 균사생장을 억제하여 자신을 방어할 수 있음을 시사한다.

잔디 예지물의 생리활성 탐색 (Screening of Biological Activities of Grass Clippings from Turfgrass)

  • This study was carried out to investigate the biological activities of grass clippings from turfgrass including Zoysia japonica (TG-1), Zoysia matella (TG-2), Agrostis palustris (TG-3), and Poa pratensis (TG-4). The 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity was relatively higher in TG-1 and -2. Especially, TG-1 exerted the strongest hydroxyl radical scavenging effect, showing 90.40% at the concentration of 100 μg/ml. In addition, TG-1 inhibited the growth of Staphylococcus aureus, Escherichia coli and Helicobactor pylori. TG-1 also showed the highest inhibitory effect of AGS human gastric adenocarcinoma cell growth and nitric oxide production against lipopolysaccharide in RAW 264.7 macrophage cells. In conclusion, among the TG extracts, TG-1 has anti-oxidative, anti-microbial, anti-cancer and anti-inflammatory effect, indicating that TG-1 may be the potential source of functional food.

Effects of Salicylate and Glucose on Biodegradation of Phenanthrene by Burkholderia cepacia PM07

  • The stimulatory effects of exogenous salicylate as a pathway inducer on phenanthrene biodegradation were investigated using Burkholderia cepacia PM07. The phenanthrene degradation rate was greatly enhanced by increasing the salicylate additions, and the maximum rate was 19.6 mg $I^{-1}\;d^{-1}$ with the addition of 200 mg $I^{-1}$ of salicylate, 3.5 times higher than that (5.6 mg $I^{-1}\;d^{-1}$) without the addition of salicylate. The degradation rate was decreased at higher concentrations of salicylate (above 500 mg$I^{-1}$), and cell growth was significantly inhibited. The phenanthrene degradation was not affected by increasing glucose up to 2 g $I^{-1}$, although dramatic microbial growth was obtained. The stimulatory effect of exogenous salicylate decreased in the presence of glucose. After the addition of 200 mg $I^{-1}$ of salicylate, approximately $60\%$ of the initial phenanthrene (50 mg $I^{-1}$) was degraded after 96 h. However, with extra addition of 200 mg $I^{-1}$ of glucose, the phenanthrene degradation rate decreased, and only $18.5\%$ of the initial phenanthrene was degraded.

  • Coffee is one of the most popular and consumed beverages in the world, which leads to a high contents of solid residue known as spent coffee grounds (SCG). As is known, coffee beans contain several classes of health related chemicals, including phenolic compounds, melanoidins, diterpenes, xanthines and carotenoids. The waste water coming out of coffee industries has high concentration of organic pollutants and is very harmful for surrounding water bodies, human health and aquatic life if discharged directly into the surface waters. Hence it is essential to treat and manage the coffee waste. Oyster shells are a waste product from mariculture that creates a major disposal problem in coastal regions of southeast Korea. In the study, the oyster shell waste was used to treat the coffee waste and its effluents. Oyster shells are calcined at $1000^{\circ}C$ for 2 h, and allowed to test the calcined CaO powder ability to inhibit the growth of bacteria in different aging coffee wastes. Calcined oyster shell powder showed anti-bacterial effect that inhibited cell growth of Escherichia coli and other bacterial forms. The antimicrobial activity of calcium oxide from oyster shell waste for biological treatment and utilization as a fertilizers with economic ecofriendly in nature.
