Quantitative Determination of Marker Compounds and Pattern Recognition Analysis for Quality Control of Alismatis Rhizoma by HPLC

  • A quantitative method for determining levels of three bioactive compounds based on pattern recognition was developed and fully validated for the quality control of Alismatis Rhizoma (AR) by HPLC. Separation conditions were optimised using an Optimapak $C_{18}$ column ($250mm{\times}4.6mm$, 5 ${\mu}m$) with a mobile phase of acetonitrile and 0.1% aqueous phosphoric acid and detection wavelengths of 205 and 245 nm. Method validation yielded acceptable linearity ($r^2$ > 0.9998) and percent recovery (98.06%-101.71%). Limits of detection ranged from 0.08 to 0.15 ${\mu}g/mL$. Levels of the three bioactive compounds, alisol C acetate, alisol B, and alisol B acetate, in AR were 0.07-0.45, 0.38-10.32, and 1.13-8.59 mg/g dried weight, respectively. Pattern analyses based on these three compounds were able to differentiate Chinese and Korean samples accurately. The results demonstrate that alisol B and its acetate may be used as marker compounds for AR quality and can be regulated to no less than 0.36 and 1.29 mg/g of dried sample, respectively. The method described here is suitable for quantitative analyses and quality control of multiple components in AR.

  • The purpose of this study was to optimize the mandarin dry chip manufacturing using a response surface methodology. The experiment was designed based on a CCD (Central Composite Design), and the independent variables were the drying temperature ($X_1$, $50-90^{\circ}C$), drying time ($X_2$, 12-36 hours), and microwave pretreat time ($X_3$, 0-4 minutes). The results of appearance ($Y_5$), color ($Y_6$), taste ($Y_8$) and overall acceptance ($Y_{10}$) were fitted to the response surface methodology model ($R^2=0.86$, 0.88, 0.89, and 0.84, respectively). Increasing the drying temperature and microwave treatment time were negatively evaluated for consumer acceptance. On the other hand, a high value of consumer acceptance was evaluated when the drying time was more than 24 hr. Therefore, the optimal conditions of $X_1$, $X_2$, and $X_3$ were $52.989^{\circ}C$, 24 hr, and 1 min, respectively. Under these optimal conditions, the predicted values of $Y_5$, $Y_6$, $Y_8$, and $Y_{10}$ were 5.066, 5.338, 5.063, and 5.339, respectively.

  • Samples of Corbicula japonica Prime of Jujin estuary in Gochang were collected from July 2000 to September 2001. Age of C. japonica was determined from the rings on the shell. The relationship between shell length and ring radius in each ring group was expressed as a regression line. Therefore, there is a correspondence in each ring formation. Based on the monthly variation of the marginal index (MI') of the shell, it is assumed that the ring of this species was formed once a year during the period of February and March. The relationship between shell length (SL; mm) and total weight (TW; g) was expressed by the following equation: TW = 1.0942 ${\times}10^{-4}SL^{3.3217}$ ($r^2$ = 0.9905). Shell length (SL) and shell height (SH; mm) was highly correlated with shell height as the following equation: SH = 0.9174 SL - 0.9935 ($r^2$ = 0.9885). The shell length (SL) - shell width (SW) relation was also expressed by the following equation; SW = 0.5925 SL - 1.1706 ($r^2$ = 0.9726). Growth curves for shell length and total weight fitted to the von Bertalanffy's growth curve were expressed as: $$SL_t = 46.4861[1-e^{-0.3383(t+0.0958)}]$$, $$TW_t = 34.54[1-e^{-0.3383(t+0.0958)}]^3.3217$$.

ICP/MS를 이용한 축산물의 유해중금속 분석방법 및 숙련도 검증 (Method and proficiency test for analysis of toxic metals in livestock products by ICP/MS)

  • The analytical method of trace toxic metals in livestock products was confirmed and validated through certified reference material (CRM) and the international proficiency tests. There are some difficulties to determine low levels of toxic metals in livestock products because of interferences due to the matrix. The recoveries of CRM (NIST 1577c) ranged from 73.9 to 119% for lead and from 86.4 to 111% for cadmium in bovine liver. The international proficiency tests were carried out with the milk powder and cocoa powder samples including metals provided by Food Analysis Performance Assessment Scheme (FAPAS$^{(R)}$, UK). The test samples were prepared by microwave digestion using solution of $HNO_3:H_2O_2:H_2O$ (v/v/v = 5 : 2 : 4) and analyzed by ICP/MS. The analytical result of cadmium in milk powder was $121{\mu}g/kg$ with -0.3 of the z-score compared to the assigned value of $131{\mu}g/kg$ by FAPAS$^{(R)}$. The analytical results of lead and cadmium in cocoa powder were $29.2{\mu}g/kg$ and $97.6{\mu}g/kg$, respectively, which satisfied the assigned values of $34.2{\mu}g/kg$ for lead and $126{\mu}g/kg$ for cadmium by FAPAS$^{(R)}$. It is verified that the analytical method is accurate and reliable to determine trace lead and cadmium in livestock products by microwave digestion and ICP/MS.

혹명나방에 대한 매실나무 추출물의 살충활성 (Insecticidal Activities of Prunus mume Extract Against Rice Leaf Folder (Cnaphalocrocis medinalis Guenee))

  • 흑명나방 3령 유충에 대한 매실나무 부위별(줄기+꽃, 열매) 메탄올 추출물의 살충활성과 용화율 및 용무게의 변화를 조사하였다. 매실나무 열매에 대한 살충률은 4,000 ppm에서 34%, 8,000 ppm에서 36%, 16,000 ppm에서 40%, 32,000 ppm에서 58% 값을 보였고, 줄기+꽃에 대한 살충률은 4,000 ppm에서 38%, 8,000 ppm에서 45%, 16,000 ppm에서 58%, 32,000 ppm에서 75%의 살충활성을 보였다. 매실나무 열매와 줄기+꽃 추출물을 혹명나방에 처리하였을 때 용화율은 농도가 증가함에 따라 서서히 낮아졌다. 용무게는 대조구과 처리구에서 별 차이점을 보이지 않았으나 용화율과 용무게에 대한 상관관계를 비교하여 분석한 결과 매실나무 열매 추출물보다 줄기+꽃 추출물에서 상대적으로 높은 유의성을 나타냈다. 또한 혹명나방의 섭식저해율은 두 가지 추출물에서 60%이상의 살충률을 보이며 섭식저해와 더불어 섭식독으로써 효과가 있음을 보였다.

품종별 감피의 화학적 성분 특성 (Chemical Composition and Quality of Persimmon Peels According to Cultivars)

  • 곶감 원료용 떫은감(상주둥시, 고종시, 월하시, 청도반시, 함안수시, 평핵무, 횡야)의 과피로부터 유용성분 및 카로티노이드, 총페놀 등 이화학적 특성을 조사하였다. 감피의 일반성분 조섬유를 제외하고는 품종별로 차이가 없었다. 유리당으로는 glucose, fructose, sucrose가 확인 되었으며, $Myristic\;acid(C_{14:0}),\;palmitic\;acid(C_{16:0}),\;palmitoleic\;acid(C_{16:1})\;oleic\;acid(C_{18:1})$$linolenic\;acid(C_{18:3})$가 주된 지방산으로 확인되었다. 총아미노산의 함량은 244.84-371.45 mg/100 g이고, 유리아미노산 함량은 3.69-28.31 mg/100 g이었다. 감피의 식이섬유중 불용성 식이섬유가 34.89-50.76 g/100 g으로 가용성 식이섬유(2.44-7.09 g/100 g)보다 월등하게 높게 나타났다. retinol equivalent(R.E.)은 20.02-60.60 mg/100 g이었으며, 비타민 C는 24.69-106.01 mg/100 g으로 품종간에 다소 차이를 보였다. 카로티노이드 함량은 상주둥시가 340.6 mg/100 g으로 가장 높은 함량을 보였으며, 총페놀 함량에서도 44.07-196.98 mg/100 g으로 품종간 차이를 보였다. 따라서 감피는 카로티노이드, 식이섬유, 페놀화합물 등 유용성분의 활용가치가 높을 것으로 생각된다.

C/N/O/S stable isotopic and chemometric analyses for determining the geographical origin of Panax ginseng cultivated in Korea

  • Background: The geographical origin of Panax ginseng Meyer, a valuable medicinal plant, is important to both ginseng producers and consumers in the context of economic profit and human health benefits. We, therefore, aimed to discriminate between the cultivation regions of ginseng using the stable isotope ratios of C, N, O, and S, which are abundant bioelements in living organisms. Methods: Six Korean ginseng cultivars (3-yr-old roots) were collected from five different regions in Korea. The C, N, O, and S stable isotope ratios in ginseng roots were measured by isotope ratio mass spectrometry, and then these isotope ratio profiles were statistically analyzed using chemometrics. Results: The various isotope ratios found in P. ginseng roots were significantly influenced by region, cultivar, and the interactions between these two factors ($p{\leq}0.001$). The variation in ${\delta}^{15}N$ and ${\delta}^{13}C$ in ginseng roots was significant for discriminating between different ginseng cultivation regions, and ${\delta}^{18}O$ and ${\delta}^{34}S$ were also affected by both altitude and proximity to coastal areas. Chemometric model results tested in this study provided discrimination between the majority of different cultivation regions. Based on the external validation, this chemometric model also showed good model performance ($R^2=0.853$ and $Q^2=0.738$). Conclusion: Our case study elucidates the variation of C, N, O, and S stable isotope ratios in ginseng root depending on cultivation region. Hence, the analysis of stable isotope ratios is a suitable tool for discrimination between the regional origins of ginseng samples from Korea, with potential application to other countries.

염장 멸치 (Salited Anchovy)의 제조조건 1. 염장방법에 따른 염장 멸치 (salted anchovy)의 제조 중 성분 변화 (Establishment of Processing Conditions of Salted Anchovy 1. Changes of Chemical Compositions during Fermentation of Salted Anchovy by Salting Methods)

  • Anchovy fillet 제조를 위하여 염장방법 및 숙성온도에 따른 염장 멸치의 숙성 중 성분변화를 조사하였다. 물간에 비하여 마른간 하였을 때 육의 탈수량이 많았으며, $20^{\circ}C$에 숙성한 것이 $5^{\circ}C$에 비하여 수분함량이 다소 낮았다. 염분함량은 물간한 것이 마른간에 비하여 높았으며, $20^{\circ}C$에서 숙성 한 것이 $5^{\circ}C$에 비하여 높았다. 총 질소는 물간이 마른간한 것에 비하여 낮았으며, 염장 초기에 총질소의 저하가 현저하였다. 마른간이나 물간 모두 $20^{\circ}C$에서 염장 멸치의 숙성 중 아미노태질소가 현저하게 증가하여 마른간은 숙성120일, 물간은 숙성 30일에 거의 최대를 나타낸 반면, $5^{\circ}C$에서는 미미한 증가를 보였다. 휘발성염기질소의 변화는 아미노태질소와 거의 비슷한 경향을 나타내었다. 물간한 것은 마른간에 비하여 동일 온도에서 가수분해가 빠르게 진행되었으며, $5^{\circ}C$에서는 가수분해가 현저하게 억제되었다. 과산화물가는 마른간이 물간에 비하여 빠르게 증가하였으며, $5^{\circ}C$에 숙성한 것이 $20^{\circ}C$에 비하여 낮은 과 산화물가를 나타내었다. 이상의 결과로 보면, 염장 멸치의 제조를 위해서는 대멸치를 가염지 한 다음, $25\%$의 식염으로 마른간하여 $5^{\circ}C$에서 숙성시키는 것이 육의 연화를 막고, 숙성이 어느 정도 진행된 염장 멸치론 얻을 수 있을 것으로 사료된다.

아마릴 정(글리메피리드 2mg)에 대한 글리메드 정의 생물학적 동등성 (Bioequivalence of Glimed Tablet to Amaryl Tablet (Glimepiride 2 mg))

  • The purpose of the present study was to evaluate the bioequivalence of two glimepiride tablets, $Amaryl^{\circledR}$ (Handok/Aventis Pharm. Co., Ltd.) and Glimed (Kuhn II Pharm. Co., Ltd.), according to the guidelines of Korea Food and Drug Administration (KFDA). The glimepiride release from the two glimepiride formulations in vitro was tested using KP VIII Apparatus II method with a variety of dissolution media (pH 1.2, 4.0, 6.8 buffer solution, water and blend of PSB 80 into each dissolution medium). Twenty six healthy male subjects, $22.65{\pm}2.19$ years in age and $66.55{\pm}8.85$ kg in body weight, were divided into two groups and a randomized $2\;{\times}\;2$ cross-over study was employed. After one tablet containing 2 mg as glimepiride was orally administered, blood was taken at predetermined time intervals and the concentrations of glimepiride in serum were determined using HPLC method with UV detector. The dissolution profiles of two formulations were similar at all dissolution media. Besides, the pharmacokinetic parameters such as $AUC_t$, $C_{max}$ and $T_{max}$ were calculated and ANOVA test was utilized for the statistical analysis of the parameters using logarithmically transformed $AUC_t$, $C_{max}$ and untransformed $T_{max}$. The results showed that the differences between two formulations based on the Amaryl were -3.70, -8.28 and 0.61% for $AUC_t$, $C_{max}$ and $T_{max}$, respectively. There were no sequence effects between two formulations in these parameters. The 90% confidence intervals using logarithmically transformed data were within the acceptance range of log(0.8) to log(1.25) (e.g., $log(0.84){\sim}log(1.04)$ for $log(0.82){\sim}log(1.03)$ for $AUC_t$ and $C_{max}$, respectively). Thus, the criteria of the KFDA guideline for the bioequivalence were satisfied, indicating Glimed tablet and Amaryl tablet were bioequivalent.

Effect of Cultivars and Cooking Methods on the Trypsin Inhibitor Activities of Potatoes

  • The trypsin inhibitor activities (TIA) of various potato cultivars were evaluated by measuring the inhibition of trypsin inhibitor activity using N-benzoyl-DL-arginine-p-nitroanilide (BAPNA) as substrate. The TIA values of 5 potato cultivars (1.99 to 2.88 mg/g) were significantly different among cultivars (p<0.05). When the TIA values of commercially processed potatoes were determined, no TIA was detected. During cooking, the $IT_{50}$ (time required to reach 50% inhibition of TIA) values were decreased as heating temperature and time increased. The ITso of moist heating was estimated to be 0.34 min at $100^{\circ}C$, whereas for deep-fat frying the $IT_{50}$ was 0.13 min at $180^{\circ}C$ and 5.28 min for oven baking at $100^{\circ}C$. The $IT_{50}$ value of microwave cooking was 0.194 min at medium heat, and which was similar to that of pressure cooking at $120^{\circ}C$ (0.185 min). Moreover, there was a negative relationship between temperature (${\geq}80^{\circ}C$) and $IT_{50}$ values ($R^2=0.99$, p<0.01). The TIA of potato was completely inactivated by moist heating at $100^{\circ}C$ within 5 min, whereas the pressure cooking at $120^{\circ}C$ and deep-fat frying at $180^{\circ}C$ within 60 and 30 sec, respectively. Based on our results, deep-fat frying is the most effective cooking method to reduce TIA in potatoes.
