The Inhibitory Effects of Acanthopanax sessiliflorum Seeman on Melanogenesis (오가피추출물의 멜라닌 생성 저해 효과)

  • To develop a new natural whitening agent for cosmetics, we investigated effects of Acanthopanax sessiliflorum Seeman on melanogenesis. We prepared phenolic acid-rich extract including two phenolic acids, chlorogenic acid and caffeic acid, as predominant constituents from Acanthopanax sessiliflorum Seeman. Phenolic acid-rich extract showed ROS scavenging activities in 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $3.43{\pm}0.35{\mu}g/mL$ and $158.91{\pm}1.57{\mu}g/mL$, respectively. Phenolic acid-rich fraction reduced melanin contents of B16 melanoma cells dose-dependantly and the decrease was $27.27{\pm}2.66%$ at a concentration of $100{\mu}g/mL$. And the phenolic acid-rich fraction reduced intracellular tyrosinase activity about $53.67{\pm}8.55%$ at a concentration of $100{\mu}g/mL$. Phenolic acid-rich extract inhibited tyrosinase and TRP-2 expression at protein level. These results suggest that phenolic acid-rich fraction reduced melanin formation by the inhibitions of tyrosinase activity and expression in B16 melanoma cells. Therefore, we suggest that phenolic acid-rich extract could be used as a whitening ingredient in cosmetics.

The Relationship between Self-esteem and Satisfaction in Major of Nursing Students (간호대학생의 자아존중감과 전공만족과의 관계)

  • Purpose: This study was performed to identify the relationship between self-esteem and satisfaction in major of nursing students. Methods: Data were collected from the 10th to the 14th of April in 2006, from 281 nursing students in Jeollabuk-Do area. The collected data were analysed by using the SPSS WIN 11.0. Results: The results of this study were as followings: 1) The mean score for self-esteem was $2.89({\pm}.42)$, self-esteem was significantly different according to the general characteristics of grade(F=4.718, p=.010), satisfaction of pocket money(F=11.241, p=.000), satisfaction of present life(F=9.875, p=. 000) and hope for the employment field(F=2.583, p=.026). 2) The mean score for satisfaction in major was $3.79({\pm}.49)$, satisfaction in major was significantly different according to the general characteristics of satisfaction of pocket money(F=5.280, p=.006), motivation of entrance(F=7.384, p=.000), satisfaction of present life(F=27.000, p=.000), hope for the employment field(F=3.019, p=.011) and one of family (t=2.416, p=.017). 3) There was a significant correlation between self-esteem and satisfaction in major(r=.353, p=.000). 4) The most important variables affected satisfaction in major could explain a total 27.9%, were accounted for 22.7% in the satisfaction of present life and 5.2% in the self-esteem by the stepwise multiple regression analysis. Conclusion: As a result, self-esteem was correlated with satisfaction in major in nursing students. Therefore we need to develop various self-esteem program for nursing students.

  • Ribose-5-phosphate isomerase A (RpiA) plays an important role in interconverting between ribose-5-phosphate (R5P) and ribulose-5-phosphate in the pentose phosphate pathway and the Calvin cycle. We have determined the crystal structures of the open form RpiA from Vibrio vulnificus YJ106 (VvRpiA) in complex with the R5P and the closed form with arabinose-5-phosphate (A5P) in parallel with the apo VvRpiA at $2.0{\AA}$ resolution. VvRpiA is highly similar to Escherichia coli RpiA, and the VvRpiA-R5P complex strongly resembles the E. coli RpiA-A5P complex. Interestingly, unlike the E. coli RpiA-A5P complex, the position of A5P in the VvRpiA-A5P complex reveals a different position than the R5P binding mode. VvRpiA-A5P has a sugar ring inside the binding pocket and a phosphate group outside the binding pocket: By contrast, the sugar ring of A5P interacts with the Asp4, Lys7, Ser30, Asp118, and Lys121 residues; the phosphate group of A5P interacts with two water molecules, W51 and W82.

siRNA Silencing EZH2 Reverses Cisplatin-resistance of Human Non-small Cell Lung and Gastric Cancer Cells

  • Clinical resistance to chemotherapeutic agents is one of the major hindrances in the treatment of human cancers. EHZ2 is involved in drug resistance and is overexpressed in drug-resistant cancer cell lines. In this study, we investigated the effects of EHZ2 on cisplatin -resistance in A549/DDP and AGS/DDP cells. EHZ2 mRNA and protein were found to be significantly overexpressed in A549/DDP and AGS/DDP cells, compared to parental cells. EHZ2 siRNA successfully silenced EHZ2 mRNA and protein expression. Proliferation was inhibited and drug resistance to cisplatin was improved. Flow cytometry showed that silencing of EHZ2 arrested A549/DDP and AGS/DDP cells in the G0/G1 phase, increasing apoptosis, rh-123 fluorescence intensity and caspase-3/8 activities. Silencing of EHZ2 also significantly reduced the mRNA and protein expression levels of cyclin D1 and MDR1,while up-regulating p15, p21, p27 and miR-218 in A549/DPP cells. Furthermore, silencing of EHZ2 also significantly increased the expression level of tumor suppressor factor miR-218. We also found down-regulating EHZ2 expression increased methylation in A549/DDP and AGS/DDP cells. This study demonstrates that drug resistance can be effectively reversed in human cisplatin-resistant lung and gastric cancer cells through delivery of siRNAs targeting EHZ2.

Multiplex PCR Assay for Simultaneous Detection of Korean Quarantine Phytoplasmas

  • Multiplex PCR assays were developed for the simultaneous detection of ten important Korean quarantine phytoplasmas. The species-specific primers were designed based on ribosomal protein, putative preprotein translocase Y, immunodominant protein, elongation factor TU, chaperonin protein and the 16S rRNA genes of 'Candidatus (Ca.) Phytoplasma' species. Three main primer sets were prepared from ten designed primer pairs to limit nonspecific amplification as much as possible. The multiplex PCR assay using the three primer sets successfully amplified the correct conserved genes for each 'Ca. Phytoplasma' species. In addition, ten important 'Ca. Phytoplasma' species could be easily determined by recognizing band patterns specific for each phytoplasma species from three primer sets. Moreover, a high sensitivity of multiplex PCR for each primer set was observed for samples containing a low DNA concentration (10 ng/${\mu}l$). This study provides the useful multiplex PCR assay as a convenient method to detect the presence of ten important quarantine phytoplasmas in Korea.

Bone Mineral Density, Body Mass Index, Stress, and Health Promotion Lifestyle of Female College Students (일 대학 여대생의 골밀도, 체질량지수, 스트레스 및 건강증진 생활양식)

  • Purpose: The purpose of this study was to examine the bone mineral density (BMD), body mass index (BMI), stress, and health promotion lifestyle of female college students and to assess relations among them. Methods: A total of 220 female college students were assessed through anthropometric measurements and bone mineral density test using quantitative ultrasound. In addition, the subjects were asked about stress and health promotion lifestyle with a self-rating questionnaire. Collected data were processed with the SPSS/WIN 12.0 program. Results: 1) Of the students, 67.8% had weight control experience. The percentage of the osteoporosis, osteopenia and normal groups were 2.8%, 53.5% and 43.8%, respectively, and the percentage of the underweight, normal and overweight groups according to BMI were 27.9%, 57.2% and 14.9%, respectively. 2) The levels of stress and health promotion lifestyle were 2.9 and 2.3, respectively. 3) There was a positive correlation between BMD and BMI (r=.196, p<.01). There was a negative correlation between stress and health promotion lifestyle (r=-.35, p<.01). 4) Weight control experience made negative effects on BMD and BMI. Conclusion: The findings of this study suggest the necessity of new health promotion programs to increase bone density of female college students. Stress management programs are also needed.

A Clinical Skill Test using OSCE Modules Developed by Nursing Students (간호학생의 OSCE모듈 개발 및 실기평가의 경험)

  • Purpose: OSCE, Objective Structured Clinical Examination, is a good way to evaluate clinical skills of nursing students. To do this, we need modules, evaluators, persons to run the examination, as well as models and standardized patients if necessary. Author coached nursing students to develop modules and ran the examination by themselves. Method: 24 third-year and 4 first-year students volunteered; third-year students developed 5 modules, and organized and ran the examination. First-year students played patient role. 60 2nd-year students participated as examinees. Modules were duplicated to finish examination in a given time. The relationship between OSCE score, conventional clinical evaluation score and credit of students was compared. Effect of module duplication on score was tested. And responses of examinees were collected. Results: There was no correlation between OSCE and conventional clinical evaluation score (r=0.07), and credit (r=0.27), And there was no difference of OSCE score between duplicated modules $(53.77{\pm}7.61$ vs $55.33\pm7.74).$ Response of examinees to OSCE was favorable. Examinee did not expressed resistance for the evaluation by OSCE developed and ran by students. Conclusion: Nursing students successfully developed and ran OSCE, which was accepted favorably by examinees. Student-developed OSCE may play a role in evaluation of clinical performance.

A study on Kudoa septempunctata infection from sashimi and sushi of olive flounder Paralichthys olivaceus in Busan, South Korea

  • Kudoa septempunctata has been reported as a new parasite in aquacultured olive flounder Paralichthys olivaceus, and also as a causative agent of food poisoning in humans. This paper investigated the infection of K. septempunctata in 216 sashimi and 20 sushi made of olive flounders in Busan, Korea. Among 236 samples, K. septempunctata was detected in eleven sashimi with 6-7 polar capsules by the microscopy. Among eleven sashimi, five sashimi were positive in Polymerase Chain Reaction (PCR) assay with the targets of 18S rDNA and 28S rDNA. The genotype of all the five PCR results is identified as the genotype ST3 which is common in Korea. K. septempunctata was found in olive flounders sashimi from Samcheonpo and Wando outside of Jeju Island. These findings would contribute to establish the standard of K. septempunctata for preventing food-borne outbreaks in advance and providing further preventive management for the seafood safety.

Factors Influencing Professional Quality of Life in Intensive Care Unit Nurses of University Hospitals (간호·간병통합서비스병동 간호사의 직무스트레스가 소진에 미치는 영향: 감성지능의 조절 및 매개효과 )

  • Purpose : This study aims to identify job stress, emotional intelligence, and exhaustion of the nurses in comprehensive nursing service units and then verify the moderating and mediating effects of emotional intelligence on the relationship between job stress and exhaustion. Methods : Participants are 118 nurses working in two general hospitals. The collected data are analyzed using the SPSS WIN 27.0 and AMOS 18.0 programs by assessing frequency and percentage, mean and standard deviation, independent t-test and one-way ANOVA, Scheffé test, Pearson correlation coefficient, and hierarchical multiple regression analysis and bootstrapping. Results : Exhaustion has a statistically significant positive correlation with job stress (r=.40, p <.001) and a statistically significant negative correlation with emotional intelligence (r=-.26, p =.004). A partial mediating effect of emotional intelligence is found between job stress and exhaustion, however, there is no moderating effect. Conclusion : It is expected that to reduce the exhaustion of nurses in comprehensive nursing service units, it is necessary to assess job stress and emotional intelligence, and strengthen emotional intelligence along with job stress intervention.

  • Cholesteryl methyl ether$(CH_3OC_{27}H_{45})$ crystallizes in the monoclinic space group $P2_1$with a = 11.740 (8), b = 7.576 (5), c = 15.492 (10)${\AA},\; {\beta}\;= 110.39 (5)^{\circ}$, Z = 2, Dc = 1.03 g/$cm^3$ and Do = 0.96 g/$cm^3$. The intensities were collected on a Nonius CAD-4 diffractometer with Nb-filtered Mo-$K_{\alpha}$ radiation. The structure was solved by direct methods and refined by full matrix least-squares methods. The final R factor was 0.085 for 1479 observed reflections. Compared with other cholesterol derivatives, the cholesteryl ring and tail region of the molecule are normal. The molecular long axes are parallel to the [101] axis and molecules are packed together in a similar way to those in cholesteryl cliloride and bromide.
