To evaluate the longevity property of Allium hookeri (Liliaceae) leaves, this study was performed. Ethanol extract of A. hookeri was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions, and lifespan-extending effects of the fractions were checked using Caenorhabditis elegans model system. The most active ethyl acetate-soluble fraction elevated heat stress tolerance, and increased expression of stress resistance protein. Further studies were performed to investigate several aging-related factors such as reproduction, food intake, growth and movement of C. elegans. The results revealed that there were no significant changes in aging-related factors including reproduction and food intake, however, ethyl acetate-soluble fraction treatment led to up-regulation of locomotory ability and growth of aged worms, suggesting ethyl acetate-soluble fraction affected healthspan as well as lifespan of nematode.

A gas chromatography/mass spectrometric assay method was developed for the simultaneous determination of ethylene glycol monomethyl ether (EGME) and diethylene glycol monomethyl ether (DEGME) in spilled aviation fuels. Ethylene glycol monobutyl ether (EGBE) and ethylene glycol monoethyl ether (EGEE) were used as internal standard and surrogate, respectively. The sample preparation consists of back-extraction with 7 mL of methylene chloride after extraction of 50 mL of fuel with 2 mL of water. The extract was concentrated to dryness and dissolved with 100L of methanol and analyzed by CC-MS (SIM). The peaks had good chromatographic properties by using semi-polar column and the extraction of these compounds from fuel also gave high recoveries of 75 and 85 % with small variations for EGME and DEGME, respectively. Method detection limits were 1.3 ng/mL for EGME and 1.0 ng/mL for DEGME in spilled fuel. The method may be useful for fuel-type differentiation between kerosene and JP-8, which may originate from the storage tank.
Five terpenes (1~5), three fatty acids (6~8), two sterols (9 and 11), and a monogalactosyldiacyl glycerol (10) were isolated from the methylene chloride extract of the aerial part of Cirsium setidens. Their chemical structures were determined to be $\alpha$-tocopherol (1), 25-hydroperoxycycloart-23-en-3$\beta$-o1 (2), 24-hydroperoxycycloart-25-en-3$\beta$-o1 (3), mokko lactone (4), transphytol (5), 9, 12, 15-octadecatrienoic acid (6), 9, 12-octadecadienoic acid (7), hexadecanoic acid (8), acylglycosyl $\beta$-sitosterol (9), (2R)-1, 2-O-(9z, 12z, 15z-dioctadecatrienoyl)-3-O-$\beta$-D-galactopyranosyl glycerol (10) and $\beta$-sitosterol glucoside (11) by spectral evidences. Compound 3 exhibited significant cytotoxic activity against five human cancer cell lines with its $ED_{50}$ values ranging from 2.66 to 11.25 $\mu$M.
In this study, an efficient ultrasound-assisted liquid-liquid extraction process was developed for recovering of paclitaxel from plant cell cultures. The optimal ultrasonic power and operating time were 250 W and 15 min at fixed ratio of bottom phase, methylene chloride to top phase, MeOH (25%, v/v). Under the optimal conditions developed in the present method, most of the paclitaxel (~92%) was recovered from crude extract by a single extraction step. Due to the synergistic effect of ultrasound by the addition of inorganic salt, an appropriate inorganic salt concentration and the ultrasonic power were found to be required for the effective recovery of paclitaxel using ultrasound-assisted liquid-liquid extraction.

Seven phenolic compounds including p-coumaric acid (1), 4-hydroxybenzoic acid (2), 4-hydroxybenzaldehyde (3), vanillic acid (4), luteolin (5), acacetin (6), and tricin (7), were isolated from the methylene chloride and ethyl acetate fractions of Echinochloa utilis grains. Compounds (1 - 4, 6) were isolated for the first time from this plant. These compounds were tested for inhibitory activities against LPS-induced NO production in RAW 264.7 cells. Compounds 5 and 6 displayed significant inhibitory effects, with $IC_{50}$ values of $27.9{\pm}2.6$ and $14.0{\pm}1.1{\mu}M$, respectively. The results suggested that E. utilis ethanolic extract may be used as a potential source of anti-inflammatory agents and functional foods for the treatment of allergic diseases.

This study was conducted to evaluate the effect of anti-obesity and antioxidant enzyme activities in vitro by different solvent fractions from the roots of Codonopsis lanceolata. The cytotoxicity of different solvent fractions of C. lanceolata on 3T3-L1 preadipocytes were evaluated using the MTT assay, the rate of cell survival progressively decreased in a dose-dependent manner. Butyl alcohol fraction at $200{\mu}g/mL$ exhibited a pronounced cytotoxic effect (75.73%) on 3T3-L1 cell comparable to that of the hexane fraction (79.82%), methylene chloride fraction (84.02%), ethyl acetate fraction (87.62%) and DW fraction (86.30%) at the same concentration. The Oil Red O solution was used to determine whether different solvent fractions of C. lanceolata induce adipocyte differentiation in 3T3-L1 preadipocytes. Confluent 3T3-L1 cells were treated with $50{\mu}g/mL$ concentration of solvent fraction extracts from C. lanceolata. Inhibitory degree of lipid accumulation against solvent fraction extracts showed a significant level compared with the control. Both lipid accumulation and adipocyte differentiation showed relatively high effect on methyl chloride fraction. The root extract of C. lanceolata had the highest SOD enzyme activity of 84.5% in ethyl acetate partition layer and while water partition layer of diploid showed the lowest SOD enzyme activity of 57.9%. The activity of CAT, APX and POD showed a significantly higher activity in ethyl acetate partition layer compared with the other fraction. These results suggested that the roots of C. lanceolata may assist in the potential biological activity on anti-obesity and antioxidant capacity.
We investigated the flavonoid and phenol contents and antioxidant effect of wine by-product extract. Antioxidant effects were measured with 1,1-diphenyl-2-picryhydrazyl (DPPH) and 2.2'-azino-bis(3-ethylbenothiazoline-6-sulfonic acid) diammonium salt radical cation (ABTS+) assays. Cellular reactive oxygen species (ROS) were measured with the dichlorofluorescein-diacetate (DCFH-DA) assay. The flavonoid and phenol contents of the methanol (MeOH) extract were greater than those of the acetone+methylene chloride (A+M) extract. Among fractions, the 85% aqueous methanol (85% aq. MeOH) fraction contained the highest flavonoid contents, while the n-BuOH fraction had more phenol contents. In the DPPH and ABTS assays, the MeOH extract showed a scavenging effect greater than that of the A+M extract (p<0.05). The n-BuOH fraction (0.5 mg/ml concentrations) showed scavenging effects of 72% and 92%, respectively, in the DPPH and ABTS assays (p<0.05). However, the 85% aq. MeOH fraction showed a 90% scavenging effect in the DPPH assay only. In 120 min ROS production assay, all tested fractions dose-dependently decreased cellular ROS production induced by H2O2 in comparison with that produced by exposure to the extract-free control. The MeOH extract showed a higher sinhibitory effect on cellular ROS producing than that of the A+M extract at all concentrations tested. Treatment with the n-BuOH fraction (0.1 mg/ml concentrations) inhibited cellular ROS production by 60%. These results indicate that the n-BuOH fraction of wine by-product extract inhibited cellular oxidation and may contain valuable bioactive compounds such as flavonoids and phenols.
To estimate the regulatory effects of Scutellaria baicalensis extracts on melanin biosynthesis, we evaluated the regulatory effects of the tyrosinase gene on B16 melanoma cells. The results revealed that methanolic extracts of Scutellaria baicalensis resulted in a high increase in the expression of the tyrosinase gene. Specifically, treatment with extracts at concentrations of $10\;{\mu}g/m{\ell}$ and $100\;{\mu}g/m{\ell}$ resulted in increases in tyrosinase gene expression rates of approximately 231% and 256%, respectively, when compared to the control. Moreover, the solvent fraction layers(methylene chloride, ethyl acetate, butyl alcohol, water) improved the expression of the tyrosinase gene, but to a lesser degree than the methanolic extracts. An MTT assay revealed, that the methanolic extract exhibited very low cytotoxicities at $10\;{\mu}g/m{\ell}$ and $100\;{\mu}g/m{\ell}$. Taken together, the results of this study indicated that the methanolic extracts of Scutellaria baicalensis was a very effective positive regulator of tyrosinase gene expression.

In this study, we investigated the antioxidative effects of AY by measuring 1.1-Diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical ($^{\cdot}OH$), and superoxide radical ($O_2{^-}$) scavenging activities in vitro. AY was extracted with ethanol and then partitioned with n-hexane, methylene chloride ($CH_2Cl_2$), ethylacetate (EtOAc) and n-butanol (n-BuOH). In the DPPH radical scavenging assay, AY at concentrations of 10 to $100{\mu}g/mL$ dose-dependently increased inhibition of DPPH oxidation, with the EtOAc fraction of AY showing the highest DPPH radical scavenging activity fractions. The $^{\cdot}OH$ radical scavenging activities of the extract and four fractions of AY increased by over 80% at a concentration of $50{\mu}g/mL$. In particular, the IC50 value of the EtOAc fraction was $0.03{\mu}g/mL$, which was the lowest value among all fractions. We also found that the EtOAc fraction of AY was better at $O_2{^-}$ radical scavenging than other fractions. Taken together, these results suggest that AY, especially the EtOAc fraction, can be used as a natural antioxidant against free radicals.
This study was carried out to discover the skin wrinkle reducing components in the seeds of Arctium lappa. The isolation of a methylene chloride-soluble fraction of 70% ethanol extract from the seeds of Arctium lappa using a procollagen type-1 synthesis and MMP-1 activity resulted in the isolation and identification of four lignin compounds: arctiin, arctigenin, matairesinol, and diarctigenin. All structures were confirmed via NMR and MS spectroscopic data. To determine cell viability and procollagen type-1 synthesis, human dermal fibroblasts were treated with 10-100 ${\mu}M$. As a result, none of the four compounds showed cytotoxicity up to 50 ${\mu}M$. We also investigated their procollagen type-1 synthesis and MMP-1 inhibition activity and found that arctiin had the highest activity in terms of both procollagen synthesis and MMP-1 inhibition among all four compounds. Putting all the data together, we suggest that arctiin be used in cosmetics as an anti-wrinkle material.
