Effect of Graphitized Carbon Supports on Electrochemical Carbon Corrosion in Polymer Electrolyte Membrane Fuel Cells (탄소 담지체의 결정성에 따른 고분자전해질형 연료전지의 내구성 평가 연구)

  • The influence of graphitization of carbon support on the electrochemical corrosion of carbon and sintering of Pt particles are investigated by measuring $CO_2$ emission at a constant potential of 1.4 V for 30 min using on-line mass spectrometry and cyclic voltammogram. In comparison to commercial Pt/C (from Johnson Matthey), highly graphitized carbon nanofiber (CNF) supported Pt catalyst exhibits lower performance degradation and $CO_2$ emission. As the more carbon corrosion occurred, the more prominent changes were detected in electrochemical characteristics of fuel cell. This indicates that the carbon corrosion affects significantly the fuel cell durability. From the observed results, CNF is considered to be more corrosion resistant material as a catalyst support. However, CNF shows higher aggregation of Pt particles under repeated cyclic voltammetry between 0 and 0.8 V where the carbon corrosion is not initiated.

Antibacterial activity from medicinal plant extracts on the Staphylococcus aureus (수종 한약재 추출물의 Staphylococcus aureus에 대한 항균활성)

  • Medicinal plant extracts including Rubus coreanus, Sanguisorba officinalis, Eriobotrya japonica, Prunus mume, Crataegus pinnatifida, Rosa leavaigate Prunus persica, Prunus japonica var. nakaii and Spiraea blumei were prepared for the test of antibacterial activity. Tryptic soy broth (TSB) containing $0{\sim}10mg/ml$ of medicinal plant extracts was inoculated with $10^6$ cells/ml of Staphylococcus aureus and incubated at $35^{\circ}C$ for 24 hours. The plate counting method and clear zone test were used to test inhibitory effect of the extracts. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) was derived from the survival curves of S. aureus. The order of antibacterial activities of medicinal plant extracts on the S. aureus was Rubus coreanus > Sanguisorba officinalis > Eriobotrya japonica > Prunus mume > Crataegus pinnatfida. Minimum inhibitory concentration of Sanguisorba ofEcmalis on the Staphylococcus aureus was 2.5mg/ml and minimum bactericidal concentration of Rubus coreanus was 1.0%. Inhibition zone of Rubus coreanus, Sanguisorba officinalis, Eriobotrya japonica, Prunus mume, and Crataegus pinnatifida was 16.5mm, 14.3mm 11.0mm, 14.0mm and 12.7mm, respectively. The morphology of S. aureus cells treated with medicinal plant extracts showed damage of cell wall and cytoplasmic membrane. Severely damaged cells of S. aureus lost electron dense material and cytoplasm. This result suggests that medicinal plant extracts can be used as an effective natural antibacterial agent in food.

Physiology and Growth Properties of Halophilic Bacteria Isolated from Jeotgal (Salted Seafood) (젓갈(염장발효식품)에서 분리한 호염세균의 생리 및 성장특성)

  • Two species of halophilic bacteria were isolated from five salted seafoods and identified by 16S rDNA sequenc­ing homology. One was identified as Halomonas subglaciescola and other four strains were belong to Halomo­nas marina. The identity of all isolates with standard organisms was above $95\%.$ H. subglaciescola, H. marina IN, and H. marina SH-2 grew in salinity condition from $3%\;to\;18\%$ NaCl but growth of H. marina SQ and H. marina SH-l grew in salinity environment from $8\%\;to\;17\%.$ Maximum biomass of H. subglaciescola, H. marina IN, H. marina SQ, H. marina SH-1, and H. marina SH-2 growing in LB medium containing $15\%$ NaCl were about 3.2, 4.5, 4.5, 5.7, and 4.2, however the maximum biomass in LB medium containing $5\%$ NaCl were about 2.2, 1.1, 0.7, 0.2, and 2.4 as optical density at 660 nm, respectively. In scanning electron micrograph, unknown material (mucus) attached to outer membrane of all isolates was observed. When mucus isolated from halophilic bacterial cell was added to culture of E. coli, E. coli grew in medium containing $15\%$ NaCl.

Ultrastructural Changes on the Secreting Cells of the Prothoracic Gland During the Larva-pupal Molt of Bombyx mori L. (누에나방(Bombyx mori L.) 종령유충일용 전환기(轉換期)의 전흉선(前胸腺) 분필세포(分泌細胞)의 미세구조적 변화)

  • In order to define the morphological changes of the secreting cells of prothoracic gland during larva-pupal molt, ultrastructural observations were carried out using Bombyx mori L. as the experimental material. At first stage of present experiment, 4 day old 5th instar larva, the polyhedral secreting cells were centrally located in the prothoracic gland surrounded by the connective sheath. The secreting cells were attached to the neighboring cells by the prominent desmosomes, and the plasma membrane contacted with connective sheath were highly infolded. In cytoplasm, the most of the cell organelles, such as rod-like mitochondria, rough surfaced endoplasmic reticulum, ribosome were developed. As the stages advance from larva to pupa, general feature of the secreting cells were retained, but structural changes of the various cytoplasmic organelles-ribosome, rough surfaced endoplasmic reticulum, mitochondria, Golgi apparatus, lamellar body, and vesicle-were noted. In the perinuclear cytoplasm of the secreting cells at the stage of 6 day old 5th instar larva, it is peculiar that only a large amount of ribosomes were distributed and the other organelles were retreated from the juxtanuclear region. Just before and after spining cocoon, these features were more remarkable. Rough surfaced endoplasmic reticulum were gradually increased from the stage just before spining cocoon to the pharate pupa. Rod-like mitochondria with irregular cristae and the matrix showing low density were distributed throughout the cytoplasm in the secreting cells of the 4 day old 5th instar larva. Sometimes, longitudinally distended and curved mitochondria were observed. At the stage of pharate pupa, most of mitochondria were deformed. The rod-like mitochondria of the secreting cells of pupal prothoracic gland were narrower than those of 4 day old 5th instar larva, and the electron density of the mitochondrial matrix is increased in pupa. Golgi apparatus were a few in number in both stages, last instar larva and spining cocoon. In stages of the pharate pupa, the Golgi apparatus were frequently observed. Cytoplasmic vesicles were observed for the first time in the secreting cells of one day after spining cocoon, and the number and the size of cytoplasmic vesicles were distinctly increased inpharate pupa and just after pupation. In the secretory cells of the PG, it in concluded that the RER was closely related to syntheting the enzymes seem to produce the ecdysone.

Effect of Type I Collagen on Hydroxyapatite and Tricalcium Phosphate Mixtures in Rat Calvarial Bony Defects

  • To repair bone defects in the oral and maxillofacial field, bone grafts including autografts, allografts, and artificial bone are used in clinical dentistry despite several disadvantages. The purpose of this study was to evaluate new bone formation and healing in rat calvarial bone defects using hydroxyapatite (HA, $Ca_{10}[PO_4]_6[OH]_2,\;Bongros^{(R)}$, Bio@ Co., KOREA) and tricalcium phosphate (${\beta}-TCP,\;Ca_3[PO_4]_2$, Sigma-Aldrich Co., USA) mixed at various ratios. Additionally, this study evaluated the effects of type I collagen (Rat tail, BD Biosciences Co., Sweden) as a basement membrane organic matrix. A total of twenty, 8-week-old, male Sprague-Dawley rats, weighing 250-300g, were divided equally into a control group (n=2) and nine experimental groups (n=2, each). Bilateral, standardized transosseous circular calvarial defects, 5.0 mm in diameter, were created. In each experimental group, the defect was filled with HA and TCP at a ratio of 100:0, 80:20, 70:30, 60:40, 50:50, 40:60, 30:70, 20:80, and 0:100 with or without type I collagen. Rats were sacrificed 4 and 8 weeks post-operation for radiographic (standardized plain film, Kodak Co., USA), histomorphologic (H&E [Hematoxylin and Eosin], MT [Masson Trichrome]), immunohistochemical staining (for BMP-2, -4, VEGF, and vWF), and elementary analysis (Atomic absorption spectrophotometer, Perkin Elmer AAnalyst $100^{(R)}$). As the HA proportion increased, denser radiopacity was seen in most groups at 4 and 8 weeks. In general radiopacity in type I collagen groups was greater than the non-collagen groups, especially in the 100% HA group at 8 weeks. No new bone formation was seen in calvarial defects in any group at 4 weeks. Bridging bone formation from the defect margin was marked at 8 weeks in most type I collagen groups. Although immunohistochemical findings with BMP-2, -4, and VEGF were not significantly different, marked vWF immunoreactivity was present. vWF staining was especially strong in endothelial cells in newly formed bone margins in the 100:0, 80:20, and 70:30 ratio type I collagen groups at 8 weeks. The calcium compositions from the elementary analysis were not statistically significant. Many types of artificial bone have been used as bone graft materials, but most of them can only be applied as an inorganic material. This study confirmed improved bony regeneration by adding organic type I collagen to inorganic HA and TCP mixtures. Therefore, these new artificial bone graft materials, which are under strict storage and distribution systems, will be suggested to be available to clinical dentistry demands.

  • A TSL (Thin Spray-on Liner) has a higher initial strength and faster construction time than conventional cementitious shotcrete. Because of its high adhesion and tensile strength, the TSL reinforced concrete show a characteristic like composite materials. In this study, to consider an application to the conventional design method, ASD (allowable stress design), numerical study was used. In the numerical analysis, material and contact properties were adopt from previous studies. Then a thickness of concrete in the tunnel was evaluated with the TSL reinforced case by the ASD concept. In other words, bending compressive stress, bending tensile stress and shearing force of the concrete were considered to determine a thickness of concrete lining by the given boundary conditions. From the numerical analysis, there was no tendency to show by the ASD because the ASD is based on the elastic theory while the TSL typically contributes to reinforcement after yielding.

  • The ultrastructure of the integumental epidermis of Korean planaria (Dugesia japonica) is studied by light microscope, scanning electron microscope and transmission electron microscope. The planaria has mono-layered integumental epidermis in which most of cells exhibit irregularly columnar shape. The epidermal cells of the integument are classified into six types on the basis of cytochemical and ultrastructural characteristics. 1) Ciliated epithelial cells: These cells have cilia in their free surfaces. The axonemes of cilia exhibits fundamental 9+2 microtubular pattern. 2) Eosinophilic cells: These cells contain a few large eosinophilic granules. The core of eosinophilic granule is consisted of sparsely dispersed fibrillar structures in relatively electron-lucent ground material. 3) Mucous cells: These cells are filled with irregularly shaped, PAS-positive mucous granules which have an average size of $0.8\\times0.3 \\muM$. 4) Rhabdite-forming cells: These cells possess a few strongly-eosinophilic large rhabdite granules. The rhabdite granules are synthesized either in the rhabdite-forming cells which constitute integumental epidermis or in the corresponding cells which are developed in the parenchyma and later transferred to epidermal cells of integumental epidermis through basement membrane. 5) A-type of basophilic granule cells: These granule cells possess round or irregularly-shaped granules which are strongly stained with Alcian blue. These electron-dense granules have an average size of $1.5\\times1.0 \\muM$. This type of cells is derived from parenchymal tissue. 6) B-type of basophilic granule cells: These basophilic granule cells with PAS-positive granules, are found in the epidermis of lateral body wall. The granules, which are about $0.7\\times0.4 \\muM$ in size, occupying most part of this cell type are originated from the parenchyma.

Isolation and Partial Physicochemical Characterization of Bile Acid-Binding Fraction from Rice Bran Protein Hydrolysates (미강 단백질 가수분해물에서 담즙산 결합 획분의 분리 및 특성구명)

  • Rice bran protein hydrolysates were prepared and some of their physicochemical properties were investigated to utilize rice bran as starting material for functional food ingredient. Rice bran proteins (RBP) were prepared from defatted rice bran by alkaline extraction and isoelectric precipitation. The enzyme for hydrolysis of RBP was selected through measuring relative activity by pH-drop method and comparing the degree of hydrolysis (DH) of hydrolysates. The enzymatic hydrolysates prepared by $Esperase^{\circledR}$ treatment were partitioned into two fractions by ultrafiltration(UF) with a 10 kDa molecular weight cut-off membrane. Each fraction was applied to a cholic acid-conjugated ${\omega}-aminohexyl$ Sepharose 4B column and the bile acid-binding components were obtained by eluting with deoxycholate. Gel permeation chromatography on a Sephadex G-50 column revealed that molecular weight of the bile acid-binding fraction of UF permeate was distributed in ranges of $2\;kDa{\sim}10\;kDa$ and $0.2\;kDa{\sim}0.6\;kDa$. Three peaks (R-1, R-2 and R-3) were obtained by prep-HPLC of bile acid-binding fraction of UF retentate and analyzed for total and free amino acid composition. The results showed that proline content of the bile-acid binding polypeptides and peptides was four times as much as that of rice bran protein and that the peak corresponding to higher average hydrophobicity had a higher free amino acid content. Average hydrophobicity slightly increased with enzymatic hydrolysis.

Extraction and verification of highly immunologenic antigen for diagnosis of bovine brucellosis (소 브루셀라병 진단용 고면역원성 항원의 추출과 검증)

  • Bovine brucellosis, an important zoonosis, is diagnosed with serological tests such as the RBT, TAT using inactivated whole bacterial cells or bacterial lipopolysaccharide (LPS) antigen in Korea. However, a strong cross-reaction between Brucella spp. and Yersinia enterocolitica O9 in these tests has seriously complicated the diagnosis of animal brucellosis because Brucella spp. shares common antigenic determinants with Y. enterocolitica O9 in the smooth LPS region. In this study, Brucella-field strains were isolated from Brucellapositive Hanwoo in Kimhae, Korea and outer membrane protein (omp) which has low cross-reaction with Y. enterocolitica O9 and high immunogenicity was extracted from the field strains Then we compared ELISA using the extract with RBT-TAT. Fifteen field strains were isolated from 47 supra-mammary-lymph nodes, which were collected from 18 farms. Isolation rate was 32%. Brucella-specific antigen was identified by performing SDS-PAGE or Western blotting on extracted omp with at 0.5% n-lauroylsarcosine One hundred and ninety-two serum-samples were used in the experiment: 142 negative and 50 positive samples verified by RBT-TAT. According to ELISA results, 127 samples were negative and 15 appeared positive among 142 negatives by RBT-TAT, while 42 samples were positive and 8 were negative among 50 positives by RBT-TAT. Therefore, it showed 89.4% of specificity and 84% of sensi-tivity. Through the current experiments, we could set up an ELISA based on the omp which has low cross-reaction and high immunogenicity and concluded that the omp could be a good material for accurate diagnosis of bovine brucellosis.

An Experimental Study and Value Analysis for Performance Assessment of the Embo-thane Membrane Waterproofing Method (엘보탄도막방수공법의 성능평가를 위한실험적 연구 및 VE분석)

  • Since 1970s, urethane waterproofing method is broadly used in rooftops, underground spaces, and sports stadium for its outstanding ultraviolet blockage, watertightness, and elasticity. However, development of slippage-resistance and endurance, improvement of function considering convenience and visually pleasing of users, urethane waterproofing method is necessary, since rooftops and underground spaces have slippage and external force risks. Therefore, many improved waterproofing methods are being developed and, recently, embo-thane waterproofing method, which applies embo-spray coating system, has been developed. This paper explains exposure, nonexposure, and floor-material of embo-thane waterproofing method, and then perform experimental study for comparison with urethane waterproofing method about tensile strength, coefficient of expansion, performance of bond, anti-abrasion, and slippage-resistance. In addition, the performance index was presented for the superiority of embo-thane waterproofing method compared to urethane by setting up evaluation criteria considering not only physical performance but also design side of embo-thane waterproofing method, and Value Analysis applying AHP. Also for an assessment considering uncertain result, Monte Carlo Simulation Method was used to operate reliability analysis through statistic approach method.
