Bacillus sp. LK-1의 Exopolysaccharides(POL-II) 생산 및 특성 (The Production and Properties of Exopolysaccharides(P0L-11) by Bacillus sp. LK-1)

  • The strain which produced highly viscous exopolysaccharides (EPS) in liquid culture was selected from soil. The strain was supposed to Bacillus sp. from the results of mophological, biochemical and physiological tests. The medium composition for EPS production was trypton 0.75%, sucrose 4%, CaCO3 0.01%, Winogradsky's nitrogen free mineral medium 5ml/l and pH 7.0. In 2-l jar fernenter, the viscosity of culture broth after 120-hr cultivation time was very high (60, 000 cps) and the amount of EPS was 6.2g/l.

Sputtering of Multifunctional AlN Passivation Layer for Thermal Inkjet Printhead

  • The aluminum nitride films were prepared by RF magnetron sputtering using an AlN ceramic target. The crystallinity, grain size, Al-N bonding and thermal conductivity were investigated in dependence on the plasma power densities (4.93, 7.40, 9.87 W/$cm^2$) during sputtering. High thermal conductivity is important properties of A1N passivation layer for functioning properly in thermal inkjet printhead. The crytallinity, grain size, Al-N bonding formation and chemical composition were observed using X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM), fourier transform infrared (FTIR) and X-ray photoelectron spectroscopy (XPS), respectively. The AlN thin film was changed from amorphous to crystalline as the power density was increased, and the largest grain size appeared at medium power density. The near stoichiometry Al-N bonding ratio was acquired at medium power density. So, we know that the AlN thin film had better thermal conductivity with crystalline phase and near stoichometry Al-N bonding ratio at 7.40 W/$cm^2$ power density.

  • Putrescine, spermidine, and spermine were found to persent in Methylobacterium extorquens AM1 growing on methanol, succinate, glucose, or nutrient broth as an energy source. Spermidine was found to be a major polyamine in cells growing on methanol or succinate, while putrescine to be the one in nutrient broth-grown cells. The overall content of polyamines in cells growing on glucose was less than that in cells growing on other substrates. Spermine was the most abundant polyamine in glucose-grown cells. Accumulation of polyamines in M. extorquens AM1 was maximal at the mid-exponential or early stationary phase during growth on each substrate. The effect of polyamines added into the medium on the polyamine composition in M. extorquens AM1 was variable. Each polyamine added into the nutrient broth medium was found to increase the amount of the respective polyamine in the cell. Exogeneously added polyamines had no effect on the growth of M. extorquens AM1.

  • The purpose of this study was to develop the new microbial bioflocculant available in a food and fer-mentation industal. This study was reported the results of the composition for optimum culture medium and elemental characteristic of crude purification bioflocculant following the previous report(I). The maximum production of the flocculant from Bacillus megaterium was observated in the culture medium containing 2% sucrose 0.3% NaNO3 0.01% tryptone 0.01% beef extract 0.05% MgSO4 ·7 H2O 0.005% CaCO3 Addition of the sucrose as carbon sources and inorganic salt such as MgSO4, CaCO3 significantly increased the production of flocculant more than nitrogen sources. In the result of color reaction of the crude purified bioflocculant it was investgated that anthrone was positive and benedict burette and nin-hydrin was negative. These result were indicated that the flocculant produced from Bacillus megaterium was a kind of exopolysaccharide.

  • To utilize hemicellulosic biomass efficiently, the microorganism producing xylanase was isolated from fermented sawdust. It was a Gram positive, aerobic and rod shape bacterium. It had endospore and secreted strong hydrolases, such as amylase and protease. Through morphological, cultural and physiological tests, it was identified as Bacillus sp. GS. To increase the productivity of xylanase from Bacillus sp. GS, the enzyme production medium was optimized. The composition of the medium and incubation conditions were like follows xylan 1.25%, yeast extract 0.1%, NaN030.2%, K2HP04 0.1%, MgSO4 0.02%, mineral salt 0.005%, pH 6.5, incubation temperature 37$^{\circ}C$.

Effect of Food Waste Compost on the Antler-Type Fruiting Body Yield of Ganoderma lucidum

  • The effects of the composition of a mixture containing food waste compost (FWC), rice bran (RB), and oak sawdust (SD) on the antler-type fruiting body (FB) yield of Ganoderma lucidum were studied. Experiments were performed using 0 (control), 5, 10, 15, 20, 25, 30, 35, and 40% (w/w) FWC added to a basal growth medium consisting of 20% (w/w) RB and 80% (w/w) SD. The content of 15% FWC gave the highest FB yield ($27.0{\pm}1.3$ g/bottle), which was 44% higher than the yield ($18.6{\pm}2.8$ g/bottle) of the control treatment. However, FWC contents of 20~40% showed reduced yield (2.4~23.0 g/bottle), partly because FWC had a high Na concentration (0.6%). These results demonstrate the potential for use of FWC as a component of a growth medium for production of G. lucidum FBs.

  • Streptococcus lactis ATCC 11454 의 protoplast 생성 및 재생에 관하여 조사하였다. Protoplast의 생성은 용균효소로써 리소짐 단독처리 만으로 충분히 가능하였으며 균체의 배양은 20mM DL-트레오닌을 첨가한 배지를 사용하는 것이 좋았으며 생육시기와 리소짐 농도등이 중요한 영양인자였다. 배지조성의 최적화 및 protoplast를 배지에 접종하는 방법을 변형시킴으로써 약 20%정도의 재생효율을 얻을 수 있었다.

절화백합의 상자 재배시 몇가지 배양토 조성이 생육과 줄기경도에 미치는 영향 (Stem Firmness and Flowering Response of Cut Lilies as Influenced by Medium Composition in Box Culture)

  • 초장과 화경장은 'Casa Blanca'의 경우 Pt:Rrh:Ve(1:1:1, v.v$^{-1}$) 처리구에서 증가되었고, 화경의 낮은 부위의 고사엽수는 Rpt:Pe:Rrh(1:1:1, v.v$^{-1}$) 처리구에서 감소되었다. 'Marco Polo'의 초장과 화경장은 Pt:Rrh(1:1, v.v$^{-1}$) 처리구에서 대조구에 비해 증가되었고, 고사엽수는 Rpt:Pe;Rrh(1:1:1, v.v$^{-1}$) 처리구에서 다른 처리구에 비해 증가되었다. 개화는 'Casa Blanca'의 경우 Pt:Pe:Ve(1:1:1, v.v$^{-1}$) 처리구에서 'Marco Polo'의 경우는 Pt:Rrh:Ve(1:1:1, v.v$^{-1}$)의 처리구에서 상당히 촉진되었다. 꽃의 길이는 'Casa Blanca'의 경우 'Rpt:Pe:Rrh(1:1:1, v.v$^{-1}$)'처리구에서, 'Marco Polo'의 경우는 Pt;Rrh(1:1, v.v$^{-1}$)처리구에서 대조구에 비해 증가되었다. 꽃의 퇴화는 두 품종 모두 Pt 단용처리시 다른 처리구에 비해 증가되었다. 화경의 경도는 'Casa Blanca'의 경우는 Pt:Pe;Ve(1:1:1, v.v$^{-1}$) 처리구에서 특히, 'Marco Polo'의 화경상단부의 경도는 Pt:Rrh(1:1, v.v$^{-1}$) 처리구에서 대조구에 비해 매우 증가되었다. 그러나 일반적으로 'Casa Blanca'의 화경 경도는 재배 배양토에 의해 크게 영향을 받지 않았다.

Microalgal Removal of $CO_2$from Flue Gases: Changes in Medium pH and Flue Gas Composition Do Not Appear to Affect the Photochemical Yield of Microalgal Cultures

  • Our research objectives are to determine under what conditions microalgal-based $CO_2$capture from flue gases is economically attractive. Specifically, our objective here was to select microalgae that are temperature, pH and flue gas tolerant. Microalgae were grown under five different temperatures, three different pH and five different flue gas mixtures besides 100% $CO_2$(gas concentrations that the cells were exposed to ranged 5.7-100% $CO_2$, 0-3504ppm SO$_2$, 0-328ppm NO, and 0-126ppm NO$_2$). Our results indicate that the microalgal strains tested exhibit a substantial ability to withstand a wide range of temperature (54 strains tested), pH (20 strains tested) and flue gas composition (24 strains tested) likely to be encountered in cultures used for carbon sequestration from smoke stack gases. Our results indicate that microalgal photosynthesis is a limited but viable strategy for $CO_2$capture from flue gases produced by stationary combustion sources.

불화 유리의 냉각 속도 효과 (Cooling Speed Effects of Fluoride Glasses)

  • The properties of the glasses are not dominantly dependent on the chemical composition, temperature, and pressure but also on the thermal history. For example, electrical, thermal, optical, and mechanical properties are all known to be strongly dependent on the thermal history. Fluoride glasses have received a great deal of attention as candidate materials for an infra-red transmitting medium. A series of fluoride glasses and fibers were prepared under a nitrogen atmosphere. Thermal history effects of the fluoride glass fibers associated with the fast cooling rate employed during the fiber drawing process were discussed in terms of the glass temperatures and the fictive temperatures on the basis of the results obtained from the Differential Scanning Calorimeter (DSC) measurements of the fiber and bulk forms of the same chemical composition.
