Effect of Soil Mixture on the Growth of Chrysanthemum zawadskii var. latilobum KITAMURA (분화용 배양토가 구절초의 생육에 미치는 영향)

  • This study was conducted to investigate the combination of proper culture medium for differentiation culture of Chrysanthemum zawadskii var. latilobum KITAMURA. The combinations of culture medium were consisted with an inorganic materials such as vermiculite, perlite, saprolite, sand and upland top soil, and with an organic materials such as peat-moss, leaf mold and compost. For the plant growth characteristics, the plant height, number of tiller, root length and root weight in the combination of leaf mold was relatively great as compared to the treatment with peat-moss and compost. It was considered that humus might be contributed to improve the physical properties of soil as well as the sequential nutrient supply. For the combination of peat-moss, the plant growth was not good because of not sufficient nutrient supply, Also, the plant growth in the treatment of compost was so bad because of enhancement of culture medium pH 8.9 and increasement of phosphorous content.

  • Optimum culture conditions for high frequency plant regeneration from leaf explants of Kalanchoe daigremontiana Hamet &Perrier were established. Shoot regeneration was achieved from leaf explant cultures using MS medium supplemented with indole-3-acetic acid (IAA) and thidiazuron (TDZ) or benzyladenine (BA). Percent regeneration was influenced by plant growth regulators and source of explants. MS medium supplemented with TDZ (1.0 mg/l) and IAA (0.4 mg/l) was the most effective, providing shoot regeneration for 76.7 % of ex vitro leaf explants associated with a high number of shoots per explant (9.5 mean shoots per explant), whereas 100% shoot regeneration associated with 12.4 shoots per explant occurred from in vitro leaf explants on the same medium. Clusters of shoots were multiplied and elongated on MS medium containing several concentrations of BA. MS medium supplemented with 0.25 mg/l BA was proved as the most effective shoot elongation medium. Elongated shoots (2-3 cm) were rooted at 100% on half-strength MS medium. Rooted plantlets were then transferred to potting soil. Regenerated plants were established in the soil with 90% success.

  • This study was carried out to investigate combination dyeing of the cationized cotton with natural colorants extracted from Gardenia and Sappan wood extract using methanol. To improve dyeing properties of cotton with natural dye, cotton fabric was preheated with cationizing agent containing chlorohydrine group in aqueous solution of sodium hydroxide. On the whole the various medium colors were developed by repeat dyeing with different colorants after dyeing with one colorant in case of natural dyeing, In this study, however in order to obtain various colors on cationized cotton fabrics with natural dyes, pre-mordanting and combination dyeing were carried out. The various colors were obtained according to various metal compounds for pre-mordants and various mixing portion of Gardenia and Sappan wood. Metal compounds containing alum, copper and iron were used for pre-mordants.

  • Explants from stem, leaf and root tissue of Cissus juttae were plased on Ms medium supplemented ifferent combination of 2.4-D, NAA, Kinetin and BA in a actorial experiment. some callus formed in every treatment, but the best callus growth was on media containing $1.0{\;}mg/{\ell}$ NAA and $1.0{\;}mg/{\ell}$ BA. Formation of advantitious emboyos was evident in callus to MS medium supplemented with $0.1{\;}mg/{\ell}$.

Medium Concentration Influencing Growth of the Entomopathogenic Nematode Heterorhabditis bacteriophora and its Symbiotic Bacterium Photorhabdus luminescens

  • The biological control potential of entomopathogenic nematodes (EPN) can be enhanced by improved culture efficiency. Optimization of the media is a key factor for improving in vitro mass production of entomopathogenic nematodes. This study reports the effect of medium concentration. The medium is a combination of carbohydrates, lipids, proteins, sats, and growth factors, on the growth of Heterorhabditis bacteriophora and its symbiotic bacterium Photorhabdus liminescens. The overall optimal medium concentration for nematode recovery, hermaphrodite size, bacterial mass, infective juveniles (IJs) yield, and doubling time was 84 g/l. At this concentration rate, the doubling time of IJs production and the biomass of symbiotic bacteria was 1.6 days and 12.8 g/l, respectively. The maximum yield of $2.4{\times}{10^5}IJs/ml$ was attained within a one-generation cycle (eight days). The yield coefficient was $2.8{\times}{10^6}$ IJs/g medium, and the maximum productivity was $3.1{\times}{10^7}$ IJs per day. Medium concentration affected two independent factors, recovery and hermaphrodite size, which in turn influenced the final yield.

  • Plant regeneration from the stem tissue of Houttuynia cordata Thunberg was investigated. The medium supplemented with the combination of 2, 4-D 1 mg/L and kinetin 0. 5 mg/L was the most effective for the embryogenic callus formation. The internode segment produced more callus formation than the leaf segment. ${\frac{1}{2}}\;MS$ medium was the most effective for the embryogenic callus formation. The medium supplemented with the 1% activated charcoal produced the whole plant directly without the callus formation from the nodes. The medium supplemented with the combination of NAA 0. 2 mg/L and BA 1 mg/L was the most effective for the plant regeneration from the embryogenic callus.

Effect of carbon sources on somatic embryogenesis and cotyledon number variations in carrot (Daucus carota L.)

  • In order to investigate the effect of carbon sources on somatic embryogenesis and cotyledon number variations in carrot, embryogenic callus were cultured in the medium supplemented with various concentrations of sucroseor glucose, and with combination of 2% sucrose and various concentrations of mannitol or sorbitol. The maximum number of somatic embryos formed per flask (1,555.70) was obtained in the medium supplemented with 2% sucrose rather than glucose alone or a combination of mannitol or sorbitol and 2% sucrose, and the number of somatic embryos was decreased with the increasing of mannitol or sorbitol concentration. The frequencies of somatic embryos with two cotyledons were 35.14% for sucrose, 19.88% for glucose, 32.55% for mannitol + 2% sucrose, and 38.59% for sorbitol + 2% sucrose, respectively, and the frequencies of abnormal somatic embryos having 3 or more cotyledons were 64.86% for sucrose, 80.12% for glucose, 67.44% for mannitol + 2% sucrose, and 61.41% for sorbitol + 2% sucrose, respectively. Particularly, the frequency of somatic embryos with two cotyledons (59.16%) was the highest in the 2% sucrose medium compared to the frequency of abnormal somatic embryogenesis with three or more cotyledons, and the frequency gradually decreased with increasing concentration of glucose, mannitol or sorbitol. According to these results, it was found that the ratio of abnormal somatic embryo was higher than the normal somatic embryo in carrot, and was shown that somatic embryogenesis and the cotyledon number was affected by the concentrations of sucrose, glucose as carbon source, and mannitol and sorbitol as osmotic agents in culture medium.

  • A serum-free medium that could be used for the large-scale culture of mouse hybridoma to produce monoclonal antibodies was developed. The medium was based on a 1:1 mixture of Iscove's Modified Dulbecco's Medium and Ham's F-12, supplemented with insulin 10$\mu\textrm{g}$/$m\ell$, transferrin 10$\mu\textrm{g}$/$m\ell$, ethanolamine 10$\mu$M and selenium 30nM (designated EBM (enriched basal medium) with the supplements). The effect of various supplements of steroid hormones, vitamins, lipid and mineral salts was investigated and their optimal concentration was determined to replace fetal calf serum (PCS). These components were added respectively and then added by way of two or three combination to discern of which component combination was effective to the culture of hybridoma. As a result, serum-free medium KM3 (EBM with BSA 100$\mu\textrm{g}$/$m\ell$, mineral cocktail and 0.05% PEG) was deter-mined. The hybridoma Alps 25-3 cultured in this medium showed almost the same growth rate as in medium added with 2% fetal bovine serum. However, the antibody concentration from KM3 cultures was 80% of that obtained from culture with FCS. KM3 was also examined for the culture of other mouse hybridomas, KW, A4W & HCGK, and it was confirmed that it could support the growth of these hybridomas and the production of monoclonal antibodies.

  • A quantitative fit test, condensation nuclei counting (Portacount 8025, TSI), was performed concurrently with a banana oil (isoamyl acetate: IAA) qualitative fit test (MSA) to evaluate reliability on IAA QLFT and correlation between two methods. One brands of quarter mask (3M model 7500 medium) was prepared for QLFT with HEPA filter and gas & vapor removing media, i.e., combination cartridge. 110 subjects (65 male, 45 female) were fit tested QNFT and QLFT each three times. For a wearer combination having a FF<10, as determined by CNC QNFT, the point es timate (${\beta}$-error) of the probability of that combination not being rejected by the banana oil QLFT was found to be 0.0 with 95% confidence that this statistic is not expected to exceed 0.15. For a wearer combination having a FF<100, as determined by CNC QNFT, the point estimate of the probability of that combination not being rejected by the banana oil QLFT was found to be 0.07 with 95% confidence that this statistic is not expected to exceed 0.13. The uncertainty associated with each estimate, however, is large due to the small number of study subjects with inadequately fitting respirators.

Effects of Medium , Plant Growth Regulators , and Explant Sources on Plant Regeneration of Rehmannia glutinosa (배지, 생장조절물질 및 치상조직이 지황 체세포조직으로부터 식물체분화에 미치는 영향)

  • When the leaf and stem tissues of Rehmannia glutinosa were cultured on MS medium with plant growth regulators, shoots were regenerated on MS medium with TDZ(0.01 to $2.0{\mu}M$), and root initiation was better on MS medium treated with NAA(0.01 to 2.0mg/$\iota$). On $B_5$ medium, shoot regeneration was better on medium with TDZ than those with Quincrac, NAA, and 2.4-D. The addition of Quincrac, NAA, and 2.4-D inhibited shoot regeneration on MS medium, but promoted shoot regeneration on $B_5$ medium. Shoot regeneration and growth was better on MS medium with the combination treatments of 2.4-D 0.01mg/$\iota$ and TDZ 0.01, 0.1 and $2{\mu}M$ compared with other treatments. Also shoot regeneration and growth on B_5 medium showed the similar results to that on MS medium.
