Synthesis and Antiviral Activity of Novel Exomethylene Cyclopropyl Pyrimidine Nucleosides

  • A series of novel exomethylene cyclopropyl nucleosides have been synthesized starting from Feist's acid. Classical nucleophilic substitution conditions ($K_2CO_3$, 18-crown-6) of the tosylate 2 as well as Mitsunobu reaction (DEAD, $PPh_3$) of alcohol 1 with pyrimidine bases afforded a series of novel cyclopropyl nucleosides. Compound 4b displayed moderate anti-HBV activity without any cytotoxicity up to $100{\;}{\mu}M$.

Inhibitory Effects of Methanol Extracts from Korean Medicinal Plants against HIV-1 Protease Activity

  • Korean medicinal plants were screened for their inhibitory activity against HIV-1 protease. The inhibitory activity of protease was determined by incubating the extracts in reaction mixtures containing protease and substrate $His-Lys-Ala-Arg-Val-Leu-(p-NO_{2}-Phe)-Glu-Ala-Nle-Ser-NH_{2}$ to perform proteolytic cleavage reactions. In this study the twenty six extracts from medicinal plants were investigated. Of the extracts tested, the extracts from the stem of Morus alba. exhibited the strongest activity with inhibition of 81% at a concentration of $100{\mu}g/ml$. The extracts of the flower of Saxjfraga stolonifera, and stems of Euonymus japonica and Castanea crenata showed appreciable inhibitory activity (>50%) against HIV-1 protease at same concentration.

독성 한약재의 법적 규제에 관한 연구 (Study on The Regulation on Poisonous Medicinal Herbs)

  • Objective: Poisonous Medicinal herbs can be considered as a risk factor to public health unless they are prescribed by Doctor of traditional Korean medicine. The proper method to manage them should be prepared to prevent risk factors caused by misuse and abuse of the poisonous medicinal herbs and enhance public health. Methods: In this dissertation (paper), the definition, scope, management status, data about pharmacology and toxicity and media release regarding adverse reaction were understood after organizing documents, laws and regulations concerning poisonous medicinal herbs. Also, management methods are suggested by analyzing related examples and regulations in China, Japan and Hong Kong, where the use of herbal medicine is general. Results: Methods for items for poisonous medicinal herbs, safety information management, management based on standardization of traditional processing methodology and reorganization and revision of related laws and regulations are established. Conclusion: Proper laws and regulations are not yet established to manage poisonous medicinal herbs in Korea. In this regard, it is urgent to establish laws and regulations which can apply independently. The purpose of the laws and regulations should be to enhance management of poisonous medicinal herbs and prevent incidence of addiction and death, improving the public health.

  • Zirconium(IV) chloride is found to be an efficient catalyst for the electrophilic addition reaction of indole with aldehydes/ketones to afford the corresponding bis(indolyl methanes in good yields. The remarkable features of this new procedure are high conversions, shorter reaction times, cleaner reaction profiles and simple experimental and work-up procedures.

Development of a single-nucleotide-polymorphism marker for specific authentication of Korean ginseng (Panax ginseng Meyer) new cultivar "G-1"

  • Background: Korean ginseng (Panax ginseng) is a well-known medicinal plant of Oriental medicine that is still in practice today. Until now, a total of 11 Korean ginseng cultivars with unique features to Korean ginseng have been developed based on the pure-line-selection method. Among them, a new cultivar namely G-1 with different agricultural traits related to yield and content of ginsenosides, was developed in 2012. Methods: The aim of this study was to distinguish the new ginseng cultivar G-1 by identifying the unique single-nucleotide polymorphism (SNP) at its 45S ribosomal DNA and Panax quinquefolius region than other Korean ginseng cultivars using multiplex amplification-refractory mutation system-polymerase chain reaction (ARMS-PCR). Results: A SNP at position of 45S ribosomal DNA region between G-1, P. quinquefolius, and the other Korean ginseng cultivars was identified. By designing modified allele-specific primers based on this site, we could specifically identified G-1 and P. quinquefolius via multiplex PCR. The unique primer for the SNP yielded an amplicon of size 449 bp in G-1 cultivar and P. quinquefolius. This study presents an effective method for the genetic identification of the G-1 cultivar and P. quinquefolius. Conclusion: The results from our study shows that this SNP-based approach to identify the G-1 cultivar will be a good way to distinguish accurately the G-1 cultivar and P. quinquefolius from other Korean ginseng cultivars using a SNP at 45S ribosomal DNA region.

  • Reaction of 2-formyl-4H-thieno [2, 3-b] benzothiopyran-4-one (1) with different heterocyclic amines afforded the corresponding Schiff's bases (2-4). Diethyl malonate, ethyl cyanoacetate and malononitrile were reached with 1 to afford compounds 5, 7 and 8, respectively. Compound 5 was cyclized to the pyrazolidin-3, 4-dione (6) by the action of hydrazine hydrate, whereas compound 7 was utilized for the synthesis of the thiazolin-4 one derivatives (9-13).

Microarray Analysis of Gene Expression by Ginseng Water Extracts in a Mouse Adrenal Cortex after Immobilization Stress

  • To investigate the effects of repeated immobilization-stress challenge on the the hypothalamic-pituitary-adrenal axis, the genomic transcriptome in the adrenal cortex of immobilization-stressed mouse was analyzed by using a cDNA microarray. Mice were subjected to immobilization stress for 2 h per day for 5 consecutive d. With a 4.0-fold cutoff of arbitrary criteria, the expression levels of 168 out of 41,174 genes were significantly modulated in the adrenal cortex by stress when comparing the control and experimental groups. These genes were related to apoptosis, cell cycle, immune response, inflammatory responses, and signal transduction, and thus may be used as potential targets for the development of therapeutics for chronic stress or depression. Six significant genes among these were selected for real time polymerase chain reaction analysis to confirm the change of their expression levels. The gene for phospho 1 was also further investigated because its expression showed the greatest fold-change.

시비처리에 따른 상동나무 용기묘의 생장 및 생리특성 (Growth and Physiological Characteristics of Containerized Seedlings of Sageretia thea at Different Fertilization Treatments)

  • 본 연구는 다양한 효능·효과가 최근에 입증된 상동나무를 대상으로 다목적 수용성 복합비료를 농도(0.5 g·L-1, 1.0 g·L-1, 1.5 g·L-1, 2.0 g·L-1)처리에 따른 생장특성(간장, 근원경 및 H/D율)과 생리특성(광합성, 엽록소 함량 및 형광반응)을 정량화하여 적정 시비 수준을 알아보고자 실시하였다. 간장생장은 시비농도가 1.5 g·L-1까지 높아짐에 따라 생장량이 증가했으나 2.0g·L-1 시비처리에서는 낮아졌다. 근원경 생장은 시비농도가 높아짐에 따라 생장량은 유의하게 낮아졌다. 광합성 반응은 1.5 g·L-1 시비처리에서 엽육세포 내 CO2 반응곡선, 최대광합성효율, 최대 카르복실화 속도가 가장 높았다. 엽록소 형광반응과 엽록소 지수는 무처리구에 비해 시비처리구에서 유의적으로 광합성 기구의 효율을 높이고 있음을 확인하였다. 따라서 우량한 묘목품질의 상동나무를 생산하기 위한 시비 수준은 1.0~1.5 g·L-1 농도라고 판단된다.

Molecular identification of oriental medicinal plant Schizonepeta tenuifolia bunge (Hyung-Gae) by multiplex PCR

  • Schizonepeta tenuifolia (Korean name "Hyung- Gae") is an oriental medicinal plant that is widely used in Korea, China and Japan. S. tenuifolia (Hyung-Gae) has many pharmacological activities and is mostly used for many medicinal preparations. The dried aerial part (spikes and stems) of three oriental medicinal plants, S. tenuifolia (Hyung-Gae), Agastache rugosa (Kwhak-Hyang) and Elsholtzia ciliata (Hyang-Yoo) belonging to the same family, mint family Labiaceae, have such similar shape and smell that it is difficult to differentiate between them. The trnL-F regions of chloroplast DNA of the three medicinal plants were sequenced and used as targets in multiplex PCR reaction to identify S. tenuifolia. After alignment of trnL-F sequences of the authenticated plant samples, one single nucleotide polymorphism (SNP) specific to S. tenuifolia was found. Based on this SNP, a new primer was designed that specifically amplifies the trnL-F region of S. tenuifolia. The established multiplex-PCR was proven to be effective in the differentiation of commercial S. tenuifolia samples from A. rugosa and E. ciliata. This rapid and accurate molecular method is highly promising for use in the food industry.

한약조제시(韓藥調劑時) 예상(豫想)되는 화학반응(化學反應) (Theoretical Chemical Reaction for Herb Medicine)

  • Objective : This study was designed to show the possible functional groups from the herb medicine in boiling water. Results : The results are summarized as follows: 1. the new functional groups can be synthesized in water solution for herb medicine. 2. The boiling water solution may change the poison materials into harmless materials. 3. The multiplication, the offset, the contradiction, etc. in terms of mixed herb medicine can be explainable by these reactions. 4. After finding the new medicinal substances for the specific disease, we can synthesize, modify, and mass produce those for that disease.
