Occurrence of Meloidogyne incognita Infecting Resistant Cultivars and Development of an Efficient Screening Method for Resistant Tomato to the Mi-virulent Nematode (뿌리혹선충 저항성 토마토를 감염하는 Meloidogyne incognita의 발생 및 이 선충을 이용한 효율적인 저항성 검정법 확립)

  • Root-knot symptoms were found on a commercial tomato cultivar carrying Mi, a resistance gene to root-knot nematodes including Meloidogyne incognita, M. arenaria, and M. javanica in 2012 at Buyeo, Chungnam Province in Korea. The isolate was identified as M. incognita based on molecular analyses using two species-specific primer sets. Pathogenicity of the isolate on one susceptible and three resistant tomato cultivars to the root-knot nematodes was tested. The nematode isolate showed strong pathogenicity on all the tested cultivars at all tested incubation temperatures. In addition, resistance degree of 33 commercial tomato cultivars, 8 susceptible and 25 resistant cultivars to root-knot nematodes, was also tested. Plants were determined as resistant when they suppressed the nematode reproduction. All the cultivars demonstrated strong susceptibility to the nematode regardless of resistance of the tomato cultivars. To our knowledge, this is the first report on the occurrence of Mi infecting M. incognita isolate in Korea. On the other hand, to construct an efficient screening method for selecting resistant breeding source to the nematode isolate, root-knot development of M. incognita on four tomato cultivars according to several conditions such as inoculum concentration, plant growth stage, and incubation period after transplant was investigated. Reproduction of the nematode on all the tested cultivars according to inoculum concentration increased in a dose-dependent manner. Except for inoculum concentration, there was no significant difference in reproduction level of the cultivars according to the other tested conditions. On the basis of the results, we suggest an efficient screening method for new resistant tomato to the nematode isolate.

Water quality and bacterial counts in hatchery of Rockbream, Oplegnathus fasciatus (돌돔(Oplegnathus fasciatus)부화장의 사육수 수질과 세균수)

  • To investigate the cause of the mass mortality during rockbream, Oplegnathus fasciatus seed production, the water quality and bacterial counts of sea water in breeding tanks was measured 20days post the hatch. During breeding of rockbream fry, the environmental factors of water quality were detected as pH, ammonia COD, phosphate at the supply of the food organisms and the seawater. pH was decreased from the 8.21 of the 1 day per hatch (dph) to 7.56 of the on the 7 dph. Ammonia was conversely increased 0.49 ppm of the 1 dph to 0.85 ppm of 10 dph. As the adding of the chlorella and the rotifer tanks, COD was increased the 3.3 times and 1.2 times than those of pre-adding respectively. The phosphate and the ammonia were also increased 1.7 and 2.3 times, with adding the chlorella respectively, which exceeded the second grade for sea water evaluation level, 0.015 ppm and 0.1 ppm respectively. Water quality was not improved by PSB (Photosynthetic Bacteria) treatment, which increased the value of COD in 1.7 times, phosphate in 2.7 times and ammonia in 1.4 times. The number of the bacteria was also increased along the dph. According to the treatment of chlorella, the number of total bacteria increased in 1.4 times and those of Vibrio sp. 1.6 times. The lethal concentration of ammonia was investigated that over than 10 ppm could killed the fry of rockbream within 28 hrs, but 40% in 2 ppm.

A New Early-Heading and High-Yielding Forage Rye Variety, 'Dagreen' (조숙 다수성 청예 조사료용 호밀 신품종 '다그린')

  • 'Dagreen' (Scale cereal L.), a new rye variety was developed from the open pollination among 10 rye lines at the National Institute of Crop Science in 1995 and mass selection were made at National Institute of Crop Science from 1996 to 1999. Recurrent selections were made at Dept. Rice and Winter Cereal Crop, NICS, RDA from 2000 to 2006. This new variety has erect plant type with medium size pale green leaves. The number of spike per $m^2$ was 713 and the plant height was 103cm. The heading date of 'Dagreen' was April 24 which was 7 days earlier than that of "Koolgrazer". It was adaptable for forage use at an early stage as a whole crop. Lodging resistance was higher than that of check variety 'Koolgrazer'. The chemical components and quality of forage showed 10.2 % crude protein, 36.6 % ADF, 62.4 % NDF and 59.9 % TDN. 'Dagreen' showed high resistance to powdery mildew and leaf rust than those of check variety in the field condition. The average dry matter (7,010kg $ha^{-1}$) of 'Dagreen' harvested at April 28 were 6 % higher than 'Koolgrazer'. This variety is recommended for all of the rye cultivation area in Korea.

Studies on Growth Characteristics and Propagation Method of Introduced Hop (Humulus lupulus L.) Cultivars (홉(Humulus lupulus L.) 도입 품종의 생육특성 및 영양번식 연구)

  • Domestic hop (Humulus lupulus L.) production has been suspended since the early 1990s due to foreign imports, but interest in local production is rising due to the recent craft beer boom in Korea. This study was conducted focusing on the development of growth characteristics and propagation technology for 6 introduced hop cultivars as a basic study for domestic hop production and breeding program. In the hop growth survey conducted in 2021 and 2022, the 5-year-old plants after planting generally showed a tendency to increase the height of strobile setting, strobile size, number and weight of strobile per hill compared to the 4-year-old plants. As a result of the experiment with hop vine cuttings, the average rooting rate of all cultivars was as high as 88% even in only water treatment that were not added with Atonik (Atonik, Arysta, Japan), a rooting agent. There were differences between cultivars in rooting length and rooting rate according to the Atonik treatment method. When checking the survival rate of the rooted cuttings seedlings after transplanting into the soil, it was confirmed that the survival rate of the cuttings in the tissue culture room was significantly lower than that of the cuttings in the greenhouse. However, in transplanting step, cutting plants from culture room condition was strongly inhibited plant growth because of changing environment conditions. As a results of tissue culture, the thidiazuron (TDZ) 1 ㎎/L treatment in the media generated 6 to 9 shoots/explant, while the 6-benzylaminopurine (BAP) 1 ㎎/L treatment generated only 1 to 2 shoots/explant. Therefore, it is more effective to culture by adding TDZ rather than BAP. These results indicated that the development of technology to manage stably after transplanting of cutting or micropropagating plants into potting soil is important for mass propagation of hops.

Determination of Color Value (L, a, b) in Green Tea Using Near-Infrared Reflectance Spectroscopy (근적외 분광분석법을 이용한 녹차의 색도 분석)

  • Near infrared spectroscopy (NIRS) is a rapid and accurate analytical method for determining the composition of agricultural products and feeds. The applicability of near infrared reflectance spectroscopic method was tested to determine the color value (L, a, b) of green tea. A total of 162 green tea calibration samples and 82 validation samples were used for NIRS equation development and validation, respectively. In the developed NIRS equation for analysis of the color value (L, a, b), the most accurate equation for L value was obtained at 2, 8, 6, 1 (2nd derivative, 8 nm gap, 6 points smoothing, and 1pointsecond smoothing), and for a, and b value were obtained at 1, 4, 4, 1 (1st derivative, 4 nm gap, 4points smoothing, and 1 point second smoothing) math treatment condition with SNVD (Standard Normal Variate and Detrend) scatter correction method and entire spectrum ($400{\sim}2,500\;nm$) by using MPLS (Modified Partial Least Squares) regression. Validation results of these NIRS equations showed very low bias (L: 0.005%, a: 0.003%, b: -0.013%) and standard error of prediction (SEP, L: 0.361%, a: 0.141%, b: 0.306%) as well as high coefficient of determination ($R^2$, L: 0.905, a: 0.986, b: 0.931). Therefore, these NIRS equations can be applicable and reliable for determination of color value (L, a, b) of green tea, and NIRS method could be used as a mass screening technique for breeding programs and quality control in the green tea industry.

Identification and Characterization of Phytochemicals from Peanut (Arachis hypogaea L.) Pods

  • Methanol extracts of peanut (Arachis hypogaea L.) pods were chromatographed, which yielded 3 phytochemicals 1-3 including 5,7-dihydroxychromone (1), eriodictyol (2), and 3',4',5,7-tetrahydroxyflavone (3). To confirm the presence of isolated phytochemicals, the pods extracts were performed by high performance liquid chromatography coupled with a photodiode array detector (HPLC-PDA) and a mass spectrometric detector (MSD) with electrospray ionization (ESI). Optimum extraction conditions for phytochemical contents using peanut germplasm were obtained by employing 90% MeOH for 12 hr at room temperature and phytochemicals 1-3 showed significant differences with concentrations of $407.56{\pm}23.35$, $52.92{\pm}5.11$, and $2,024.34{\pm}134.18\;{\mu}g/g$, respectively. Under this optimal conditions, the contents of phytochemicals 1-3 in peanut pods of 3 Korea cultivars including 'Jakwang', 'Daekwang', and 'Palkwang' exhibited phytochemical 3 was the highest range of $1,338.01-5,162.93\;{\mu}g/g$, followed by phytochemical 1 ($590.13-1,382.10\;{\mu}g/g$), and phytochemical 2 ($25.12-186.85\;{\mu}g/g$), respectively. Moreover, 'Jakwang' exhibited the highest contents of phytochemical (1: $1,362.10{\pm}52.49$, 2: $186.85{\pm}17.69$, and 3: $5,162.93{\pm}148.64\;{\mu}g/g$, respectively), whereas the lowest contents was found in the 'Daekwang' (1: $590.13{\pm}22.23$, 2: $25.12{\pm}2.45$, and 3: $1,338.01{\pm}62.17\;{\mu}g/g$, respectively). These results suggest that the methanol extracts of peanut pods may possess health related benefits to humans owing to various known biological activities of phytochemicals 1-3.

Proteomic Responses of Diploid and Tetraploid Roots in Platycodon grandiflorum (2배체와 4배체 도라지의 단백질 발현양상 비교 분석)

  • The roots of Platycodon grandiflorum species either dried or fresh, are used as an ingredient in salads and traditional cuisine in Korea. To interpret the root proteins, a systematical and targeting analysis were carried out from diploid and tetraploid roots. Two dimensional gels stained with CBB, a total of 39 differential expressed proteins were identified from the diploid root under in vivo condition using image analysis by Progenesis Same Spot software. Out of total differential expressed spots, 39 differential expressed protein spots (${\geq}\;1.5$-fold) were analyzed using LTQ-FTICR mass spectrometry. Except two proteins, the rest of the identified proteins were confirmed as down-regulated such as Isocitrate dehydrogenase, Proteasome subunit alpha type-2-B. However, the most of the identified proteins from the explants were mainly associated with the oxidoreductase activity, nucleic acid binding, transferase activity and catalytic activity. The exclusive protein profile may provide insight clues for better understanding the characteristics of proteins and metabolic activity in various explants of the economically important medicinal plant Platycodon grandiflorum.

Water Temperature and Food on Growth and Survival of Parrot Fish Larvae, Oplegnathus fasciatus (수온 및 먹이계열에 따른 돌돔, Oplegnathus fasciatus의 초기 성장과 생존율)

  • This study was to investigate the effects of water temperature and food on the mass seed production of larval parrot fish, Oplegnathus fasciatus. Growth of the larvae reared at heated water temperature ($25.3^{\circ}C$) was significantly higher than that of the larvae reared at natural water temperature ($20.5^{\circ}C$). In addition, survival rate of the larvae at heated and natural sea water temperature were $24.0\%$ and $12.3\%$, respectively (P<0.05). Growth and survival rate of the larvae fed mixed diets of Nannochloropsis oculata, rotifer (Brachionus rotundiformis). Artemia nauplii, Tigriopus japonicus and artificial diets were significantly higher than that of the larvae fed only either rotifer or artificial diets (P<0.05). The average survival rate and total length of the larvae reared for 50 days after hatching were $22.5\%$ and 62.0$\pm$4.0 mm, respectively. Amount of consumed rotifers at one time feeding by a larva was rapidly increased with growth from 10.3 individuals per larva 5 day old to 65.5 individuals per larva 20 day old.

Cloning of Farm Animals in Japan; The Present and the Future

  • 1. About fifty thousand of cattle embryos were transferred and 16000 ET-calves were born in 1999. Eighty percents of embryos were collected from Japanese Black beef donors and transferred to dairy Holstein heifers and cows. Since 1985, we have achieved in bovine in vitro fertilization using immature oocytes collected from ovaries of slaughterhouse. Now over 8000 embryos fertilized by Japanese Black bull, as Kitaguni 7~8 or Mitsufuku, famousbulls as high marbling score of progeny tests were sold to dairy farmers and transferred to their dairy cattle every year. 2. Embryo splitting for identical twins is demonstrated an useful tool to supply a bull for semen collection and a steer for beef performance test. According to the data of Dr. Hashiyada(2001), 296 pairs of split-half embryos were transferred to recipients and 98 gave births of 112 calves (23 pairs of identical twins and 66 singletons). 3. A blastomere-nuclear-transferred cloned calf was born in 1990 by a joint research with Drs. Tsunoda, National Institute of Animal Industry (NIAI) and Ushijima, Chiba Prefectural Farm Animal Center. The fruits of this technology were applied to the production of a calf from a cell of long-term-cultured inner cell mass (1988, Itoh et al, ZEN-NOH Central Research Institute for Feed and Livestock) and a cloned calf from three-successive-cloning (1997, Tsunoda et al.). According to the survey of MAFF of Japan, over 500 calves were born until this year and a glaf of them were already brought to the market for beef. 4. After the report of "Dolly", in February 1997, the first somatic cell clone female calves were born in July 1998 as the fruits of the joint research organized by Dr. Tsunoda in Kinki University (Kato et al, 2000). The male calves were born in August and September 1998 by the collaboration with NIAI and Kagoshima Prefecture. Then 244 calves, four pigs and a kid of goat were now born in 36 institutes of Japan. 5. Somatic cell cloning in farm animal production will bring us as effective reproductive method of elite-dairy- cows, super-cows and excellent bulls. The effect of making copy farm animal is also related to the reservation of genetic resources and re-creation of a male bull from a castrated steer of excellent marbling beef. Cloning of genetically modified animals is most promising to making pig organs transplant to people and providing protein drugs in milk of pig, goat and cattle. 6. Farm animal cloning is one of the most dreamful technologies of 21th century. It is necessary to develop this technology more efficient and stable as realistic technology of the farm animal production. We are making researches related to the best condition of donor cells for high productivity of cloning, genetic analysis of cloned animals, growth and performance abilities of clone cattle and pathological and genetical analysis of high rates of abortion and stillbirth of clone calves (about 30% of periparutum mortality). 7. It is requested in the report of Ministry of Health, labor and Welfare to make clear that carbon-copy cattle(somatic cell clone cattle) are safe and heathy for a commercial market since the somatic cell cloning is a completely new technology. Fattened beef steers (well-proved normal growth) and milking cows(shown a good fertility) are now provided for the assessment of food safety.

Characterization of Bacillus amyloliquefaciens DA12 Showing Potent Antifungal Activity against Mycotoxigenic Fusarium Species

  • In an attempt to develop a biological control agent against mycotoxigenic Fusarium species, we isolated Bacillus amyloliquefaciens strain DA12 from soil and explored its antimicrobial activities. DA12 was active against the growth of mycotoxigenic F. asiaticum, F. graminearum, F. proliferatum, and F. verticillioides both in vitro and in planta (maize). Further screening using dual culture extended the activity range of strain DA12 against other fungal pathogens including Botrytis cinerea, Colletotrichum coccodes, Endothia parasitica, Fusarium oxysporum, Raffaelea quercus-mongolicae, and Rhizoctonia solani. The butanol extract of the culture filtrate of B. amyloliquefaciens DA12 highly inhibited the germination of F. graminearum macroconidia with inhibition rate 83% at a concentration of $31.3{\mu}g/ml$ and 100% at a concentration of $250{\mu}g/ml$. The antifungal metabolite from the butanol extract was identified as iturin A by thin layer chromatography-bioautography. In addition, volatile organic compounds produced by DA12 were able to inhibit mycelial growth of various phytopathogenic fungi. The volatile compounds were identified as 2-heptanone, 5-methyl heptanone and 6-methyl heptanone by gas chromatography-mass spectrometry (GC-MS) analysis. These results indicate that the antagonistic activity of Bacillus amyloliquefaciens DA12 was attributable to iturin A and volatile heptanones, and the strain could be used as a biocontrol agent to reduce the development of Fusarium diseases and mycotoxin contamination of crops.
