The mechanism for the elimination of xenobiotics undergoes three different phases of reactions in organisms. Among these, glutathione S-transferases (GSTs) are classified as phase II detoxification enzymes, catalyzing the conjugation of electrophilic substrates to glutathione or reduced hydroperoxides. This study aimed to investigate the molecular characteristics, detoxification functions, and immune responses of GST omega (LhGSTO1) and kappa (LhGSTK1) in redlip mullet. The open reading frames of LhGSTO1 (720 bp) and LhGSTK1 (687 bp) encoded proteins of 239 and 228 amino acids, respectively. Sequence analysis revealed that LhGSTO1 and LhGSTK1 possessed GSH-binding sites in their N-terminal domains. Substrate-binding sites in the C-terminal domain were exclusively identified in LhGSTO1. In the tissue-specific transcription profile analysis, both LhGSTO1 and LhGSTK1 were ubiquitously expressed in all tissues of healthy mullets. Temporal expression analysis of LhGSTO1 and LhGSTK1 in the blood showed that their expression was significantly modulated by polyinosinic:polycytidylic (poly I:C), lipopolysaccharide (LPS), and Lactococcus garvieae. Different chemical and cellular assays were performed to assess the detoxification and cellular protective abilities of the two proteins. A substrate specificity test using the recombinant proteins revealed that both proteins possessed specific activity toward 1-chloro-2,4-dinitrobenzene (CDNB). In the disk diffusion assay, the smallest clearance zones were observed for LhGSTO1 and LGSTK1 against CdCl2. In the cell protection assay, both LhGSTO1 and LhGSTK1 showed significant Cd detoxification ability compared to the control. Collectively, these results demonstrate that GST omega and kappa are involved in host defense against immune stimulants and xenobiotics in redlip mullet.

Carbon dioxide capture and storage (CCS) technology has been regarded as one of the most possible and practical option to reduce the emission of carbon dioxide ($CO_2$) and consequently to mitigate the climate change. Korean government also have started a 10-year R&D project on $CO_2$ storage in sea-bed geological structure including gas field and deep saline aquifer since 2005. Various relevant researches are carried out to cover the initial survey of suitable geological structure storage site, monitoring of the stored $CO_2$ behavior, basic design of $CO_2$ transport and storage process and the risk assessment and management related to $CO_2$ leakage from engineered and geological processes. Leakage of $CO_2$ to the marine environment can change the chemistry of seawater including the pH and carbonate composition and also influence adversely on the diverse living organisms in ecosystems. Recently, IMO (International Maritime Organization) have developed the risk assessment and management framework for the $CO_2$ sequestration in sub-seabed geological structures (CS-SSGS) and considered the sequestration as a waste management option to mitigate greenhouse gas emissions. This framework for CS-SSGS aims to provide generic guidance to the Contracting Parties to the London Convention and Protocol, in order to characterize the risks to the marine environment from CS-SSGS on a site-specific basis and also to collect the necessary information to develop a management strategy to address uncertainties and any residual risks. The environmental risk assessment (ERA) plan for $CO_2$ storage work should include site selection and characterization, exposure assessment with probable leak scenario, risk assessment from direct and in-direct impact to the living organisms and risk management strategy. Domestic trial of the $CO_2$ capture and sequestration in to the marine geologic formation also should be accomplished through risk management with specified ERA approaches based on the IMO framework. The risk assessment procedure for $CO_2$ marine storage should contain the following components; 1) prediction of leakage probabilities with the reliable leakage scenarios from both engineered and geological part, 2) understanding on physio-chemical fate of $CO_2$ in marine environment especially for the candidate sites, 3) exposure assessment methods for various receptors in marine environments, 4) database production on the toxic effect of $CO_2$ to the ecologically and economically important species, and finally 5) development of surveillance procedures on the environmental changes with adequate monitoring techniques.
Sexual dimorphism is the most conspicuous difference between the sexes. This study examines possible sexual dimorphism and the relative growth patterns of morphometric characteristics in the marine medaka, Oryzias dancena for their potential to help differentiate between males and females of this species. The von Bertalanffy growth parameters estimated by a non-linear regression method were $L_{\infty}=30.2mm$, K=3.22/year, and ${\tau}_0=-0.05$. All 18 characteristics measured showed a difference between males and females from 70 days after hatching. Each of these characteristics were significantly different between sexes (ANCOVA, P<0.05), and the ratio of standard length between sexes showed that males were larger than females for all five morphometric measurements. Fin length measurements were taken for 21 distances of anal fin and 7 distances of dorsal fin between landmarks. There were all differences for all dorsal fin rays between the males and the females and there is significant difference in 70 days after their hatch when the sexual dimorphism is presented. The significant difference (P<0.05) in fin ray for male and female was more greatly seen as they grow. Male marine medaka showed more rapid growth than females, with longer length, dorsal fins and anal fins. Differences in these characteristics will be useful during experiments when it is necessary to differentiate between sexes of marine medaka.
Microplastic particles are ubiquitous in the environment and not standardized particles of size, shape, or type. Therefore, it is very limited to establish a risk assessment framework that accurately evaluated and manage the multi-dimension of marine environment including seawater and sediment based on toxic data. In the study, we review the characteristics and effects of marine environmental microplastic and suggest risk assessment framework (draft) based on the distribution and impact of marine environmental microplastics. Although, the characteristics of environmental microplastic are very widely but the most abundant toxic data are concentrated on unique shape and type, and there are also large gaps of test organism between laboratory-exposed organisms and resident species. Great limitations with respect to toxicity data quality also exist for traditional effect assessment methods, which in reliability of the resulting risk characterizations. However, considering the fact that the international community's movement on microplastics management is gradually strengthening and the pollution level of microplastics in marine environment is increasing, further research on environmental relevant risk assessment technique should be proposed based on the characteristics of microplastics in the marine environment.
Mixotrophy in marine organisms is an important aspect of ecology and evolution. The discovery of mixotrophic abilities in phototrophic dinoflagellates alters our understanding of the dynamics of red tides. In the phototrophic dinoflagellate genus Alexandrium, some species are mixotrophic, but others are exclusively autotrophic. There are differences in the ecological roles of autotrophic and mixotrophic Alexandrium in marine food webs. However, of the 34 known Alexandrium species, the mixotrophic ability of >20 species has yet to be explored. In this study, the mixotrophic capabilities of Alexandrium insuetum CCMP2082, Alexandrium mediterraneum CCMP3433, Alexandrium pacificum CCMP3434, Alexandrium tamutum ATSH1609, and Alexandrium margalefii CAWD10 were investigated by providing each species with 22 diverse prey items including bacterium-sized microbeads (1 ㎛), the cyanobacterium Synechococcus sp., algal prey species, and the ciliate Mesodinium rubrum. None of the 5 Alexandrium species fed on any of the prey items. These results increase the number of Alexandrium species lacking mixotrophic abilities to 9, compared to the 7 known mixotrophic Alexandrium species. Furthermore, the Alexandrium phylogenetic tree based on the large subunit ribosomal DNA contained 3 large clades, each of which had species with and without mixotrophic abilities. Thus, the acquisition or loss of mixotrophic abilities in Alexandrium might readily occur.
In 2009, United Nations Educational, Scientific and Cultural Organization (UNESCO) recognized the unique outstanding ecosystem biodiversity and distinct ecocultural values of the Shinan Dadohae Biosphere Reserve in the island region. The Dadohae area, which has been sustainably conserved for scores of years, boasts not only a unique ecosystem, but also has residents with a wide range of traditional ecological knowledge. In terms of understanding the soundness of the ecosystem network known as the landscape system, the recent expansion of environmental development has served to heighten the degree of consideration given not only to biodiversity, which has long been used as an indicator to assess ecosystem soundness, but also to assess cultural diversity. Man has used the surrounding landscape and living organisms as his life resources since the beginning. Moreover, whenever necessary, man has developed new species through cultivation. Biodiversity became a foundation that facilitated establishing cultural diversity such as food and housing. Such ecological knowledge has been conveyed not only to adjacent regions, but also at the international level. The recent rapid changes in the Dadohae area island ecosystem caused by the transformation of fishing grounds by such factors as climate change, excess human activities, and marine pollution, is an epoch event in environmental history that shows that the balance between man and nature has become skewed. Furthermore, this issue has moved beyond the biodiversity and landscape diversity level to become an issue that should be addressed at the cultural diversity level. To this end, the time has come to pay close attention to this issue.
Environmental changes caused by the abrupt climatic change are one of the important issues in the scientific community. In the East Sea, abrupt climatic shift, called Younger Dryas, is identified. The age of the Younger Dryas cold episode occurred at 11.2 ka. Overall, changes in circulation and bottom water conditions occurred during the Younger Dryas cold episode in the study area. Especially, climatic transition from meltwater spike to the Younger Dryas cold episode is characterized by significant shifts of oxygen isotope values, the coiling ratios of Neogloboquadrina pachyderma, and the planktonic foraminifers abundances. The impact of abrupt climate change on the ecosystem is very significant. In the East Sea, the calcium carbonate secreting organism(foraminifers) is replaced by silicon dioxide secreting organisms(diatom, radiolarian) after the abrupt and severe cold climatic event. Based on the Doctrine of Uniformitarianism, at least climate change for the next 100 years would be severely influence on the marine ecosystem.
Mussels have been commonly used in bioassay for quality assessments of environment. Moreover, several standard protocols for the developmental bioassay of bivalves have been proposed. In this study, the EC50 of polycyclic aromatic hydrocarbons (PAHs) was determined using mussel, Mytilus galloprovincialis embryonic developmental bioassay. To determine the sensitivity of M. galloprovincialis, their fertilized eggs were exposed to five PAHs (2-metylnaphthalene, fluorene, dibenzothiophene, phenanthrene, pyrene). The EC50 of 2-metylnaphthalene, fluorene, dibenzothiophene, phenanthrene, and pyrene were 232, 273, 67.9, 43.2, and $33.1{\mu}g/L$, respectively. The overall sensitivity of M. galloprovincialis in the present developmental bioassay was similar to or more sensitive than that of other marine organisms commonly used in aquatic bioassays. The results of this study could be provide with fundamental data of setting standard for protection of marine life and or can use prediction the aquatic toxicity of PAHs.
The marine dinoflagellate genus Alexandrium, which produces PSP toxins, has a global distribution. As human-assisted dispersal of the species has been suggested, it is important to develop molecular tools to trace the dispersal pathway. To screen population-specific DNA sequences that differentiate Korean and Japanese A. catenella, we targeted the area downstream of the chloroplast psbA gene using PCR with population-specific DNA primers followed by RFLP (restriction fragment length polymorphism) analysis and sequencing. The RFLP patterns of the PCR products divided Korean and Japanese A. catenella regional isolates into three types: Korean, Japanese, and type CMC3, isolated from Korea. We sequenced the PCR products, but found no similar gene in a homology search. The molecular phylogeny inferred from the sequences separated the Korean and Japanese A. catenella strains, as did the RFLP patterns. However, the Japanese isolates included two slightly different sequences (types J and K), while the Korean sequence was the same as the Japanese K type. In addition, a unique sequence was found in the Korean strains CMC2 and CMC3. Population-specific PCR amplification with Japanese A. catenella type-specific PCR primers designed from the type J sequence yielded PCR products for Japanese strains only, showing that the unknown gene can be used for a population analysis of Korean and Japanese A. catenella.
Temporal and spatial variation in the freshwater region, created by river runoff, of a small bay, caused by the passage of typhoons was examined using a three-dimensional primitive equation model (the Princeton Ocean Model, POM). Numerical experiments were implemented focusing on temporal evolution in the freshwater region in association with typhoon tracks. The model domain covered most of the estuary around the Nakdong River, including Noksan Bay, where river water is periodically released from upstream (Noksan dam). The model showed that the extension of the freshwater region outside of the bay depended strongly on the tracks of typhoons, specifically the associated wind directions and inner flow fields that are accompanied by new clockwise eddies. The model also showed that entrainment from typhoon passage frequently creates salt wedges in the estuary, indicating that organisms in the bay are biologically and chemically influenced with variation in the freshwater region.
