  • Catfish is one of the most important freshwater fish farming commodities in Indonesia. Higher catfish production can be achieved by cultivating transgenic catfish carrying the growth hormone (GH) gene of African catfish (Clarias gariepinus GH, CgGH). This research focuses on analysis of the presence of the CgGH gene in transgenic G1, G2, and G3 mutiara catfish broodstock, as an indication of stable CgGH inheritance. CgGH gene was isolated using the RNeasy mini kit and RT-PCR. RT-PCR revealed amplicons measuring approximately 600 bp in transgenic G0, G1, G2, and G3 mutiara catfish. The CgGH consensus sequence similarities ranged from 93.76% to 97.06%, with four functional domain sites (somatotropin-1, somatotropin-2, four α-helix, N-glycosylation, four cysteine residues) of fish GH proteins. The functional domains of fish GH proteins are conserved in G1, G2, and G3 and indicate stable exogenous GH inheritance to produce transgenic catfish strains in each generation.

  • The aim of this study was to compare the efficacy of cryopreservation methods for ex situ conservation of spermatozoa from far eastern catfish, Silurus asotus. The spermatozoa activity index (SAI) and hatching rates were higher in spermatozoa stored in Alserver's solution than those of spermatozoa stored in glucose solution. The SAI and hatching rates in all experimental groups gradually decreased with increasing duration of storage. Additionally, the SAI and hatching rates gradually decreased with increasing thawing temperatures at all storage durations (P<0.05). Based on the SAI and hatching rates, our results suggest that the optimal cryopreservation conditions of catfish spermatozoa involve storage in Alserver's solution with 15% ethylene glycol, and thawing at $25^{\circ}C$. Cryopreservation of spermatozoa is a useful and reliable technique for conserving gene resources and for artificial propagation of far eastern catfish.

  • Histological observations of the barbels of torrent catfish (Amblycipitidae) in Korea are reported. The bullhead torrent catfish (Liobagrus andersoni) and Korean torrent catfish (L. obesus) have eight obvious barbels in two pairs each on the maxilla and mandible. Each barbel is composed of an epidermis, dermis, and central cartilagerod. The epidermis of the maxillary and mandibular barbels of the bullhead torrent catfish is thicker than that of the Korean torrent catfish (P<0.05) and is formed of stratified epithelium that contains many cutaneous taste buds and a few small club cells. There are many taste buds on the upper part of each barbel. The dermis consists of loose connective tissue fibers that enclose blood vessels and bundles of nerve fibers. The barbels of these fish can be categorized into stiff and flexible types and are accessory, feeding, and sensory structures. Our results validate the barbels as gustatory receptor organs for these fishes.

  • For the 2 years of farming, at the indoor circulating aquaculture system, four kinds of skeletal deformities were found among 60 Far Eastern catfish, Silurus asotus. Deformities saw jawbone's luxation, abnormality of upper lip and malocclusion. Spinal deformity was most fatal deformities with low weight and small length. Jawbone's luxation had 1 maxilla and 2 mandibles. Abnormality of upper lip had just lip was back over. Malocclusion's left maxilla and right maxilla were not balanced. This experiment was any deformities in this species through the deformity can grasp how it affects.

  • Of the 20 arbitrarily chosen primers, six oligonucleotides decamer primers were used on the basis of the number of the polymorphisms generated in catfish (Silurus asotus) from Yesan and bullhead (Pseudobagrus fulvidraco) from Dangjin in Korea. Six primers were used generating a total of 602 scorable bands in catfish and 195 in bullhead population, respectively, ranging in size of DNA fragments from less than approximately 100 to larger than 2,000 base pairs (bp). Six primers yielded 199 polymorphic fragments (33.1 %) in catfish and 47 (24 %) in bullhead, respectively. In the present study, a total of 328 common fragments (an average of 54.7 per prime.) were observed in catfish population, whereas 84 (an average of 14.0 per prime.) in bullhead. The total number of specific fragments in catfish and bullhead population were 76and 64, respectively.

  • This study identified the differences in the morphometric characteristics of the truss and classical dimensions between diploid and triploid Far Eastern catfish, Silurus asotus, and provided methods for sorting diploid and triploid Far Eastern catfish based on morphometric observations. The significant variables were the direct distance between the anterior edge of the lower lip and the anterior insertion of the dorsal fin(DALAD), the horizontal distance between the anterior edge of the lower lip and the anterior insertion of the ventral fin (HALAV), the direct distance between the anterior edge of the upper lip and the first nostril(DAUF), the direct distance between the anterior edge of the upper lip and the second nostril(DAUS), the interorbital width(IW), and the mandible barbel length (ManBL). The more significant variables were HALAV, DALAD, DAUF, IW, and DAUS. The most useful combination of variables for separating the two groups was DALAD, IW, and DAUF, which correctly classified 85% of the catfish as triploid or diploid, and that percentage was the maximum degree of value possible (p<0.05). Triploid Far Eastern catfish had a high rate of growth in the head region and body depth during the first year after hatching. Triploid Far Eastern catfish had smaller heads and shorter mandible barbels than diploid Far Eastern catfish.

  • A closely associated set of characteristics was analyzed using both classical and truss dimensions to discriminate the morphometric traits of three species of torrent catfish (Amblycipitidae) in Korea. After arcsine square root transformation, 18 measurements relative to the standard length and three measurements relative to the head length of each species of torrent catfish were compared statistically. Classical and truss dimensions of each of four morphometric traits showed higher values in the bullhead torrent catfish, Liobagrus obesus, than the Korean and south torrent catfish (L. andersoni and L. mediadiposalisa, respectively; P<0.05). Truss dimensions of three morphometric traits were higher in the bullhead and Korean torrent catfish than in the south torrent catfish (P<0.05). There were no significant differences in six morphometric traits among these Amblycipitidae. The dimensions used in this study may be useful as taxonomic indicators to discriminate species of Liobagrus in Korea.

  • The egg development of various fishes is studied by various methods including electron microscopy(Matsuyama et al, 1991) and hormone treatment(Schoonbee et al. 1980). In addition to attempt to induce final maturation and spawning, the present experiments were conducted to investigate the influence of hCG upon reproductive cycles in catfish(Silurus asotus) with developing gonads and to stimulate the reproductive system during the rapid phase of gonadal development. • Ileal and total tract digestibility and nitrogen (N) balance of diets with four different protein sources were determined in growing pigs. The diets were based on rice bran, broken rice and maize meal and contained Tra catfish by-product (CBP), processed using three different methods, and marine fish meal (FM). The CBP diets consisted of the by-product in meal form, ensiled with molasses, and CBP waste water (WWBD). The four diets were fed to four growing pigs fitted with post-valve T-cecum (PVTC) cannulas in a $4{\times}4$ Latin Square design. All experimental diets included $Cr_2O_3$ at 5 g/kg feed as an indigestible marker. The ileal apparent digestibility of organic matter and ether extract was higher on diet WWBD than on the other three diets (p<0.05), and the total tract apparent digestibility was higher on diet WWBD than on the FM diet (p<0.05). The ileal and total tract apparent digestibility of crude protein and amino acids was not significantly different among diets (p>0.05). No significant effects of diet were found on N-retention and N utilization. In conclusion, the catfish by-product meal, ensiled catfish by-product and processing waste water diets and the fish meal diet had similar ileal and total tract apparent digestibility, and similar N utilization in growing pigs.
