Antifibrotic Activity of Manassantin B from Saururus chinensis in HSC-T6 Hepatic Stellate Cells

  • Manassantin B, a dilignan isolated from Saururus chinensis, significantly inhibited proliferation in HSC-T6 cells in concentration- and time-dependent manners. In addition, treatment of HSC-T6 cells with manassantin B changed cell morphology from flattened myofibroblastic membranous morphology, representing activation state, to slender shape, representing quiescent state. Furthermore, manassantin B effectively reduced collagen content in HSC-T6 cells. These results suggested that manassantin B exerted antifibrotic activity in HSCT6 cells, in part, via inhibition of cell proliferation and decrease of collagen production.

Qualities and Lignans Contents of Saururus chinensis Baill. Rhizome by Storage Methods (삼백초 근경의 저장방법에 따른 품질과 Lignans 함량 변화)

  • This study was carried out to investigate the qualities of Saururus chinensis Baill. rhizomes and it content of active ingredient accenting to the storage methods. As the storage period increased the rates of weight loss, rotten rhizomes and the number of sprouts increased. Refrigerator storage resulted in the lowest weight reduction and sprouts rates, while storage in soil-filled box in a store-house was most effective in reducing spoilage rate. Five lignans from rhizome were determined by HPLC Retention time ranged $18{\sim}36$ minutes and showed saucernetin sauchinone, manassantin A, saucerneol D, and manassantin B in that order. Regardless of storage methods, the lignan content was lower after 120 days than after 30 days of storage, and increased manassantin B, manassantin A, saucernetin sauchinone, and saucerneol D in that order.

Inhibition of NF-IL6 Activity by Manassantin B, a Dilignan Isolated from Saururus chinensis, in Phorbol Myristate Acetate-stimulated U937 Promonocytic Cells

  • Mannasantin B, a dilignan structurally related to manssantin A, is an inhibitor of NF-${\kappa}B$ transactivation. In the present study, we found that it inhibited PMA-induced expression of IL-$1{\beta}$, IL-$1{\beta}$ mRNA, and IL-$1{\beta}$ promoter activity in U937 cells with $IC_{50}$ values of about 50 nM. It also inhibited NF-IL6- and NF-${\kappa}B$-induced activation of IL-$1{\beta}$, with $IC_{50}$ values of 78 nM and $1.6{\mu}M$, respectively, revealing a potent inhibitory effect on NF-IL6. Electrophoretic mobility shift assays showed that manassantin B had an inhibitory effect on DNA binding by NF-IL6, but not by NF-${\kappa}B$. Further analysis revealed that transactivation by NF-IL6 was also inhibited. Our results indicate that manassantin B suppresses expression of IL-$1{\beta}$ in promonocytic cells by inhibiting not only NF-${\kappa}B$ but also NF-IL6 activity. Furthermore, our observations suggest that manassantin B may be clinically useful as a potent inhibitor of NF-IL6 activity.

  • In our search for NF-kB inhibitors from natural resources, we have previously identified two structurally related dilignans, manassantin A and B as specific inhibitors of NF-kB activation from Saururus chinensis. However, their molecular mechanism of action remains unclear. We here demonstrate that manassantin A and B are potent inhibitors of NF-kB activation by the suppression of transciptional activity of RelA/p65 subunit of NF-kB. These compounds significantly inhibited the induced expression of NF-kB reporter gene by LPS or TNF-a in a dose-dependent manner. (omitted)

  • Manassantin B classified into dineolignans have been isolated from Saururus chinensis Manassantin B was found to induce apoptosis in human promyelocytic leukaemia HL -60 cells with characteristic apoptotic features like increase of nucleosomalladder. apoptotic body ormation. flipping of membrane phosphatidylserine. Manassantin B induced FAS and FAS ligand expression, and activated caspase 8 which cleaved bid to tbid in cytosol. The release of cytochrome c to sytosol was accompanied with decrease of bcl-2 protein and incresase of tbid and bax protein in mitochondria. Released xytochrome c activated caspase 9 and-3. but these effects were completely attenuated by the treatment of broad caspses ingibitor. Z-VAD fmk. These results indicate that manassatin B induce apoptosis through upregulation of FAS. caspase family and mitochondria-related proteins.

  • In the course of our search for intercellular adhesion molecule-1 (ICAM-1)/leukocyte function-associated antigen-1 (LFA-1) mediated cell adhesion inhibitors from natural sources, new type of cell adhesion inhibitors were isolated from the MeOH extract of Saururus chinensis roots. On the basis of spectral evidence, the structures of the active compounds were identified as manassantin A and B. Manassantin A and B inhibited phorbol 12-myristate 13-acetate (PMA)-induced homotypic aggregation of the human promyelocytic leukemia HL-60 cells without cytotoxicity with MIC value of 1.0 and 5.5 nM, respectively.   • In order to find the new hepatoprotective agents from natural products, the isolation and identification of biological active components of the roots of Saururus chinensis has been carried out. A MeOH extract of this plant showed the significant hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells. Five lignans including sauchinone, manassantin A, manassantin B, saucerneol B, and di-O-methyltetrahydrofuriguaiacin B were isolated and identified by spectroscopic evaluation. (omitted)

  • Cancer-inducing PMA stimulates cells to increase the expression of transcription factor c-Jun/c-fos and then increase the activity of AP-1 in the nucleus. The activity of AP-1 has been reported to cause cancer. In this study, We conducted cytotoxicity experiments to assess the safety of natural marker compounds and also observed inhibition of activator protein(AP-1) activity to predict cancer-preventing effects. The results of this experiment indicated that arctigenin, manassantin A, and B can predict the development of cancer prevention agents.

Manassantin A and B Isolated from Saururus chinensis Inhibit $TNF-{\alpha}-Induced$ Cell Adhesion Molecule Expression of Human Umbilical Vein Endothelial Cells

  • Leukocyte adhesion to the vascular endothelium is a critical initiating step in inflammation and atherosclerosis. We have herein studied the effect of manassantin A (1) and S (2), dineolignans, on interaction of THP-1 monocytic cells and human umbilical vein endothelial cells (HUVEC) and expression of intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin in HUVEC. When HUVEC were pretreated with 1 and 2 followed by stimulation with $TNF-{\alpha}$, adhesion of THP-1 cells to HUVEC decreased in dose-dependent manner with $IC_{50}$ values of 5 ng/mL and 7 ng/mL, respectively, without cytotoxicity. Also, 1 and 2 inhibited $TNF-{\alpha}-induceda$ up-regulation of ICAM-1, VCAM-1 and E-selectin. The present findings suggest that 1 and 2 prevent monocyte adhesion to HUVEC through the inhibition of ICAM-1, VCAM-1 and E-selectin expression stimulated by $TNF-\alpha$, and may imply their usefulness for the prevention of atherosclerosis relevant to endothelial activation.
