금호강 오염의 종합적 조사 (A Survey of the Kumho River Pollution)

  • 금호강 수계의 수질판정과 낙동강 본류에 미치는 영향을 조사하기 위해 2000년 5월부터 2001년 2월까지 5월, 8월 및 12월 3회에 걸쳐 16개 지점에서 BOD 등 총 24개 항목의 수질을 측정하였다. 각 지점별 항목별 대표적인 특징을 조사기간 전 3년간의 연구결과("전 연구결과")와 비교하여 다으과 같은 결론을 얻었다. 조사기간의 금호강 전 지점에 대한 각 항목의 평균값은 다음과 같다. 수온 $17.71^{\circ}C$, pH 8.06, BOD $2.26{\mu}g/mL$ COD $5.58{\mu}g/mL$, DO $7.13{\mu}g/mL$ SS $4.73{\mu}g/mL$ 전기전도도 $570.0{\mu}S/cm$ 유지류 $0.20{\mu}g/mL$ ABS $0.016{\mu}g/mL$ Phenol $0.29{\mu}g/mL$ 영양염류인 Total-N $5.03{\mu}g/mL$ Total-P $0.27{\mu}g/mL$ 이 었다. 중금속류인 Cu $0.005{\mu}g/mL$, Zn $0.007{\mu}g/mL$ Fe $0.044{\mu}g/mL$, Mn $0.001{\mu}g/mL$ 이었다. 한편 As, Cd, Cr 및 Pb는 불검출되었다. 금호강 전 지점에서 평균값으로 "전 연구결과" 와 비교할 때에 오염이 증가된 항목은 페놀 $NH_3-N$$NO_2-N$이며, 감소된 항목은 BOD, COD, SS, 유지류 ABS 등이고, 기타 항목은 비슷한 결과를 보였다. 금호강 수질이 낙동강의 수질에 미치는 영향의 정도는 다음과 같다. 금호강 하천수의 유입 전 지점 14와 유입 후 지점 16의 평균값으로서 BOD 1.07 에서 $1.42{\mu}g/mL$ COD 1.99에서 $2.44{\mu}g/mL$ 전기 전도도 221에서 $392{\mu}S/cm$ 총질소 2.21에서 $2.81{\mu}g/mL$ 총인 0.08에서 $0.19{\mu}g/mL$ 유지류 0.15에서 $0.16{\mu}g/mL$ ABS 0.006에서 $0.015{\mu}g/mL$ 페놀 0.06에서 $0.07{\mu}g/mL$으로 변화를 보였다.

  • Molecular targeting may be defined as the specific concentration of a diagnostic or therapeutic tracer by its Interaction with a molecular species that is distinctly present or absent in a disease state. Monoclonal antibody (mAb) is one of the successful agents for targeted therapy in cancer. To enhance the therapeutic effect, the concept of targeting radionuclides to tumors using radiolabeled mAbs against tumor-associated antigens, radioimmunotherapy, was proposed. The efficacy of radioimmunotherapy, however, has to be further optimized. Several strategies to improve targeting of tumors with radiolabeled mAbs have been developed, such as the use of mAb fragments, the use of high-affinity mAbs, the use of labeling techniques that are stable in vivo, active removal of the radiolabeled mAb from the circulation, and pretargeting strategies. Until now, however, there are many kinds of obstacles to be solved in the use of mAb for the targeted therapy. Major technical challenges to molecular targeting are related to the rapid and specific delivery of tracers to the target, the elimination of unwanted background activity, and the development of more specific targets to create a cytocidal effect. further development of this field will be determined by success in solving these challenges.

미생물학적 모니터링 분석방법 개발을 위한 생물학적 수용체로서 살모넬라에 특이적인 다중클론 항체의 생산 및 특성 검토 (Production and characterization of anti-Salmonella polyclonal antibodies as bio-recognition element for developing a microbial monitoring method)

  • 살모넬라는 주요한 식중독균의 하나로 미생물학적 식품품질을 보증하기 위해 신속 정확하게 현장에서 관리되고 모니터링되어야 한다. 본 연구팀에서는 광학 현미경 기반의 이미징 시스템과 바이오센서가 결합된 모니터링 시스템을 개발하였고, 이를 살모넬라에 적용하기 위해서는 바이오센서에 적용되는 생물학적 수용체의 개발이 절대적으로 필요한 실정이다. 따라서 본 연구에서는 살모넬라에 반응하는 특이적인 anti-Salmonella 다중클론 항체를 토끼로부터 성공적으로 생산 정제하였으며, 최종 항체의 농도는 2 mg/mL로 결정되었다. 또한, 정제된 anti-Salmonella 다중클론 항체의 역가가 상업용 anti-Salmonella 다중클론 항체보다 전 항체 농도 범위에서 우수함을 알 수 있었다. 정제된 항체 특이성 검토를 위해, 정제된 anti-Salmonella 다중클론 항체를 20 종의 살모넬라 serotypes과 20 종의 non-Salmonella strains과 반응시킨 결과, 20 종의 non-Salmonella strains과는 특이성을 보여주지 않은 반면 19 종의 살모넬라 serotypes와 유의적으로 높은 반응성으로 보여 주었다. 최종적으로, anti-Salmonella 다중클론 항체가 고정화된 골드센서를 살모넬라 현탁액에 노출시켰을 때, 수 백여 개의 살모넬라가 센서 표면에 capture되어 있음을 확인할 수 있었다. 따라서, 본 연구를 통해 생산 정제된 anti-Salmonella 다중클론 항체는 새로운 생물학적 수용체로서 충분한 역가와 특이성 그리고 센서에 적용 시 살모넬라와의 높은 결합력을 보여주었으므로 광학현미경 기반 이미징 시스템을 활용한 바이오센서 모니터링 시스템에 새로운 생물학적 수용체로 적용될 수 있으리라 판단된다.

  • In this study, to investigate a characteristics of slip ratio control of H.C.U for ABS, half car model tester were developed and a new H.C.U. was compactly designed comparing to the commercical H.C.U. for ABS. In half car model tester, variable inertia wheel has been used to load the car weights and braking forces according to the road surface conditions which were realized by pneumatic cylinder. And solenoid valves using P.W.M. (Pulse Width Modulation) method were installed in the new H.C.U The slip ratio characteristics of tire had been measured using half car model tester and the results were used in the control simulation for a new H.C.U.

IMMUNOREGULATORY EFFECTS OF A MONOCLONAL ANTIBODY TO HUMAN 4-1 BB MOLECULE ON ALLOANTIGEN-MEDIATED IMMUNE RESPONSES.

  • 4-lBB molecule is expressed on the surface of activated CD4$\^$+/ and CD8$\^$+/ T cells. We generated a panel of anti-4-1 B5 murine mAbs using a fusion protein consisting of the extracellular domain of human 4-1 BB fused to Glutathione S-transferase. The binding activity against cell surface 4-1 BB molecule was assessed by flow cytometry analysis. These studies showed that several anti-4-1 BB mAbs bound to 10-30% of CD4$\^$+/ and CD8$\^$+/T cells in PHA or Con A stimulated PBLs, although these mAbs interacted with only, l-2% of CD4$\^$+/ and CD8$\^$+/ T cells in normal PBLs, indicating the specificity of mAbs to the 4-l BB molecule on activated CD4$\^$+/ and CD8$\^$+/ T cells. Next, we examined the effect of an anti-4-l BB mAb (4B4-1-1) on allogeneic mixed lymphocyte reactions (MLRs). The data indicated that the antibody significantly inhibited the proliferative response at higher concentrations. When tested with several T cell mitogens, the antibody had no stimulatory or inhibitory effects on the mitogen-mediated T cell proliferation. These data suggest that 4-1 BB molecule may play a role in the regulation of antigen-mediated immune response.

Effects of PP-g-MAH on the Mechanical, Morphological and Rheological Properties of Polypropylene and Poly(Acrylonitrile-Butadiene-Styrene) Blends

  • The effects of maleic anhydride-grafted polypropylene (PP-g-MAH) addition on polypropylene (PP) and poly(acrylonitrile-butadiene-styrene) (ABS) blends were studied. Blends of PP/ABS (70/30, wt%) with PP-g-MAH were prepared by a twin-screw extruder. From the results of mechanical testing, the impact, tensile and flexural strengths of the blends were maximized at a PP-g-MAH content 3 phr. The increased mechanical strength of the blends with the PP-g-MAH addition was attributed to the compatibilizing effect of the PP and ABS blends. In the morphological studies, the droplet size of ABS was minimized (6.6 ${\mu}m$) at a PP-g-MAH content of 3 phr. From the rheological examination, the complex viscosity was maximized at a PP-g-MAH content of 3 phr. These mechanical, morphological and rheological results indicated that the compatibility of the PP/ABS (70/30) blends is increased with PP-g-MAH addition to an optimum blend at a PP-g-MAH content of 3 phr.

Development of a Novel Immunochromatographic Assay for Rapid Detection of OXA-23 β-lactamase-producing Acinetobacter baumannii

  • Among the several agents causing carbapenem resistance of Acinetobacter baumannii, the most common cause is OXA-23 ${\beta}$-lactamase, which is known to hydrolyze carbapenem. To effectively control dissemination of carbapenem-resistant Acinetobacter baumannii (CRAB), development of both rapid and easy-to-use detection methods are required. The aim of this study is to develop a novel immunochromatographic assay (ICA) for rapid detection of OXA-23 ${\beta}$-lactamase. Of the seven monoclonal antibodies (mAbs) screened by ELISA, four mAbs (4G6, 4H6, 6G4, 9A4) exhibited high reactivity. Of these four specific antibodies, the combination of 6G4/4G6 showed the greatest reactivity and this combination of mAbs (6G4/4G6 mAbs) was used to develop the OXA-23 ${\beta}$-lactamase ICA. Of 102 A. baumannii isolates tested, the OXA-23 ${\beta}$-lactamase ICA results were consistent with PCR analysis except one false positive and one false negative isolate. The overall sensitivity and specificity were 98.36% and 97.56%, respectively. In conclusion, to the best of our knowledge, we have developed the first specific antibody set to detect OXA-23 ${\beta}$-lactamase using an ICA kit. This novel ICA can be used as a reliable and easy-to-use immunological assay for detection of OXA-23 ${\beta}$-lactamase producing CRAB in clinical laboratories.
