Comparative Studies on the Effects of Fresh, White and Red Ginseng Their Effects on the Content of Several Components in Rat Serum and the Production of Lipid Peroxide in Rat Liver (수삼, 백삼 및 홍삼의 효과에 대한 비교 연구 흰쥐의 혈청내 몇가지 성분 함량 및 과산화지질생성에 미치는 영향)

  • Seventy percent ethanol extracts were prepared from 6-year-old fresh, white and red ginseng cultivated in Kangwha, and administered orally into rats for the comparison of their effects. And their effects on the production of lipid peroxide in the livers of ethanol-administered rats were measured. Red ginseng administered group showed the largest body weight increase. However, fresh, white, red ginseng extract administered group showed no significant change in concentration of total lipid, triglyceride and free cholesterol in serum. White and red ginseng extract administeration decreased blood-sugar levels significantly. Lipid peroxide content in livers of white and red ginseng administered groups was decreased significantly. Red ginseng administered group showed greater decrcase in lipid peroxide content than that of white ginseng administred group. The increase of lipid peroxide content in the livers of ethanol-administered group was inhibited by administration of fresh, white, and red ginseng extract. And the strongest-inhibitory action was observed in red ginseng administered group. Therefore, it is supposed that red ginseng has the most powerful antiaging effect.

Bioimpedance Changes in Rats with CCl4-Induced Liver Fibrosis

  • To characterize the relationship between the stage of hepatic fibrosis and bioimpedance, several electric parameters were estimated in rats with $CCl_4$-induced liver fibrosis. Thirty three Sprague-Dawley rats were intraperitoneally injected with a $CCl_4$-mineral oil mixture (1:1, 0.2 mL/100 g) twice a week for 8 weeks. The resistance(R), reactance(X), impedance(Z), and dissipation factor(D) between 1 kHz and 100 kHz were then evaluated in the livers of the rats under pentobarbital anesthesia using an HP4294A Impedance Analyzer. The rats were killed 2, 4, 6, and 8 weeks later, and their livers were classified in accordance with Ishak's scoring system. R, X, and Z changed in accordance with the progression of hepatic fibrosis and the changes were greater at lower frequencies than at higher frequencies. In comparison, the D spectrum was biphasic; D increased initially then decreased with increasing frequency. All of the parameters(R, X, Z, and D) changed in accordance with the stage of fibrosis in the livers, but D changed specifically with the progression of fibrosis. These results indicate that hepatic fibrosis may be evaluated by determining the changes in D.

Histomoniasis of chuckar partridge in Korea : A case report (바위 자고새의 흑두병 발생예)

  • The diseased Chuckar partridges that were raised at the wild bird-raising farms located in Ockchun, Chungbuk province, were submitted for diagnosis in the laboratory in July. 1990. The diseased partridges aged 3~4 weeks old were shown clinical signs such as anorexia, depression, white-colored or transparent-watery diarrhea and dropping of the head and wings. At necropsy, major gross lesions were found in the livers, ceca and lungs. Histopathologically, numerous histomonads were observed in the necrotic tissues of the livers and ceca. In the cecal specimens, it was found that the ameboid histomonads have flagellum and the size of 10~20 micrometer. The case was confirmed as histomoniasis due to the infection of Histomonas meleagridis.

Effects of TongBiYeum (TBY) on the Fatty Liver

  • Purpose : This study examined the effect of Tongbiyeum (TBY) on rats with fatty livers. Methods : After administration of TBY extracts to rats with fatty livers, body weight, liver weight, serum, cholesterol, TG, HDL-cholesterol were measured, and histopathological changes were observed. Results : In proportion to density, TBY decreased liver weight and serum TG levels and suppressed fat accumulation and liver cell death, but increased serum HDL levels in comparison with those of controls. Conclusion : TBY might be considered to be used to control fatty liver by inhibiting absorption and storage of TG.

Effect of Methanethiol on the Xanthine Oxidase Activity of Liver and Serum in Rats (흰쥐에 Methanethiol 투여가 간 및 혈청 Xanthine Oxidase 활성에 미치는 영향)

  • To evaluate the effect of methanethiol on the activity of xanthine oxidase in both liver and serum, the rats intraperitoneally received methoanethiol. Injection of methoanethiol in rats showed the more decreased xanthine oxidase activity of boty liver and serum than that of the control group. Concomitantly, the xanthine oxidase activity in livers preincubated with methanethiol was decreased in vitro. The xanthine oxidase in livers preincubated with the methoanethiol also showed more increased Km and similar Vmax value than those of the control. These results suggest that the methanethiol may induce a change in substrate binding affinity of xanthine oxidase.

  • The present study was to investigate Pb and Cd concentrations in tissues of feral pigeons in urban areas. The Pb levels in bones and livers did not differ between Seoul and Kwangju areas. However, there were significant differences in kidney and liver Cd concentrations between the study sites. These are thought to be a reflection of the metal exposure conditions in the local environment. In particular, metal levels from Seoul were compared with those obtained in the same area between 1991 and 2000 in terms of the long-term biomonitoring approaches. The mean Pb concentrations in bones decreased from 59.1 $\mu$g/wet g in 1991 to 29.5 $\mu$g/wet g in 2000, totaling a reduction in concentrations of approximately 50 percent. However, there were no significant differences in the liver Pb concentrations. Concentrations of Cd in livers and kidneys did not differ between 1991 and 2000.

Effects of Dietary Lipid on Ethoxycoumarin Metabolism in Isolated Perfused Rat Liver (식이지질의 조절이 흰쥐 적출관류간장에 의한 Ethoxycoumarin 대사기능에 미치는 영향)

  • Using isolated perfused livers obtained from rats that have been fed saturated and unsatu-rated fatty acid diets the rates of hepatic microsomal oxidation of 7-ethoxycoumarin(EC) to 7-hydroxycoumarin(HC) and the rates of subsequent conjugation of the produced HC to its glucuronide and sulfate esters have been determined. Prior to preparing the isolated perfused livers. rats were fed either fat free diet 10% beef tallow diet or 10% corn oil diet for 3 weeks. The rates of oxidation from EC to HC and also of the subsequent glucuronidation of HC were higher in the corn oil diet group than those found for the fat free and beef tallow diet groups. When the concentrations of infusing EC were increased stepwise there was a dose-dependnet increase for the release of the glucuronide form of HC metabolites at the expense of the sulfate ester form. This dose dependant shift observed for the corn oil group was more significnat than those found for other groups. These results indicate that corn oil feeding has produced enhancement in the rates of hepatic microsomal drug oxidation and glucuronide conjugation the reactions catalyzed by enzymes embedded in the hepatic microsomal membranes.

  • This study was carried out to determine the sulfamethazine residues in liver and kidney of chickens. For this experiment total 80 samples of livers and kidneys were collected at random 4 points(east area 2, west area 2) meat markets in Kyong-nam area 2 and were analysed by HPLC system. The results were as follows : 1. The average concentration of sulfamethazine residues in liver and kidney were 0.056 ppm and 0.035 ppm, respectively, the sulfamethazine residues in chicken tissue was higher in liver than kidney. 2. The sulfamethazine residues of livers were exceed 0.1 ppm in three samples and no samples were exceed than 0.1 ppm in kidney. 3. No sulfamethazine residues in liver and kidney were 14 and 25 samples respectively.

Effects of Polyacetylenes from Panax ginseng on Some Microsomal and Mitochondrial Enzymes

  • Effects of panaxydol, panaxynol and panaxytriol isolated from Panax ginseng C.A. Meyer on some enzyme activities were determined. Activities of ATPase, membrane-bound enzyme from Sarcoma 180 and rat liver were slightly inhibited by panaxydol. Activities of 5'-nucleotidase, membrane-bound enzyme and succinate cytochrome c reductase in mitochonidria from sarcoma 180 and rat livers were significantly inhibited in a dose-dependent manner by panaxynol. The inhibitory effects of panaxydol and panaxynol on succinate cytochrome c reductase activities were more potent than those on 5'-nucleotidase activities and panaxynol was found to be a very potent inhibitor of succinate cytochrome c reductase. Activities of glucose-6-phosphatase in endoplasmic reticulum from Sarcoma 180 and rat livers were not affected by all three polyacetylenes. These results suggested that the inhibitory effects of panaxydol and panaxynol on enzyme activities might contribute to their biological activities.

Effect of Restrict Feeding, Roxarsone or Its Analogues in Inducing Fatty Livers in Mule Ducks

  • This study is aimed at understanding the role of arsenic in Roxarsone in causing fatty livers in mule ducks. One hundred 10-week-old mule ducks were randomly divided into 5 groups. Ducks received 2 weeks of various treatments followed by 2 weeks of withdrawal. The treatments were non-treatment (control), 300 mg/kg Roxarsone inclusion for 2 weeks ($1^{st}$ and $2^{nd}$ week), Roxarsone inclusion for one week ($2^{nd}$ week only), restrict feeding, or Roxarsone analogue (3-nitro-4-hydroxyphenyl acid) inclusion. Results showed that feed intake and body weight in the Roxarsone groups and the restrict feeding group decreased significantly during the treatment period. However only the liver and heart weights were significantly decreased (p<0.05) in the restrict feeding group. Fatty acid synthetase (FAS) activity showed a significant decrease (p<0.05) in the Roxarsone groups and the restrict feeding group, two-week-Roxarsone treatment significantly increased NADP-malic dehydrogenase (MDH) activity compared to the restrict (p<0.05). After 2 weeks drug withdrawal, the 1-week-Roxarsone or restrict feeding group showed significantly increased (p<0.05) glucose-6-phosphate dehydrogenase (G-6-PDH) activity (p<0.05). Two-week-Roxarsone treatment significantly decreased (p<0.05) the high density lipoprotein (HDL) and increased (p<0.05) the low density lipoprotein (LDL) and very low density lipoprotein (VLDL) ratio. After drug withdrawal, the 1-week-Roxarsone or restrict feeding group showed significantly increased (p<0.05) creatine kinase (CK) activity. The 2-week-Roxarsone treatment group showed significantly increased (p<0.05) aspartate aminotransferase (AST) activity. The restrict feeding treatment group showed significantly decreased (p<0.05) total protein (TP) concentration. After drug withdrawal, the related enzyme activities in the blood that reflected the liver function were restored to the normal physiological range, except for the total bilirubin concentration and CK activity in the 1-week-Roxarsone group. This group showed a significant increase (p<0.05). Thus, the reasons for liver enlargement in the Roxarsone and restrict feeding groups were different.
