The hepatotherapeutic effect of the extract of Lycii fructus has been studied in rats against $CCl_4$ induced liver toxicity. The rats were orally treated with $CCl_4$ (corn oil/$CCl_4$ 1:1, $1m{\ell}/kg$) and then $CCl_4$ ($0.5m{\ell}/kg$) administered four times for 2 weeks. The extracts of L. fructus have been administered every day for 2 weeks after the last $CCl_4$ injection. The experimental groups consisted of negative control (G1), positive control ($CCl_4$ alone; G2), extract of L. fructus (50 mg/kg; G3, 100 mg/kg; G4, 200 mg/kg; G5), respectively. There was a significant decrement to G2 on the serum level of aspartate aminotransferase, alanine aminotransferase, and alkaline phosphatase in G5. Also, the content of thiobarbituric acid reactive substance, and phosphatidylcholine hydroperxidase, a marker of lipid peroxidation, in the liver were decreased significantly G5 and G4 compared with G2. Although, catalase or superoxide dismutase, antioxidant enzyme, in the liver were decreased significantly too, it would not be a good sign for the liver. In histopathological findings, such a hepatocellular vacuolar degeneration, lobular restructure, cellular infiltration, necrosis, and so on were shown severely in G2. However, G4 and G5 was shown a mild cytoplasmic vacuolation and inflammatory cell. In conclusion, as a protection against cell damage, lipid peroxidation and serum level, it suggested that the extract of Lycii fructus would have been a therapeutic effect of liver injury directly.
Aristolochic acid (AA), extracted from Aristolochiaceae plants, plays an essential role in traditional herbal medicines and is used for different diseases. However, AA has been found to be nephrotoxic and is known to cause aristolochic acid nephropathy (AAN). AA-induced acute kidney injury (AKI) is a syndrome in AAN with a high morbidity that manifests mitochondrial damage as a key part of its pathological progression. Melatonin primarily serves as a mitochondria-targeted antioxidant. However, its mitochondrial protective role in AA-induced AKI is barely reported. In this study, mice were administrated 2.5 mg/kg AA to induce AKI. Melatonin reduced the increase in Upro and Scr and attenuated the necrosis and atrophy of renal proximal tubules in mice exposed to AA. Melatonin suppressed ROS generation, MDA levels and iNOS expression and increased SOD activities in vivo and in vitro. Intriguingly, the in vivo study revealed that melatonin decreased mitochondrial fragmentation in renal proximal tubular cells and increased ATP levels in kidney tissues in response to AA. In vitro, melatonin restored the mitochondrial membrane potential (MMP) in NRK-52E and HK-2 cells and led to an elevation in ATP levels. Confocal immunofluorescence data showed that puncta containing Mito-tracker and GFP-LC3A/B were reduced, thereby impeding the mitophagy of tubular epithelial cells. Furthermore, melatonin decreased LC3A/B-II expression and increased p62 expression. The apoptosis of tubular epithelial cells induced by AA was decreased. Therefore, our findings revealed that melatonin could prevent AA-induced AKI by attenuating mitochondrial damage, which may provide a potential therapeutic method for renal AA toxicity.
The hepatoprotective effect of DA-9601, a quality-controlled extract of artemisisa asiatica, on liver damage induced by acetaminophen (APAP) and carbon tetrachloride ($CCI_{4}$) was investigated by means of serum-biochemical, hepatic-biochemical, and histopathological examinations. Doses of Da-9601 (10, 30, or 100 mg/kg) were administered intragastrically to each rat on three consecutive days i.e. 48 h, 24 h and 2 h before a single administration of APAP (640 mg/kg, i.p.) or $CCI_{4}$ (2 ml/kg, p.o.). Four h and 24 h after hepatotoxin treatment, the animals were sacrificed for evaluation of liver damage. Pretreatment of Da-9601 reduced the elevation of serum ALT, AST. LDH and histopathological changes such as centrilobular necrosis, vacuolar degeneration and inflammatory cell infiltration dose-dependently. Da-9601 also prevented APAP- and $CCI_{4}$-induced hepatic glutathione (GSH) depletion and $CCI_{4}$-induced increase of hepatic malondialdehyde (MDA), a parameter of lipid peroxidation, in a chemically induced liver injury by complex mechanisms which involve prevention of lipid peroxidation and preservation of hepatic GSH.
목적: 비침습적으로 간의 상태를 예측하기 위하여 여러 검사법들이 시도되고 있으나 각각의 제한점이 있다. 간 신티그라피는 방사성교질과 방사능 표지 iminodiacetic acid (IDA) 화합물이 가장 널리 사용되고 있으나, 실제 간세포의 상태를 나타내는데는 부족한 점이 있다. 최근에는 간세포 표면에 발현되는 asialoglycoprotein (ASGP) 수용체 (ASGP receptor: ASGPR)에 특이적으로 결합 할 수 있는 제제인 galactosylated serum albumin (GSA)이 간 신티그라피에 유용한 방사성의약품으로 성장하고 있으나 제한점을 가지고 있다. 본 연구는 현재 상용화된 GSA보다 유용한 ASGPR 영상용 방사성의약품인 lactosylated serum albumin (LSA)이 간세포의 ASGPR 발현 정도의 평가에 이용될 수 있는지와 조직학적 간손상정도 비침습적 평가 하는데 이용될 수 있는 방사성의약품인지를 알아보고자 시행하였다. 대상 및 방법: $^{99m}Tc$-LSA의 간기능 평가 성능을 알아보기 위하여 diethylnitrosamine (DEN)과 thioacetamide (TAA) 투여로 간손상을 일으킨 생쥐에서 생체내 분포변화를 알아보았으며, DEN 투여로 간손상을 일으킨 흰쥐에서 영상을 통하여 간 및 혈액내 방사능 분포 변화 양상을 알아보았다. 방사성의약품의 체내 분포 변화 및 간 및 혈액내 분포 변화가 간손상 여부를 잘 반영하는 지를 알아보기 위하여 간조직 검사를 시행하여 비교하였다. 결과: 체중 kg당 DEN 60 mg이 주당 1회 5번 투여된 생쥐는 광학현미경상 간손상 정도가 미약하였으며, 면역조직화학검사상 ASGPR의 발현이 높았으며, $^{99m}Tc$-LSA의 체내 분포는 정상생쥐와 유의한 차이가 없었다. 체중 kg당 DEN 180 mg이 주당 1회 2번 투여된 생쥐는 조직검사상 간조직의 괴사가 광범위하였으며, 면역조직화학검사상 ASGPR의 발현이 감소되어 있었고, $^{99m}Tc$-LSA의 체내 분포는 정상생쥐에 비해 간섭취가 감소되어 있었으며, 혈액에서의 제거나 늦었다. TAA를 투여하여 간조직의 괴사가 발생한 생쥐에서도 $^{99m}Tc$-LSA의 체내 분포는 정상생쥐에 비해 간섭취가 감소되어 있었으며, 혈액에서의 제거가 늦었다. 결론: 새로이 개발된 $^{99m}Tc$-LSA는 정상 간조직에 섭취정도가 높으며, ASGPR 발현정도에 비례하여 간섭취를 나타내며, 간손상 정도에 따라 섭취가 감소하여, 간손상 정도를 비침습적으로 잘 반영해 주는 것으로 나타나 향후 간기능 평가용 방사성의약품으로 임상에 손쉽게 쓰일 수 있을 것으로 기대된다.
본 연구는 산겨릅나무 세포배양 추출물이 D-galactosamine에 의해 유발된 간독성에 따른 보호 효과를 살펴보았다. 간 조직 내 국소적 지방 변성과 염증세포 침윤은 산겨릅나무 세포배양 추출물을 처리한 실험군에서 크게 감소되는 경향을 보였다. 또한 산겨릅나무 세포배양 추출물을 처리한 실험군은 간 손상에 의해 급격히 증가된 AST와 ALT, LDH 및 ALP의 활성과 조직 내 지질함량과 과산화지질함량이 감소되는 것으로 나타나 산겨릅나무 세포배양 추출물이 D-galactosamine으로 인한 혈중 효소활성과 조직 내 지질함량을 개선하는 것으로 조사되었다. 이와 더불어 산겨릅나무 세포배양 추출물을 처리한 실험군은 염증반응을 촉진시켜 조직 상해 및 괴사를 유도하는 TNF-α의 발현 수준이 간독성을 유발한 실험군에 비해 낮은 것으로 확인되었고 항산화효소의 활성을 효과적으로 조절하였다. 이러한 결과로 미루어 보아 산겨릅나무 세포배양 추출물은 D-galactosamine에 의한 조직 괴사를 감소시키고 혈중 효소의 활성과 조직 내 지질함량을 개선할 뿐만 아니라 염증 반응 인자의 발현과 항산화효소 활성을 조절하고 있어 간독성에 대한 보호효과가 매우 높은 것으로 사료된다.
Objectives : Alcoholic fatty liver is an early and reversible consequence of excessive alcohol consumption. The initial hepatocyte cell death stimulates subsequent inflammatory responses, leading to further liver injury and fibrosis. The objective of this study is to investigate the effects of Injinsaryung-san extract on the alcoholic fatty liver by chronic EtOH administration. Method : Male Sprague Dawley rats were used in this study. All animals were randomly divided into Normal group, treated with saline (n=10); EtOH group, treated with ethanol (n=10); EtOH+IS group, treated with ethanol+Injinsaryung-san extract (n=10). For oral administration of ethanol in Control and Sample group, the ethanol was dissolved in distilled water in concentrations of 25%(v/v). Throughout the experiment of 8 week, the rats were allowed free access to water and standard chow. Sample group were administrated by Injinsaryung-san extract daily for 8 weeks. Results : The levels of hepatic marker such as aspartate aminotransferase and alanine aminotransferase were altered. Histopathological changes were reduced and the expression of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) was markedly attenuated by Injinsaryung-san extract. Conclusion : These data suggest that Injinsaryung-san extract could be effective in protecting the liver from alcoholic fatty liver. The hepatoprotective mechanisms of Injinsaryung-san may be related to attenuation of $TNF-{\alpha}$ protein, as well as to the inhibition of inflammatory response in the liver. Therefore, Injinsaryung-san can be a candidate to protect against alcoholic fatty liver.
The purpose of this study is to evaluate the effect of Injinchunggantang-derivative on proliferation of hepatocyte in rats. Cell viability is studied by MTI assay. The gene related to cell replication such as p53, waf1, bcl-2 and $bcl-_{X_L}$ is quantitized by quantitative RT-PCR and the proteins coded by these genes are studied by Western blotting. The results are as follows. 1. The hepatocytes cultured in medium with lnjinchunggantang-derivative showed better viability compared with control grroup in MTI assay, and the hepatocytes cultured in medium with the Injinchunggantang-derivative-and-ethanol-mixed group showed better viability than the hepatocytes cultrued in 10% ethanol culture medium(control group), noting that Injinchunggantang-derivative has protective effect on hepatocyte injury. There was no dose- and time-dependence. 2. In quantitative RT-PCR, i) Bel-2 gene increased significantly both in Injinchunggantang-derivative group and in Injinchunggantang-derivative-and-ethanol-mixed group, while it showed no significant increase or decrease in other group. ii) $Bcl-_{X_L}$ gene increased significantly in Injinchunggantang-derivative group as well as in Injinchunggantang-deri vative-and-ethanol -mixed group. iii) P53 gene showed no significant increase or decrease in hepatocytes cultured in medium with 10% ethanol and in hepatocytes cultured in medium with Injinchunggantang-derivative-and-ethanol-mixed group, suggesting that 10% ethanol induced cell toxicity, thus increased p53 gene expression. iv) Wafl gene showed no significant increase or decrease in hepatocytes cutured in medium with Injinchtrnggantang-derivative, while increased in hepatocytes cultured in medium with 10% ethanol and in hepatocytes cultured in medium with Injinchtrnggantang-derivative-andethanol-mixed group, suggesting that 10% ethanol induced cell toxicity increased wafl gene expression. 3. In the study on protein by western blotting, the band of bcl-2 and $bcl-_{X_L}$ were widened in Injinchtrnggantang-derivative group. Especially the amount of $bcl-_{X_L}$ increased significantly compared with other groups. But in the study on p53 and wafl, there was no significant difference among those groups. Above study shows that Injinchunggantang-derivative has good effect on cell viability and that the genes resistant to cell death such as bcl-2 and $bcl-_{X_L}$ are induced by Injinchunggantang-derivative to resist to cell death by toxic agent And this is reconfirmed in protein study using' western blotting: These results suggest that Injinchunggantang-derivative has inhibitory effect on cell death as well as protective effect on hepatocyte. Therefore this prescription is recommended in various liver diseases such as chronic liver disease and-induced hepatic injury.
Background: Rb3 is a ginsenoside with anti-inflammatory properties in many cell types and has been reported to attenuate inflammation-related metabolic diseases such as insulin resistance, nonalcoholic fatty liver disease, and cardiovascular disease. However, the effect of Rb3 on podocyte apoptosis under hyperlipidemic conditions, which contributes to the development of obesity-mediated renal disease, remains unclear. In the current study, we aimed to investigate the effect of Rb3 on podocyte apoptosis in the presence of palmitate and explore its underlying molecular mechanisms. Methods: Human podocytes (CIHP-1 cells) were exposed to Rb3 in the presence of palmitate as a model of hyperlipidemia. Cell viability was assessed by MTT assay. The effects of Rb3 on the expression of various proteins were analyzed by Western blotting. Apoptosis levels were determined by MTT assay, caspase 3 activity assay, and cleaved caspase 3 expression. Results: We found that Rb3 treatment alleviated the impairment of cell viability and increased caspase 3 activity as well as inflammatory markers in palmitate-treated podocytes. Treatment with Rb3 dosedependently increased PPARδ and SIRT6 expression. Knockdown of PPARδ or SIRT6 reduced the effects of Rb3 on apoptosis as well as inflammation and oxidative stress in cultured podocytes. Conclusions: The current results suggest that Rb3 alleviates inflammation and oxidative stress via PPARδ-or SIRT6-mediated signaling, thereby attenuating apoptosis in podocytes in the presence of palmitate. The present study provides Rb3 as an effective strategy for treating obesity-mediated renal injury.
Objectives : Present study investigated hepatoprotective effect of Haegan-jeon extract (HE) and tried to elucidate molecular mechanism involved. According to molecular mechanism, present study optimized herbal composition of HE (op-HE) and compared in vitro and in vivo hepatoprotective effects of op-HE to HE. Methods : For in vitro experiments, HepG2 cells were exposed to arachidonic acid (AA, $10{\mu}M$) and iron ($5{\mu}M$) for inducing oxidative stress. Cell viability, GSH contents, $H_2O_2$ production, mitochondrial membrane potential, immunoblot and reporter gene assay were performed to investigate cytoprotective effects and responsible molecular mechanisms. For in vivo experiments, hepatoprotective effect of HE and op-HE were assessed on $CCl_4-induced$ liver injury mice model. Results : HE pretreatment prevented AA+iron-mediated hepatocytes apoptosis. In addition, AA+iron-induced mitochondrial dysfunction, $H_2O_2$ production, glutathione depletion were reduced by HE pretreatment. In addition, nuclear factor erythroid 2-related factor 2 (Nrf2) phosphorylation, antioxidant response element (ARE)-driven reporter gene activity, and antioxidant genes expression were increased by HE. Based on reporter gene and MTT assays, we found that op-HE consisting three medicinal herbs also significantly increased transactivation of Nrf2 and reduced the AA+iron-mediated cytotoxicity. Moreover, in $CCl_4-induced$ liver injury mice model, HE-op had an ability to ameliorate $CCl_4-mediated$ increases in serum alanine transferase and aspartate aminotransferase activity, hepatic degeneration, inflammatory cell infiltration, and collagen deposition. Hepatoprotective effects of op-HE were comparable to those of HE. Conclusions : Present study suggests that op-HE as well as HE exhibit hepatoprotective effect against oxidative stress-mediated liver injury via Nrf2 activation.
Granulocyte macrophage-colony stimulating factor (GM-CSF)는 과립구 및 대식세포뿐만 아니라 상피세포의 증식과 분화를 자극하는 당단백질이며, 최근 생산된 벼세포 유래 재조합 GMCSF(rrhGM-CSF)는 감염원으로부터 안전하고 당사슬이 매우 풍부하여 물질의 안정성 혹은 효과의 지속성을 높여 주는 것으로 알려졌다. 본 실험에서는 간 재생 능력이 우수한 흰쥐를 실험모델로 하여 간의 78%를 제거한 후, 간 재생을 유도하는 과정에서 rrhGM-CSF를 처리하고, 시간 경과에 따라 형태변화의 차이와 더불어 단백질 발현 분석법과 면역조직화학법을 이용하여 PCNA 발현에 미치는 효과에 대해서 알아보고자 하였다. rrhGMCSF는 간 재생 속도를 뚜렷이 증가시키지는 못하였으나, 대조군에 비해 실험군에서는 재생 초기에 간 세포판의 붕괴와 재구성 시기를 다소 앞당기는 것으로 관찰되었다. 증식 중인 세포에서 증가하는 것으로 알려져 있는 핵단백질인 proliferating cell nuclear antigen (PCNA)의 간 조직에서의 분포와 발현 정도를 보면 부분간절제 후 12시간과 24시간에서는 PCNA 단백질이 두 그룹에서 조금씩 발현되다가 간 절제 3일과 5일이 경과한 실험군에서 단백질이 높게 발현되었다. 간 재생이 진행될수록 간 조직 전체에서 고르게 PCNA 양성반응이 나타났으며, 대조군 보다 실험군에서 반응성이 더 뚜렷한 것으로 나타났다. 이러한 결과들로 보아 부분 간절제 후 간 재생을 유도하는 과정에서 rrhGM-CSF가 세포분열을 촉진시키는 인자들 중의 하나로 작용하여 간 재생에 효과를 나타낼 수 있을 것으로 사료된다.

④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
