수삼, 연질 홍삼, 수액 홍삼의 항산화 효과 (Antioxidant Effects of Raw Ginseng, Soft Red Ginseng, and Red Ginseng Sap)

  • 고려 인삼(Panax ginseng)은 일반적으로 안전성이 우수하고 ginsenosides or panaxosides 같은 많은 생활성적인 물질을 함유하고 있다. 특히 고려 홍삼은 교감 신경계를 안정시키고 인지력을 향상시키고 개인의 의학적 효과를 높이는 데 도움을 줄 수 있다. 새로운 가공 기술로 부드러운 홍삼을 생산하였다. 본 연구는 새로운 가공 기술에 따라 생산된 연질 홍삼이 항산화 효과를 감소시키거나 개선했는지 여부를 조사하는 데 초점을 맞췄다. 연질 홍삼과 시판 홍삼액은 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) 소거활성능에 있어 유의한 차이를 나타내지 않았다(p<0.05). 연질 홍삼은 수삼과 시판 홍삼액에 비해 DPPH (22-Diphenyl-1-pichrylic)와 OH(hydroxyl radical) 소거활성능이 높게 나타났다. OH 소거활성능은 세 그룹(수삼, 연질 홍삼과 시판 홍삼액) 간 유의한 차이를 나타내었다(p<0.05). NO (nitric oxide) 소거활성능 역시 수삼, 연질 홍삼, 홍삼 수액생산품 사이에 유의한 차이가 있었다(p<0.05). 연질 홍삼에서 많은 칼슘 결정체가 전자현미경상에서 관찰되었다. 마그네슘과 칼륨은 연질 홍상과 경질 홍삼 사이에 유의한 차이가 없었다. 연한 홍삼의 추출물은 DPPH와 OH의 존재로 인해 서로 다른 활성산소를 효율적으로 소거할 수 있어 자유 라디칼로 유도된 질환에 도움이 될 수 있다.

Comparison of physiological activities and of useful compounds between new and waste bulbs of different lily (Lilium davidii) varieties

  • Lily (Lilium davidii) is a high-yielding flowering plant. Besides roses and chrysanthemums, lily bulbs have long been used as food and in oriental medicine. However, the usage and value of cut lily bulbs has not been recognized. A bulb whose yield has been decreased is called a waste bulb, and a large amount of such bulbs is discarded every year. In this study, the functionality of waste bulbs from cut lilies was investigated to explore their potential use as a value-added product. We divided lily bulbs into two groups, one group with six varieties of new bulbs (Medusa, Siberia, Woori Tower, Yelloween, Le Reve, and Morning Star) used for cultivation and the other group with six varieties of waste bulbs (Medusa, Siberia, Woori Tower, Yelloween, Sorbonne, and Sheila). Physiological activities (${\alpha},{\alpha}$-diphenyl-${\beta}$-picrylhydrazyl: DPPH) and 3-ethlbenzthiazolne-6-sulfonic acid (ABTS) radical scavenging capability and tyrosinase inhibiting activity), the amount of total as well as eight individual phenolic compounds (chlorogenic acid, epicatechin, rutin hydrate, p-coumaric acid, kaempferol 3-O-${\beta}$-rutinoside, phloridzin dihydrate, myricetin, and quercetin), and total flavonoid content were measured in the bulbs by high performance liquid chromatography. We detected high amounts of total phenol and total flavonoid as well as high DPPH and ABTS radical scavenging ability. More tyrosinase inhibiting activity was detected in the new bulbs than in the waste bulbs. However, both the new and waste bulbs showed a higher inhibitory activity than the standard (100 ppm ascorbic acid). Although the content of phenolic compounds differed among varieties, under the conditions of the experiment, the most abundant phenolics were epicatechins, followed by chlorogenic acid, and rutins. Overall, the waste bulbs had a higher content of these compounds than the new bulbs. Based on these results, we concluded that bulbs from cut lilies could be used as functional foods in the future and farmers could expect economic gain from the hitherto neglected waste bulbs.

High Serum Level of Retinol and α-Tocopherol Affords Protection Against Oral Cancer in a Multiethnic Population

  • Background: A comparative cross-sectional study involving oral cancer patients and healthy individuals was designed to investigate associations between retinol, ${\alpha}$-tocopherol and ${\beta}$-carotene with the risk of oral cancer. Materials and Methods: This study included a total of 240 matched cases and controls where subjects were selected from the Malaysian Oral Cancer Database and Tissue Bank System (MOCDTBS). Retinol, ${\alpha}$-tocopherol and ${\beta}$-carotene levels and intake were examined by high-performance liquid chromatography (HPLC) and food frequency questionnaire (FFQ) respectively. Results: It was found that results from the two methods applied did not correlate, so that further analysis was done using the HPLC method utilising blood serum. Serum levels of retinol and ${\alpha}$-tocopherol among cases ($0.177{\pm}0.081$, $1.649{\pm}1.670{\mu}g/ml$) were significantly lower than in controls ($0.264{\pm}0.137$, $3.225{\pm}2.054{\mu}g/ml$) (p<0.005). Although serum level of ${\beta}$-carotene among cases ($0.106{\pm}0.159{\mu}g/ml$) were lower compared to controls ($0.134{\pm}0.131{\mu}g/ml$), statistical significance was not observed. Logistic regression analysis showed that high serum level of retinol (OR=0.501, 95% CI=0.254-0.992, p<0.05) and ${\alpha}$-tocopherol (OR=0.184, 95% CI=0.091-0.370, p<0.05) was significantly related to lower risk of oral cancer, whereas no relationship was observed between ${\beta}$-carotene and oral cancer risk. Conclusions: High serum levels of retinol and ${\alpha}$-tocopherol confer protection against oral cancer risk.

  • For evaluating the boar semen quality, sperm motility is an important parameter because the movement of sperm indicates active metabolism, membrane integrity and fertilizing capacity. Phospholipase C zeta (PLCz) is important enzyme in spermatogenesis, but the effect has not been confirmed in pigs yet. Therefore, this study was aimed to analyze their association with sperm motility and kinematic characteristics. DNA samples from 124 Duroc pigs with records of sperm motility and kinematic characteristics [total motile spermatozoa (MOT), curvilinear velocity (VCL), straight-line velocity (VSL), the ratio between VSL and VCL (LIN), amplitude of lateral head displacement (ALH)] were subjected. A SNP in non-coding region of PLCz g.158 A > C was associated with MOT (p < 0.05), VCL (p < 0.01), LIN (p < 0.01) and ALH (p < 0.05) in Duroc population. Therefore, we suggest that the intron region of the porcine PLCz gene may be used as a molecular marker for Duroc boar semen quality, although its functional effect was not defined yet. Whether the association is due to the candidate gene or not require further verification. Thus, it will be of interest to continue association studies in the regions surrounding those genes.

캥거루 꼬리의 영양학적 특성 (Nutritional Characteristics of Water Extract from Kangaroo Tail.)

  • 캥거루의 수입육 중 꼬리의 식품학적 이해를 위해 콜라겐 및 콘드로이친황산 함량, 단백질 및 지질 함량, 미량원소 함량 그리고 CLA 함량을 조사한 결과 황산분해시료의 콜라겐 함량은 캥거루꼬리, 소꼬리 시료 모두에서 고기보다 뼈에 월등히 함량이 높았다. 캥거루꼬리와 소꼬리의 시료 사이에는 캥거루 꼬리가 고기 및 뼈에서 모두 약 1.7배 1.2배로 높은 함량을 나타냈다. 캥거루 꼬리 고기시료 수용액 추출의 콘드로이친황산 함량 성분이 2.4배 이상 높았다. 캥거루꼬리에서 다양한 기능을 가지는 CLA는 4가지이성체가 주성분이고, 함량은 소고기꼬리의 0.92%에 비하여 5.3배 정도나 많은 4.9%의 높은 비율로 존재하였다.

미강유중(米糠油中)의 Sterol조성(組成) (Sterol Composition of Rice Bran Oil)

  • 미강유(米糠油)의 부(不)검화물(化物)을 분석(分析)하여 sterol조성(組成)을 살펴본 결과(結果)는 다음과 같았다. 4-Desmethylsterol로서는 10개가 동정(同定)되었으며 이 중(中) sitosterol이 주성분(主成分)을 이루고 있었다. 2개의 소량성분(少量成分)인 ${\Delta}^7$-stigmastenol(3%)과 ${\Delta}^7$-avenasterol(1%)를 제외(除外)하면 모두가 ${\Delta}^5$-sterol였으며 소위(所謂) ${\Delta}^5$형(型) sterol분포(分布)를 보이고 있었다. 4-monomethylsterol fraction에서는 9개가 확인(確認)되었으나 cycloeucalenol, citrostadienol, gramisterol가 주성분(主成分)이었다. 환계(環系)의 구조가 다양(多樣)하여 sterol의 생합성경로(生合成經路)의 중간단계(中間段階)임을 보여주고 있다. 4,4-Dimethylsterol(triterpene alcohol)로서는 4개가 동정(同定)되었으나 이들은 모두 9,19-cyclopropanering을 가지고 있다. 이들중(中) sterol생합성(生合成)의 초기단계(初期段階)의 물질(物質)인 cycloartenol와 24-methylenecyclo-artenol가 이 fraction의 96%를 점(占)하고 있는 것으로 봐서 미(米)강의 4,4-dimethylsterol는 짧은 경로(經路)를 거쳐 쉽게 4-monomethylsterol로 전환(轉換)되는 것 같다. Side chain에서 E-, Z-관계(關係)인 이성체(異性體)가 4-desmethylsterol로서는 fucosterol와 ${\Delta}^5$-avenasterol가 그리고 4-monomethylsterol로서는 28-isocitrostadienol와 citrostadienol로서 각각 1쌍식이 검출(檢出)된 것으로 미루어 볼때 sterol의 생합성과정(生合成過程)에서 side chain에서 보다 환계(環系)에서 먼저 변환(變換)이 일어나는 것으로 보아진다.

개선된 HPLC분석법을 이용한 세파클러 모노하이드레이트 250 mg 캡슐의 생물학적동등성 (Bioequivalence of Cefaclor Monohydrate 250mg Capsules Using an Improved HPLC Analytical Method)

  • A bioequivalence study of CKD $Cefaclor^{(R)}$ capsule (Chong Kun Dang Pharm Co., Ltd) to $Ceclor^{(R)}$ capsule (Lilly Korea Co., Ltd.) was conducted according to the guidelines of Korea Food and Drug Administration (KFDA). Twenty four healthy male Korean volunteers received each medicine at the cefaclor dose of 250 mg in a $2{\times}2$ crossover study. There was a one-week washout period between the doses. An improved high-performance liquid chromatorgraphy (HPLC) analytical method with UV detection was used to determine plasma cefaclor concentration in human volunteers for 8 hr after oral drug administration. The area under the plasma concentration-time curve from time zero to 8 hr ($AUC_{0-8hr}$) was calculated by the linear trapezoidal rule. the $C_{max}$ (maximum plasma drug concentration) and $T_{max}$ (time to reach $C_{max}$) were compiled from the plasma concentration-time data. Analysis of variance was carried out using logarithmically transformed $AUC_{0-8hr}\;and\;C_{max}$. No significant sequence effect was found for all of the bioavailability parameters indicating that the cross-over design was properly performed. The $90{\%}$ confidence intervals of the $AUC_{0-8hr}$ ratio and the $C_{max}$ ratio for CKD $Cefaclor^{(R)}$ and $Ceclor^{(R)}$ were $0.9400{\leq}{\delta}{\leq}1.0345$ and $0.8858{\leq}{\delta}{\leq}1.1021$, respectively. These values were within the acceptable bioequivalence intervals of 0.80-1.25. Thus, our study demonstrated the of CKD $cefaclor^{(R)}$ capsule was bioequivalent to $Cefaclor^{(R)}$ capsule with respect to its bioavailability.

HPLC와 DPPH radical 소거능 측정 방법의 결합에 의한 약용 식물 추출물의 항산화 활성 비교 (Measurement of the Anti-oxidative Properties of Extract from Medicinal Plants Using an On-line HPLC-DPPH Assay)

  • 본 연구에서는 일반적으로 분리 및 분석에 가장 빈번히 사용되고 있는 C18 column과 UV 검출기가 장착된 액체크로마토그래피(HPLC)와 항산화 활성 측정에 사용되는 1, 1-diphenyl-2-picryl hydrazyl (DPPH) 라디칼 소거능 측정 방법을 결합한 HPLC-DPPH 동시 측정법의 최적화와 유용성 확인을 약용식물의 추출물을 대상으로 실시하였다. 최종적으로 적용된 HPLC-DPPH 동시 측정법의 유용성은 갈근, 건강, 유근피, 모과, 황기 등 5종 약용식물의 열수추출물과 대조군으로서 ascorbic acid의 라디칼 소거능을 측정하여 확인하였다. HPLC-DPPH 동시 측정에 앞서 추출액 중 고형분 함량을 refractometer를 사용하여 측정함으로써 추출 수율에 따른 활성 차이를 보정할 수 있도록 하였다. 갈근, 모과, 유근피 열수추출물의 라디칼 소거능이 대조군으로 사용된 ascorbic acid와 비교하여 7.77%, 4.71%, 4.19%로서 다른 열수추출물보다 상대적으로 높은 것으로 확인되었다. 이와 같은 측정법은 실제 활성 성분의 분리 및 분석에 있어서 불필요한 시간 및 시약의 낭비를 줄일 수 있는 유용한 수단이 될 수 있을 것으로 판단된다.

청상견통탕(淸上甄痛湯))이 고지혈증에 미치는 영향 (The Experimental Study on the Effect of Chungsanggyuntong-tang on Hyperlipidemia)

  • Objective : In order to study effects of Chungsanggyuntong-tang on Hyperlipidemia which causes Hypertension, Arteriosclerosis, Cerebral vascular disease and Ischemic heart disease Methods : The changes of serum total lipid, triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol, body weight and organ weight were observed after the liquid extracts of Single-dosage Chungsanggyuntong-tang and Double-dosage Chungsanggyuntong-tang were administered p.o to the Hyperlipidemic rats induced by 1% cholesterol diet for 10, 20 and 30 days. Results : The food consumption and the body weight were significantly decreased in both Chungsanggyuntong-tang group compared with control group, except Double-dosage 30days. The contents of total lipid were significantly decreased in Double-dosage Chungsanggyuntong-tang group for 20, 30 days, but Single-dosage Chungsanggyuntong-tang group did not show any significant change compared with control group. The contents of total cholesterol were decreased in Single-dosage Chungsanggyuntong-tang group and Double-dosage Chungsanggyuntong-tang group, but did not show any significant changes compared with control group. The contents of triglyceride were significantly decreased in Double-dosage Chungsanggyuntong-tang group for 10, 30 days, but Single-dosage Chungsanggyuntong-tang group did not show significant changes compared with control group. The contents of HDL-cholesterol were significantly increased in Single-dosage Chungsanggyuntong-tang group for 10 days compared with control group. The contents of LDL-cholesterol were significantly decreased in both Chungsanggyuntong-tang group for 10 and 30 days compared with control group. The liver and spleen weight were significantly increased in control group compared with normal group. The liver weight was significantly decreased in Single-dosage Chungsanggyuntong-tang group compared with control group. The liver, spleen and kidney weight were significantly decreased in Double-dosage Chungsanggyuntong-tang group compared with control group. Conclusions : It was thought that Single-dosage Chungsanggyuntong-tang and Double-dosage Chungsanggyuntong-tang could be applied effectively to the Hyperlipidemia.

  • A bioequivalence study of $Ambrect^{TM}$ tablets (Dong Wha Pharm. Ind. Co., Ltd.) to $Mucopect^{TM}$ tablets (Boehringer Ingelheim Korea, Ltd.) was conducted according to the guideline of Korea Food and Drug Administration (KFDA). Twenty four healthy male Korea volunteers received each medicine at the ambroxol hydrochloride dose of 30 mg in a $2{\times}2$ crossover study. There was a one-week wash out period between the doses. Plasma concentrations of ambroxol were monitored by a high-performance liquid chromatography for over a period of 24 hours after the administration. $AUC_t$ (the area under the plasma concentration-time curve from time zero to 24 hr) was calulated by the linear trapezoidal rule method. $C_{max}$ (maximum plasma drug concentration) and $T_{max}\;(time\;to\;reach\;C_{max})$ were compiled from the plasma concentration-time data. Analysis of variance was carried out using logarithmically transformed $AUC_t\;and\;C_{max}$. No significant sequence effect was found for all of the bioavailability parameters indicating that the crossover design was properly performed. The 90% confidence intervals of the $AUC_t$ ratio and the $C_{max}$ ratio for $Ambrect^{TM}/Mucopect^{TM}$ were 0.89-1.01 and 0.89-1.02, respectively. These values were within the acceptable bioequivalence intervals of 0.80-1.25. Thus, our study demonstrated the bioequivalence of $Ambrect^{TM}\;and\;Mucopect^{TM}$ with respect to the rate and extent of absorption.
