Analysis of residual neomycin in honey by LC-MS/MS (LC-MS/MS에 의한 벌꿀 중 잔류 네오마이신의 분석)

  • An effective and specific procedure for confirmation of neomycin, aminoglycoside antibiotic in honey was developed and validated. Honey was adjusted to pH 2 with 0.1M HCl and applied to weak cation-exchange SPE cartridge. Neomycin was eluted with basified methanol. Following separation by ion-pairing liquid chromatography, neomycin was analysed with positive electrospray ionization and MRM mode. Quantification was linear over the range of $5.0{\sim}250.0{\mu}g/kg$ ($r^2$ >0.9951). The precision (R.S.D.) and accuracy (as a bias) of quality control samples in honey ranged 11.5~18.7% and 10.9~20.9%, respectively. Established method can be applied to analysis of neomycin in honey.

  • Streptomycin, which is one of aminoglycoside antibiotics, has been widely used in the rearing of food-producing animals to prevent and treat diseases in cattle, pigs and poultry. Although not licensed in South Korea, streptomycin has also been used for the treatment of bacterial honeybee disease, such as European foulbrood in Third World countries. A reliable and effective method using liquid chromatography-tandem mass spectrometry (LC-MS/MS) was developed for the determination of streptomycin in honey. A established method was optimized the clean-up and extraction procedure for the trace determination, good precision and accuracy. And the chromatographic and tandem mass spectrometric parameters were also optimized. The precision (RSD) and accuracy (bias) in the concentration range of 5.0~50.0 ug/kg were 5.5~14% and -10.0~8.0%, respectively. Limit of detection was 0.75 ug/kg and recovery of streptomycin spiked at level of 10 ug/kg in honey was 74%. The established and validated method was applied to determine streptomycin in honey which was on the market.

  • The analysis method of 3-Monochloropropane-1,2-diol (3-MCPD) compound in water was developed using liquid chromatography with mass spectrometric detection. Aqueous solution was controlled in strong basic condition with sodium hydroxide, and then $25{\mu}L$ of benzoyl chloride was added to the solution for the derivatization of 3-MCPD. The derivative was extracted using pentane and analyzed by the selected ion monitoring (SIM) method of LC-MS. The results of analyses showed that the calibration curves was in the range of 1.0 to $100{\mu}g/mL$ with a good linearity (correlation coefficient of $r^2=0.992$) and limit of detection was below $0.01{\mu}g/mL$. The recoveries of this analysis method by LC-MS were 92.3-98.0 %.

  • Objectives : Lysimachia mauritiana Lam. is known as a medicinal plant native to Korea that has antioxidant, anticancer, antibacterial, and antiviral activities. However, until now, research on the cultivation technology of L. mauritiana is insufficient, and there are no research data on the systematic cultivation method and mass production of L. mauritiana. Therefore, this study aims to establish a cultivation system of L. mauritiana. Methods : The cultivation environment of open land and facilities according to the growth of L. mauritiana was compared and tested. In addition, the equivalence of the origin collection extract and the cultivation extract was evaluated through Ultra high performance liquid chromatography (UHPLC) patterns analysis according to cultivation and comparison of the effect of inhibiting respiratory inflammation using BEAS-2B human bronchial epithelial cells. Results : The cultivation technology system was established through cultivation research of L. mauritiana raw materials. In addition, as a result of comparing and evaluating the equivalence of cultivated plants and L. mauritiana raw materials for suppressing respiratory inflammation, the same results were confirmed, and the equivalence was confirmed as a result of analyzing the UHPLC pattern with L. mauritiana raw materials. Conclusions : This study suggests that extract from cultivation research of L. mauritiana plants, which are native to Korea, can be used as a health functional food or medicine to improve respiratory health.

Isolation of Cesium and Radiation Resistance Bacteria for Bioremediation (생물정화를 위한 세슘 및 방사선 저항성 세균의 분리)

  • The global problem of handling radioactive materials is facing limitations. Eco-friendly bioremediation methods using microorganisms are being studied. This study was conducted to screen cesium-resistant microbial strains. M1 strain was selected from the soil sample by enriched culture in R2A medium containing 100 mM CsCl. In liquid medium containing above 40 mM of CsCl, the growth of M1 was inhibited in a concentration-dependent manner. Otherwise, M1 can survive up to 80mM CsCl in solid medium although the growth rate was slow and colony size was small. M1 strain was genetically identified as a strain of the genus Acinetobacter through 16S rRNA sequencing, and radiation resistance (D10 value) of M1 was found to be 0.307 kGy. These results showed that M1 strain is highly resistant to cesium and can grow in radiation environment. It was considered that M1 strain is useful in the field of biological decontamination of cesium.

Contamination Rates in Duodenoscopes Reprocessed Using Enhanced Surveillance and Reprocessing Techniques: A Systematic Review and Meta-Analysis

  • Background/Aims: Multiple outbreaks of multidrug-resistant organisms have been reported worldwide due to contaminated duodenoscopes. In 2015, the United States Food and Drug Administration recommended the following supplemental enhanced surveillance and reprocessing techniques (ESRT) to improve duodenoscope disinfection: (1) microbiological culture, (2) ethylene oxide sterilization, (3) liquid chemical sterilant processing system, and (4) double high-level disinfection. A systematic review and meta-analysis was performed to assess the impact of ESRT on the contamination rates. Methods: A thorough and systematic search was performed across several databases and conference proceedings from inception until January 2021, and all studies reporting the effectiveness of various ESRTs were identified. The pooled contamination rates of post-ESRT duodenoscopes were estimated using the random effects model. Results: A total of seven studies using various ESRTs were incorporated in the analysis, which included a total of 9,084 post-ESRT duodenoscope cultures. The pooled contamination rate of the post-ESRT duodenoscope was 5% (95% confidence interval [CI]: 2.3%-10.8%, inconsistency index [I2]=97.97%). Pooled contamination rates for high-risk organisms were 0.8% (95% CI: 0.2%-2.7%, I2=94.96). Conclusions: While ESRT may improve the disinfection process, a post-ESRT contamination rate of 5% is not negligible. Ongoing efforts to mitigate the rate of contamination by improving disinfection techniques and innovations in duodenoscope design to improve safety are warranted.

Advancing the discovery of bioactive compounds, its extraction and identification from the underexplored mollusc, Cipangopaludina lecythis (W. H. Benson, 1836)

  • The enormous diversity of molluscs has provided humans with food, colours, medicines and shells, among other resources. Molluscs have long been utilized in traditional medicine in several countries and they are a valuable source of medical supplies for many diverse communities worldwide. The purpose of this study was to identify and assess Cipangopaludina lecythis bioactive compounds in order to determine the nature of the primary ingredient that gives its medicinal properties. C. lecythis flesh and shell extracts using polar and lipophilic solvents were analyzed using liquid chromatography-mass spectrometry (LC-MS) and gas chromatography-mass spectrometry (GC-MS). This study provides the first chemical assessment of flesh and shell-opercula of C. lecythis. Chemical analysis of flesh and shell-opercula of C. lecythis clearly showed the presence of major compounds such as chitin, allantoin, linoleic acid, dihydrotachysterol, cyclotrisiloxane hexamethyl and 6-gingerol besides other minor compounds with bioactive properties of medicinal significance. Overall, this research provides good evidence that C. lecythis produce secondary metabolites with a variety of intriguing pharmacological characteristics. They have also long been a part of traditional medicine in many human cultural groups. Its usage by traditional practitioners to treat a range of human diseases is justified by the presence of numerous medicinally significant bioactive chemicals. However, more research on the bioactive compounds found in snails is necessary to standardize the extraction techniques for their detection, quantification and formulations, to validate their in vivo efficacy, and confirm their safety.

  • To investigate the physiological effect of Hibiscus sabdariffa, in vitro antioxidant activities and neuroprotective effects against high glucose-induced oxidative stress were examined. The ethyl acetate fraction (EtOAc-Fr) from H. sabdariffa contained high total phenolic contents compared with other fractions but total anthocyanin contents were lower than 80% Ethanol extract showed the highest 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical scavenging activity and malondialdehyde inhibitory effect. Furthermore, the EtOAc-Fr decreased the intracellular reactive oxygen species level, and protected the neuron-like PC12 cells from high glucose-induced cytotoxicity. The EtOAc-Fr also presented inhibitory effects against acetylcholinesterase as an acetylcholine hydrolase enzyme. Finally, chlorogenic acids as main phenolics by high performance liquid chromatography analysis.

Isolation and In Vitro Antimicrobial Activity of Low Molecular Phenolic Compounds from Burkholderia sp. MP-1 (Brukholderia sp. MP-1 에서의 페놀화합물의 분리와 항균활성의 측정)

  • An antagonistic strain, Burkholderia MP-1, showed antimicrobial activity against various filamentous plant pathogenic fungi, yeasts and food borne bacteria (Gram-positive and Gram-negative). The nucleotide sequence of the 16S rRNA gene (1491 pb) of strain MP-1 exhibited close similarity (99-100%) with other Burkholderia 16S rRNA genes. Isolation of the antibiotic substances from culture broth was fractionated by ethyl acetate (EtOAc) solvent and EtOAc-soluble acidic fraction. The antibiotic substances were purified through a silica gel, Sephadex LH-20, ODS column chromatography, and high performance liquid chromatography, respectively. Four active substances were identified as phenylacetic acid, hydrocinnamic acid, 4-hydroxyphenylacetic acid and 4-hydroxyphenylacetate methyl ester by gas chromatographic-mass spectrum analysis. The minimum inhibition of concentration (MIC) of each active compound inhibited the growth of the microorganisms tested at 250 to $2500{\mu}g\;ml^{-1}$. The antimicrobial activity of crude acidic fraction at 1 mg of dry weight per 6 mm paper disc was more effective than authentic standard mixture (four active substances were mixed with the same ratio as acidic fraction) over a wide range of bacterial test.

Historical Record of Mushroom Research and Industry in Korea

  • Two kinds of mushrooms, Gumji (金芝; Ganoderma) and Soji, were described in old book of Samguksagi (History of the three kingdoms; B.C 57~A.D 668; written by Bu Sik Kim in 1145) in Korea-dynasty. Many kinds of mushrooms were also described in more than 17 kinds of old books during Chosun-dynasty (1392~1910) in Korea. Nowadays, mushroom cultivation has been increased through out the world last decade years. Production of mushrooms has also been increased 10-20% and many varieties have been cultivated. Similar trends were also observed in Korea. Approximately two hundred commercial strains of 37 species in mushrooms were developed and distributed to cultivators. Somatic hybrid variety of oyster mushroom 'Wonhyeong-neutari' were developed by protoplast fusion, and distributed to grower in 1989. The fruiting body yield index of somatic hybrids of Pleurotus ranged between 27 and 155 compared to parental values of 100 and 138. In addition, more diverse mushroom varieties such as Phellinus baumi, Auricularia spp., Pleurotus ferulae, Hericium erinaceus, Hypsizigus marmoreus, Grifola frondosa, Agrocybe aegerita and Pleurotus cornucopiae have been attempted to cultivate in small scale cultivation. Production of mushrooms as food was 190,111 metric tons valued at 800 billion Korean Won (one trillion won if include mushroom factory products; 1dollar = 1,040 Won) in 2011. Major cultivated species are Pleurotus ostreatus, Pleurotus eryngii, Flammulina velutipes, Lentinula edodes, Agaricus bisporus, and Ganoderma lucidum, which cover 90% of total production. Since mushroom export was initiated from 1960 to 1980, the export and import of mushrooms have been increased in Korea. Technology developed for liquid spawn production and automatic cultivation systems lead to the reduction of the production cost resulting in the increasement of mushroom export. However some species were imported because of high production cost for these mushrooms requiring the effective cultivation methods. Developing of effective post-harvest system will be also directly related to mushroom export. In academic area, RDA scientists have been conducting mushroom genome projects. One of the main results is the whole genome sequencing of Flammulina velutipes for molecular breeding. An electrophoretic karyotype of of F. velutipes was obtained using CHEF with 7 chromosomes, with a total genome size of approximately 26.7 Mb. The mususcript of the genome of F. velutipes was published in PLOS ONE this year. For medicinal mushrooms, we have been conducting the genome research on Cordyceps and its related species for developing functional foods using this mushroom. In 2013, Korea Food and Drug Administraion (KFDA) approved Cordyceps mushroom for its value as an immune booster.
