  • 액체 추진제 안의 헬륨과 얼리지 안의 헬륨에 질량보존법칙을 적용하여 추진제 탱크의 평형압력을 구하는 간단하고 직관적인 방법을 제안하고, 이를 이용하여 추진제 충전 해석 프로그램을 개발하였고 기존의 참고자료와 비교하여, 검증하였다. 또한 개발된 프로그램을 현재 개발 중인 통신해양기상위성의 자료와 비교하였고, 다음 정지궤도 복합위성의 추진계 기술개발에 사용하고자 한다.

  • The present study has been conducted to simulate dynamics of a gas engine-driven heat pump (GHP) for design of control algorithm. The dynamics modeling of a GHP was based on conservation laws of mass and energy. For automatic control of refrigerant pressures, actuators such as engine speed, outdoor fans, coolant three-way valves and liquid injection valves were PI or P controlled. The simulation results were found to be realistic enough to apply for control algorithm design. The model can be applied to build a virtual real-time GHP system so that it interfaces with a real controller in purpose of prototyping control algorithm.

Simultaneous Determination of Four Compounds from Cercidiphyllum japonicum Using HPLC-UV Analysis

  • Cercidiphyllum japonicum is being used for the treatment of obesity and liver fibrosis in Korean local clinics. In the present study, we tried to develop an analytical methodology for the determination of the chemical markers of Cercidiphyllum japonicum. Four chemicals, maltol (1), chlorogenic acid (2), quercetin (3), and avicularin (4), were selected for method validation, and the analytical conditions were optimized and validated using high-performance liquid chromatography coupled with an ultraviolet detector (HPLC-UV). Additionally, the seasonal variations of four markers were monitored every month for six months. The contents of four chemicals markers were most detected in a sample collected in June.

  • Artemisia keiskeana and A. stolonifera are plants of the genus Artemisia, distributed in various regions, especially China and Korea. They are renowned as medicinal plants with biological and pharmacological activities. Fraxidin, isofraxidin, and daphnoretin are coumarins present in Artemisia spp.; however, research on them is limited. Therefore, this study was carried out to quantify the content of these compounds in the aerial parts of A. keiskeana and A. stolonifera in different regions in Korea. High-performance liquid chromatography was performed with a photodiode array detector and a reverse-phase INNO column. A. stolonifera only contained fraxidin with the highest amount found in Yongmun commune. A. keiskeana cultivation in Soyang commune gave the highest fraxidin and daphnoretin content. However, isofraxidin was not present in all samples. The findings suggest that the concentrations of the three compounds may differ depending on the growth site and provide a foundation for future studies.

Purification and Identification of Antioxidant Peptides from Enzymatic Hydrolysate of Spirulina platensis

  • The aim of this study was to isolate antioxidant peptides from an enzymatic hydrolysate of Spirulina platensis. A novel antioxidant peptide was obtained by ultrafiltration, gel filtration chromatography, and reverse-phase high-performance liquid chromatography, with the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay used to measure the antioxidant activity, and the sequence was determined to be Pro-Asn-Asn (343.15 Da) by electrospray ionization tandem mass spectrometry. This peptide was synthesized to confirm its antioxidant properties, and it exhibited 81.44 ± 0.43% DPPH scavenging activity at 100 μg/ml, which was similar to that of glutathione (82.63 ± 0.56%). Furthermore, the superoxide anion and hydroxyl free-radical scavenging activities and the SOD activity of the peptide were 47.84 ± 0.49%, 54.01 ± 0.82%, and 12.55 ± 0.75%, respectively, at 10 mg/ml. These results indicate that S. platensis is a good source of antioxidant peptides, and that its hydrolysate may have important applications in the pharmaceutical and food industries.

Regeneration of Cryopreserved Pear Shoot Tips Grown in Vitro by Encapsulation-Dehydration

  • The preservation of pear germplasm, like that of other clonal germplasms, is difficult because it requires conservation of whole plants or their tissues. Among the currently available methods for long-term conservation of clonal germplasm, cryopreservation of shoot tips is the most reliable and cost- and space-effective option. Alginate-coated axillary shoot tips from in vitro-grown pear were conserved successfully in liquid nitrogen (LN) following dehydration. Shoot recovery from cryopreserved shoot tips was improved greatly after 8 weeks of cold acclimation, but recovery decreased slightly after then. The highest regeneration rate was observed when in vitro shoot tips were preincubated in MS (Murashige and Skoog) medium with 0.3 M sucrose for 48 h, and when alginate-coated shoot tips were precultured in MS medium with increasing sucrose concentrations (0.5 M and 0.7 M) for 8 and 16 h, respectively. When the encapsulated beads were dehydrated for up to 7 h [25% water content (fresh weight basis)] under laminar flow, the highest regeneration rate was observed in "BaeYun No. 3" (55.7%) and "Whanggeum" (43.3%) after warming from LN. This technique is useful as a practical procedure to cryopreserve plant material that is sensitive to freezing of the surrounding cryoprotectant medium. Therefore, this technique appears to be promising for the cryopreservation of shoot tips from in vitro-grown plantlets of pear germplasm.

Numerical and experimental studies of cryogenic reciprocating expander without inner piston

  • It is difficult to fabricate and maintain moving parts of expander at cryogenic temperature. This paper describes numerical analysis and experimental investigation on a cryogenic reciprocating expander without moving piston. An intake valve which takes high-pressure gas, and an exhaust valve which discharges low-pressure gas, are connected to a tube. The inside pressure of the tube is pulsated for work production. This geometric configuration is similar to that of pulse tube refrigerator but without regenerator. An orifice valve and a reservoir are installed to control the phase of the mass flow and the pressure. At the warm end, a heat exchanger rejects the heat which is converted from the produced work of the expanded gas. For the numerical analysis, mass conservation, energy conservation, and local mass function for valves are used as the governing equations. Before performing cryogenic experiments, we carried out the expander test at room temperature and compared the performance results with the numerical results. For cryogenic experiments, the gas is pre-cooled by liquid nitrogen, and then it enters the pulse tube expander. The experiments are controlled by the opening of the orifice valve. Numerical analysis also found the expander conditions that optimize the expander performance by changing the intake pressure and valve timing as well as the opening of the orifice valve. This paper discusses the experimental data and the numerical analysis results to understand the fundamental behavior of such a newly developed non-mechanical expander and elucidate its potential feature for cryogenic application.

초저온 저장이 다릅나무 종자의 발아와 유묘의 생장특성에 미치는 영향 (Effects of Cryopreservation on the Seed Germination and Growth Properties of Seedlings of Maackia amurensis)

  • 본 연구는 산림 수목 종자를 대상으로 다양한 초저온 동결 조건을 적용하고, 초저온 저장 후에 나타나는 종자의 발아 특성 및 유묘의 생장 특성 조사하기 위해 수행되었다. 유리화 과정을 기초로 한 초저온 저장은 다릅나무 종자의 발아율을 크게 감소시켰으나, 초저온 저장 전 동결보호제 처리와 초저온 저장 후의 빠른 해빙은 종자의 발아율 감소를 완화시켰다. 초저온 저장 전 동결보호제 노출 시간은 종자의 발아율에 영향을 주었다. 즉 동결보호제 노출시간이 길어짐에 따라 종자의 발아율은 감소하였다. 그러나 초저온 저장 기간은 종자의 발아율에 영향을 주지 않았다. 또한 동결보호제의 노출 시간과 초저온 저장 기간은 유묘의 생장 특성에 영향을 주지 않았다. 따라서 수목 종자의 초저온 저장은 조직의 손상이나 변형이 없이 장기간 저장할 수 있는 현지 외 보존 기술로 적절하다고 판단된다. 그러나 산림 종자의 보다 효율적인 초저온 저장을 위해서는 각 수종의 종자 특성에 맞는 동결보호제 및 처리 기술이 개발되어야 할 것으로 판단된다.

부식철편에 있어서 철환원능력을 갖춘 세균의 활동에 의한 부식생성물의 변화 (A Study on the Change of the Corrosion Products by the Activity of Iron Reducing Bacteria for Corrosion Carbon Steel)

  • 토양 중에 존재하는 철환원능력을 갖춘 세균은 철제유물의 부식생성물을 에너지원으로 이용할 수 있다. 이러한 세균의 활동은 부식생성물의 변화를 초래하여 유물의 부식을 촉진시키는 부식생성물의 판단을 어렵게 할 수 있다. 본 연구의 목적은 철환원세균이 부식생성물에 일으키는 변화를 조사하여, 철제유물의 부식에 관한 이해를 높이고자 한다. 실험은 출토철제유물을 재현하기 위해서 부식촉진인자 중에서 가용성염류(염화물이온, 황산이온)를 이용하여 부식시킨 철편을 준비하였다. 이 부식철편을 세균이 존재하는 배지에서 42일간 배양하였다. 실험 후, 부식철편의 부식생성물은 SEM, EDS, XRD를 이용하여 관찰, 분석을 실시하였다. 관찰결과, 부식철편이 세균의 활동으로 인해 녹색으로 변화하였으며 부식철편에 판상 결정과 마름모꼴 결정이 새롭게 생성된 사실을 알게 되었다.

사막화방지(沙漠化防止) 및 방사기술개발(防沙技術開發)에 관한 연구(硏究)(IV) - 중국(中國)의 사구고정(沙丘固定) 및 방사공법분석(防沙工法分析) - (Studies on the Desertification Combating and Sand Industry Development(IV) - Technology Development for Sanddune Fixation and Sandy Land Conservation in China -)

  • 이 연구에서는 지난 3년간 북경임업대학교와 공동연구과제로 중국의 사막화방지 및 방사기술개발을 위한 연구의 일환으로, 특히 중국의 사구고정 및 방사공법을 조사 분석 평가하고자 수행하였다. 사구의 주요 형태에는 관목총사구, 사랑사구, 지상사구, 봉소상사구, 격자상사구, 양설상사구, 초생달사구, 포물선상사구, 초생달사연사구, 초생달사롱, 피라미드상사구, 복합형초생달형사구, 복합형종사롱사구, 돔상사구, 불규칙한 사구 사지 등 매우 다양하다. 이들 사구의 분포비율은 높이 5m이하 13%, 6~10m 17%, 11~25m 18%, 26~50m 14%, 51~100m 28%, 100m 이상 10% 등이다. 중국 건조지역에서 사구의 이동방향은 주로 주풍방향에 따르나 지역 지형 사구의 크기 및 형태 등에 따라서 이동사구형태에 차이가 많다. 사막화지에서 주요 비사방지기술(방사공법)은 생물 생태적 방사공법(식물피복방법 식재에 의한 방법), 물리적 방사공법, 화학적 방사공법 등으로 구분할 수 있는데, 생물 생태적 방사공법에는 초생법(봉사육초(封沙育草)), 관목 초생고사법(草生固沙法), 사구간 방사림대, 기간방사림대, 경지방풍림대조성 등이 있고, 물리적 방사공법에는 고방사원(高防沙垣) 및 저방사원 조성, 방사제방, 석력점토피복법 등이 있으며, 화학적 방사공법에는 고화제뿜어붙이기, 유사개량화학역제혼용법 등이 효과적인 것으로 분석되었다. 그 밖에도 관개 침사공법, 비사포착수로 구방사공법(溝防沙工法) 등이 효과적인 것으로 판단된다.
