It is well known that oxidative stress of reactive oxygen species(ROS) may be a causative factor in the pathogenesis of bone disorder. The purpose of this study was to evaluate the cytotoxicity of oxidative stress. Cell viability by MTS assay or INT assay, activity of glutathione peroxidase(GPx), lipid peroxidation(LPO) activity and cell viablity. And also protctive effect of glutamate receptors against ROS-induced osteotoxicity was examined by protein synthesis, alkaline phosphatase (ALP) activity and lactate dehydrogenase (LDH) activity in cultured rat osteoblasts and osteoclasts. XO/HX decreased cell viability and GPx activity, protein synthesis and ALP activity, but increased LPO activity and LDH activity. In the protective effect, N-methyl-D-aspartate (NMDA) receptor antagonists or AMPA/kainate receptor antagonists such as D-2-amino-5-phosphonovaleric acid (APV), 7-chlorokynurenic acid (CKA), 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX) and 6,7-dinitroquinoxaline-2,3-dione (DNQX), NMDA receptor antagonists but AMPA/kainate receptor antagonists showed protective effect on xanthine oxidase (XO) and hypoxanthine (HX) in these cultures by the increse of protein synthesis, ALP activity.

Cell viability by MTS assay or !NT assay, activity of glutathione peroxidase (GPx), lipid peroxidation (LPO) activity, protein synthesis by sulforhodamine B (SRB) assay, alkaline phosphatase (ALP) activity, lactate dehydrogenase (LDH) activity, MTS assay for NMDA (N-methyl-D-aspartate) receptor antagonist or AMPA/kainate receptor antagonist, measurement for $TNF-\alpha$, and c-fos gene expression were performed after these cells were treated with or without various cocentrations of xanthine oxidase (XO), hypoxanthine (HX), D-2-amino-5-phosphonovaleric acid (APV), 7-chlorokynurenic acid (CKA), 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX) and 6,7-dinitroquinoxaline-2,3-dione (DNQX), respectively. In this study, XO/HX showed decreased cell viability and glutathione peroxidase (GPx) activity, but it showed increased LPO activity, $TNF-\alpha$ secretion and c-fos expression. APV and CKA incresed protein sythesis and ALP activity. While, CNQX or DNQX did not show any protective effect in LDH activity or cell viability. From these results, XO/HX showed cytotoxic effect in cultured rat osteoblast or osteoclast, and also NMDA receptor antagonist such as APV or CKA was effective in blocking XO/HX-induced osteotoxicity in these cultures.
간장해에 미치는 녹각의 효과를 밝힐 목적으로 녹각을 성분별로 추출 분리하여 급여하고 galactosamine으로 간장해를 유도한 후 녹각추출물이 간기능 유지에 관여하는 효소활성 및 지질 대사에 미치는 영향을 생화학적 측면에서 고찰하였다. Cytochrome P-450은 녹각추출물 급여로 증가하였으며 갈락토사민 투여로 감소하였으나 neutral-ext.군에서 높은 증가를 보였다. Glutathione peroxidase 활성은 갈락토사민 투여로 감소하였으나, 녹각 추출물 급여로 활성이 증가하였다. 간조직의 글루타티올 함량은 녹각추출물 급여로 유의적 변동이 없었으며 갈락토사민 투여로 감소하였다. 과산화지질 함량은 대조군에 비해 각각의 녹각추출물 급여군에서 감소하여 특히 water-ext. 군에서 감소정도가 가장 현저하였다. 간조직중의 총 지방질, 총 콜레스테롤 및 트리글리세리드의 함량은 대조군에 비해 각각의 녹각추출물 급여군에서 감소하였으며, water-ext.군에서 가장 많이 하게 감소하였다. 이상의 결과로 미루어 볼 때 녹각의 water-ext.과 neutral-ext.에 함유되어 있는 성분이 간기능 유지에 관여하는 효소합성 증가를 통해 활성증가를 유도하고, 이로써 독성의 대사를 촉진시키므로 간독성을 예방할 수 있을 것으로 생각되며, 이와 같은 녹각추출물의 작용이 생체에 흡수되어진 xenobiotics의 해독작용과 연관될 것으로 생각된다.
Objectives : Oldenlandiae Diffusae herba has been used as a natural drug for tumor, inflammation and liver disease in traditional medicine. This study was performed in order to investigate the antioxidative effects of Oldenlandiae Diffusae herba methanol extract(ODHM) on acetaminophen induced acute liver injury in mice. Methods : In order to investigate the protective effect of ODHM on acute hepatic injury in vivo, ICR mice were pretreated with ODHM, and then treated with acetaminophen(500mg/kg). And the levels of LPO and glutathione(GSH), antioxidative enzyme activities were measured. The levels of LPO were measured by TBA method. And catalase activity was measured as the decrease in hydrogen peroxide absorbance at 240nm on spectrophotometer using 30mM hydrogen peroxide. Superoxide dismutase(SOD) was assayed by recording the inhibition of nitro blue tetrazolium reduction with xanthine and xanthine oxidase. Glutathione peroxidase(GPX) activity was determined by the modified coupled assay developed by Paglia and Lawrence. The reaction was started by addition of 2.2mM hydrogen peroxide as substrate. The change in absorbance at 340nm was measured for 1min on spectrophotometer. Glutathione-S-transferase(GST) activity was assayed with CDNB as substrate and enzyme activity of GST towards the glutathione conjugation of CDNB. And Total SH and GSH levels were measured. Results : In vivo study, LPO levels of acetaminophen treatment group were significantly higher than other groups. This increased level was significantly reduced by ODHM pretreatment. The acetaminophen treatment resulted in a decrease of catalase, GPX, SOD and GST activities. By contrast, ODHM pretreatment markedly increased compare to those of untreated groups. Total SH and GSH levels were reduced by of acetaminophen treatment, and ODHM pretreatment significantly increased GSH levels.
This study was designed to investigate the effects of mulberry (Morus alba L.) leaf extract (MLE) on oxygen radicals and their scavenger enzymes in brain membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (MLE-100 and MLE-300 groups) added 100 and 300 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) lecels resulted in significant decreases (13.4% and 21.1%, 12.0% and 13.4%, respectively) in brain mitochondria and microsome of MLE-100 and MLE-300 groups compared with control group. Superoxide radical ($O_2$) levels were significantly decreased about 12% in brain cytosol of MLE-300 group compared with control group. Lipid peroxide (LPO) levels were effectively inhibited (18.1% and 12.3%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Oxidized protein (OP) levels were significantly decreased (14.2%, and 10.9%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Mn-SOD activities in brain mitochondria were significantly increased (13.5% and 18.6%, respectively) in MLE-100 and MLE-300 groups, and Cu/Zn-SOD activities in brain cytosol were also effectively increased (about 17.7%) in MLE-300 groups compared with control group. GSHPx activities in brain cytosol were remarkably increased (17.2% and 23.9%, respectively) in MLE-100 and MLE-300 groups compared with control group. These results suggest that anti-aging effect of mulberry leaf extract (MLE) may play a pivotal role in attenuating a various age-related changes in brain.
This study was designed to investigate the effects of silkworm (Bombyx mori L.) powder (SWP) on oxygen radicals and their scavenger enzymes in liver membranes of rats. Sprague-Dawley(SD) male rats(160$\pm$10g) were fed basic diet (control group), and experimental diets(SWP-200 and SWP-400 groups) added 200 and 400 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) levels resulted in a consistent decreases (4.0% and 7.2%, 5.0% and 14.1%, respectively) in liver mitochondria and microsomes of SWP-200 SWP-400 groups compared with control group, and O2 radical level was significantly decreased about 12% in liver cytosol of SWP-400 group compared with control group. Lipid peroxide(LPO) levels were significantly decreased (14.4% and 9.1%, respectively) in liver mitochondria and microsomes of SWP-400 group only compared with control group. Oxidized protein (OP) levels were remarkably decreased about 12.7% and 16.3% in liver microsomes only of SWP-200 and SWP-400 groups, but significant difference between liver mitochondria could not obtained. Mn-SOD activities were remarkably increased (15.8% and 25.2%, respectively) in mitochondria of SWP-200 and SWP-400 groups, but significant difference between Cu, Zn-SOD activities in these group could be not obtained. GSHPx activity was significantly increased in liver cytosol of SWP-400 group compared with control group. These results suggest that silkworm powder may play an effective role in a attenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.
본 실험은 streptozotocin(STZ)으로 당뇨병이 유도된 쥐의 혈장 요소질소량, 지질수준 및 간 지질수준, 간 효소 활성에 천연 기능성 소재 화합물이 미치는 영향을 평가하는 것이 목적이었다. 총 콜레스테롤은 기능성 소재를 보충 급여한 실험군에서 유의적으로 낮은 수치(70.69 mg/dL)를 보였으며, 이로 인해 총 콜레스테롤에 대한 HDL 콜레스테롤의 비율이 증가(42.6에서 51.5%로) 하였다. 그러나, SOD, CAT, GSH-Px, GSH 및 LPO 활성에는 변화가 없었으며, 이는 기능성 소재의 보충 급여가 당뇨 쥐의 산화적 스트레스에는 큰 영향을 주지 않음을 의미한다. 보충 급여가 당뇨 쥐의 AST 수치 감소시켰으며, 이는 천연 기능성 소재 화합물 섭취가 간 기능 손상을 입은 STZ 유발 당뇨 쥐에 회복작용을 함을 알 수 있었다.
White-skinned sweet potato (WSSP, Ipomoea batatas L.)는 인도네시아 및 다른 나라 등에서 전통약제로 당뇨병 치료에 널리 사용되고 있다. 본 실험에서는 흰 쥐를 streptozotocin (45 mg/kg체중, i.p.)으로 당뇨병을 유발시킨 후 WSSP의 메탄올 추출물을 체중 1 kg당 Dose 100; 200 mg/kg을 경구로 투여하였다. 산화적 스트레스에 대한 보호효과를 평가하였고 그 효능을 인슐린 분비촉진제인 glimepiride (50 mg/kg 체중)와 비교해 보았다. 산화적 스트레스 평가는 WSP 메탄올 추출물과 glimepiride를 2주 투여 한 후 간장조직의 지질 과산화물(LPO)함량, 혈청 AST, ALT, total triglyceride (TG), total cholesterol (TC), 그리고 항산화효소들인 superoxide dismutase (SOD), 카탈라아제(CAT), 글루타치온 과산화물 분해효소(GPx), 글루타치온 S-전이효소(GST)활성도 등을 간장에서 측정하여 시행하였다. 당뇨 흰쥐에서 혈당, LPO 함량, AST, ALT, TG, TC 함량 등은 정상군에 비하여 그 값이 증가하였고, SOD, CAT, GPx, GST 활성도 값은 감소하였다. 당뇨 흰쥐에 WSSP 메탄올 추출물(200 mg/kg)을 2주일 동안 투여한 결과 의미있는 혈당 감소를 볼 수 있었고, LPO, TG, TC, AST, ALT 함량에서도 개선효과를 볼 수 있었다. 또한 SOD, GPx, 그리고 CAT등 항산화효소들의 활성도 증가도 나타났다. 따라서 WSSP 메탄올 추출물은 당뇨쥐의 혈당을 낮추어 산화적 스트레스를 약화시키고 당뇨로 유발된 손상을 보호해 주는 효과가 있다는 결과를 얻었다.
This study was designed to investigate the effects of mulberry leaf extract (MLE) on oxygen radicals and their scavenger enzymes in serum of rats. Sprague-Dawley (SD) male rats (160${\pm}$10g) were fed experimental diets (MLE-100 and MLE-300 groups) added 100 and 300mg/kg BW/day for 6weeks. Triglyceride (TG) levels were significantly inhibited (10% and 20%) in MLE-100and MLE-300 groups, but there were no significant differences in total, LDL-and HDL- cholesterol levels in both MLE-100 and MLE-300 groups. Hydroxyl radical ($.$OH) formations resulted in a marked decreases(20∼25%) in MLE-100 and MLE-300 groups compared with control group, while superoxide radical (O2.-)and hydrogen peroxide formations resulted in a considerable decreases(7∼10% and 5∼10%) in MLE-100 and MLE-300 groups compared with control group. Lipid peroxide (LPO)and oxidized protein(>C=O group) productions resulted in a significant decreases (10% and 6∼10%) in MLE-100 and MLE-300 groups compared with control group. Superoxide dismutase (SOD)and catalase (CAT) activities were remarkably increased (30% and 40∼55%) in MLE-100 and MLE-300 groups, but glutathione peroxidase (GSHPX) activities were significantly increased (10∼15%) in MLE-100 and MLE-300 groups compared with control group. These results suggest that anti-aging effect of mulberry leaf extract (MLE) may play a pivotal role in attenuating a various agerelated changes.
SD계 랫트를 사용하여 6주 동안 탄수화물 대용으로 50% 및 100%의 Lentinus tuber-regium(LTR)-첨가 투여그룹의 산화적 스트레스 및 방어체계에 미치는 영향을 평가하였다. LTR-50 및 LTR-100 투여그룹은 대조그룹 대비 21.7% 및 16.4%의 매우 유의적인 ${\cdot}OH$ 라디칼의 생성 억제효과가 인정되었고, 이들 두 투여그룹은 대조그룹 대비 약 10%의 $H_2O_2$의 생성 억제효과 및 $6{\sim}10%$ 정도의 바람직한 NO의 생성 억제효과가 인정되었다. 그렇지만 이들 LTR 투여에 의하여 ${O_2}^-$ 라디칼의 생성 억제효과는 인정할 수 없었다. 이들 활성산소의 공격에 의한 산화적 스트레스로서 LPO의 생성은 LTR 투여에 의하여 대조그룹 대비 $6{\sim}12%$의 유의적인 LPO의 생성 억제효과가 인정되었고, OP의 생성은 이들 LTR의 부여에 의하여 대조그룹 대비 5.0% 및 12.8%의 생성 억제효과가 인정되었다. 이들 LTR-50, LTR-100 투여그룹은 대조그룹에 비해 $15{\sim}50%$의 매우 유의적인 SOD의 활성 증가효과가 인정되었고, 또한 이들 LTR투여그룹의 GSHPx은 대조그룹 대비 $10{\sim}25%$의 매우 유의적인 GSHPx의 활성 증가효과가 인정되었다. 한편 CAT의 활성에 미치는 LTR투여의 영향도 대조그룹 대비 $60{\sim}90%$의 매우 유의적인 CAT의 활성 증가효과가 인정되었다. 이상의 결과에서 탄수화물 대용으로 사용된 유용버섯 LTR의 투여는 ${\cdot}OH$ 라디칼, $H_2O_2$ 및 NO의 생성 및 그로 인한 LPO 및 OP 같은 산화적 스트레스를 효과적으로 억제할 수 있을 뿐만 아니라 SOD, GSHPx 및 CAT 같은 방어효소의 활성을 매우 효과적으로 증가할 수 있기 때문에 노화를 매우 효과적으로 억제할 수 있을 것으로 기대된다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
