Objective : The types and functions of lipids involved in glioblastoma (GB) are not well known. Lipidomics is a new field that examines cellular lipids on a large scale and novel aplication of lipidomics in the biomedical sciences have emerged. This study aimed to investigate the potential of blood lipids for use as biomarkers for the diagnosis of GB via untargated lipidomic approach. Gaining a deeper understanding of lipid metabolism in patients with GB can contribute to the early diagnosis with GB patiens and also development of novel and better therapeutic options. Methods : This study was performed using blood samples collected from 14 patients (eight females and six males) and 14 controls (eight females and six males). Lipids were extracted from blood samples and quantified using phosphorus assay. Lipid profiles of between patients with GB and controls were compared via an untargeted lipidomics approach using 6530 Accurate-Mass Q-TOF LC/MS mass spectrometer. Results : According to the results obtained using the untargeted lipidomics approach, differentially regulated lipid species, including fatty acid (FA), glycerolipid (GL), glycerophospholipid (PG), saccharolipid (SL), sphingolipid (SP), and sterol lipid (ST) were identified between in patients with GB and controls. Conclusion : Differentially regulated lipids were identified in patients with GB, and these lipid species were predicted as potential biomarkers for diagnosis of GB.
The present study evaluated the effects of pomegranate peel (Granati pericarpium) extract on the lipid profiles and antioxidant biomarkers in mice fed a high fat and cholesterol diet: the measured biomarkers included the TBARS value, GPx, GR, SOD and GST activities. Body fat depositions were significantly decreased in the group that received pomegranate peel. In addition, the activities of GPx, GST and SOD were significantly higher in the liver and plasma of the pomegranate peel group than in the control group. Also, the pomegranate peel diet decreased lipid peroxidation of the liver and kidney. Alkaline single-cell gel electrophoresis (comet assay) showed that the DNA damage in the plasma of the pomegranate peel group was decreased compared to that of control. The present results show that a diet with added pomegranate peel exerts protective effects against oxidative DNA damage and lipid peroxidation possibly via effects on the free radical levels in mice fed a high fat and cholesterol diet.
Dietary lipid biomarkers (fatty acids, fatty alcohols, and sterols) in adult specimens were analyzed to compare and understand the feeding ecology of the euphausiid, Euphausia pacifica, from three geographically and environmentally diverse Korean waters (Yellow Sea, East China Sea, and East Sea). Total lipid content of E. pacifica from Korean waters was about 10% dry weight (DW) with a dominance of phospholipids (>46.9% of total lipid content), which are known as membrane components. A saturated fatty acid, C16:0, a monounsaturated fatty acid, C18:1(n-9), and two polyunsaturated fatty acids, C20:5(n-3) and 22:6(n-3), were most abundant (>60% of total fatty acids) in the fatty acid composition. Some of the fatty acids showed slight differences among regions although no significant compositional changes of fatty acids were detected between these regions. Phytol, originating from the side chain of chlorophyll and indicative of active feeding on phytoplankton, was detected all samples. Trace amounts of various fatty alcohols were also detected in E. pacifica. Specifically, krill from the Yellow Sea showed relatively high amounts of longchain monounsaturated fatty alcohols (i.e. 20:1 and 22:1), generally found in herbivorous copepods. Three different kinds of sterols were detected in E. pacifica. The most dominant of these sterols was cholest-5-en-$3{\beta}$-ol (cholesterol). The lipid compositions and ratios of fatty acid trophic markers are indicative of herbivory in E. pacifica from the Yellow Sea and East Sea (mainly feeding on dinoflagellates and diatoms, respectively). The lipid compositions and ratios of fatty acid trophic markers are indicative of carnivory or omnivory in E. pacifica from the East China Sea, mainly feeding on microzooplankton such as protozoa. In conclusion, lipid biomarkers provide useful information about krill feeding type. However, further analyses and experiments (i.e. gut content analysis, in situ grazing experiment, etc.) are needed to better understand the feeding ecology of E. pacifica in various marine environments.
In order to ascertain latitudinal variation of lipid contents and compositions in copepods, we collected warm water copepod species (Euchaeta sp. and Pleuromamma spp.) from four different regions from low (sub-tropical) to mid (temperate) latitudes in the Northwest Pacific Ocean. Total lipid contents of Pleuromamma spp. were about 11 $ug{\cdot}ind^{-1}$ with little latitudinal variation, whereas Euchaeta sp. showed slightly higher lipid content (20 $ug{\cdot}ind^{-1}$) than Pleuromamma spp. with latitudinal gradient (low at subtropic and high at temperate). Wax esters, known as the major storage lipid classes, were found to be the dominant lipid classes (accounting for more than 35% of total lipids) in Euchaeta sp., whereas in Pleuromamma spp., phospholipids, known as cellular membrane components, were the dominant lipid classes. However, the exception was specimens from warm pool region exhibiting dominance in storage of lipids as a form of triacylglycerols. Among fatty acids, polyunsaturated fatty acids (PUFA), especially docosahexaenoic acid (DHA : 22:6(n-3)) (about 35% of total fatty acids), were most abundant in Euchaeta sp., while saturated fatty acids (SAFA), specially hexadecanoic acid (16:0) (about 30% of total fatty acids), were most abundant in Pleuromamma spp.. Among the neutral fraction of lipids, phytol, originated from the side chain of chlorophyll, was found in all samples which generally indicate active copepods feeding on algae. While only trace amounts of short-chain fatty alcohols were found in Pleuromamma spp., significant amounts of fatty alcohols were found in Euchaeta sp.. Particularly, significant amounts of long chain monounsaturated fatty alcohols (20:1 and 22:1), generally found in cold water species, were found in Euchaeta sp. from low latitudes. The latitudinal variation of trophic lipid markers in these copepods could be significantly related with in-situ food availability and species-specific diet preference. The result of this study suggests that the lipid contents and compositions in copepods may not only indicate their nutritional condition and feeding ecology but also provide insight into species-specific living strategies under different environmental conditions (i.e. water temperature, food availability).
Different lifestyles have an impact on useful metabolic functions, causing disorders. Different lipids are involved in the metabolic functions that play various vital roles in the body, such as structural components, storage of energy, in signaling, as biomarkers, in energy metabolism, and as hormones. Inter-related disorders are caused when these functions are affected, like diabetes, cancer, infections, and inflammatory and neurodegenerative conditions in humans. During the Covid-19 period, there has been a lot of focus on the effects of metabolic disorders all over the world. Hence, this review collectively reports on research concerning metabolic disorders, mainly cardiovascular and diabetes mellitus. In addition, drug research in lipid metabolism disorders have also been considered. This review explores lipids, metabolism, lipid metabolism disorders, and drugs used for these disorders.

The present study was carried out with the hypothesis that combination of canagliflozin and omega-3 fatty acid may have potential effect on insulin level, insulin resistance, cardiac biomarkers, inflammatory cytokines and histological studies in type 2 diabetes mellitus (DM). Type 2 DM was induced by injecting nicotinamide (120 mg/kg, i.p.) 15 min before STZ (60 mg/kg) injection. Canagliflozin (5 and 10 mg/kg) and omega-3 fatty acid (300 mg/kg) were given for 28 days after confirmation of diabetes. Biochemical estimations revealed elevated levels of glucose, insulin, HOMA-R and inflammatory cytokines in diabetic group. Daily dosing of alone canagliflozin and omega-3 fatty acid slightly reduced elevated levels of glucose, insulin, HOMA-R and inflammatory cytokines ($IL-1{\beta}$, IL-2, and $TNF{\alpha}$), whereas canagliflozin and omega-3 fatty acid combination has reduced these biochemical parameters significantly when compared with diabetic group. Similarly in diabetic group the levels of cardiac biomarkers such as lipid profile, LDH, CKMB and troponin were significantly increased. Elevated levels of cardiac biomarkers were significantly reduced after daily dosing of alone canagliflozin and omega-3 fatty acid. Canagliflozin and omega-3 fatty acid combination has offered better improvement in cardiac biomarkers compared to alone canagliflozin and omega-3 fatty acid. Histopathological analysis also supported the above hypothesis that combination therapy (canagliflozin and omega-3 fatty acid) offered better protection against degenerative changes in ${\beta}-cells$ of pancreas as compared to alone treatment with these drugs. Thus the present study revealed that canagliflozin and omega-3 fatty acid can be used as potential combination therapy in type 2 DM along with cardiac complication.
Objectives: This study was designed to evaluate lipid-lowering and antioxidant effects of Artemisia capillaris(A. capillaris) using a model of hyperlipidemic rats induced by poloxamer-407. Methods: Rats were previously treated by A. capillaris water extract, and intraperitoneally injected by poloxamer-407 to induce hyperlipidemia. Parameters of serum lipid and oxidative stress biomarkers were determined. Results: 1. A. capillaris ameliorated the elevation of serum total cholesterol, triglyceride, LDL-cholesterol and MDA level. 2. A. capillaris ameliorated the reduction of serum TAC and SOD activities. 3. A. capillaris ameliorated the reduction of serum GSH and GSH-reductase level. Conclusions: According to these results, A. capillaris can be used to treat hyperlipidemia or as basis for making new drugs for treating hyperlipidemia in the future.
A lipidomic study on extensive plasma lipids in bacterial peritonitis (cecal ligation and puncture, CLP)-induced sepsis in mice was done at 24 h post-CLP. The effects of administration of lysophosphatidylcholine (LPC) and lysophosphatidic acid (LPA), compounds known to have beneficial effects in CLP, on the sepsis-induced plasma lipid changes were also examined. Among the 147 plasma lipid species from 13 lipid subgroups (fatty acid [FA], LPA, LPC, lysophosphatidylethanolamine [LPE], phosphatidic acid [PA], phosphatidylcholine [PC], phosphatidylethanolamine [PE], phosphatidylinositol [PI], monoacylglyceride [MG], diacylglyceride [DG], triacylglyceride [TG], sphingomyelin [SM], and ceramide [Cer]) analyzed in this study, 40 and 70 species were increased, and decreased, respectively, in the CLP mice. Treatments with LPC and LPA affected 14 species from 7 subgroups, and 25 species from 9 subgroups, respectively. These results could contribute to finding the much needed reliable biomarkers of sepsis.
We evaluated the effects of butterbur (Petasites japonicus Maxim) addition to the diet on lipid profiles and antioxidant biomarkers such as total glutathionine, TBARS value, carbonyl value, GPx, GR, SOD and paraoxonase activity in the plasma or liver of mice. The distribution of body fat deposition, total cholesterol (TC) contents, and atherogenic index in the plasma were significantly decreased in the butterbur group. The levels of GSH and the activity of GR and SOD were significantly higher in the liver of the butterbur group than in that of the control group. Lipid oxidation of the liver and kidney and protein oxidation of the liver and heart were decreased in the butterbur group. Additionally, the DNA damage, as determined using the comet assay (single cell gel assay) with alkaline electrophoresis and as quantified by measuring the tail length (TL), was decreased in the butterbur group. The results of the present study showed that a diet with added butterbur exerts degenerative disease-protective effects on oxidative DNA damage and lipid peroxidation.
