Characterization of a Stress-Responsive Ankyrin Repeat-Containing Zinc Finger Protein of Capsicum annuum (CaKR1)

  • We isolated many genes induced from pepper cDNA microarray data following their infection with the soybean pustule pathogen Xanthomonas axonopodis pv. glycines 8ra. A full-length cDNA clone of the Capsicum annuum ankyrin-repeat domain $C_3H_1$ zinc finger protein (CaKR1) was identified in a chili pepper using the expressed sequence tag (EST) database. The deduced amino acid sequence of CaKR1 showed a significant sequence similarity (46%) to the ankyrin-repeat protein in very diverse family of proteins of Arabidopsis. The gene was induced in response to various biotic and abiotic stresses in the pepper leaves, as well as by an incompatible pathogen, such as salicylic acid (SA) and ethephon. CaKR1 expression was highest in the root and flower, and its expression was induced by treatment with agents such as NaCl and methyl viologen, as well as by cold stresses. These results showed that CaKR1 fusion with soluble, modified green fluorescent protein (smGFP) was localized to the cytosol in Arabidopsis protoplasts, suggesting that CaKR1 might be involved in responses to both biotic and abiotic stresses in pepper plants.

The Comparison of dietary behavior and nutrients intakes with different kinds of Stress at university students of outer region of Seoul (대학생의 스트레스와 식행동 및 영양 섭취 상태 비교 - 수도권 1개 대학 재학생을 중심으로 -)

  • This study was performed to investigate the degree of different stresses in university students of outer region of Seoul and to search the influence of food habit and nutrient intakes in stress. The data was collected by questionnaire method for self-diagnosed different stresses, dietary habit, and physical status and 24 hours recall for nutrient intakes. The subjects were 282 students attended at the lecture Food and Health. Diet balance was no difference with gender but food intakes were more diverse in female. Nutrient intake of male students was lower in vitamin A and riboflavin, but that of female was low in vitamin A and iron. The university students outer region of Seoul had more stress situation in frustration, deprivation and self-efface than in the stress of the noise and Type-A behavioral stress. Total self-diagnosed dietary habit score was better in the group of no susceptive overload stress than that of the high susceptive group. Same trend of dietary behavior score was in the group of no noise and anxiety-response stress than that in the susceptive groups. The quantity of nutrient intakes was correlated with different kind of stress. Niacin intake was positively related with the stress from life-events, and food induced stress. Thiamin intake was positively related with food induced stress. But the intake of iron, calcium, niacin and riboflavin were negatively related with the stress of noise. Therefore, we could concluded that different kind of stresses were influenced in dietary behavior and vitamin-mineral nutrient intakes. More research would be needed at the aspects of health promotion in the stressful situation of modern society.

Extrapolation of extreme traffic load effects on bridges based on long-term SHM data

  • In the design and condition assessment of bridges, it is usually necessary to take into consideration the extreme conditions which are not expected to occur within a short time period and thus require an extrapolation from observations of limited duration. Long-term structural health monitoring (SHM) provides a rich database to evaluate the extreme conditions. This paper focuses on the extrapolation of extreme traffic load effects on bridges using long-term monitoring data of structural strain. The suspension Tsing Ma Bridge (TMB), which carries both highway and railway traffic and is instrumented with a long-term SHM system, is taken as a testbed for the present study. Two popular extreme value extrapolation methods: the block maxima approach and the peaks-over-threshold approach, are employed to extrapolate the extreme stresses induced by highway traffic and railway traffic, respectively. Characteristic values of the extreme stresses with a return period of 120 years (the design life of the bridge) obtained by the two methods are compared. It is found that the extrapolated extreme stresses are robust to the extrapolation technique. It may owe to the richness and good quality of the long-term strain data acquired. These characteristic extremes are also compared with the design values and found to be much smaller than the design values, indicating conservative design values of traffic loading and a safe traffic-loading condition of the bridge. The results of this study can be used as a reference for the design and condition assessment of similar bridges carrying heavy traffic, analogous to the TMB.

The Effect of Stress and Stress Coping Method on Health Related Behavior in Female University Students (여대생의 스트레스와 대처방식이 건강관련 행동에 미치는 영향)

  • Purpose : The purpose of this study was to investigate the effects of stresses to female university students and their habits of dealing with stresses through drinking alcohol and smoking. Methods : Subjects, 313 students, were selected through convenience sampling method from the 2 four-year universities in Chonbuk and Chonnam province from May to June, 2005. Data were collected through the structured questionnaires that include general characteristics, Quantity Frequency methods, the number of cigarette per day, campus stress scale, and stress coping style scale, and they were analyzed by Cronbach' alpha, descriptive statistics, ANOVA and t-test by using SPSS/PC+ program. Results: In this study, 80% of subjects have drunk alcohol. The mean frequency of drinking alcohol per month was 4.68 times and the mean number of alcohol consumption amount per drinking 6.16 glasses. The rate of smoking was 13.7%, and the mean number smoking cigarette per day in the previous month was 12.4. The mean score of stress was 2.20. Among 8 sub-factors of stress, study related stress scored highest among the sub-factors. The mean score of coping styles was 2.50. Among 4 coping styles, hopeful thought was mostly used. Among stresses, the concern of one's future affected her drinking habits. Faculty relationship, academic problem, and value affected smoking habits. Hopeful thought comes out to affect smoking, while drinking, perceived health status, and practice time showed no relationship with coping style. Conclusion: Based on the results, developing a life stress counseling program and effective coping program for women's university students is imperative, especially for those of who show passive attitude toward stress and solve it emotionally instead of using problem-oriented methods. Also, it will be necessary to study further nursing intervention to curb university females' drinking alcohol and smoking.

TGF-β1 protects colon tumor cells from apoptosis through XAF1 suppression

  • Transforming growth factor-β1 (TGF-β1) is a multifunctional cytokine that functions as a growth suppressor in normal epithelial cells and early stage tumors, but acts as a tumor promoter during malignant progression. However, the molecular basis underlying the conversion of TGF-β1 function remains largely undefined. X-linked inhibitor of apoptosis-associated factor 1 (XAF1) is a pro-apoptotic tumor suppressor that frequently displays epigenetic inactivation in various types of human malignancies, including colorectal cancer. The present study explored whether the anti-apoptotic effect of TGF-β1 is linked to its regulatory effect on XAF1 induction in human colon cancer cells under stressful conditions. The results revealed that TGF-β1 treatment protected tumor cells from various apoptotic stresses, including 5-fluorouracil, etoposide and γ-irradiation. XAF1 expression was activated at the transcriptional level by these apoptotic stresses and TGF-β1 blocked the stress-mediated activation of the XAF1 promoter. The study also demonstrated that mitogen-activated protein kinase kinase inhibition or extracellular signal-activated kinase (Erk)1/2 depletion induced XAF1 induction, while the activation of K-Ras (G12C) led to its reduction. In addition, TGF-β1 blocked the stress-mediated XAF1 promoter activation and induction of apoptosis. This effect was abrogated if Erk1/2 was depleted, indicating that TGF-β1 represses XAF1 transcription through Erk activation, thereby protecting tumor cells from apoptotic stresses. These findings point to a novel molecular mechanism underlying the tumor-promoting function of TGF-β1, which may be utilized in the development of a novel therapeutic strategy for the treatment of colorectal cancer.

  • A vehicle structure needs to be more precisely analyzed because of complexities and varieties. Structural fatigue which is generated by fluctuations of stresses during the service life of a mechanical system is the primary concern in the structural design for safety. A fatigue life is difficult to obtain in structural components during the service life of mechanical systems since the fluctuating stress contributes to fatigue. This study introduces new procedures to measure the lethargy coefficient and to predict the fatigue life of a mechanical structure by using molecular dynamic simulation. A lethargy coefficient is the total defect-estimating coefficient, which was obtained by using the results of a simple tensile test in this study. With this lethargy coefficient, fatigue life was estimated. The proposed method will be useful in predicting the fatigue life of a structurally-modified vehicle design. The effectiveness of the proposed method using lethargy coefficient measurement to predict the fatigue life of a structure was examined by applying this method to predict the fatigue life of SS41 steel, used extensively as material of vehicle structures. Two types of specimen such as pre-cracked plate and simple plate is discussed. equation of fatigue life using the lethargy coefficient and failure time, both obtained from a simple tensile test, will be useful in engineering. This measurement and prediction technology will be extended for use in analysis of any geometric shapes of modified automotive structures.

Study on the Autofrettage Pressure for SCBA Type3 Cylinder (공기호흡기용 Type3 용기의 자긴압력과 수명에 관한 연구)

  • In this study, experiments and finite element method analysis were used to determine the autofrettage pressure that is optimal and then maximizes the cycling life of Type3 composite cylinders used in self-contained breathing apparatus. For both approaches, the cylinders were pressurized at 100, 110, ${\ldots}$, 290 % of the test pressure, respectively. The stresses were computed by the FEM analysis; while the strains of cylinders were recorded and the failure modes were monitored during the cycling test. As a result, from the good agreements between the simulations and experiments, it was concluded that at least 70 % of the test pressure should be applied as the autofrettage pressure in order to takes visible effect on the cycling life, and 160 % of the test pressure induces the maximum cycling life and the desired failure mode.

  • This paper on planning constant accelerated life test is assumed that parameters for a lognormal life distribution are depended on changes of stresses. The proposed test plans are optimum in that they minimize the asymptotic variance of maximum likelihood estimator of a specified quantile at the design stress. The optimal amount of low stress level ${\xi}_{L}$ and optimal sample proportion ${\pi}$ to be allocated at low stress level are obtained when the ratio of scales at high stress level and design stress level is unknown.

  • This paper deals with the variability analysis of short term creep rupture test data based on the previous creep rupture tests and the possibility of the creep life prediction. From creep tests performed by constant uniaxial stresses at 600, 650 and $700^{\circ}C$ elevated temperature, in order to investigate the variability of short-term creep rupture data, the creep curves were analyzed for normalized creep strain divided by initial strain. There are some variability in the creep rupture data. And, the difference between general creep curves and normalized creep curves were obtained. The effects of the creep rupture time (RT) and steady state creep rate (SSCR) on the Weibull distribution parameters were investigated. There were good relation between normal Weibull parameters and normalized Weibull parameters. Finally, the predicted creep life were compared with the Monkman-Grant model.

  • A cam ring gear (CRG) system based on a hypotrochoid curve consists of a pinion with roller teeth and its conjugated internal CRG. In this study, we investigated contact forces, contact stresses, and load stress factors to predict the surface pitting life using an exact CRG profile by introducing the profile modification coefficient. The results show that the pitting life can be extended significantly by increasing the profile modification coefficient without any other change of parameters in the CRG system.
