The Influence of Light Reduction on the Growth of Microcystis aeruginosa and Variation of Environmental and Chemical Parameters in Large-scale Cultivation System

  • Large-scale cultivation of Microcystis aeruginosa in different light conditions was conducted for verifying the cell growth in a greenhouse system. Environmental and chemical parameters of the large-scale culture medium were measured for analyzing the interaction between M. aeruginosa and its symbiotic bacteria. During cultivation, a difference in cell growth pattern was observed between control (natural light) and light-limited groups (reduction of blue, green, and blue/green light, respectively). Comparing the control group, the light reduced groups showed slow and delayed cell growth through the cultivation period. Also, there is differences in the consuming pattern of total nitrogen and total phosphorus which indicated that the possibility of interaction between M. aeruginosa and symbiotic bacteria.

고구마재배의 경제성 분석 (An Economic Analysis of the Sweet Potato Cultivation)

  • This paper is aims at studying the economics of scale of sweet potato cultivation. Sweet potato is one of the decreasing crop. Its cultivated areas has been decreasing more rapidly than soybean or corn. In this paper 23 farmers were surveyed, including 8 families with less than 1 ha, 7 families with 1-5 ha, and 8 families with more than 5 ha. The results are as follows; First, sweet potato cultivation is good for specialization, considering the high income per acre. Second, there is no good evidence explaining for the economy of scale of sweet potato cultivation, but there is an evidence for the profit of specialization. Large scale farmers have more incomes per acre, lather than more production or less management costs, by means of good sales methods. Third, mechanization of sweet potato cultivation is one of the key factor for successful large scale sweet potato cultivations.

  • 대단위 오리농법 지역에서 오리방사 대신 두과작물인 헤어리베치를 사용하여 유기 벼재배 현장에서 효과 검정을 수행한 결과는 다음과 같다. 1) 유기벼 재배시 휴한기에 헤어리베치를 재배함으로서 오리농법 재배 외에 친환경 비료절감 기술로도 이용이 가능하였음. 2) 두과작물인 헤어리베치를 활용함으로서 토양의 물리성 및 작물 생산성 확보가 가능하였음. 3) 두과작물인 헤어리베치 시용으로 대단위 오리농법에 의한 농업용수 수질오염에 대한 효과도 있었음.

일본의 양식어장 이용제도에 관한 연구 -구획어업권을 중심으로- (A Study on the Utilizing System of Aquaculture Farms in Japan)

  • The objects of this study are to consider the current features and the development process of the utilizing system of aquaculture farms in Japan, and to suggest the theoretical basis for improvement of aquaculture system of Korea in the future through the analysis of background of the liberalization theory proposed recently. The aquaculture-fishery system of Japan was begun from the Meiji Era Fishery Act and New Fishery Act of the World war II. A small sum of fishery fee is paid to the fishery cooperative having fishing rights for securing fishing area newly, because the aquaculture of Japan belong to fishery cooperative not individual ownership of union membr of cooperative society, the other words, cultivation fisheries household. In case of Korea, there are several differences with Japan as follows; almost cultivation fisheries household has a individual license, the lisence of fishing rights are recognized as an article, the license of fishing lights are able to do sale. Therefore, it is needed to paid a lots of money for securing fishing area newly. On the other hand, advanced countries in the marine aquaculture such as Norway have reached the stage where the managing abilities of marine aquaculture are similar to those in the manufacturing industry. And the number of large scale aquaculture farms with developed technologies and advanced marketing strategies in those countries is increasing. Considering that the marine aquaculture in Japan under the similar fishery systems of Korea has developed the state-of-the-art management skills or lead to large scale management, it is difficult to expect the decrease in the production costs under the small scale family business in Korea and this will lead to the decreasing competitive advantage over the imported seafood. Therefore marine aquaculture in Korea needs to increase the economy of scale to acquire the competitive advantage.

지속가능한 친환경농업을 위한 광역친환경농업단지의 발전방향 (Developing Strategies of the Large-scale Districts for a Sustainable Environment-friendly Agriculture)

  • The objective of this study is to analyze the properness, effectiveness and sustainability on the large-scale environment-friendly agricultural districts (LEAD). For this, we survey 11 public officers who manage the LEAD. Most of them aware necessities of LEAD project. They have a higher satisfaction and think that the project affect ripple effect and contribution on environment-friendly agriculture. Main performance expected of the LEAD project is a spread of natural circulation agriculture in connection with cultivation and stockbreeding. For continuance of environment-friendly agriculture after the project, it is necessary to find markets of products. Also policy making related with environment-friendly agricultural technique is required. The results of this study show that the LEAD project need a modification based on field and change of agricultural issues.

Light Stress after Heterotrophic Cultivation Enhances Lutein and Biofuel Production from a Novel Algal Strain Scenedesmus obliquus ABC-009

  • Scenedesmus obliquus ABC-009 is a microalgal strain that accumulates large amounts of lutein, particularly when subjected to growth-limiting conditions. Here, the performance of this strain was evaluated for the simultaneous production of lutein and biofuels under three different modes of cultivation - photoautotrophic mode using BG-11 medium with air or 2% CO2 and heterotrophic mode using YM medium. While it was found that the highest fatty acid methyl ester (FAME) level and lutein content per biomass (%) were achieved in BG-11 medium with CO2 and air, respectively, heterotrophic cultivation resulted in much higher biomass productivity. While the cell concentrations of the cultures grown under BG-11 and CO2 were largely similar to those grown in YM medium, the disparity in the biomass yield was largely attributed to the larger cell volume in heterotrophically cultivated cells. Post-cultivation light treatment was found to further enhance the biomass productivity in all three cases and lutein content in heterotrophic conditions. Consequently, the maximum biomass (757.14 ± 20.20 mg/l/d), FAME (92.78 ± 0.08 mg/l/d), and lutein (1.006 ± 0.23 mg/l/d) productivities were obtained under heterotrophic cultivation. Next, large-scale lutein production using microalgae was demonstrated using a 1-ton open raceway pond cultivation system and a low-cost fertilizer (Eco-Sol). The overall biomass yields were similar in both media, while slightly higher lutein content was obtained using the fertilizer owing to the higher nitrogen content.

Effect of Inoculum Size on Biomass Accumulation and Ginsenoside Production by Large-Scale Cell Suspension Cultures of Panax ginseng

  • Cell growth and ginseng saponin production by large-scale suspension (bioreactor) cultures of Panax ginseng were investigated under various inoculum sizes. Cell growth was low at an inoculum size of 40 g FW/L, and the maximum cell growth was obtained with increasing inoculum size up to 100 g FW/L. The cell density of 333 g FW/L and 12.7 g DW/L was obtained at inoculum size of 100 g FW/L after 30 days of cultivation. Maximum saponin production of $4.40\;\cal{mg/g}$ DW was achieved at 60 g FW/L of inoculum size. Thus, inoculum size 60 g FW/L was suitable for optimum biomass accumulation as well as saponin production during bioreactor cultivation of ginseng suspension cells.

암모니아 산화균 및 아나목스균의 배양을 통한 파일롯 규모 단일 아나목스 반응기의 성공적인 시운전 (Successful start-up of pilot-scale single-stage ANAMMOX reactor through cultivation of ammonia oxidizing and ANAMMOX bacteria)

  • The lack of seed sludges for Ammonium Oxidizing Bacteria (AOB) and slow-growing ANaerobic AMMonium OXidation (ANAMMOX) bacteria is one of the major problem for large-scale application. In this study, $24m^3$ of single-stage SBR (Sequencing Batch Reactor) was operated to remove nitrogen from reject water using AOB and ANAMMOX bacteria cultivated from activated sludge in the field. The ANAMMOX activity was found after 44 days of cultivation in the ANAMMOX cultivation reactor, and then $0.66kg\;N/m^3/d$ of the nitrogen removal rate was achieved at $0.78kg\;N/m^3/d$ of the nitrogen loading rate at 153 days of cultivation. The AOB cultivation reactor showed $0.2kg\;N/m^3/d$ of nitrite production rate at $0.4kg\;N/m^3/d$ of nitrogen loading rate after 36 days of operation. The cultivated ANAMMOX bacteria and AOB was mixed into the single-stage SBR. The feed distribution was applied to remove total nitrogen stably in the single-stage SBR. The nitrogen removal rate in the single-stage SBR was gradually enhanced with an increase of specific activities of both AOB and ANAMMOX bacteria by showing $0.49kg\;N/m^3/d$ of the nitrogen removal rate at $0.56kg\;N/m^3/d$ of the nitrogen loading rate at 54 days of operation.

Beet western yellows virus (BWYV): Aspect of Outbreak and Survey, and First Complete Genome Sequence of a Korea Isolate of BWYV

  • In 2010, foliar symptoms were observed in the paprika leaves in Jinju city, Korea. Beet western yellows virus (BWYV) was identified in paprika by using the large-scale oligonucleotide chip assay. To investigate the occurrence of BWYV, a survey was performed on various crops, including paprika, from 2011 to 2014. Further, the presence of BWYV was consistently verified through literature survey from 2015 to 2017. BWYV infection has been identified in Solanaceae crops (bell pepper, hot pepper, and paprika), various weeds, and green peach aphids and it occurs on a nationwide scale. Cultivation using organic methods involved natural enemies and showed a high BWYV infection rate, which was more than that for conventional cultivation methods in greenhouse. The complete genome sequence of BWYV isolated from paprika was determined for the first time. The genome of the BWYV-Korea isolate consists of 5750 nucleotides and has six open reading frames. Sequence identity results showed maximum similarity between the BWYV-Korea isolate and the BWYV LS isolate (identity > 90%). This study is the first report of BWYV infecting paprika in Korea. The survey revealed that BWYV is naturalized in the domestic ecology of Korea.

미생물에 의한 생물 계면활성제 대량생산을 위한 기술개발 (Development of a Microbial Biosurfactant Production Process)

  • The microbial biosurfactants can be substituted to the chemical detergents in some industrial processes. In this study we developed a biotechnological processes for the biosurfactants with microorganisms. The biosurfactants have a lot of advantages in comparision with the chemical surfactants. They are proenvironmental even during and after industrial use. But there are not so many kinds of biosurfactants. The production cost and the end price is much higher than the chemical surfactants. But nowdays there are many kinds of microorganisms, which can produce the surfactants in large quantity and fast. We tried to develop a production process for the large scale with some microorganisms. At first Candida bombicola KCTC 7145, Sphingomonas chungbukensis KCTC 2955 and Sphingomonas yanoikuyae KCTC 2818 are cultivated and studied. For the large scale production process we used molasses as a complex medium and tried to optimize the process. Molasses contains 17 to 25% of water, 45 to 50% of sugar and 25% of carbohydrate, it can be fully used as a substrate. The microorganisms have been cultivated in the diluted media with molasses 2, 5, 8 and 10%, respectively, The optimal conditions for the cultivation and the production process have been studied. For the study the optical density, glucose concentration and the surface tension were measured. Candida bombicola KCTC 7145 and the 5% molasses media was selected as an optimal condition for the production process of a biosurfactant. During cultivation of Candida bombicola KCTC 7145 in the 5% molasses medium kerosene and corn oil were added for promoting the biosurfactants.
