Isolation and Culture of Methanotrophs in Inorganic Medium and Characterization of COD Production, Nutrient Removal (무기배지에서 메탄산화균의 분리배양과 COD 생성 및 탈질.탈인 특성 연구)

  • The objectives of this study were to isolate and culture methanotrophs and to apply them for biological removal of nitrogen and phosphorous. Methanotrophs (dominant species: Methylomonas methanica) were isolated from a landfill cover soil, cultured in a NMS medium, and analyzed to reveal their characteristics of growth and nutrient removal. The methanotrophs themselves can produce substantial amount of organic substances(as COD) including methanol, formaldehyde, and formate, as carbon sources required for denitrification. For instance, the production rate for methanol was $8\;mg/L{\cdot}hr$. Moreover, the analysis of nitrogen and phosphorous in the sludge suggested that the methanotrophs assimilate nitrogen and phosphorous as growth substances.

Comparative Evaluation of Three Culture Methods for the Isolation of Mycobacteria from Clinical Samples

  • We assessed the capacity of two liquid-medium culture methods with automated incubation and reading systems (MB/BacT ALERT 3D System and BACTEC MGIT 960 System) and one solid-medium culture method ($L\ddot{o}wenstein$-Jensen) to detect mycobacteria in different types of clinical samples. Out of 1,770 cultured clinical samples (1,519 of respiratory origin and 251 of non respiratory origin), mycobacteria were isolated in 156 samples (135 M. tuberculosis complex, 8 M. chelonae, 6 M. kansasii, 4 M. fortuitum, 2 M. gordonae, and 1 M. marinum) by at least one of the methods used. The BACTEC MGIT 960 System proved to be the most sensitive method (86.5%), especially in the detection of M. tuberculosis complex (89.1%). However, $L\ddot{o}wenstein$-Jensen culture was the most sensitive (76.2%) to detect nontuberculous mycobacteria. The BACTEC MGIT 960 System showed the lowest mean detection time for mycobacterial growth (15.3 days), significantly shorter than the other two methods. Highest sensitivity (95.5%) and specificity (99.6%) values were obtained using the BACTEC MGIT 960 System with the $L\ddot{o}wenstein$-Jensen culture method, which was also the only combination capable of detecting 100% of the nontuberculous mycobacteria.

Bacterial Communities of Biofilms Sampled from Seepage Groundwater Contaminated with Petroleum Oil

  • The diesel-degrading activities of biofilms sampled from petroleum-contaminated groundwaters in urban subway drainage systems were examined in liquid cultures, and the microbial populations of the biofilms were characterized by denaturing gel gradient electrophoresis (DGGE) and 16S rDNA sequence analysis. Biofilm samples derived from two sites (19 K and 20 K) at subway Station N and Station I could degrade around $80\%$ of applied diesel within 20 and 40 days, respectively, at $15^{\circ}C$, and these results were strongly correlated with the growth patterns of the biofilms. The closest phylogenetic neighbor of a dominant component in the 19 K biofilm was Thiothrix fructosivorans strain Q ($100\%$ similarity). Four dominant strains in the 20 K biofilm were closely related to Thiothrix fructosivorans strain Q ($100\%$ similarity), Thiothrix sp. CC-5 ($100\%$ similarity), Sphaerotilus sp. IF14 ($99\%$ similarity), and Cytophaga-Flexibacter-Bacterioides (CFB) group bacterium RW262 ($98\%$ similarity). Three dominant members in the Station I biofilms were very similar to uncultured Cytophagales clone CRE-PA82 ($91\%$ similarity), Pseudomonas sp. WDL5 ($97\%$ similarity), and uncultured CFB group bacterium LCK-64 ($94\%$ similarity). The microbial components of the biofilms differed depending on the sampling site. This is the first report on the isolation of clones highly similar to Thiothrix fructosivorans and Thiothrix sp. from biofilms in petroleum-polluted groundwaters, and the first evidence that these organisms may play major roles in petroleum degradation and/or biofilm-development.

Isolation of Chondromyces crocatus in Pure Culture (Chondromyces crocatus의 순수 분리)

  • We have isolated Chondromyces crocatus KYC2823 in pure culture and five other strains in mixed culture with companion bacteria from Korean soil samples. The strain KYC2823, which was isolated from the soil sample collected in Cheongdo-gun, Gyeongsangbuk-do, showed typical characteristics of C. crocatus, including the shape of fruiting bodies and production of a peculiar odor. In addition, the 16S rDNA sequence was 99.8% identical to that of the strain Cm c5, the proposed neotype strain of C. crocatus. Cloning and sequence analysis of the polyketide biosynthetic genes from KYC2823 by performing PCR have revealed that this strain has biosynthetic gene clusters for ajudazols (inhibitors of electron transport systems) and chondramides (substances affecting the function of the actin cytoskeleton), and biosynthetic genes for other polyketide compounds that have not been cloned yet.

Nonlinear response of stiffened triceratops under impact and non-impact waves

  • Dynamic response analysis of offshore triceratops with stiffened buoyant legs under impact and non-impact waves is presented. Triceratops is relatively new-generation complaint platform being explored in the recent past for its suitability in ultra-deep waters. Buoyant legs support the deck through ball joints, which partially isolate the deck by not transferring rotation from legs to the deck. Buoyant legs are interconnected using equally spaced stiffeners, inducing more integral action in dispersing the encountered wave loads. Two typical nonlinear waves under very high sea state are used to simulate impact and non-impact waves. Parameters of JONSWAP spectrum are chosen to produce waves with high vertical and horizontal asymmetries. Impact waves are simulated by steep, front asymmetric waves while non-impact waves are simulated using Stokes nonlinear irregular waves. Based on the numerical analyses presented, it is seen that the platform experiences both steady state (springing) and transient response (ringing) of high amplitudes. Response of the deck shows significant reduction in rotational degrees-of-freedom due to isolation offered by ball joints. Weak-asymmetric waves, resulting in non-impact waves cause steady state response. Beat phenomenon is noticed in almost all degrees-of-freedom but values in sway, roll and yaw are considerably low as angle of incidence is zero degrees. Impact waves cause response in higher frequencies; bursting nature of pitch response is a clear manifestation of the effect of impact waves on buoyant legs. Non-impact waves cause response similar to that of a beating phenomenon in all active degrees-of-freedom, which otherwise would not be present under normal loading. Power spectral density plots show energy content of response for a wide bandwidth of frequencies, indicating an alarming behaviour apart from being highly nonlinear. Heave, being one of the stiff degrees-of-freedom is triggered under non-impact waves, which resulted in tether tension variation under non-impact waves as well. Reduced deck response aids functional requirements of triceratops even under impact and non-impact waves. Stiffened group of buoyant legs enable a monolithic behaviour, enhancing stiffness in vertical plane.

Everyday Information Practices of 'Isolated' Adolescents: A Case Study of New Korean Immigrant Adolescents in the U.S. (소외 청소년의 일상적 정보행태: 재미 한인 청소년을 사례로 하여)

  • The purpose of the study is to explore how isolated immigrant adolescents seek and use necessary information when they are unable to use significant information sources-their peer groups-in the period of transition before new peer groups are established. Sixteen recently arrived Korean immigrant adolescents were recruited and a mixed method including surveys and in-depth interviews was used through three research phases. This study gained a preliminary understanding of isolated immigrant adolescents' information world: how they interpret their current situations and daily hassles, seek (or do not seek), and utilize information to cope with their daily life problems, and evaluate their use of information, including library systems and interpersonal sources. Five main emergent themes were analyzed from the findings and pertinent theories/models to interpret these unique features were suggested and discussed. The contribution and limitation of the study and future study are suggested and discussed.

  • A communication is one of the most important factors for the human beings. Therefore, people who have some problems in communication need an Augmentative & Alternative Communication (AAC) intervention to live in the human societies and several AAC systems have been developed to help the people. Lately, smart-phone is widely used to people as a communication device which can connect between persons. It can provide many useful methods and Apps to the user. If the AAC system is unified to the smart-phone, it is very effective device to the people with disabled because they can reduce the excessive expense and easily accommodate themselves to the AAC system. In this paper, an AAC program which is installed in the smart-phone or tablet PC is introduced. It has basic AAC solutions and can send SMS message to other people. We expect that the developed device can help the communication of the people with disabled and solve the social isolation of the disabled person.

Bacterial regrowth in biofilms formed in granular activated carbon filter adsorbers and the bacterial isolation and identification (입상 활성탄 여과지에서 세균의 재성장과 생물막 형성 세균의 분리 및 동정)

  • This study aimed to investigate the biofilm formation, bacterial regrowth, and bacterial community structure in the granular-activated carbon (GAC) filter adsorbers (FAs) used in water treatment plants. In 2005 and 2006, raw water, settled water, GAC FA by depth, and filtered water were collected twice a year from water treatment plants (WTPs) B and S. The number of heterotrophic bacteria, including mesophilic and psychrophilic bacteria, in such collected waters was investigated along with the total number of coliforms therein. Heterotrophic bacteria were detected in most samples, mainly at the surface layers of the GAC FAs, and fewer such bacteria were found in the lower and bottom layers. An increase in the bacterial number, however, was observed in the samples from various depths of the GAC FAs in WTPs B and S compared with the surface layers. An increase in the bacterial number was also detected in the filtered water. This may indicate that there is a regrowth of the bacteria in the GAC FA. Considering, however, that heterotrophic bacteria were not found in the filtered water, it can be deduced that most bacteria are removed in the chlorination process. Coliforms were detected at the surface layer of the GAC FAs, but their regrowth was not observed. MicroLog systems were used to identify the bacteria community distribution. Eight genera and 14 species, including Pseudomonas spp., were detected in WTP B, and 8 genera and 9 species, including Aeromonas spp., in WTP S. Further studies are required to elucidate their role in the biofilms in water treatment processes.

Design of Wideband Facility Power-line EMI filter (광대역 설비용 전원선 EMI 필터의 설계)

  • Facility power-line electromagnetic interference (EMI) filters are used to provide radio frequency(RF) isolation between dedicated power distribution services and noise sensitive local power distribution systems. This type of EMI filters generally needs a high current capacity and, in some special applications, a high insertion loss of a minimum 100dB from 14kHz through 10GHz per MIL-STD 220 is required. This paper deals with an analysis and design of a wideband facility power-line EMI filter with the above requirements. The characteristics of the inductor and capacitor at high frequencies are investigated. The characteristics and design method of the facility EMI filter with a high order LC network are also presented. The prototype filter is finally implemented and its performance is verified from the experimental results.

A New Photovoltaic System Architecture of Module-Integrated Converter with a Single-sourced Asymmetric Multilevel Inverter Using a Cost-effective Single-ended Pre-regulator

  • In this paper, a new architecture for a cost-effective power conditioning systems (PCS) using a single-sourced asymmetric cascaded H-bridge multilevel inverter (MLI) for photovoltaic (PV) applications is proposed. The asymmetric MLI topology has a reduced number of parts compared to the symmetrical type for the same number of voltage level. However, the modulation index threshold related to the drop in the number of levels of the inverter output is higher than that of the symmetrical MLI. This problem results in a modulation index limitation which is relatively higher than that of the symmetrical MLI. Hence, an extra voltage pre-regulator becomes a necessary component in the PCS under a wide operating bias variation. In addition to pre-stage voltage regulation for the constant MLI dc-links, another auxiliary pre-regulator should provide isolation and voltage balance among the multiple H-bridge cells in the asymmetrical MLI as well as the symmetrical ones. The proposed PCS uses a single-ended DC-DC converter topology with a coupled inductor and charge-pump circuit to satisfy all of the aforementioned requirements. Since the proposed integrated-type voltage pre-regulator circuit uses only a single MOSFET switch and a single magnetic component, the size and cost of the PCS is an optimal trade-off. In addition, the voltage balance between the separate H-bridge cells is automatically maintained by the number of turns in the coupled inductor transformer regardless of the duty cycle, which eliminates the need for an extra voltage regulator for the auxiliary H-bridge in MLIs. The voltage balance is also maintained under the discontinuous conduction mode (DCM). Thus, the PCS is also operational during light load conditions. The proposed architecture can apply the module-integrated converter (MIC) concept to perform distributed MPPT. The proposed architecture is analyzed and verified for a 7-level asymmetric MLI, using simulation results and a hardware implementation.
