Relationships between Pork Quality Traits and Growth Factor Concentrations in Serum and Longissimus dorsi Muscle before and at Slaughter in Female Market Pigs

  • The present study was conducted to test a hypothesis that pork quality traits would be influenced by the systemic and/or local bioavailability of insulin-like growth factor-I (IGF-I), transforming growth factor-${\beta}1$ (TGF-${\beta}1$), or epidermal growth factor (EGF) before or at slaughter. To this end, 60 cross-bred female market pigs weighing approximately 110 kg were slaughtered, after which Longissimus dorsi muscle (LM) samples taken at slaughter (D 0) and blood samples taken at D -7 and D 0 were analyzed. The 60 carcasses rendered 36 RFN (reddish-pink, firm, and non-exudative), 16 RSE (reddish-pink, soft, and exudative), and 6 PSE (pale, soft, and exudative); 2 DFD (dark, firm, and dry) also were found but were excluded in subsequent experiments. The $L^*$ and drip loss were greater in PSE vs. RFN and RSE and in PSE and RSE vs. RFN, respectively, as they should (P<0.05). The $pH_{45min}$ was less in PSE vs. RFN (P<0.05); $pH_{24h}$ tended to be less in the former (P=0.09). The LM IGF-I and TGF-${\beta}1$ as well as serum EGF concentrations were less in PSE than in RFN. None of the other LM and serum concentrations of the three growth factors differed across the three pork quality categories. The LM IGF-I and TGF-${\beta}1$ concentrations and serum EGF concentration at D 0 were negatively correlated with drip loss [r = -0.36(P<0.01), -0.44 (P<0.01), and -0.32 (P<0.05), respectively]. However, none of the serum and LM growth factor variables was correlated with $L^*$ or $a^*$ (redness) of LM. Taken together, results suggest that locally expressed IGF-I and TGF-${\beta}1$ and blood-borne EGF may have a beneficial effect on postmortem water holding capacity of the muscle and that pork quality traits could be predicted to some extent from concentrations of IGF-I and TGF-${\beta}1$ in muscle and EGF in serum at slaughter.

예측성인신장이 작은 조기사춘기 여아에서 성선자극호르몬 방출호르몬 효능약제와 성장호르몬 병합치료의 성장획득 효과 (Growth promoting effect of combined gonadotropin releasing hormone analogue and growth hormone therapy in early pubertal girls with predicted low adult heights)

  • 목 적 : 조기사춘기 여아에게 성선자극호르몬 방출호르몬 효능약제(gonadotropin releasing hormone analogue, GnRHa)와 성장호르몬(growth hormone, GH)의 병합치료를 시행하여 성인신장획득에 대한 효과를 알아보고자 하였다. 방 법 : 2001년 1월부터 2004년 1월까지 조기사춘기로 소아과 외래를 내원한 여아 중 예측성인신장이 -2 표준편차점수 이하로 표적키보다 훨씬 작은 23명($9.73{\pm}1.59$세)을 대상으로 초경을 시작하지 않은 군(Group 1, n=19, $9.59{\pm}1.59$세)과 초경을 시작한 군(Group 2, n=4, $10.38{\pm}1.44$세)으로 나누어 GnRHa와 GH의 병합치료를 시행하고, 치료 시작 전후 및 두 군 간의 역연령, 골연령, 성장속도, 신장, 예측성인신장, 혈청 IGF-1, IGFBP-3 치, BMI를 비교 분석하였다. 결 과 : 치료 시작 시의 두 군의 역연령, 성장속도, BMI는 유의한 차이는 없었으나 골연령 및 골연령과 역연령의 차이는 Group 2에서 Group 1보다 유의하게 증가되어 있었다. 신장은 Group 1의 평균이 $130.7{\pm}7.4cm$, Group 2의 평균이 $140.7{\pm}6.0cm$으로 유의하게 차이가 있었으나 표적키, 예측성인신장, 혈청 IGF-1과 IGFBP-3은 유의한 차이가 없었다. 치료 종료 시의 Group 1의 골연령은 평균 $12.18{\pm}1.02$세, Group 2의 골연령은 평균 $13.56{\pm}0.83$세로 치료 시작 전과 마찬가지로 Group 2에서 유의하게 높았으나, 골연령과 역연령의 차이, 신장, 그리고 IGF-1은 차이가 없었다. 전체에서 골연령과 역연령의 차이가 치료 시작 전에 평균 $2.50{\pm}2.46$세, 치료 종료 후에 평균 $1.10{\pm}0.97$세로 감소되었고, 역연령에 대한 골연령의 비도 $1.22{\pm}0.16$에서 $1.10{\pm}0.10$으로 감소되어서 결국 골연령의 증가는 현저히 감소하였다. 성장 속도 역시 평균 $7.46{\pm}2.44cm$/년에서 $5.65{\pm}2.08cm$/년으로 감소하였다. 예측성인신장은 치료 전에 평균 $146.6{\pm}3.50cm$, 치료 후에 평균 $153.8{\pm}4.7cm$으로 현저히 증가하였으며 신장의 표준편차점수도 $-0.66{\pm}1.02$ SDS에서 $-0.34{\pm}1.04$ SDS로 유의하게 증가하였다. BMI는 치료 전 평균 $18.80{\pm}2.35kg/m^2$, 치료 후에 평균 $20.4{\pm}2.53kg/m^2$로 유의하게 증가하였고, 혈청 IGF-1은 치료 후에 유의하게 증가하였으나 IGFBP-3는 유의한 변화가 없었다. 치료 전후에 두 군에서 각 성장 변수의 변화를 보면 골연령과 역연령의 차이는 유의하게 감소하였고 신장, 예측성인신장, BMI는 유의하게 증가하였다. 그러나 혈청 IGF-1의 경우, Group 1에서는 치료 후에 현저히 증가하였으나 Group 2에서는 별 차이가 없었다. 혈청 IGFBP-3는 모두에서 유의한 차이가 없었다. 결 론 : 조기사춘기 여아에게 GnRHa와 GH의 병합치료를 시행했을 때, 골연령의 증가 속도와 성장속도가 감소하고, 혈청 IGF-1이 증가함으로써 예측성인신장이 의미 있게 증가하며 표적키에 가깝게 증가한다. 또한 IGF-1의 변화를 보면 초경을 시작하지 않은 조기사춘기 여아가 초경을 시작한 경우보다 성장획득에 대한 가능성이 더 클 것으로 생각된다.

한약제제의 골성장 효과에 대한 국내외 실험 연구 고찰 (A Literature Review of Experimental Study about Herbal Medicine for Bone Growth)

  • Objectives The purpose of this study is to investigate the effect of herbal medicine on bone growth and growth factors by collecting and analyzing domestic and foreign experimental research papers to establish a platform for future research. Methods Experimental studies published before March 2021 were searched using 7 Korean databases and 2 foreign databases. Among the numbers of studies searched, 48 experimental studies met the criteria and were selected. The results of those 48 studies upon herbal medicine administration were analyzed by bone growth, growth plate, weight. Results 23 studies mainly used monotherapy of either Astragali Radix and Cervi Parvum Cornu. 28 studies used combination of Poria Sclerotium, Dioscoreae Rhizoma, and Acanthopanacis Cortex. Among all studies selected, weight, bone growth, Insulin-like growth factor-1 (IGF-1) and Growth hormone (GH) were increased, but the increase was significant only in some of the studies. Conclusions The result of this study can be utilized as a background information for further studies and treatment in pediatric growth. Further randomized control studies are needed to underscore the finding.

Matrix Degradative Enzymes and Their Inhibitors during Annular Inflammation : Initial Step of Symptomatic Intervertebral Disc Degeneration

  • Objective : Symptomatic disc degeneration develops from inflammatory reactions in the annulus fibrosus (AF). Although inflammatory mediators during annular inflammation have been studied, the roles of matrix metalloproteinases (MMPs) and their inhibitors have not been fully elucidated. In this study, we evaluated the production of MMPs and tissue inhibitors of metalloproteinase (TIMPs) during annular inflammation using an in vitro co-culture system. We also examined the effect of notochordal cells on annular inflammation. Methods : Human AF (hAF) pellet was co-cultured for 48 hours with phorbol myristate acetate-stimulated macrophage-like THP-1 cells. hAF pellet and conditioned media (CM) from co-cultured cells were assayed for MMPs, TIMPs, and insulin-like growth factor (IGF)-1 levels using real-time reverse-transcriptase polymerase chain reaction and enzyem-linked immunosorbent assay. To evaluate whether notochordal cells affected MMPs or TIMPs production on annular inflammation, hAF co-cultured with notochordal cells from adult New Zealand White rabbits, were assayed. Results : MMP-1, -3, -9; and TIMP-1 levels were significantly increased in CM of hAF co-cultured with macrophage-like cells compared with hAF alone, whereas TIMP-2 and IGF-1 levels were significantly decreased (p<0.05). After macrophage exposure, hAF produced significantly more MMP-1 and -3 and less TIMP-1 and -2. Interleukin-$1{\beta}$ stimulation enhanced MMP-1 and -3 levels, and significantly diminished TIMP-2 levels. Co-culturing with rabbit notochordal cells did not significantly influence MMPs and TIMPs production or COL1A2 gene expression. Conclusion : Our results indicate that macrophage-like cells evoke annular degeneration through the regulation of major degradative enzymes and their inhibitors, produced by hAF, suggesting that the selective regulation of these enzymes provides future targets for symptomatic disc degeneration therapy.

특발성 저신장증 소아에서 성장호르몬의 치료효과 (Effects of Growth Hormone Therapy in Children with Idiopathic Short Stature)

  • 목 적 : 최근까지 성장호르몬 결핍증으로 진단 받은 소아에서의 성장호르몬 치료는 그 효과를 인정받아 왔으나 성장호르몬 자극 유발 검사에서 유의한 결과를 보이지 않는 특발성 저신장증은 그 원인이 성장호르몬의 부분적 결핍 또는 감수성의 부족 때문이므로 성장호르몬의 치료 효과가 기대되나 성장호르몬 투여가 인정되어 오지 못한 현실이다. 특발성 저신장 소아에서의 성장호르몬의 효과에 대하여 몇몇 국외 연구 논문들이 발표되어 왔으나 국내에서는 거의 없는 실정으로 이에 저자들은 국내 소아를 대상으로 국내에서 생산된 성장호르몬을 투여하기 전과 후의 여러 성장 관련인자들 및 신장을 비교 분석하여 특발성 저신장증에서의 성장호르몬 치료효과에 대하여 검증하고자 하였다. 방 법 : 충북대학교병원에서 최소 1년 이상 성장호르몬 투여를 받은 특발성 저신장증 환아 15명을 대상으로 하여 1년 치료군과 2년 치료군으로 나누어 투여 전 및 후의 여러 성장 관련변수들 및 신장의 변화를 검토하여 성장호르몬의 효과를 비교 분석하였다. 결 과 : 15명의 소아 중 남아가 7명, 여아가 8명이었고 평균 연령은 $11.44{\pm}2.81$세(4.6-12.73세)이었다. 성장호르몬을 사용하기 전에 1년 치료군과 2년 치료군 사이의 역 연령, 골 연령, 신장 표준 편차치, 예측 성인 신장의 표준 편차치, 표적 신장의 표준 편차치, IGF-I, 그리고 IGFBP-3의 차이는 없었다. 또한 성장호르몬 투여 1년 후의 각종 성장 관련변수들의 비교 결과 1년 치료군과 2년 치료군 사이의 유의한 차이는 없었다. 그러나 성장호르몬 투여 전후의 신장의 변화를 비교하였을 때 1년 치료군과 2년 치료군 모두에서 의미 있는 증가를 보였다. 또한 각종 성장 관련 변수들도 두 군 모두에서 증가를 보였으나 2년 치료군에서는 IGF-I과 IGFBP-3가 통계학적으로 관련성이 미미한 것처럼 보였는데 이는 환아의 수가 적기 때문으로 보인다. 결 론 : 통상적으로 최대 자극 성장호르몬 치를 성장호르몬 치료의 기준으로 사용하여 왔다. 그러나 이 연구에서는 성장호르몬 자극 검사상 성장호르몬 결핍증의 범위에서 벗어나는 특발성 저신장의 치료에도 성장호르몬이 효과가 있으며 또한 치료의 지표로서 IGF-I과 IGFBP-3를 사용할 수 있다는 것을 보여주고 있다.

조기 사춘기 여아에서 단기간 성선자극호르몬 방출호르몬 효능약제와 성장호르몬 병합투여의 성장촉진 효과 (Growth promoting effect of short-term gonadotropin releasing hormone agonist with or without growth hormone in girls with early puberty)

  • 목 적: 성조숙증에서 관찰된 최근의 연구 결과를 보면 사춘기 발육이 빠른 당시에는 신장이 커보일 수 있으나 결국 최종성인신장은 감소하게 된다. 이에 사춘기를 중단시킴으로써 최종 성인신장을 증가시킬 수 있을 것이라는 기대로 성선자극호르몬 방출호르몬 효능약제를 사용하여 골성숙의 기간을 연장하고자 하였다. 또한 조기 사춘기로 인하여 최종성인신장의 예후가 불량한 소아에서 성선자극호르몬 방출호르몬 효능약제 및 성장호르몬의 병합요법 효과를 성선자극호르몬 방출호르몬 효능약제 단독 사용 시의 효과와 비교하여 각각의 성장 촉진 효과를 알아보고자 하였다. 방 법: 역연령에 비하여 골연령이 증가되어 있으며 조기 사춘기를 보이는 여아를 대상으로 GnRHa 및 성장호르몬을 단기간 투여하였다. 제 1군은 triptorelin을 단독으로 4주에 한 번씩 근육주사하였고, 제 2군은 성장호르몬을 병합하여 치료하였다. 치료 기간은 약 1년이었으며 치료 시작시와 치료 후의 두 군간의 역연령, 골연령, 역연령과 골연령의 차이, 예측성인신장, 표적키, 표적키와 예측성인신장과의 차이, 혈청 IGF-1 및 IGFBP-3치를 비교하였다. 결 과: 치료 시작시 두 군 간의 골연령, 표적키, 예측성인신장, 예측성인신장과 표적키의 차이는 없었으나 골연령과 역연령의 차이 및 역연령은 유의한 차이를 보였다. 혈청 IGF-1 및 IGFBP-3값의 차이는 없었고 평균 치료기간은 $1.06{\pm}0.93$년이었다. 치료 후의 골연령과 역연령의 차이는 전체적으로 감소하였으며 두 군간에도 유의한 차이를 보였다. 치료 후의 예측성인신장은 치료전과 비교했을 때 모두 증가하여 단기간의 치료에도 효과가 있었으며 두 군 모두 의미 있는 값을 가지나 특히 성장호르몬을 같이 사용한 2군에서 더욱 큰 차이를 보여 병합요법의 성장촉진효과를 알 수 있었다. 표적키와 예측성인신장의 차이는 전체적으로 치료전보다 의미 있는 감소를 보였으며 두 군을 비교했을때 역시 2군에서 더욱 의미 있게 감소하였다. 혈청 IGF-1과 IGFBP-3는 GnRHa 사용 후에 성장호르몬-IGF 축이 억제되어 1군에서 모두 감소하였으나 성장호르몬을 병합한 2군에서는 모두 증가하였고 이때 통계적인 의미는 없었다. 결 론: 조기 사춘기로 인하여 역연령에 비해 골연령이 증가함으로써 성인신장에 나쁜 영향을 끼치는 경우에는 단기간의 GnRHa의 사용으로 사춘기의 빠른 진행을 억제하고 골성숙의 기간을 연장시킴으로써 예측성인신장을 증가시킬 수 있다. 또한 성장호르몬 병합요법으로 예측성인신장이 표적키에 도달하는데 있어 GnRHa 만으로 성장호르몬-IGF 축이 억제된 경우보다 성장촉진 효과를 나타낸다. 그러나 이는 단기간 사용 시의 치료효과이므로 최종성인신장의 비교를 위해서는 향후 장기적인 추적관찰이 필요하다.

Expression of Serum and Muscle Endocrine Factors at Antemortem and Postmortem Periods and Their Relationship with Pig Carcass Grade

  • Carcass weight and backfat thickness are primary yield grading factors. Insulin-like growth factor (IGF)-I/-II, transforming growth factor $\beta$1 (TGF-$\beta$1), and epidermal growth factor (EGF) regulate the proliferation and differentiation of cells including adipocytes. Also, interleukin (IL)-2/-6, cortisol, and dehydroepiandrosterone-sulfate (DHEA-S) are known to be related to muscle growth and fat depth. However, the relationships between endocrine factors and carcass grade have not been studied. Therefore, this study aimed to measure the concentrations of endocrine factors in serum and muscle, and to investigate the relationship of endocrine factors with carcass grade. A total of 60 crossbred gilts (Duroc${\times}$Yorkshire${\times}$Landrace) were used. Blood from the jugular vein was collected at antemortem (7 days before slaughter) and postmortem periods, and M. Longissimus was collected at 45 min and 24 h after slaughter. The concentrations of IGF-I/-II, EGF, TGF-$\beta$1, IL-2/-6, cortisol and DHEA-S were analyzed by radioimmunoassay (RIA) or enzyme-linked immunosorbent assay (ELISA). In general, IGF and EGF concentrations in serum and muscle of grade A carcasses were found to be higher than those of grade C carcasses at antemortem and postmortem periods, whereas the pattern of TGF-$\beta$1 concentration was reversed. In particular, the concentrations of muscle IGF-I (24 h postmortem) and serum TGF-$\beta$1 (antemortem) were significantly different between grades A and C (p<0.05). The present results indicate that serum and muscle growth factors affect carcass weight and backfat thickness, and indirectly suggest the possibility that carcass grade could be predicted by expression of serum and/or muscle growth factors.

Nutritional and Tissue Specificity of IGF-I and IGFBP-2 Gene Expression in Growing Chickens - A Review -

  • Nutritional regulation of gene expression associated with growth and feeding behavior in avian species can become an important technique to improve poultry production according to the supply of nutrients in the diet. Insulin-like growth factor-I (IGF-I) found in chickens has been characterized to be a 70 amino acid polypeptide and plays an important role in growth and metabolism. Although it is been well known that IGF-I is highly associated with embryonic development and post-hatching growth, changes in the distribution of IGF-I gene expression throughout early- to late-embryogenesis have not been studied so far. We revealed that the developmental pattern of IGF-I gene expression during embryogenesis differed among various tissues. No bands of IGF-I mRNA were detected in embryonic liver at 7 days of incubation, and thereafter the amount of hepatic IGF-I mRNA was increased from 14 to 20 days of incubation. In eyes, a peak in IGF-I mRNA levels occurred at mid-embryogenesis, but by contrast, IGF-I mRNA was barely detectable in the heart throughout all incubation periods. In the muscle, no significant difference in IGF-I gene expression was observed during different stages of embryogenesis. After hatching, hepatic IGF-I gene expression as well as plasma IGF-I concentration increases rapidly with age, reaches a peak before sexual maturity, and then declines. The IGF-I gene expression is very sensitive to changes in nutritional conditions. Food-restriction and fasting decreased hepatic IGF-I gene expression and refeeding restored IGF-I gene expression to the level of fed chickens. Dietary protein is also a very strong factor in changing hepatic IGF-I gene expression. Refeeding with dietary protein alone successfully restored hepatic IGF-I gene expression of fasted chickens to the level of fed controls. In most circumstances, IGF-I makes a complex with specific high-affinity IGF-binding proteins (IGFBPs). So far, four different IGFBPs have been identified in avian species and the major IGFBP in chicken plasma has been reported to be IGFBP-2. We studied the relationship between nutritional status and IGFBP-2 gene expression in various tissues of young chickens. In the liver of fed chickens, almost no IGFBP-2 mRNA was detected. However, fasting markedly increased hepatic IGFBP-2 gene expression, and the level was reduced after refeeding. In the gizzard of well-fed young chickens, IGFBP-2 gene expression was detected and fasting significantly elevated gizzard IGFBP-2 mRNA levels to about double that of fed controls. After refeeding, gizzard IGFBP-2 gene expression decreased similar to hepatic IGFBP-2 gene expression. In the brain, IGFBP-2 mRNA was observed in fed chickens and had significantly decreased by fasting. In the kidney, IGFBP-2 gene expression was observed but not influenced by fasting and refeeding. Recently, we have demonstrated in vivo that gizzard and hepatic IGFBP-2 gene expression in fasted chickens was rapidly reduced by intravenous administration of insulin, as indicated that in young chickens the reduction in gizzard and hepatic IGFBP-2 gene expression in vivo stimulated by malnutrition may be, in part, regulated by means of the increase in plasma insulin concentration via an insulin-response element. The influence of dietary protein source (isolated soybean protein vs. casein) and the supplementation of essential amino acids on gizzard IGFBP-2 gene expression was examined. In both soybean protein and casein diet groups, the deficiency of essential amino acids stimulated chickens to increase gizzard IGFBP-2 gene expression. Although amino acid supplementation of a soybean protein diet significantly decreased gizzard IGFBP-2 mRNA levels, a similar reduction was not observed in chickens fed a casein diet supplemented with amino acids. This overview of nutritional regulation of IGF-I and IGFBP-2 gene expression in young chickens would serve for the establishment of the supply of nutrients to diets to improve poultry production.

  • This study was conducted to investigate the effects of extrusion pressure and particle size of feed ingredients on the growth performance and plasma hormone activity in juvenile olive flounder Paralichthys olivaceus. Experimental diets were prepared with extrusion pressure manipulated by screw speed [low pressure (LP), 885 rpm/min; high pressure (HP), 708 rpm/min] and different dietary particle sizes [specific surface area: small (SS), $169.9m^2/kg$; large (LS), $67.4m^2/kg$] in a two-level factorial design. Four experimental diets (LP+SS, LP+LS, HP+SS, and HP+LS) were randomly assigned to 12 tanks (3 replicates) stocked with 20 fish (initial weight, 57 g) per tank. After a 4-week feeding trial, the observable trends of the main effects of extrusion pressure and particle size on growth performance showed that LP and SS enhanced fish weight gain. The plasma insulin-like growth factor-I level was significantly higher in fish fed the LP+SS diet than in fish fed the HP+SS diet. These results indicate that manipulation of the physical qualities of feed through adjustment of extrusion pressure and feed ingredient particle size may influence the growth performance of juvenile olive flounder, which should be considered in feed manufacture.

Association Between Insulin-like Growth Factor-2 Expression and Prognosis after Transcatheter Arterial Chemoembolization and Octreotide in Patients with Hepatocellar Carcinoma

  • Objective: To investigate the association between the change of IGF-2 level in serum after transcatheter arterial chemoembolization (TACE) and hepatocellular carcinoma (HCC) progression, especially in relation to metastasis. Methods: IGF-2 in serum was measured by quantitative sandwich enzyme-linked immunosorbent assaybefore, 3 days and 4 weeks after TACE in 60 patients with HCC. The occurrence of HCC metastasis was also evaluated, 3 months after TACE. Results: (1) The average serum level of IGF-2 in the 60 patients with HCC was $136.5{\pm}87.3$ pg/ml; (2) A tendency for increase was observed with heterogenous uptake of octreotide and portal vein thrombosis. Metastatic foci were found in 37/38 patients in the group with IGF-2 increasing (97.0%), in contrast to 3/22 (13.6%) patients with IGF-2 decrease. Conclusion: The increase of IGF-2 level in serum appears to be associated with the occurrence of metastatic HCC after TACE and chemotherapy.
