Detergent and Phospholipid Mixed Micelles as Proliposomes for an Intravenous Delivery of Water-Insoluble Drugs

  • A novel drug delivery system, detergent-phospholipid mixed micelles as proliposomes, for water-insoluble compounds was developed by investigating (i) spontaneous formation of small unilamellar vesicles (SUV) from bile salt-egg phosphatidylcholine mixed micelles, (ii) the molecular mechanism of micelle-to-vesicle transition in aqueous mixtures of detergent-phospholipid, (iii) preparation and screening of a suitable liposomal formulation for a lipophilic drug: solubilization of the drug within the lipid bilayer, evaluation of the solubility limit, and characterization of the resulting product with respect to the physical properties and stability of the drug in the system, and (iv) testing antitumor activity in vitro. The results showed that the new carrier had a strong possibility to be a biocompatible universal formulation for water-insoluble drugs.

불용성 세리신의 알칼리 가수분해 (Alkali Hydrolysis of Insoluble Sericin)

  • When insoluble sericin was hydrolyzed by treatment of NaOH solution, the solubility was increased with higher treatment temperature and longer treatment time. Whereas it was decreased in addition of NaHSO$_3$. As the results of electrophoresis in sericin powder obtained by the NaOH treatment, a distinguishable band was not confirmed. Average degree of polymerizations(A.D.P.) of sericin hydrolyzed by NaOH solutions were about 19.6∼22.1 and average molecular weight(M.W.) were about 2,200∼2,500. The longer hydrolysis time increased the whiteness of sericin powder. As the results of amino acid analysis, the contents of Thr., Tyr., and Ser. were decreased in NaOH hydrolysis as compared to HCl hydrolysis. In DSC analysis, thermal deformation and pyrolysis peak located at near 230$\^{C}$ and 320$\^{C}$, respectively.

  • On the performance of the hydrogen peroxide method for Th-determination we encountered some difficulties, namely the formation of the insoluble matter occured in the nearly neutral solution after evaporation. We carried out the Th-determination in the mixture of the Th and some kind of the rare earth, and found that the insoluble matter was not formed in case of the mixture of Th and some kind of rare earth, but formed in case of Monazite. The formation of the insoluble matter in the case of Monazite sand, however, could be avoid by using the beaker instead of the evaporating dish and by adding the water repeatedly before it was completely evaporated to dryness.

  • The 'Mixing Index($D_I$)' is used as a conventional guidance measuring the degree of mixing for multiphase flows. For the case when insoluble solutions flow in a passive micromixer, a new method to calculate $D_I$ is proposed. The 'Vortex Index(${\Omega}_I$)' is suggested and formulated. We infer that ${\Omega}_I$ relates to the degree of chaotic advection. Various arbitrary shaped microchannels were tested to calculate the $D_I\;and\;{\Omega}_I$, and then a simple algebraic equation, $D_I=Aexp(B{\Omega}_I)$, is obtained. This equation may be used instead of the conventional partial differential equation, concentration equation, to estimate the degree of mixing.

시금치에서 분이한 엽록체의 인산대사에 관한 연구 (A Study on Phosphate Metabolism of Chloroplast Isolated from Spinach)

  • In order to observe the phosphate metabolism in chloroplast, the contents of inorganic phosphate and various compounds in chloroplast from spinach leaf tissues were investigated during the reaction in the light and dark in the reaction mixture and the turnover of phosphate in chloroplast was compared with that of whole cell system: 1. The phosphorus of DNA in chloroplast appears to be transferred from inorganic phosphate, while in whole cell system from phosphate pool. 2. $^{32}P-phosphate$ content of acid soluble fraction in chloroplast as well as in whole cell system was more increased in the light than dark during the reaction. It was noted to be caused by the stimulation of sugar phosphate synthesis in the light. 3. It was confirmed that polyphosphate exists in chloroplast as well as whole cell. Acid insoluble polyphosphate content in whole cell system was significantly decreased during the reaction and the similar tendency was also observed in chloroplst. It is, therefore, considered that acid insoluble polyphosphate also play an most important role as a phosphate pool respectively in chloroplast and in cytoplasm. 4. Protein and lipid phosphorus in chloroplast as well as whole cell system were transferred from acid insoluble polyphosphate.

불용성 세리신의 효소 가수분해 (Enzyme Hydrolysis of Insoluble sericin)

  • To hydrolyze insolule sericin the enzyme hydrolysis was used, and then obtained the results as given belows. When insoluble sericin was hydrolyzed by enzyme treatment, the solubility was best at pH 7, 60$\^{C}$ and was slightly increased both above 2 hours treatment and above 10% of enzyme concentration. As the results of electrophoresis, the distribution of molecular weight of sericin powder obtained by enzyme hydrolysis was very weak and showed in the wide range having no distinguishable band. Average degree of polymerzations (A.D.P.) of sericin hydrolyzed by enzyme were about 4.1∼6.3, average molecular weight were about 470∼730. The whiteness of the sericin powder obtained by enzyme hydrolysis was high and increased slightly with higher treatment concentration of enzyme. As the results of amino acid analysis, the amino acid analysis, the amino acid composition of the sericin powder from the enzyme treatment were similar to which located at near 230$\^{C}$ and 320$\^{C}$. The peak of near 230$\^{C}$ could not been found in the sericin powder obtained by enzyme hydrolysis.

Ni-W 합금도금용 불용성 양극의 제조 및 특성 연구 (Preparation and Characterization of Insoluble Anodes for Electrodeposition of Ni-W Alloys in Ammoniacal Citrate Bath)

  • Insoluble anodes of the Ta/Ir mixed metal oxide for electrodeposition of Ni-W alloy in ammoniacal citrate bath were prepared by thermal decomposition method. Ti plate was etched in boiling oxalic acid solution and coated with ethanol solution of $TaCl_{5}$ and $IrCl_4$ mixed in a fixed ratio, followed by drying and treating at various temperatures. The coating layer of these insoluble anode was characterized by SEM, EDX, XRD and DSC. The decomposition rate of citric acid in plating bath was determined by measuring the $CO_2$ gas evolved at the anodes with Gas Chromatography. Evolution of $CO_2$ gas from Ta/Ir oxide anodes decreased about 5% compared with that of Pt. The $CO_2$ gas evolution was increased with the amount of Ir-oxide in the coatings. The coatings which have more than 40% ratio of Ta content and heat-treated at the temperature higher than $400^{\circ}C$ showed better efficiency

  • The catalytic dehydrocoupling of di-n-butylstannane n-Bu2SnH2 by the Cp2MCl2/Red-Al (M = Ti, Zr, Hf) and M(CO)6/Red-Al in situ combination catalysts yielded a mixture of two kinds of catenated products: one is a cross-linked insoluble solid, and the other is a non-cross-linked soluble solid (≒Sn5) or viscous oil (≒Sn2). The soluble oligostannanes could be produced by simple dehydrocoupling of n-Bu2SnH2, whereas the insoluble polystannanes could be obtained via disproportionation/dehydrocoupling combination process of n-Bu2SnH2. The disproportionation/dehydrocoupling combination process may initially produce a low-molecular-weight oligostannane with partial backbone Sn-H bonds which could then undergo an extensive cross-linking reaction of backbone Sn-H bonds, resulting in the formation of an insoluble polystannane.

한국산 소맥과 수입 소맥의 단백질 특성 비교 (A Comparison of Protein Characteristics of Korean and Imported Wheat Varieties)

  • 그루, 은파 및 탑동 등의 한국산 소맥분의 단백질 함량은 수입 소맥과 비교하여 상당히 많은 양 함유되어 있으나 탑동을 제외한 한국밀의 반죽 물성은 매우 약하여 단백질의 함량이 많음에도 불구하고 연질맥과 같은 반죽물성을 나타내었고, 특히 그루는 가장 많은 양의 단백질을 함유하지만 전형적인 sticky dough의 문제점을 나타내었다. 수입산과 국내산의 분류에 관계없이 단백질 함량이 많은 그루, 은파, 및 탑동 등의 한국 밀들은 DNS와 같이 acid insoluble protein의 함량이 상대적으로 많으며 water soluble과 salt soluble 단백질의 함량은 상대적으로 적었다. 특히 수입 DNS 밀은 전체 단백질의 함량에 대한 40% 이상이 acid insoluble protein이며 SDS insoluble protein 함량 또한 가장 많은 양 함유되어 있었다. 반면 한국산 소맥은 대부분 연질맥인 WW와 같이 SDS insoluble protein의 함량이 상대적으로 적었다. 또한 품종이나 제품성에 관계없이 밀가루의 단백질 함량이 높을 수록 단백질의 S-S와 SH 함량이 증가하였으며, 이러한 결과는 외국문헌에서 알려진 밀가루의 특성과 특별한 차이가 없었다. 그러나 탑동밀을 제외한 4종의 한국 밀에서 gluten의 특징적인 116 kD 이상의 high molecular weight protein subunit이 관찰되었으며, 이러한 subunit은 high molecular weight glutenin subunit에 해당되는 것으로 외국문헌의 경우 subunit 2.2로 분류하기도 한다. 또한 116 kD영역에 나타나는 triplet band는 탑동을 제외한 4 종의 우리밀은 doublet로 나타나므로 단백질의 high molecular weight subunit region에서 경질맥인 탑동밀의 구조가 가장 수입밀과 일치하는 것을 관찰할 수 있었다.
