Screening Plant Growth-Promoting Bacteria with Antimicrobial Properties for Upland Rice

  • This study explores beneficial bacteria isolated from the roots and rhizosphere soil of Khao Rai Leum Pua Phetchabun rice plants. A total of 315 bacterial isolates (KK001 to KK315) were obtained. Plant growth-promoting traits (phosphate solubilization and indole-3-acetic acid (IAA) production), and antimicrobial activity against three rice pathogens (Curvularia lunata NUF001, Bipolaris oryzae 2464, and Xanthomonas oryzae pv. oryzae) were assessed. KK074 was the most prolific in IAA production, generating 362.6 ± 28.0 ㎍/ml, and KK007 excelled in tricalcium phosphate solubilization, achieving 714.2 ± 12.1 ㎍/ml. In antimicrobial assays using the dual culture method, KK024 and KK281 exhibited strong inhibitory activity against C. lunata, and KK269 was particularly effective against B. oryzae. In the evaluation of antimicrobial metabolite production, KK281 and KK288 exhibited strong antifungal activities in cell-free supernatants. Given the superior performance of KK281, taxonomically identified as Bacillus sp. KK281, it was investigated further. Lipopeptide extracts from KK281 had significant antimicrobial activity against C. lunata and a minimum inhibitory concentration (MIC) of 3.1 mg/ml against X. oryzae pv. oryzae. LC-ESI-MS/MS analysis revealed the presence of surfactin in the lipopeptide extract. The crude extract was non-cytotoxic to the L-929 cell line at tested concentrations. In conclusion, the in vitro plant growth-promoting and disease-controlling attributes of Bacillus sp. KK281 make it a strong candidate for field evaluation to boost plant growth and manage disease in upland rice.

Plant Growth Promotion and Biocontrol Potential of Various Phytopathogenic Fungi Using Gut Microbes of Allomyrina dichotoma Larva (장수풍뎅이 유충의 장내 미생물을 이용한 다양한 식물 균류병의 생물적 방제 및 생장촉진)

  • This research was executed to select beneficial antagonists from digestive organ of Allomyrina dichotoma larva that can be put on environment friendly control against phytopathogenic fungi. We screened 38 bacterial strains inhibiting mycelial growth against eight plant pathogens through dual culture assay. The 10 strains among 38 bacterial strains were selected as beneficial microbes showing antifungal activity against Botrytis cinerea, Plasmodiophora brassicae, Colletotrichum acutatum and Phytophthora capsici through under greenhouse pot trials. The 10 bacterial strains that shown strongest antifungal activity were classified into 3 genera and 10 species, and identified as the genus Bacillus (DM146, DM152, DH2, and DH16), Paenibacillus (DF30, DH14, and DM142) and Streptomyces (DF137, DM48, and DH92) by morphological characteristics and 16s rRNA gene sequence. The 10 bacterial strains had solubilizing activity of insoluble phosphates, production of IAA (indole-3-acetic acid), β-1,3-glucanase and protease. Among the 10 bacterial strains, DM152 strain was produced significant enhancement of all growth parameters of chili pepper and tomato seedlings under greenhouse condition. Thus, this study demonstrated that gut microbes of Allomyrina dichotoma larva will be useful as a potential biocontrol agent against plant pathogens and biofertilizer.

  • This study was conducted to investigate both plant growth-promoting and plant disease- controlling activities of bacterial strains isolated from soil. All the isolated strains were able to grow at various temperatures. All the strains, except ANG40, showed antagonistic effects against various phytopathogenic fungi. This antagonism can be ascribed to the production of siderophores and antibiotic substances. In addition, all the strains showed abilities such as nitrogen fixation, phosphate solubilization, and siderophore production. These results suggest that nitrogen, phosphorus, and iron can be converted into forms that can be easily absorbed by the plants for their growth. Analysis of the growth-promoting properties revealed that ANG51 produced 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase and indole-3-acetic acid (IAA) both of which are related to ethylene production. In contrast, the other strains were found to have only IAA-producing ability. Therefore, this study suggests that Pseudomonas geniculata ANG3, Exiguobacterium acetylicum ANG40, and Burkholderia stabilis ANG51, which were selected through analysis of comparative advantages for both plant growth promotion and disease-controlling activity, may be used as biological agents.

Changes of Root Physiology of Tissue Cultured M.9 Apple Rootstock after Layering (기내 배양 사과 대목 M.9의 순화 후 휘묻이 번식 시 발근 관련 생리적 특성 변화)

  • This work was conducted to evaluate the effects of rooting on tissue cultured M.9 (Malus domestica Bork. cv, tcM.9) after layering in field. We investigated an appearance period of first root in shoot, rooting ratio, contents of indole-3-acetic acid (IAA), abscisic acid (ABA), inorganic matters, sugars, and lignin in rooting areas of stems by layering. First root in shoot of tcM.9 and natural M.9 appeared 25 and 30 days after layering (DAL), respectively. Rooting ratio was much higher in tcM.9 than in natural M.9. The content of IAA was higher in tcM.9 than in natural M.9 before layering, but it was reversed at 20 and 30 DAL. In contrast, the content of ABA was much higher in natural M.9 than in tcM.9 in case of both before and 10 and 20 DAL. The contents of N, B, Mn, and Zn were significantly higher in tcM.9 than in natural M.9 both before and 10 and 20 DAL. The contents of sugars in tcM.9 had the similar pattern of the contents of inorganic materials. There were statistically significant differences in the contents of sucrose and glucose at 30 DAL as well as the content of maltose at 20 and 30 DAL. The content of lignin was significantly higher in tcM.9 than in natural M.9 before layering and 10 and 30 DAL while there was no difference 20 DAL. Therefore, improvement of rooting ability in the tissue cultured root stock M.9 might be due to the changes of inorganic matters or lignin rather than that of sugars and hormones.

  • 농업현장에서 활용되는 농업용미생물의 효능을 평가하고 현장실용화를 제고할 목적으로 본 시험을 수행하였다. 8개 지자체를 대상으로 친환경농업 특성화 지원사업 현황을 조사하고, 지자체별 미생물 활용수준을 평가하였으며, 각 지자체별로 활용되는 미생물을 수집하여 항균활성, siderophore 생성, IAA 생성, 질소고정, 인산가용화 능력 및 작물생육에 미치는 영향을 분석하였다. 지자체에서 활용하는 미생물은 주로 유산균, 고초균, Bacillus속 세균, 광합성균, 효모 및 EM 등으로 몇가지 종류의 미생물을 활요하는 것으로 조사되었고, 배양을 위한 배지는 대부분 균주를 개발한 대학 또는 산업체에서 공급받는 것으로 나타났으며, 배양기술 측면에서 볼 때 단순 배양 보급하는 지자체가 2개소, 산-학-연간 공급네트워크 및 대량배양체계를 구축하여 미생물을 활용하는 지자체가 7개소, 미생물 대량배양체계를 구축하고 병해충 관리연구를 수행하는 지자체가 1개소 그리고 미생물 선발, 특성검정 및 효과 평가 등 개발 및 응용을 추진하는 지자체가 1개소인 것으로 나타나 지자체에 따라 활용기술 수준은 큰 차이를 보였다. 지자체 공급 농업미생물의 항균활성을 조사한 결과, 고초균, 효모, 유산균, 광합성균, Bacillus속 세균 모두에서 균주에 따라 항균활성을 보였는데, 특히 Bacillus 속 세균이 높은 빈도로 황균활성을 보이는 것으로 나타났다. 철 킬레이트 능력 (Siderophore 형성능)에 있어서는 5개 광합성 균주 중 1균주가, 5종의 효모 중 1균주만이 활성을 보인 반면, Bacillus속 세균(고초균 포함)의 경우에는 11개 균주 모두 활성을 보였다. 작물 생육과 밀접한 관계가 있는 IAA(Indole acetic acid)의 생성능에 있어서는 광합성균이 5균주 중 4균주가 활성을 보여 비교적 생성능이 높은 것으로 나타났으며, Bacillus 속 세균의 경우에는 11균주 중 8개 균주가 활성을 보여 광합성균 다음으로 높은 반도를 나타냈다. 효모의 경우에는 5균주 중 1개 균주가 IAA를 생성하는 것으로 나타났다. 질소고정능에 있어서는 광합성균은 5균주 중 3균주, Bacillus 속세균 11균주 중 6균주가, 효모는 5균중 1균주가 활성을 보인 반면, 유산균 중에는 단 한 균주도 질소 고정능을 보이지 않았다. 연작재배에 따라 토양 내에 축적되는 인산의 가용화능을 분석한 결과, 수집한 대부분의 유산균은 높은 활성을 보인 반면 기타 미생물은 인산가용화능이 거의 없는 것으로 나타났다. 결론적으로 Bacillus 속 세균은 항균활성이 높고, 작물생육과 밀접한 IAA 생성 및 질소고정능력을 보유 하고 있는 것으로 나타나 친환경 작물재배시 병 발생을 줄이고 작물생장을 촉진할 수 있는 미생물로 평가되었다. 농업현장에서 기능성 및 효능을 고려하여 미생물을 잘 활용하면 병 발생 관리 및 생산성 향상에 도움이 되리라 판단된다.

Plant Growth-promoting Ability by the Newly Isolated Bacterium Bacillus aerius MH1RS1 from Indigenous Plant in Sand Dune (해안 사구에서 서식하는 토착식물로부터 분리된 근권미생물 Bacillus aerius MH1RS1의 식물성장 촉진 능력 연구)

  • Coastal sand dunes have been seriously damaged caused by the development thoughtless for the environment and coastal erosion and destruction due to artificial structures like coast roads and breakwater. Hereupon, in this study we made a library of rhizobacteria that have the plant growth-promoting ability for plant rhizosphere of indigenous plants inhabiting in a coastal sand dune as well as the strong tolerance to salt, and evaluated the plant growth-promoting ability of these strains. Furthermore, we evaluated the effect of rhizobacteria on the growth rate of saline tolerant plants in sandy soil; selected out the most useful micro-organism for the restoration of a damaged sand dune. The effect of inoculation of strains selected from the first experiment on the growth of Peucedanum japonicum and Arundo donaxes planted in a coastal sand dune was evaluated. As a result, Bacillus aerius MH1RS1 had plant growth promoting activities: indole acetic acid (IAA) production, siderophores and 1-aminocyclopropane-1-carboxylate deaminase (ACC deaminase) activity, and also had a salinity tolerance. Also, in case of Peucedanum japonicum, the length of stems and weights of roots were enhanced by the inoculation of B. aerius MH1RS1. Fresh weights of stems and weights of roots in experimental group were, in particular, increased by 25% comparing with the control group. For an Arundo donax in experimental group, plant length increased by 18%, and weight of roots by 20% which is significant.

Isolation and Characteristics of Bacteria Showing Biocontrol and Biofertilizing Activities (생물방제 및 생물비료 활성을 가지는 세균의 분리 및 특성)

  • To develop multifunctional microbial inoculant, microorganisms with antagonistic activity and biofertilizing activity were screened. Pantoea agglomerans and Bacillus megaterium from our laboratory culture collection, and strain MF12 from soil near poultry farm in Miryang were selected. On the basis of morphological, physiological studies and 16S rDNA sequence analysis, isolate MF12 was identified as the Bacillus pumilis. Three strains were studied for insoluble phosphate solubilization, indole-3-acetic acid (IAA) and siderophore production, ammonification ability, hydrolytic enzyme production and antifungal activity against phytopathogenic fungi. P. agglomerans did not produce any visible clear zone on agar plate containing 0.5% $Ca_3(PO_4){_2}$ as a sole phosphorus source. However, this strain could solubilize insoluble phosphate in liquid medium. All strains produced IAA ranged from $3{\sim}639{\mu}g/ml$ depending on culture time and had ammonification ability. Among three strains, only P. agglomerans produced siderophore. P. agglomerans produced pectinase and lipase, B. megaterium produced amylase, protease and lipase while B. pumilis produced protease and lipase. P. agglomerans showed antifungal activities against phytopathogenic fungi, Fusarium oxysporum and Colletotrichum gloeosporioides. B. pumilis showed antifungal activities against Botrytis cinerea, Sclerotinia sclerotiorum and Phythium ultimum.

Mass Cultivation and Characterization of Multifunctional Bacillus velezensis GH1-13 (복합기능성 Bacillus velezensis GH1-13 균주의 대량배양 최적화 및 특성)

  • Bacillus genus are found abundantly in various sites and their secondary metabolites were used as potential agents in agriculture, notably plant growth promoting and bio-control. The objective of this study was to develop the culture conditions of GH1-13 strain including higher cell growth, stable endospore-forming and enhancement of potential agents which are related with plant growth promoting and phytopathogen suppression. The optimal carbon and nitrogen sources were determined by glucose and soy bean flour, respectively, then resulted in $7.5{\times}10^9cells/mL$, $6.8{\times}10^9\;endospore\;cells/mL$ and sporulation yield of 90% after 30 h cultivation in 500 L submerged fermenter at $37^{\circ}C$, pH 7.0. Cells and cell-free supernatant of GH1-13 strains showed the potent antifungal activity against phytopathogenic fungi of Colletotrichum gloeosporioides. It was also confirmed that indole-3-acetic acid (IAA) production of GH1-13 strain was greatly increased by addition of 0.3% tryptophan.

Biological Control of Anthracnose (Colletotrichum gloeosporioides) in Red Pepper by Bacillus sp. CS-52 (Bacillus sp. CS-52를 이용한 고추 탄저병 (Colletotrichum gloeosporioides) 방제 특성)

  • This study was carried out in order to develop a biological control of anthracnose of red pepper caused by fungal pathogens. In particular, this study focuses on the Colletotrichum species, which includes important fungal pathogens causing a great deal of damage to red pepper. Antagonistic bacteria were isolated from the soil of pepper fields, which were then tested for biocontrol activity against the Colletotrichum gloeosporioides anthracnose pathogen of pepper. Based on the 16S rRNA sequence analysis, the isolated bacterial strain CS-52 was identical to Bacillus sp. The culture broth of Bacillus sp. CS-52 had antifungal activity toward the hyphae and spores of C. gloeosporioides. Moreover, the substances with antifungal activity were optimized when Bacillus sp. CS-52 was grown aerobically in a medium composed of 0.5% glucose, 0.7% $K_2HPO_4$, 0.2% $KH_2PO_4$, 0.3% $NH_4NO_3$, 0.01% $MnSO_4{\cdot}7H_2O$, and 0.15% yeast extract at $30^{\circ}C$. The inhibition of spore formation resulting from cellulase, siderophores, and indole-3-acetic acid (IAA), were produced at 24 h, 48 h, and 72 h, respectively. Bacillus sp. CS-52 also exhibited its potent fungicidal activity against anthracnose in an in vivo test, at a level of 70% when compared to chemical fungicides. These results identified substances with antifungal activity produced by Bacillus sp. CS-52 for the biological control of major plant pathogens in red pepper. Further studies will investigate the synergistic effect promoting better growth and antifungal activity by the formulation of substances with antifungal activity.
