Components and Their Antioxidative Activities of Methanol Extracts from Sarcocarp and Seed of Zizyphus jujuba var. inermis Rehder (생대추(풋대추) 과육 및 씨 추출물의 항산화 효과 및 항산화 성분)

  • Antioxidant activities of methanol extracts from sarcocarp and seed of Zizyphus jujuba var. inermis Rehder were investigated in vitro. Contents of total polyphenols in methanol extracts from sarcocarp and seed were 98.83, $138.99\;{\mu}g/mg$, respectively. Radical-scavenging activities of methanol extracts were examined using ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$ and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) radicals, and hydrogen peroxide assay. Inhibition effects of methanol extracts on peroxidation of linoleic acid were examined by ferric thiocyanate and thiobarbituric acid methods. Both sarcocarp and seed of Zizyphus jujuba var. inermis Rehder showed relatively high antioxidant activities in various systems.

FINITE ELEMENT ANALYSIS OF MAXILLARY CENTRAL INCISORS RESTORED WITH VARIOUS POST-AND-CORE APPLICATIONS (여러가지 post-and-core로 수복된 상악 중절치의 유한요소법적 연구)

  • The purpose of this study was to investigate the effect of rigidity of post core systems on stress distribution by the theoretical technique, finite element stress-analysis method. Three-dimensional finite element models simulating an endodontically treated maxillary central incisor restored with a zirconia ceramic crown were prepared and 1.5 mm ferrule height was provided. Each model contained cortical bone, trabecular bone, periodontal ligament, 4 mm apical root canal filling, and post-and-core. Six combinations of three parallel type post (zirconia ceramic, glass fiber, and stainless steel) and two core (Paracore and Tetric ceram) materials were evaluated, respectively. A 50 N static occlusal load was applied to the palatal surface of the crown with a $60^{\circ}$angle to the long axis of the tooth. The differences in stress transfer characteristics of the models were analyzed. von Mises stresses were chosen for presentation of results and maximum displacement and hydrostatic pressure were also calculated. An increase of the elastic modulus of the post material increased the stress, but shifted the maximum stress location from the dentin surface to the post material. Buccal side of cervical region (junction of core and crown) of the glass fiber post restored tooth was subjected to the highest stress concentration. Maximum von Mises stress in the remaining radicular tooth structure for low elastic modulus resin core (29.21 MPa) was slightly higher than that for high elastic modulus resin core (29.14 MPa) in case of glass fiber post. Maximum displacement of glass fiber post restored tooth was higher than that of zirconia ceramic or stainless steel post restored tooth.

In vitro Screening of Jeju Island Plants for Customerized Cosmetics (맞춤형화장품 소재 개발을 위한 제주 식물 탐색)

  • In this study, we investigated collagen production and hyaluronic acid production effects for wrinkle improvement test on 50 kinds of land plants and 10 kinds of marine plants native to Jeju Island as a part of developing customized cosmetic materials. Collagen and hyaluronic acid are recognized as major factors affecting skin aging. Cerastium holosteoides var. hallaisanense Mizushima extract ($100{\mu}g/mL$) produced more than 190% of collagen in the extracts of 50 kinds of land plants. Vicia angustifolia var. segetilis K. Koch. extract ($100{\mu}g/mL$) produced more than 160% of collagen. Ftsia japonica Decne. et Planch. extract ($100{\mu}g/mL$), Euonymus japonica Thunb. extract ($100{\mu}g/mL$), Suaeda malacosperma H.Hara extract ($100{\mu}g/mL$), Elaeagnus umbelellata Thunb. extract ($100{\mu}g/mL$), Sedum oryzifolium Makino extract ($100{\mu}g/mL$), Vicia unijuga A. Br. extract ($100{\mu}g/mL$), and Brassica juncea var. integrifolia Sinsk. extract ($100{\mu}g/mL$) showed more than 140% collagen production effect. Among the 10 species of marine plants, Sargassum macrocarpum C. Agardh extract ($50{\mu}g/mL$) produced more than 190% of collagen, and Carpopeltis angusta (Harvey) Okamura extract ($100{\mu}g/mL$), Codiumcoactum Okamura extract ($100{\mu}g/mL$), and Codium tenuifolium S. Shimada, T. Tadano & J. Tanaka extract ($100{\mu}g/mL$) showed more than 140% collagen production. Suaeda malacosperma H.Hara extract ($100{\mu}g/mL$) showed the effect of producing hyaluronic acid more than 140%, and Ftsia japonica Decne. et Planch. extract ($20{\mu}g/mL$) and Wistaria floribunda A.P. DC extract ($100{\mu}g/mL$) showed more than 130% hyalunonic acid production effect. Among the 10 species of marine plants, Peyssonnelia capensis Montagne extract ($100{\mu}g/mL$) was the most effective. Carpopeltis angusta (Harvey) Okamura extract ($100{\mu}g/mL$), Codiumcoactum Okamura extract ($100{\mu}g/mL$), and Codium tenuifolium S. Shimada, T. Tadano & J. Tanaka extract ($100{\mu}g/mL$) showed more than 120% hyalunonic acid production. Jeju resources, which have good collagen and hyaluronic acid production, showed the potential to be applied to solve the skin troubles of customized cosmetics in the future.

Effects of Different Natural Extracts and Plant Growth Regulators on Plant Regeneration and Callus Induction from Pseudobulbs Explants through in vitro Seed Germination of Endangered Orchid Bulbophyllum auricomum Lindl.   • Bulbophyllum auricomum Lindl. is a rare orchid and has flowers with an attractive fragrance. The present study investigated the tissue culture method for micropropagation. Capsules derived from artificial self-pollination were obtained for the best seed germination in MS basal medium. Plant growth regulators (1.0 mg·L-1 of BAP and 2.0 mg·L-1 of NAA) were affected by callus induction from subcultured pseudobulb explants. For the callus subculture, different natural plant extracts were tested in 11 treatment media. Among them, MS medium with 150 mL·L-1 of coconut water was generally effective in fresh weight (1.75 ± 0.08) and (3.01 ± 0.20) of callus proliferation and PLBs induction at 1 and 2 months, respectively, followed by an MS combination of 30 g·L-1 of banana and 20 g·L-1 of potato extract. The results of a comparative study of different MS mediums containing plant growth regulators with a natural extract combination and MS medium supplemented with natural extract only showed that MS medium supplemented with a combination of natural extracts (150 mL·L-1 of coconut water) and plant growth regulators (2.0 mg·L-1 of BAP and 1.0 mg·L-1 of NAA) obtained the highest shoot regeneration (3.37 ± 0.17) and (6.41 ± 0.68) after 1 month and 2 months of culturing, respectively.

Physicochemical properties and microencapsulation process of rice fermented with Bacillus subtilis CBD2 (Bacillus Subtilis CBD2로 배양된 백미 발효물의 미세캡슐 제조 및 물리화학적 특성)

  • This study was conducted to examine the physicochemical properties and micro-encapsulation process of rice fermented with Bacillus subtilis CBD2. The viable bacterial cell, pH, and amylase activity of the rice liquid culture were 7.61 log CFU/mL, pH 5.08 and 159.43 units/mL, respectively. The micro-encapsulated rice liquid culture was manufactured via spray drying with different forming agents: i.e., alginic acid 1.0% and chitosan 0.3%, 0.5%, and 1.0%. The moisture contents of the spray-dried powders were approximately 2.90~3.68%. The color of the L and a value decreased whereas that of the b and ${\Delta}E$ value increased. The particle size and outer topology of the spray-dried rice liquid culture were $48.13{\sim}68.48{\mu}m$ and globular, respectively. The water absorption index of the spray-dried powder (2.40~2.65) was lower than that of the freeze-dried powder (2.66). The water solubility index of the spray-dried powder (9.17~10.89%) was higher than that of the freeze-dried powder (7.12%). The in vitro dissolution was measured for five hours in pH 1.2 simulated gastric fluid, and pH 6.8 and pH 7.4 simulated intestinal fluids, using a dissolution tester at $37^{\circ}C$ with 50 rpm agitation. The amylase survival in the fermented rice was 85.93% through the spray-drying and it was very effectively controlled.

A comparative study on the fit and screw joint stability of ready-made abutment and CAD-CAM custom-made abutment (기성 지대주와 맞춤형 CAD-CAM 지대주의 적합 및 나사 안정성 비교)

  • Purpose: The purpose of this study was to investigate the fit and screw joint stability between Ready-made abutment and CAD-CAM custom-made abutment. Materials and methods: Osstem implant system was used. Ready-made abutment (Transfer abutment, Osstem Implant Co. Ltd, Busan, Korea), CAD-CAM custom-made abutment (CustomFit abutment, Osstem Implant Co. Ltd, Busan, Korea) and domestically manufactured CAD-CAM custom-made abutment (Myplant, Raphabio Co., Seoul, Korea) were fabricated five each and screws were provided by each company. Fixture and abutments were tightening with 30Ncm according to the manufacturer's instruction and then preloding reverse torque values were measured 3 times repeatedly. Kruskal-Wallis test was used for statistical analysis of the preloading reverse torque values (${\alpha}=.05$). After specimens were embedded into epoxy resin, wet cutting and polishing was performed and FE-SEM imaging was performed, on the contact interface. Results: The pre-loading reverse torque values were $26.0{\pm}0.30Ncm$ (ready-made abutment; Transfer abutment) and $26.3{\pm}0.32Ncm$ (CAD-CAM custom-made abutment; CustomFit abutment) and $24.7{\pm}0.67Ncm$ (CAD-CAM custom-made abutment; Myplant). The domestically manufactured CAD-CAM custom-made abutment (Myplant abutment) presented lower pre-loading reverse torque value with statistically significant difference than that of the ready-made abutment (Transfer abutment) and CAD-CAM custom-made abutment (CustomFit abutment) manufactured from the same company (P=.027) and showed marginal gap in the fixture-abutment interface. Conclusion: Within the limitation of the present in-vitro study, in domestically manufactured CAD-CAM custom-made abutment (Myplant abutment) showed lower screw joint stability and fitness between fixture and abutment.

Effect of Extraction Conditions on in vitro Antioxidant Activities of Root Bark Extract from Ulmus pumila L. (추출조건에 따른 유근피 추출물의 항산화 활성)

  • This study investigated optimal extraction conditions for application of Ulmus pumila L. as a natural antioxidant. U. pumila L. was extracted using ethanol (EtOH) at various concentrations (0, 40, and 80%) and extraction times (1, 2, and 3 h) at $70^{\circ}C$ and then evaluated for extraction yield, total phenolic contents, total flavonoid contents, as well as antioxidant activities [2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, reducing power, and oxygen radical absorbing capacity (ORAC)]. Antioxidant activities were correlated with total phenolic and flavonoid contents. Of the solvent conditions, 80% EtOH extracts for 3 h at $70^{\circ}C$ showed the highest total phenolic and flavonoid contents with strong antioxidant activities, although there were no significant time effects on DPPH and ABTS radical scavenging activities and reducing power. However, ORAC values of all EtOH extracts remarkably increased in a time-dependent manner. In addition, 80% EtOH extract for 3 h exhibited strong antioxidant effects on HDF and 3T3-L1 cells. Therefore, the antioxidant capacity of U. pumila L., may due to phenolic and flavonoid contents, and extraction conditions were 80% EtOH for 3 h at $70^{\circ}C$. This extract could be a good source for natural antioxidants.

An in vitro study of a few crystal growth solutions on the bracket shear bond strength (수종의 실험 결정형성용액에 의한 브라켓 전단결합강도의 비교)

  • The purpose of this study was to compare the bracket shear bond strengths of the crystal growth solutions with those of the $37\%$ phosphric acid etch technique. The 4 crystal growth solutions were made experimentally in the lab, that is, (1) $30\%$ polyacrylic acid solution containing 0.3 M sulfuric acid (ES 1), (2) $30\%$ polyacrylic acid solution containing 0.6M sulfuric acid (ES 2), (3) $30\%$ polyacrylic acid solution containing 0.3 M sulfuric acid and 0.6 M lithium sulfate(ES 3), and (4) $30\%$ polyacrlic acid solution containing 0.3 M sulfuric acid and $5\%$ phosphoric acid(ES4). The $37\%$ phosphoric acid solution used as a control. Bovine lower incisor tooth enamel was treated by the above solutions for 60 sec, washed out for 20 sec with slow water stream, and bonded lower anterior edgewise bracket with the light curing orthodontic composite resin adhesives. The teeth bonded brackets were stored in the distilled water at room temperature for 24 h, and followed to test the bracket shear bond strength. The acid etch technque showed 177.6 kg/$cm^2$ of mean shear bond strength which was the highest among the enamel treatment solutions. ES 1 shown 58.4 kg/$cm^2$ of mean shear bond strength and that of ES 4 showed 66.5 kg/$cm^2$. There was no significant difference between the two(p>0.05). ES2 showed 110.6kg/$cm^2$ of mean shear bond strength which was $62.3\%$ of that of acid etch technique. ES 3 showed 131.1 kg/$cm^2$ of mean shear bond strength which was the highest among experimental crystal growth solutions and which was $74\%$ of that of acid etch technique. The shear bond strengths of the crystal growth solutions were significantly lower that that of acid etch technique(p<0.05). The results sugest that although bracket shear bond strength of $30\%$ polyacrylic acid solution containing 0.3M sulfuric acid and 0.6 M lithium sulfate were showed the highest, it is low for the clinical application of this solution.

Effect of Harvest Stage, Wilting and Crushed Rice on the Forage Production and Silage Quality of Organic Whole Crop Barely (수확시기, 예건 및 싸라기 처리가 유기 청보리의 사초 생산성 및 사일리지 품질에 미치는 영향)

  • Although whole crop barley (Hordeum vulgare L.) is now widely grown as a silage crop in Korea, forage production and silage quality of that for organic farm have not been published. Therefore, this experiment was conducted to evaluate the effect of harvest stage on forage production and quality of organic barley, and effect of field wilting and crushed rice for shortening of harvest date and improvement of forage quality. The experiment was split-plot design with three replications. Main plots were heading, milking and yellow stages, and sub-plot were field wilting for I day, crushed rice 10% (CR10%) and 15% (CR15%) treatments. The dry matter (DM) contents of heading, milking and yellow stages were 12.8%, 21.9% and 29.8%, respectively. The DM yields of heading, milking and yellow stages were 10,346, 15,819 and 18,336 kg/ha, respectively, and the total digestible nutrients (TDN) of these were 6,288, 9,550 and 10,178 kg/ha, respectively. The pH of milking stage showed low 4.00 pH values. The crude protein, ether extract, crude ash, non-fiber carbohydrate (NFC) and TDN contents were decreased as harvest stage progressed, while neutral detergent fiber (NDF) and acid detergent fiber (ADF) were increased. The crude ash (CA) of milking stage showed the lowest among harvest stages. Field wilting and crushed rice treatments decreased CA, NDF and ADF contents, and increased NFC and TDN contents. In vitro dry matter digestibility (IVDMD) decreased with progressed harvest stage, while field wilting and crushed rice treatments increased that of barley silage. Lactic acid and total organic acid contents of milking stage were the highest, and butyric acid of milking stage was the lowest among harvest stage. The good effect of field wilting and crushed rice was observed in heading stage. The experiment results indicate that optimum harvest stage of barley silage for organic was milking stage. The field wilting and crushed rice additive could be recommended as effective method for shortening harvest date and increasing forage quality of organic barley silage

Biological Activities of Hot Water Extracts from Euonymus alatus Leaf (화살나무 잎 열수추출물의 생리활성)

  • This study was performed to investigate the biological activities of roasted (RE) and non-roasted (NRE) hot water extract samples from Euonymus alatus leaf by measuring DPPH radical scavenging, total polyphenol content, hydroxyl radical scavenging, superoxide dismutase (SOD)-like activity, anti-inflammatory activity, and inhibitory effects on ${\alpha}-amylase$ and ${\alpha}-glucosidase$ in vitro. The $IC_{50}$ values fur DPPH free radical scavenging activity of the NRE and RE hot water extracts were $19.1{\mu}g/mL$ and $21.9{\mu}g/mL$, respectively, and their total polyphenol contents were estimated as 9.6 mg/g for NRE and 10.6 mg/g for RE. Both NRE and RE scavenged the hydroxyl radical in a concentration-dependent manner, but their activities were lower than that of BHA. It was also shown that SOD-like activity was dependent on NRE and RE concentration, and the SOD-like activity of NRE was slightly higher than that of RE. The highest SOD-like activity obtained from NRE was 20% at a concentration of 3 mg/mL. Neither NRE nor RE seemed to have an effect on ${\alpha}-amylase$ and ${\alpha}-glucosidase$ inhibition. Finally, the hot water extracts of NRE and RE significantly decreased the concentration of LPS-induced NO in RAW 264.7 cells, indicating anti-inflammatory activity.
