EFFECT OF DIFFERENT SOURCES OF FIBER ON THE INTESTINAL MORPHOLOGY OF DOMESTIC GEESE

  • Experiment was conducted to study the effect of sources of dietary fiber on the intestinal morphology of geese. Sixty white Roman geese of two-week-old were divided randomly into six groups and were fed with isoenergetic and isonitrogenous diets which contain alfalfa meal, barley hull, rice hull, purified cellulose, lignin, or pectin as the major dietary source of fiber. Different sources of dietary fiber significantly influenced the villi height and the crypt depth in the duodenum, and the villi height and the muscle layer thickness in the ileum (p < 0.05). The duodenal villus in the geese that fed diets with alfalfa meal, rice hull or pectin supplemented were significantly longest ($968.5{\mu}m$), whereas the lignin group was significantly shortest villus and deepest crypt depth (p < 0.05). The thicknesses of the ileal and caecal muscle layer were significantly thicker in the geese fed with cellulose supplemented diets than in those fed with the other treatment diets. The caecum of the barley bran fed geese possessed significantly longest villi and the most thick muscle layers (p < 0.05). From scanning electronic microscopic observation, the leafy and plate-like in the duodenal villi morphology of geese represented a more effective nutrient absorption in the small intestine. The morphology of ileal villi in geese was similar from herbivorous rabbit and from the African Green monkey.

Effect of Fibre Level and Fibre Source on Gut Morphology and Micro-environment in Local (Mong Cai) and Exotic (Landrace×Yorkshire) Pigs

  • The effect of genotype, fibre level and fibre source on gut morphology, environment and microflora was studied using 18 Mong Cai (MC) and 18 Landrace${\times}$Yorkshire (LY) pigs, aged around 60 d. The diets were based on maize, rice bran, soybean meal, fish meal and soybean oil, and cassava residue (CR) or brewer's grain (BG) as fibrous ingredient sources in the high-fibre diets (HF). A low-fibre diet (LF), containing around 200 g NDF/kg dry matter (DM), was formulated without CR and BG as feed ingredients. The HF diets (HF-CR and HF-BG) were formulated to contain around 270 g NDF/kg DM. The experiment was arranged according to a $2{\times}3$ factorial completely randomized design with six replications, and lasted 30 d. Crypt density in ileum was lowest (p<0.05) and villus height in jejunum and ileum were the greatest (p<0.05) in pigs fed diet HF-BG. Villus width in ileum was greatest in pigs fed diets HF-CR and HF-BG (p<0.05). Lactic acid bacteria (LAB) counts in stomach were greatest (p<0.05) and E. coli counts in ileum and colon were lowest (p<0.05) in pigs fed diet HF-CR. The concentration of total organic acids in ileum, caecum and colon were greatest (p<0.05), and pH in ileum and colon were lowest (p<0.05) in pigs fed diet HF-CR. Crypt density in ileum was lowest, and villus height in ileum and villus width in jejunum and ileum was greatest in LY pigs (p<0.05). LAB counts in stomach and ileum were greatest, and E. coli counts in ileum were lowest in MC pigs (p<0.05). The concentration of total organic acids in ileum, caecum and colon were greatest (p<0.05) and pH lowest (p<0.05) in MC pigs.

  • The experiment was conducted to determine the effects of dietary supplementation of enzyme(Endopower) into corn-soybean meal diet on the performance of broiler chicks. Three hundred sixty 3 day-old male broiler chicks(Ross) were divided into 4 groups with 3 replicates of 30 birds per replicate and assigned at random to each of four treatment groups for two energy levels(3100 and 2980 kcal TMEn/kg) with 0 or 0.1% Endopower in the diet. The body weight gain of birds fed the low energy diet with 0% Endopower was lower significantly than the other groups(P<0.05). There were no significant differences in feed intake and feed conversion rate among the treatments. The relative sizes(% B. wt) of the liver and leg muscle were not different significantly among the treatments. The breast muscle weights of the low energy diet birds were higher than those of the high ones, and those of the lower energy group with 0.1% Endopower were highest. The intesinal lengths(cm/B.wt) of low energy diet group without Endopower were lower than those of the others. No significant differences in the relative jejunum and ileum weights were found, but % duodenum weights of high energy group were higher than those of the low energy group. The villi of the ileum of birds fed corn-soybean diet without Endopower were shortened and thickened compared to those of the birds fed with Endopower diet regardless of energy level. The results demonstrated that dietary Endopower improved body weight gain reduced abdominal fats and developed intestinal villi. Therefore it can be concluded that dietary supplementation of Endopower improves nutritive value of corn-soybean diet in broiler chicks.

Study on Radioprotective Effects of Bujeongsaengjintang (부정생진탕(扶正生津湯)이 방사선(放射線) 조사(照射) 부작용(副作用)에 미치는 영향(影響))

  • To evaluate the radioprotective effects of Bujeongsaengjintang studies were done experimentally. The results were obtained as follows: 1. WBC, Platelet and RBC were significantly increased in Bujeongsaengjintang treated group as compared with control group after exposure to radiation by Liniac. 2. By FACS analysis of splenic leukocyte after exposure to radiation by Liniac, T cell, T-helper cell and macrophase were significantly increased in Bujeongsaengjintang treated group. 3. In histological changes of ileum and jejunum of Balb/C mice after exposure to radiation by Liniac, exclusion and fusion of villi were decreased in Bujeongsaengjintang treated group as compared with control group. From above results, it is suggested that Bujeongsaengjintang is available to a clinic for the protection from damage by radiotherapy to cancer.

Effect of Dietary Lactobacillus on Growth Performance, Intestinal Microflora, Development of Ileal Villi, and Intestinal Mucosa in Broiler Chickens (유산균의 급여가 육계 생산성, 장내 미생물 균총, 회장 융모 발달 및 장점막에 미치는 영향)

  • This experiment was conducted to investigate the effects of dietary supplementation of various Lactobacilli strains on growth performance, intestinal microflora, ileal villi development and ileal mucosal surface of broiler chickens. Six hundred 1-d-old male chicks(Avian) were randomly divided into five groups of four replicates, each replicates containing 30 birds and fed corn-soybean meal diets containing Latobaillus were isolated from cecum of chichens at 107cfu/g diet for five weeks. The treatments were control(antibiotics-free diet), Lactobacillus crispatus avibro1(LCB), Lactobacillus reuteri avibro2(LRB), Lactobacillus crispatus avihen1(LCH) or Lactobacillus vaginalis avihen2(LVH). The body weight gain in groups fed Lactobacillus significantly improved as compared to those of control(P<0.05). Feed intake was not statistically different among the groups. The number of Lactobacillus in ileum and cecum of chicks fed various Lactobacillus tended to be increased or significantly increased as compared to those of the control(P<0.05), but there was difference by age of chicks and species of Lactobacillus. The number of yeast was significantly increased in cecum and ileum at three weeks old chicks fed Lactobacilli compared with the control(P<0.05). The anaerobes’ number of ileum and cecum tended to increase or significantly increased in Lactobacillus treatments compared with the control(P<0.05). The ileal villi length extended greatly at three weeks of age in groups fed Lactobacillus compared with the control(P<0.05). The length of ileal villi in chicks fed Lactobacillus was continuously increased up to five weeks of age, but did not increased in the control(P<0.05). Lactobacillus was found on ileal mucosal surface. And ileal mucosal surface was maintained better with Latobacillus feeding. These results suggest the possibility that cecal Lactobacillus of chickens could be used as probiotics by improving the growth performance and promoting development of ileal villi in broiler chicks.

Prevalence and pathologic study of porcine epidemic diarrhea in Jeju (제주도의 돼지유행성설사 발생 상황과 병리학적 고찰)

  • From April 2014 to September 2015, 153 piglets from 52 farms in Jeju were diagnosed with porcine epidemic diarrhea (PED). The major PED cases were focused on suckling piglets (144 piglets, 94.1%), particularly in 1-7-day-old piglets. Histopathologically, severe villous atrophy was observed in the small intestine, especially in the jejunum and ileum. The mean villous height to crypt depth ratios of the jejunum and ileum were 1.4:1 and 1.5:1, respectively. The major histopathologic findings of the small intestine were cytoplasmic vacuolation, cuboidalization, squamation, and exfoliation of the mucosal enterocytes in the villi. The cytoplasmic vacuolations in the enterocytes were the most prevalent lesions in the small intestine and were more severe in the ileum than in the jejunum. According to immunohistochemistry methods, the PED virus (PEDV) antigens were presented in the cytoplasms of the enterocytes, and were distributed more prevalently in the ileum than in the jejunum. PEDV antigens were also detected in the colon of 26 piglets (19.5%). Sequence comparison and phylogenetic analysis indicated that 12 PEDV had more than a 98.9% homology with each other. These PEDV strains were highly homologous with the genogroup 2 North American group.

Effect of Two Doses of Different Zinc Sources (Inorganic vs. Chelated form) on the Epithelial Proliferative Activity and the Apoptotic Index of Intestinal Mucosa of Early-weaned Pigs Orally Challenged with E. coli K88

  • The effect of two doses of different sources of zinc, inorganic (zinc oxide) or chelated (zinc glutamate chelate), on morphology and turn-over of the small intestine was assessed in early-weaned pigs orally challenged with enterotoxigenic E. coli K88 (ETEC). Sixty pigs weaned at 21 days were assigned to one of the following 5 diets: control (C); C+Zinc oxide (ZnO), either a 200 or a 2,500 mg Zn/kg dose; or C+zinc chelate with glutamic acid (Glu-Zn), either a 200 or a 2,500 mg Zn/kg dose. On d 2, the pigs were orally inoculated with 1.5 ml of a $10^{10}$ CFU/ml E. coli K88ac O148 suspension. Zinc supplements did not improve the performance of the pigs, but on d 5 faecal excretion of ETEC was reduced, and this was mainly due to high zinc doses (p<0.05). The villous height in the duodenum was improved by the zinc supplements (p<0.01) whatever the source and the level, whereas no effect was seen in the other two tracts of small intestine. The diet did not affect apoptosis and mitosis counts, while ETEC-susceptible pigs had more mitotic cells in the villi than non-susceptible pigs, particularly in the jejunum (p<0.01). The duodenum had fewer mitotic cells in the villi (p<0.05) and in the crypts (p<0.01) and more apoptotic cells in the villi. High dietary doses of ZnO or Zn-Glutamate improve villous height of the duodenum, but not of the jejunum and the ileum, and do not affect the epithelial proliferative activity and apoptotic index of intestinal mucosa of early-weaned pigs orally challenged with ETEC.

Effects of vitamin A supplementation in the diet of breeding geese on offspring intestinal tissue morphology and immune performance

  • Objective: The effects of maternal and offspring dietary vitamin A (VA) supplementation on early body weight, digestive tract function and immune function in goslings were studied. Methods: Yangzhou geese (180 d old) were randomly divided into 5 experimental groups of 15 females and 3 males (the males were kept until slaughter). Eggs were collected for hatching during the peak laying period. A total of 96 goslings were selected from each treatment group (each fed a basic diet supplemented with 0, 4,000, 8,000, 12,000 or 16,000 IU/kg VA) and randomly divided into 2 groups, with 6 replicates in each group and 8 goslings in each replicate. The gosling diet was supplemented with 0 or 9,000 IU/kg VA. Results: i) Villus length, villus width and the muscle thickness of the duodenum, jejunum and ileum were increased and the crypt depth was reduced after adding 12,000 IU/kg VA to the goslings' diet (p<0.05). Adding 9,000 IU/kg VA to the offspring diet increased the length of the duodenal villi and width of the ileum and decreased the crypt depth of the ileum (p<0.05). ii) Supplementing the maternal diet with 12,000 IU/kg VA increased immune organ weight, the immune organ index and immunoglobulin content in goslings (p<0.05). The bursa weight and immunoglobulin G content of offspring were higher in the 9,000 IU/kg VA supplementation group than in the group with no supplementation (p<0.05). Conclusion: Offspring growth and development were affected by the amount of VA added into maternal diet. The negative effect of maternal VA deficiency on offspring can be compensated by adding VA to the offspring diet. Continued VA supplementation in the offspring diet after excessive VA supplementation in the maternal diet is unfavorable for gosling growth and development.

Pathogenesis on enteritis induced by Cryptosporidium parvum alone and combined with porcine rotavirus in piglets (Cryptosporidium parvum 단독 및 돼지 rotavirus와 혼합 감염시킨 자돈 장염의 병원성)

  • The purpose of the present study was to understand the pathogenesis of infections in piglets inoculated with C parvum isolated from mice alone and combined with porcine rotavirus (S-80). Thirteen 10-day piglets were divided in four groups; Three, A group, were only given by C parvum. Four, B group, were orally administrated with firstly porcine rotavirus and then C patvum. Three, C group, were orally inoculated with porcine rotavirus alone. The rest, D group, were used as controls. During the experiment, there were daily recorded clinical signs including diarrhea to each pig. According to the periodic intervals for necropsy, all pigs were sacrificed from 4 to 12 days after the final inoculation of C parvum. Location and distribution of two pathogens, C parvum and rotavirus, in the intestinal mucosa of piglets tested were examined by pathological and immunohistological means. In addition, parasitological test using the feces of piglets was applied for the detection of cryptosporidial oocysts as well. A group showed diarrhea from 4 to 6 days post-inoculation(PI) and also discharged C parvum oocysts in feces during the day 4 to 7 PI. In tissue sections of jejunum and ileum, cryptosporidial oocysts were observed a few on the top of villi with slightly fusion. B group represented sign of diarrhea and discharge of oocysts from 2 to 11 days PI. There were some cryptosporidial oocysts both in the jejunal lumen and in the lumen of mucosal glands. As progressed, oocysts were most commonly distributed on the tip of villi of jejunum. Histopathologically there were also mild to moderately fused, attenuated focal desquamated, congested villi and mononuclear cell infiltration of varying degrees in the lamina propria of small intestine and colon at the day 4 and 7 PI. C group showed slightly to mildly attenuated and fused top of villi and mildly mucosal congestion. D group as controls was grossly and histopathologically normal in all parts of intestine. The present results indicate that the piglets inoculated with C parvum only are certainly milder in pathogenesis including duration of clinical course and severity of lesion than those in piglets concurrently infected with porcine rotavirus and C parvum. Also the strain (VRI-CN91) of C parvum used in the study has very low pathogenicity to occur enteritis of piglets.

Distributions of proliferative epithelial cells in gastrointestinal tracts by anti-bromodeoxyuridine monoclonal antibody (Anti-bormodeoxyuridine monoclonal antibody를 이용한 랫드 위(胃)와 장(腸)의 분열 상피세포의 분포에 대하여)

  • The purpose of this stady was to investigate division cells by in vivo bromodeoxyuridine(Brdur) immunohistochemistry for labeling the proliferative epithelial cells in the gastrointestinal tracts of rats. Rats were administrated intraperitonially by twice consecutive injections of 24 hr interval with Brdur(0.05mg/g BW/time) and then were sacrificied at 1 hour after last injection. The specimens were taken from the stomach, small intestine(ileum), and large intestine(colon). The well-oriented crypts and villi in the preparations were examined, The crypt columns and villi were devided into 10 segments from crypt base to surface of the lumen or to villis top. Labeling index(LI) was measured by counting the number of Brdur-positive cells against the total number of crypt column cells in the stomach and large intestine and also against the total numbers of crypt column and it's villi epiterial cells in the small intestine. 1. In the stomach, the LI in each part from segment 1 to segment 10 of the crypt column were 4.2%, 5.0%. 6.6%, 9.0%, 11.3%, 15.3%, 9.3%, 15.6%, 11.3%, 0%, respectively and it's mean LI were 8.7%. The Brdur-positive epithelial cells were predominantly located in the middle regions and middle-upper regions of the crypt columns. 2. In the small intestine, the LI in each part from segment 1 to segment 10 of were 62.4%, 50.9%, 27.8%, 22.5%, 18.6%, 12.1%, 7.5%, 4.3%, 2.5%, 1.4%, respectively and it's mean LI were 21.0%. The Brdur-positive epithelial cells were predominantly located in the lower regions of the crypt columns and tended to be less in the higher regions of the villi than that in the crypt column. 3. In the large intestine, the LI in each part from segment 1 to segment 10 of the crypt column were 19.4%, 29.9%, 34.1%, 41.6%, 41.2%, 32.4%, 25.4%, 15.4%, 10.8%, 1.2%, respectively and it's mean LI were 25.1%, The Brdur-positive epithelial cells were predominantly in the middle and middle-lower regions of the crypt columns. 4. The organs with higher LI were ordered as the large intestine(25.1%), small intestine(21.0%) and stomach(8.7%).
