Development of a Multicopy Integration Vector in Yarrowia lipolytica (Yarrowia lipolytica의 Multicopy Integration Vector 개발)

  • Multicopy integration vector is a very useful vector system in that they can be integrated into chromosomal DNA in several copies and stably maintained under non-selective conditions. To develop a multicopy integration vector system in the yeast Yarrowia lipolytica, P-type ribosomal DNA was cloned from Y lipolytica. A HindIII-BglII fragment of the cloned rDNA and a promoterless URA3 gene were inserted into pGEM1, generating multicopy integration vectors, pMIYL-1 and pMIYL-2. The rDNA fragment is for targeted homologous recombination between the vector and the chromosomal DNA of Y. lipolytica, and the promoterless URA3 gene is a defective selection marker for inducing multicopy integration. pMIYL-1 and pMIYL-2 have an unique restriction enzyme site, KpnI, and two unique restriction enzyme sites, KpnI and EcoRI, repectively, which can be used for targeting of the vectors into the rDNA of Y. lipolytica chromosomal DNA. After transformation of the vectors into Y. lipolytica, copy number and stability were analyzed by Southern hybridization. The vectors were found to be present in less than 5 copies per cell and were stably maintained during growth in non-selective media.

  • Given vectors x and y in a Hilbert space $\cal{H}$, an interpolating operator is a bounded operator T such that Tx = y. An interpolating operator for n vectors satisfies the equations $Tx_i=y_i$, for i = 1, 2, ${\cdots}$, n. In this paper the following is proved : Let $\cal{L}$ be a subspace lattice on a Hilbert space $\cal{H}$. Let x and y be vectors in $\cal{H}$ and let $P_x$ be the projection onto sp(x). If $P_xE=EP_x$ for each $E{\in}\cal{L}$, then the following are equivalent. (1) There exists an operator A in Alg$\cal{L}$ such that Ax = y, Af = 0 for all f in $sp(x)^{\perp}$ and $A=A^*$. (2) sup $sup\;\{\frac{{\parallel}E^{\perp}y{\parallel}}{{\parallel}E^{\perp}x{\parallel}}\;:\;E\;{\in}\;{\cal{L}}\}$ < ${\infty}$, $y\;{\in}\;sp(x)$ and < x, y >=< y, x >.

GORENSTEIN SEQUENCES OF HIGH SOCLE DEGREES

  • In [4], the authors showed that if an h-vector (h0, h1, …, he) with h1 = 4e - 4 and hi ≤ h1 is a Gorenstein sequence, then h1 = hi for every 1 ≤ i ≤ e - 1 and e ≥ 6. In this paper, we show that if an h-vector (h0, h1, …, he) with h1 = 4e - 4, h2 = 4e - 3, and hi ≤ h2 is a Gorenstein sequence, then h2 = hi for every 2 ≤ i ≤ e - 2 and e ≥ 7. We also propose an open question that if an h-vector (h0, h1, …, he) with h1 = 4e - 4, 4e - 3 < h2 ≤ (h1)(1)|+1+1, and h2 ≤ hi is a Gorenstein sequence, then h2 = hi for every 2 ≤ i ≤ e - 2 and e ≥ 6.

  • Since one property(i.e. coarseness, orientation, regularity, granularity etc.) of ultrasound liver images was not sufficiently enough to classify the characteristics of livers, we used the multi-feature vectors from ultrasound images to diagnose the liver disease. The proposed classifier, which uses the multi-feature vectors and Bayes decision rule, performed well for the classification of normal, fat and cirrhosis liver. In our simulation, we used the Battacharyya distance and Hotelling Trace Criterion to select the best multi-feature vectors for the classifier and obtained less classification errors than other methods using single feature vector.

SECOND ORDER TANGENT VECTORS IN RIEMANNIAN GEOMETRY

  • This paper considers foundational issues related to connections in the tangent bundle of a manifold. The approach makes use of second order tangent vectors, i.e., vectors tangent to the tangent bundle. The resulting second order tangent bundle has certain properties, above and beyond those of a typical tangent bundle. In particular, it has a natural secondary vector bundle structure and a canonical involution that interchanges the two structures. The involution provides a nice way to understand the torsion of a connection. The latter parts of the paper deal with the Levi-Civita connection of a Riemannian manifold. The idea is to get at the connection by first finding its.spary. This is a second order vector field that encodes the second order differential equation for geodesics. The paper also develops some machinery involving lifts of vector fields form a manifold to its tangent bundle and uses a variational approach to produce the Riemannian spray.

Text Independent Speaker Verficiation Using Dominant State Information of HMM-UBM (HMM-UBM의 주 상태 정보를 이용한 음성 기반 문맥 독립 화자 검증)

  • We present a speaker verification method by extracting i-vectors based on dominant state information of Hidden Markov Model (HMM) - Universal Background Model (UBM). Ergodic HMM is used for estimating UBM so that various characteristic of individual speaker can be effectively classified. Unlike Gaussian Mixture Model(GMM)-UBM based speaker verification system, the proposed system obtains i-vectors corresponding to each HMM state. Among them, the i-vector for feature is selected by extracting it from the specific state containing dominant state information. Relevant experiments are conducted for validating the proposed system performance using the National Institute of Standards and Technology (NIST) 2008 Speaker Recognition Evaluation (SRE) database. As a result, 12 % improvement is attained in terms of equal error rate.

Connecting the Inner and Outer Product of Vectors Based on the History of Mathematics (수학사에 기초한 벡터의 내적과 외적의 연결)

  • In this paper, I investigated the historical development process for the product of two vectors in the plane and space, and draw implications for educational guidance to internal and external product of vectors based on it. The results of the historical analysis show that efforts to define the product of the two line segments having different direction in the plane justified the rules of complex algebraic calculations with its length of the product of their lengths and its direction of the sum of their directions. Also, the efforts to define the product of the two line segments having different direction in three dimensional space led to the introduction of quaternion. In addition, It is founded that the inner product and outer product of vectors was derived from the real part and vector part of multiplication of two quaternions. Based on these results, I claimed that we should review the current deployment method of making inner product and outer product as multiplications that are not related to each other, and suggested one approach for connecting the inner and outer product.

Selection of Voltage Vectors in Three-Level Five-Phase Direct Torque Control for Performance Improvement

  • This paper presents a Direct Torque Control (DTC) strategy for the five-phase induction motor driven by a three-level five-phase inverter in order to improve the performance of the five-phase induction motor. In the proposed DTC technique, only 22 voltage vectors out of 243 available voltage vectors in a three-level five-phase inverter are selected and are divided in 10 sectors each with a width of $36^{\circ}$. The four different DTC combinations (DTC-I, II, III and IV) for a three-level five-phase induction motor drive are investigated for improving the performance of five-phase induction motor. All four of the DTC strategies utilize a combination of the same large and zero voltage vectors, but with different medium voltage vectors. Out of these four techniques, DTC-II gives the best performance when compared to the others. This DTC-II technique is analyzed in detail for improvements in the performance of five-phase induction motor in terms of torque ripple, x-y stator flux and Total Harmonics Distortion (THD) of the stator phase current when compared to its two-level counterparts. To verify the effectiveness of the proposed three-level five-phase DTC control strategy, a DSP based experimental system is build. Simulation and experimental results are provided in order to validate the proposed DTC technique.
