  • PAU is the block copolymer consists of a small amount of a small amount of poly(${\gamma}-methyl-L-glutamate$) (PMLG) and the polyurethane. The urethane segments are hydrophobic and then strongly interact with the other hydrophobic materials such as PTFE, and the PMLG segments with the ${\alpha}-helix$ structure possess the cytocompatibility. Therefore, PAU can be easily coated onto the PTFE fiber and acts as an artificial extracellular matrix with the high cytocompatibility Results shows, the immobilization, cultured and functions of porcine hepatocytes is greatly improved.

Crystal Structure of MJ0684 from Methanococcus jannaschii, a Novel Archaeal Homolog of Kynurenine Aminotransferase

  • MJ0684 from Methanococcus jannaschii is a hypothetical protein belonging to the subfamily Ig of amino acid aminotransferases. In the present study, the crystal structure of MJ0684 has been determined at 2.2 resolution. It reveals that MJ0684 has an overall structure similar to subfamily Ig aminotransferases and its active site architecture is most similar to that of kynurenine aminotransferases among several kinds of aminotransferases in the subfamily Ig?. It has two hydrophobic active site residues conserved in the kynurenine aminotransferases for recognizing hydrophobic substrates. In addition, the absence of any basic residue for recognizing the side chain carboxylic group of the aspartate in the active site rules out the possibility that MJ0684 would act as an aspartate aminotransferase. These structural observations collectively imply that MJ0684 is a novel archaeal homolog of the subfamily Ig kynurenine aminotransferase.

  • The interaction of mastoparan B, a cationic tetradecapeptide amide isolated from the hornet Vespa basalis, with phospholipid bilayers was studied with synthetic mastoparan B and its analogue with Ala instead of hydrophobic 12th amino acid residue in mastoparan B. MP-B and its derivative, [12-Ala]MP-B were synthesized by the solid-phase peptide synthesis method. MP-B and its analogue, [12-Ala]MP-B adopted an unordered structure in buffer solution. In the presence of neutral and acidic liposomes, the peptides took an $\alpha$-helical structure. The two peptides interacted with neutral and acidic lipid bilayers. These results indicated that the hydrophobic face in the amphipathic $\alpha$-helix of MP-B critically affected the biological activity and helical content.

  • The effect of the presence of dissolved organic matters(DOM) on the binding of phenanthrene to cetylpyridinium chloride(CPC) coated sand was investigated. The distribution coefficient of phenanthrene increased with increase of sufactant coverage, and decreased with the presence of dissolved organic matters except for the 1.600mg/g coverage case. Both Aldrich humic acid and extracted dissolved organic matter showed the similar tendency. For the quantification of the overall distribution coefficient, this study presented mass distribution model and estimated the sorption equilibrium coefficients of hydrophobic organic compounds(HOCs) in multi system. The suggested model combined a series of sorption equilibrium relationships including the adsorption of DOMs on sorbents, the binding between HOCs and DOMs, and the sorption of HOCs on sorbents with or without DOMs.

  • Branched oligopeptides prepared by coupling a very hydrophobic aspartic diamino acid ethyl ester such as Asp $(LeuEt)_2$, $Asp(IleEt)_2$ and $Asp(PheEt)_2$ to glycine or glycylphenylalanine were grafted to the cyclotriphosphazene backbone bearing an equimolar hydrophilic poly(ethylene glycol) (PEG). The properties of the resultant amphiphiles were examined in comparison with those of the linear oligopeptide analogues previously reported. All cyclic phosphazene trimers grafted with the branched tetra- and pentapeptides displayed a normal trend of thermosensitivity depending on their hydrophilic to hydrophobic balance, but the stability and particle size of their micelles were found to be greatly dependent on the fine structure of the branched oligopeptides grafted. The trimers bearing branched tetrapeptides with a low hydrophobicity were found to form unstable micelles initially, which reassemble into thermodynamically more stable polymersomes.

Anti-diabetic Effects of Banaba Leaf Extracts (Lagerstroemia speciosa Pers.) through Solvents (추출용매 변화에 따른 바나바(Lagerstroemia speciosa Pers.) 잎 추출물의 항당뇨 효과)

  • This study investigated that the antidiabetic effects of banaba extracts with variety solvents selectivity in vitro and in vivo. Banaba extracts were prepared with water, 70% ethanol, 90% ethanol, 100% ethanol and water-ethanol that of extract twice times sequentially water and ethanol. Cell toxicity and insulin secretion of banaba extracts was tested by MTT (3-[4,5-dimetylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay on hamster insulinoma cell line, HIT-T15. Also we tested that insulin, body weight, blood glucose, total cholesterol, HDL-cholesterol, triglyceride and free fatty acid in streptozotocin diabetic rats. Water-ethanol extract has remarkable antidiabetic effect compare with the other banaba extracts. For water-ethanol extract has both of hydrophilic and hydrophobic antidiabetic materials from banaba. Expecially, corosolic acid, as known as unique polyphenol, has antidiabetic effect studied by many researchers till nowadays. But corosolic acid does not solve in water. Otherwise, we suggest that banaba extract of hydrophilic and hydrophobic materials (polyphenol and antioxidants) mixture more increased antidiabetic effects.

  • Release characteristics of salicylic acid from ethylcellulose(EC) films containing varying ratio of dieththylphthalate (DEP) were studied. Mathematical analysis of the release data showed that the release behavior actually conforms with the Higuchi's diffusion-controlled model. The release rate constants(k) were independent from the film thickness and the pH of release medium, but were proportional to the concentration of salicylic acid itself. The logarithm of the release rate constant (log k) increased as the concentration of DEP was increased. In conclusion, hydrophobic plastisizer DEP seemed to be very useful in controlling release rate constant of slightly soluble drugs as like salicylic acid without changing it's release characteristics.

The Physical Properties of Wheat Flour Extrudates with Added Phenolic Acids (페놀산 첨가 밀가루 압출성형물의 물리적 특성)

  • The effects of phenolic acids on the physical properties of wheat flour extrudate were investigated. Ferulic acid, fumaric acid, and p-coumaric acid were mixed with hard wheat flour, respectively, and extruded under a twin screw extruder. We found that by adding the phenolic acids, longitudinal expansion at the die increased, textural hardness decreased, and the water absorption capacity of the extrudate decreased. The results showed that the addition of phenolic acids produced a softer textured, more longitudinally puffed and hydrophobic extrudate compared to the control extrudate. Moreover, the addition of phenolic acids did not significantly affect the color of the extrudate: oxidative browning of the phenolic acids was not observed, due to inactivation of the browning enzymes under the hot temperature and reduced oxygen conditions of the extrusion process.

Characteristics and Application of the Highly-Durable and Highly-Sensitive Super Hydrophobic Acid-gas Sensing Dye (고내구성 고감도 강산감지기능 초소수성 색소의 특성 및 응용)

  • In order to detect gas phase strong acid on fabrics, a hexyl-substituted monoazo yellow dye, which was the modified form of a conventional pH-indicating dye, Methyl Yellow, was studied in view of acid-gas sensing properties and its fastness. The dye was printed on polypropylene non-wovens for protective coveralls and examined under various conditions of strong acid such as hydrochloric acid. The dye showed color change from yellow to red on exposure to gas phase hydrochloric acid as low concentration as 1~3 ppm very instantly. Considering reuse of the dye-printed non-wovens, the repeatability of color change was tested on the same sample for 50 repeats and 100 days. The acid-gas sensing function was maintained almost the same level of initial performance. The color fastness of the dye on polypropylene non-wovens was very good showing higher than ratings 4 except for 3~4 to rubbing under wet condition.

Mechanism of Fatty Acid Transfer between Fatty Acid Binding Proteins and Phospolipid Model Membranes (지방산 결합단백질과 인지질막 사이의 지방산이동기전)

  • Fatty acid binging proteins(FABP) are distinct but related gene productes which are found in many mamalian cell types. FABP bind long chain fatty acids in vitro. However, their functions and mechanisms of action, in vivo, remain unknown . Also not known is whether all FABP function similaryly in their respective cell types. or whether different FABP have unique functions. The puropose of the present study was to assess whether different members of the FABP family exhibit different structural and function properties. A comparison was made between heart(H-FABP) and liver (L-FABP). The results show that the binding sites of both FABP are hydrophobic in nature, although the L-FABP site is more nonpolar than the H-FABP site. Additionally, the bound ligand experiences less motional constraint within the H-FABP binding site than within the L-FABP binding site. In accordance with these differences in structural properties, it was found that anthroyloxy-fatty acid transfer from H-FABP to membranes is markedly faster than from L-FABP. moreover, the mechanism of fatty acid transfer to phospholipid membranes appears to occur via transient collisional interactions between H-FABP and membranes. In contrast , transfer of fatty acid from L-FABP occurs via an aqueous diffusion mechanism.
