  • The shell of Reeve's turtle has been used as a traditional folk medicine in Korea. We produced a hot water extract from Reeve's turtle shell according to the traditional medical practice. To release bioactive peptides, the hot water extract was enzymatically hydrolyzed with various proteases, and the free radical scavenging activity of the hydrolysate was investigated against 1,1-diphenyl-2-picryl-hydrazyl (DPPH), hydroxyl and peroxyl radicals. The free radical scavenging activity of the enzymatic hydrolysates varied from 1 to 79% depending on the enzymes, free radical species, and concentration. The $EC_{50}$ values demonstrated that the enzymatic hydrolysates of hot water extract from the shell of Reeve's turtle are potential antioxidants.

  • In order to investigate the hypocholesteremic effect of soybean perptides, soybean protein(ISP), casein(CNP), their peptic hydrolyzates fractionated by acid precipitation at different pH's(SHT, SH8, SH6, SH4, CHT, CH5, CH4) and amino acid mixtures of the same composition as the proteins(SAA, CAA) were fed to rats and the concentration of serum cholesterol was measured. Then in vitro digestibility and molecular weight distribution of the peptides by pepticpancreatic hydrolysis was measured by FPLC. The lower the in vitro digestibility of peptides is, the lower the concentration of serum cholesterol becomes(r=0.986) and the higher the ratio of macropeptides is, the lower the concentration of serum cholesterol becomes(r=-0.932) in rats. These results suggest that the in vitro digestibility of peptides has close relationship to the concentration of serum cholesterol in rats and non-digestible meacropeptides or polypeptides especially more than 1 kDa, formed through digestion in gut, may lower the serum cholesterol in rats.

  • Petroleum-based adhesive resins have extensively been used for the production of wood panels. However, it is necessary to develop environmentally friendly adhesive resins due to the increase of manufacturing cost and the environmental issue, such as the emission of volatile organic compounds, of the pertroleum-based adhesive resins. This study was conducted to formulate environmentally friendly adhesive resins using by rapeseed flour (RSF), which is the by-product of bio-diesel produced from rapeseed, for replacing petroleum-based adhesives with them. To formulate RSF-based adhesive resins, RSF was hydrolyzed in de-ionized water, 1% and 3% sodium hydroxide solutions. As a crosslinking agent, PF prepolymers were prepared with 1.8, 2.1 and 2.4 mol formaldehyde and 1 mol phenol (1.8-, 2.1- and 2.4-PF), and then mixed with RSF hydrolyzates to complete the formulation of RSF-based adhesive resins. The RSF-based adhesive resins were applied to fabricate 3-ply plywood panels. The solid content of RSF-based adhesive resins were ranged from 26.08% to 36.12% depending on the hydrolysis condition of RSF and PF prepolymer type with a high viscosity. The tensile shear strength and wood failure of plywood fabricated with RSF-based adhesive resins exceeded a minimum requirement of KS standard for ordinary plywood regardless of the hydrolysis condition of RSF and PF prepolymer type. Formaldehyde emissions of the plywood panels fabricated with 1.8-PF and RSF hydrolyzates were lower than that of E0 specified in the KS standard. Based on the results, RSF might be used as a raw material of environmentally friendly adhesives for the production of plywood panels, but further researches - the increase of solid content of RSF-based adhesives for reducing press time and the microscopic observation of plywood specimen for identifying the relationship between tensile shear strength and the penetration of adhesives into wood structure - are required to commercialize the RSF-based adhesives.

  • In our study, the potential of okara as an ingredient of new bio-based adhesives was investigated for the production of fancy-veneered flooring boards. Okara was hydrolyzed by 1% sulfuric acid solution (AC) and 1% sodium hydroxide solution (AK). Phenol formaldehyde (PF) prepolymers were prepared as a cross-linker of okara hydrolyzates. Then, okara-based adhesive resins were formulated with 35% AC, 35% AK and 30% PF prepolymer on solid content basis. The adhesive resins were applied on high-density fiberboards (HDF) with the spread rate of $300g/m^2$. After that, oak fancy veneers are covered on the HDF, and then pressed with the pressure of $7kg/m^2$ at $120^{\circ}C$. The experimental variables were three mole ratios of formaldehyde to phenol (1.8, 2.1, 2.4), three assembly time (0, 10, 20 min), and two press time (90 sec, 120 sec), respectively. The fancy-veneered high-density fiberboards were tested by dry tensile strength, glueline failure by wetting and formaldehyde emission. Tensile strength of the boards exceeded the requirement of KS standard. The formaldehyde emissions were approached at the E0 level specified in KS standard. Based on these results, okara can be used as an ingredient of environmentally friendly adhesive resin systems for the production of flooring boards.

  • As a substitute for CCA, which is inhibited due to its environmental pollution and human harmfulness, and CuAz and ACQ with a high cost, okara-based wood preservatives were formulated with okara hydrolyzates using copper sulfate and/or borax as a metal salt. The efficacy of the preservatives and X-ray microanalysis of wood specimens treated with the preservatives were examined to confirm the potential of the okara-based wood preservatives. Most of the preservatives showed excellent decay resistance against brown-rot fungi, Postia placenta and Gloeophyllum trabeum. The efficacy was improved when the acid concentration and temperature used for the hydrolysis of okara increased. In addition, when borax was added into copper sulfate/okara hydrolyzates preservative formulations, any decay was not found in the specimens. From the microscopic observation of the specimens treated with okara-based wood preservatives, it seems that okara is contributed to the fixing of metal salts in wood blocks. Therefore, it is speculated that okara-based wood preservatives can effectively protect wood against fungal attack as CuAz, and that the preservatives are sufficient to use as an alternative wood preservative of CCA, ACQ and CuAz.

  • Abstract Extracellular chitinase was purified from the culture liquid of the marine bacterium, Bacillus sp. LJ-25 , and its enzymatic properties were examined. The purified chitinase exhibited a single band on SDS-PAGE and the molecular weight was estimated to be approximately 50 kDa. The optimum pH and temperature for the enzymatic activity were 7.0 and $35^{\circ}C$, respectively. The activity of the chitinase was strongly inhibited by $Zn^{2+}$ and slightly inhibited by $Ba^{2+},{\;}Co^{2+},{\;}Mn^{2+},{\;}and{\;}Cu^{2+}$. The purified chitinase did not hydrolyze $p-nitrophenolN-acetyl-{\bata}-D-glucosaminide{\;}(GlcNAc)_2$ and Micrococcus lysodeikticus cells, which are known to be the substrates for exo-type chitinase. Among the hydrolyzates of colloidal chitin, $(GlcNAc)_2$ was in the highest concentration with small amounts of GlcNAc and $(GlcNAc)_3$..

  • Two strains (No. 26 and 143) of bacteria which secrete both pectinase and raw-starch-digesting amylase simultaneously, were isolated from various domestic soil samples. The two bacteria were identified as Pasteurella ureae judging by their morphological and physiological characteristics. The optimal culture conditions for the production of raw-starch-digesting enzyme by the Pasteurella ureae 26 were using $NH_4NO_3$ as the nitrogen source at $37^{\circ}C$ with the pH of 7.5, and 15 of C/N ratio. Since the enzyme was produced only when raw or soluble starch was used as a carbon source, but not when glucose or other sugars was used, the enzyme was considered to be an inducible enzyme by starch. Thin layer chromatography of the hydrolyzed product of starch by the raw-starch-digesting enzyme of the strain No. 26 showed that glucose, maltose and other oligosaccharides were present in the hydrolyzates, and therefore the enzyme seemed to be ${\alpha}-amylase$. The enzyme had adsorbability onto raw com starch in the pH range of 3 to 9.

  • Various peptides derived from food are among the most potent physiologically active agents known, and include anticancer peptides, angiotensin converting enzyme(ACE) inhibitor exhibiting antihypertension action, opioid peptides, antithrombotic peptides, hypocholesterolemic peptides, immunomodulators, calcium absorption enhancers, and other peptides. Hydrophobic peptides extracted from a Cheddar-type cheese slurry were fractionated by gel chromatography and repeated HPLC. A peptide fraction from HPLC showed high cytotoxicity on the tumor cell lines such as a human colon carcinoma, and comprised of Tyr, Ser, Leu, Gly, and others. Hypocholesterolemic peptides were isolated from peptic hydrolyzates of casein and soy proteins. Macropeptides of 1,000${\sim}$5,000 dalton were effective on reducing the cholesterol level of mouse serum. Peptides showing high Krigbaum hydrophobicity and ANS surface hydrophobicity resulted in high hypocholesterolemic effect and fecal steroid concentrations. Caseinomacropeptides(CMP) were isolated from whey powder and treated with soluble and immobilized trypsin to obtain antithrombotic peptides. One fraction from the CMP hydrolyzed with immobilized trypsin for 24h exhibited high antithrombotic activity with 52.5% inhibition of platelet aggregation. These result suggested that peptides from various milk products could be utilized as a good bioactive agents for developing health functional foods.
