Korean Lentinus edodes SR-1 was cultured to multiply the mycelia in the complete broth medium (C/N=13.1) for mushroom, and protein-bound polysaccharides were extracted from the cultured broth containing mycelia (The whole cultured broth was used to increase the yields: 80% of protein-bound polysaccharides were existed at the cell wall of mycelia and 20% of those were secreted extracellularily in this culture). Protein-bound polysaccharides in the cultured broth containing mycelia were extracted by using three different methods: 1) Extraction with hot water, 2) Disintegration of cell wall by glass bead mill treatment before extraction with hot water, and 3) Cellulase treatment before extraction with hot water. The highest yield was obtained (930 mg polysaccharides/100 mL culture broth) when protein-bound polysaccharides were extracted with 2) method. The extracted crude protein-bound polysaccharides were purified using protease, DEAE-cellulose and Sephadex G-100. The growth inhibition activity for $P_{388}$, mouse leukemic cell, increased (53.7, 62.2, 93.7% and 97.4%) as the purification level increased. Protein-bound polysaccharides contained 46.1% of polysaccharides, 7.3% of protein, and trace amounts of minerals. Polysaccharides contained glucose, galactose, xylose and mannose. The content of proline and glycine were high, however, methionine and leucine were not found. The major minerals were Na, K, Zn, and Ca.
To find antitumor components in liquid cultured cell-free broth and the hot water extract from the fruiting body of Hericium erinaceus, polysaccharides were purified and fractionated by DEAE-cellulose ion exchage column chromatography and Sepharose CL-6B gel filtration chromatography. Among various crude polysaccharides and purified polysaccharides the fractions showing immune-activity were determined by NO assay.
Physiochemical properties, such as yield and molecular weight distribution of polysaccharide fractions, of polysaccharides in the enzymatic hydrolysates of purslane were investigated and characterized. A higher amount of micro nutrients, such as potassium (9,413 mg/100 g), phosphorus acid (539 mg/100 g), leucine, alanine, lysine, valine, glycine, and isoleucine, was present in whole purslane. The yield of water soluble polysaccharides (WSP) was 0.29, 7.01, and 7.94% when extracted using room temperature water (RTW), hot-water (HW), and hot temperature/high pressure-water (HTPW), respectively, indicating that HW or HTPW extraction may be effective to obtain WSP from purslane. The average ratio of L-arabinose:D-galactose in the WSP was 37:49, 34:37, and 27:29, when extracted using RTW, HW, and HTPW, respectively. These results indicate that water was a suitable extraction solvent for preparation of the arabinogalactan component of whole purslane. A higher yield and total carbohydrate content was obtained by using Viscozyme L instead of Pectinex 5XL during extraction of the WSP, which indicates that enzymatic treatment of purslane may be an effective method to control the Mw of polysaccharides. Finally, it was confirmed that Viscozyme L is a suitable enzyme for the hydrolysis and separation of polysaccharides obtained from purslane.
To examine the pharmacological activity of the polysaccharide extracted from phellinus linteus, we obtained the polysaccharide treated by the alkali and hot water, molecular weights of hot water was 10kD and 225 kD. Anticomplementary activity were highly observed in hot water fraction than alkali fraction. The tumor inhibition ratio of the polysaccharide extracted by hot water and alkali of Phellinus linteus against sarcoma 180 were 72.5% and 67.6%, respectively. These results suggest that protein bound polysaccharides extracted by a hot water and alkali of Phellinus linteus were similar to the pharmacological activities and chemical properties.
Polysaccharide FHW was extracted from the fruiting bodies of Fomitella fraxinea with hot-water treatment and then fractionated into FHW-I and FHW-II on DEAE-Cellulose chromatography. FHW-I and FCW-II were further purified into FHW-Ia and Ib, FHW-IIa and IIb on gel permeation chromatography, respectively. A small amount of uronic acid was detected and glucose, galactose, fucose, and mannose were found to be main sugars in the polysaccharides. Protein was detected in FHW-Ia, FHW-IIa, and FHW-IIb, but not in FHW-Ib. FHW-Ia was identified to be a fuco-gluco-mannogalactan with molecular weight of 19,000 and FHW-Ib was a gluco-fuco-mannogalactan of 15,000. FHW-IIa and FHW-IIb were galacto-mannoglucan and their molecular weights were estimated to be 31,000 and 9,000, respectively. Both FHW-Ib and FHW-IIb did not show an absorption band characteristic of the ${\beta}-glycosidic$ linkage in IR spectra. FHW-IIb showed a strong immuno-stimulating activity but the other three polysaccharides showed a weak activity.

Laver, Porphyra, is distinctive for its high content of proteins and polysaccharides such as porphyran and hemicellulose. The chemical properties of the polysaccharides extracted with different extraction methods such as hot water, dilute acid(pH 4.0) or alkali solution(2N NaOH) were examined to investigate the suitable extraction conditions for porphyran and hemicellulose from laver. For porphyran extraction, dilute acid solution was more preferable to hot water and alkali solution because of its higher 3,6-anhydrogalactose content and lower protein content. However, alkali solution was more suitable to extract the hemicellulose because of higher mannose content indicating the extraction of mannan. To decrease contamination of the polysaccharides with protein, the dried lavers were pretreated with enzymes (Protamex, Flavourzyme, Alcalase, Viscozyme) before hot water extraction. All enzyme pretreatments increased the yield of polysaccharides by compared with control (enzyme unpretreated) and Flavourzyme pretreatment was most effective to decrease protein contamination in the polysaccharide. All viscosities of porphyran solutions pretreated by enzymes were lower compared to the control porphyran solution and showed pseudoplastic behavior with yield stress. In case of alkali extraction of residues obtained after enzyme hydrolysis and hot water extraction, protease pretreatment increased the mannose contents in the polysaccharide while the xylose content was increased by Viscozyme pretreatment.
To investigate constituents of Russula pseudodelica Lange and Microporus affinis(Blume et Nees) Kuntze, quantitative analyses of free and total amino acids were carried out with G. L.C. and an amino acid autoanalyzer. Polysaccharides of the two mushrooms were extracted with hot water and the filtrate was concentrated. The addition of three volumes of ethanol to the concentrate formed precipitation of crude polysaccharides which were analyzed by G.L.C. and H.P.L.C. Sixteen amino acids and four monosaccharides were identified in the protein-bound polysaccharides of the two mushrooms. The polysaccharides of M. affinis showed antineoplastic activity against sarcoma 180 implanted in mice.
The polysaccharides from fruit body of Auricularia auricula and Tremella fuciformis were extracted using hot water, and partially purified through ethanol precipitation and dialysis. Free radical scavenging activities of the crude and purified polysaccharides were examined and compared each other. Free radical scavenging activities of the partially purified polysaccharides were higher than those of crude polysaccharides. DPPH free radical, ABTS radical and SOD-like activities of partially purified polysaccharide at 1 mg/mL of concentration from A. auricula were 61.7, 9.6 and 38.9%, respectively, while those of T. fuciformis were 9.6, 5.7 and 15.3%, respectively. Results of site and non-site specific hydroxyl radical scavenging activities indicated that the partially purified polysaccharide fractions from A. auricula and T. fuciformis exhibited the hydroxyl radical scavenging effect by hydrogen donating ability and iron ion chelating ability. Also, reducing powers of A. auricula and T. fuciformis were 77.1 and 14.7% of BHT (0.1%) as standard, respectively. It was suggested that antioxidant activities of A. auricula were about 1.4~6.4 times higher than those of T. fuciformis due to different levels of polyphenol content.
The biological activity of water-soluble polysaccharide (WSP) fractions extracted from Cedrela sinensis was examined in this study. Cedrela sinensis was extracted using hot water, ultrasonication, and enzymes (Viscozyme, Shearzyme) and precipitated using ethanol to produce crude polysaccharides. The yield (3.51%) and total polysaccharide content (28.03 g/100 g) of WSP extracted using Shearzyme (WSPs) were highest compared to other extracts. The antioxidant activity of WSP extracted using hot water was highest and had the lowest $IC_{50}$ values in DPPH, ABTS, hydroxyl radical scavenging activity, reducing power, and superoxide dismutase-like activity. Tyrosinase inhibitory activity increased as the concentration increased. All extracts showed higher retardation effects on glucose and bile acid compared to the control; particularly, WSPs showed a similar glucose retardation effect to carboxymethyl cellulose. This study suggests that WSP from C. sinensis can be used as a functional food material.

To confirm the possibility of seaweed extracts for functional food, water or ethyl alcohol solubles were extracted from laver, Porphyra yezoensis and evaluated those food components such as proteins, carbohydrates, nucleic acids, taurine, pigments and browning extent. The amount of proteins, polysaccharides and nucleic acids extracted decreased with increasing ethyl alcohol concentration, which was maximal when water was used as extraction solvent. The extractability of proteins, polysaccharides and nucleic acids was different between the dried and the roasted laver. Taurine was extracted about 1% from the dried and the roasted laver in the range of o~7o% ethyl alcohol concentration. The amount of carotenoids extracted by 95% ethyl alcohol from the dried and the roasted laver were 146.6 and 138.4mg%, respectively, which was 66 ~ 80% of yield extracted by methanol/acetone(1/1) solvent. The browning value of 50 ~6o% ethyl alcohol extraction group from roasted laver was highest among water/ethyl alcohol extraction group. The extraction yield was maximum when laver was extracted with water, and the value was 26.3% for the dried laver and 27.5% for the roasted layer. Organoleptic characteristics from four kinds of extraction groups containing hot water extraction showed that extracts from the roasted laver were evaluated most eminent and thought to be applicable to various preparation of food.
