Infection and Immune Response in the Nematode Caenorhabditis elegans Elicited by the Phytopathogen Xanthomonas

  • Xanthomonas oryzae pv. oryzae (Xoo) strains are plant pathogenic bacteria that can cause serious blight of rice, and their virulence towards plant host is complex, making it difficult to be elucidated. Caenorhabditis elegans has been used as a powerful model organism to simplify the host and pathogen system. However, whether the C. elegans is feasible for studying plant pathogens such as Xoo has not been explored. In the present work, we report that Xoo strains PXO99 and JXOIII reduce the lifespan of worms not through acute toxicity, but in an infectious manner; pathogens proliferate and persist in the intestinal lumen to cause marked anterior intestine distension. In addition, Xoo triggers (i) the p38 MAPK signal pathway to upregulate its downstream C17H12.8 expression, and (ii) the DAF-2/DAF-16 pathway to upregulate its downstream gene expressions of mtl-1 and sod-3 under the condition of daf-2 mutation. Our findings suggest that C. elegans can be used as a model to evaluate the virulence of Xoo phytopathogens to host.

A Cache Consistency Scheme to Consider Period of Data in Mobile Computing Environments (이동 컴퓨팅 환경에서 데이터의 주기성을 고려한 캐쉬 일관성 기법)

  • Due to the rapid development of wireless communication technology, demands on data services in mobile computing environment are gradually increasing. In mobile computing environments, a mobile host began to use cache to overcome the weak point. Caching at mobile host could reduce the bandwidth consumption and query response time, but mobile host must maintain cache consistency. In this paper, we propose a cache consistency strategy to consider the period of data usage. The server allows an effective broadcasting by classifying data into two groups of periodic and non-periodic. By classifying the data, it prevent from elimination of cache after disconnection of periodic data by using expired time. By storing IR(Invalidation Report) messages, this scheme divides cached data by selection after disconnection. Consequently, we have shown much improvement in total consumption of bandwidth than the conventional scheme.

  • Acute respiratory virus infectious diseases are a growing health problem, particularly among children and the elderly. Much effort has been made to develop probiotics that prevent influenza virus infections by enhancing innate immunity in the respiratory tract until vaccines are available. Lactobacillus plantarum GB-LP2, isolated from a traditional Korean fermented vegetable, has exhibited preventive effects on influenza virus infection in mice. To identify the molecular basis of this strain, we conducted a whole-genome assembly study. The single circular DNA chromosome of 3,284,304 bp was completely assembled and 3,250 protein-encoding genes were predicted. Evolutionarily accelerated genes related to the phenotypic trait of anti-infective activities for influenza virus were identified. These genes encode three integral membrane proteins, a teichoic acid export ATP-binding protein and a glucosamine - fructose-6-phosphate aminotransferase involved in host innate immunity, the nonspecific DNA-binding protein Dps, which protects bacteria from oxidative damage, and the response regulator of the three-component quorum-sensing regulatory system, which is related to the capacity of adhesion to the surface of the respiratory tract and competition with pathogens. This is the first study to identify the genetic backgrounds of the antiviral activity in L. plantarum strains. These findings provide insight into the anti-infective activities of L. plantarum and the development of preventive probiotics.

  • Viruses are obligate intracellular parasites which cause infection by invading and replicating within cells. The immune system has mechanisms which can attack the virus in extracellular and intracellular phase of life cycle, and which involve both non-specific and specific effectors. The survival of viruses depends on the survival of their hosts, and therefore the immune system and viruses have evolved together. Immune responses to viral infection may be variable depending on the site of infection, the mechanism of cell-to-cell spread of virus, physiology of the host, host genetic variation, and environmental condition. Viral infection of cells directly stimulates the production of interferons and they induce antiviral state in the surrounding cells. Complement system is also involved in the elimination of viruses and establishes the first line of defence with other non-specific immunity. During the course of viral infection, antibody is most effective at an early stage, especially before the virus enters its target cells. The virus- specific cytotoxic T lymphocytes are the principal effector cells in clearing established viral infections. But many viruses have resistant mechanism to host immune responses in every step of viral infection to cells. Some viruses have immune evasion mechanism and establish latency or persistency indefinitely. Furthermore antibodies to some viruses can enhance the disease by the second infection. Immune responses to viral infection are very different from those to bacterial infection.

Ultrastructures of Colletotrichum orbiculare in the Leaves of Cucumber Plants Expressing Induced Systemic Resistance Mediated by Glomus intraradices BEG110

  • The colonization of an arbuscular mycorrhizal fungus Glomus intraradices BEG110 in the soil caused a decrease in disease severity in cucumber plants after fungal inoculation with Colletotrichum orbiculare. In order to illustrate the resistance mechanism mediated by G. intraradices BEG110, infection patterns caused by C. orbiculare in the leaves of cucumber plants and the host cellular responses were characterized. These properties were characterized using transmission electron microscopy on the leaves of cucumber plants grown in soil colonized with G. intraradices BEG110. In the untreated plants, inter- and intra-cellular fungal hyphae were observed throughout the leaf tissues during both the biotrophic and necrotrophic phases of infection. The cytoplasm of fungal hyphae appeared intact during the biotrophic phase, suggesting no defense response against the fungus. However, several typical resistance responses were observed in the plants when treated with G. intraradices BEG110 including the formation of sheaths around the intracellular hyphae or a thickening of host cell walls. These observations suggest that the resistance mediated by G. intraradices BEG110 most often occurs in the symplast of the host cells rather than in the apoplast. In addition, this resistance is similar to those mediated by biotic inducers such as plant growth promoting rhizobacteria.

Concurrency Control Method using the Asynchronous Data Broadcast in Mobile Computing Environments (이동 컴퓨팅 환경에서 비동기적 데이터방송을 사용한 동시성 제어 기법)

  • In mobile computing environments, a mobile host caches the data to use the narrow bandwidth efficiently and improve the response time. A server periodically broadcasts the update data for maintaining the cache consistency of a mobile host. However, a method for maintaining cache consistency using periodic invalidation report has a problem that defers the commit decision of a mobile transaction to guarantee its serializable execution. In this paper, we propose CCM-AD method that guarantees the serializable execution of a mobile transaction even in the case that it is executed using cached data. As CCM-AD method guarantees the serializable execution of mobile transactions by using the intersection between the data broadcast to mobile host and the data updated at server. Then the CCM-AD method can reduce the delay of commitment decision of a mobile transaction. Also our method can reduce the size of invalidation report.

  • In case of environment requiring safety such as human body and requiring flexible shape, a conventional mechanical actuator system does not satisfy requirements. Therefore, in order to solve these problems. a research of various smart material such as EAP (Electro Active Polymer), EAC (Electro Active Ceramic) and SMA (Shape Memory Alloy) is in progress. Recently, the highest preferring material among various smart material is EP (Electrostictive Polymer), because it has very fast response time, powerful force and large displacement. The previous researches have been studied properties of polymer and simple control, but present researches are studied a polymer actuator. An EP (Electostrictive Polymer) actuator has properties which change variably ils shape and environmental condition. Therefore, in order to coincide with a user's purpose, it is important not only to decide a shape of actuator and mechanical design but also to investigate a efficient controller. In this paper, we constructed the control logic with an adaptive fuzzy algorithm which depends on the physical properties of EP that has a dielectric constant depending on time. It caused for a sub-actuator to operate at the same time that a sub-actuator system operation increase with a functional improvement and control efficiency improvement in each actuator, hence it becomes very important to manage it effectively and to control the sub-system which Is operated effectively. There is a limitation on the management of Main-host system which has multiple sub-system, hence it brings out the Multi-Vehicle Control process that disperse the task efficiently. Controlling the multi-dispersion system efficiently, it needs the research of Main-host system's scheduling, data interchange between sub-actuators, data interchange between Main-host system and sub-actuator system, and data communication process. Therefore in this papers, we compared the fuzzy controller with the adaptive fuzzy controller. also, we applied the scheduling method for efficient multi-control in EP Actuator and the algorithm with interchanging data, protocol design.

Impact of High-Level Expression of Heterologous Protein on Lactococcus lactis Host

  • The impact of overproduction of a heterologous protein on the metabolic system of host Lactococcus lactis was investigated. The protein expression profiles of L. lactis IL1403 containing two near-identical plasmids that expressed high- and low-level of the green fluorescent protein (GFP) were examined via shotgun proteomics. Analysis of the two strains via high-throughput LC-MS/MS proteomics identified the expression of 294 proteins. The relative amount of each protein in the proteome of both strains was determined by label-free quantification using the spectral counting method. Although expression level of most proteins were similar, several significant alterations in metabolic network were identified in the high GFP-producing strain. These changes include alterations in the pyruvate fermentation pathway, oxidative pentose phosphate pathway, and de novo synthesis pathway for pyrimidine RNA. Expression of enzymes for the synthesis of dTDP-rhamnose and N-acetylglucosamine from glucose was suppressed in the high GFP strain. In addition, enzymes involved in the amino acid synthesis or interconversion pathway were downregulated. The most noticeable changes in the high GFP-producing strain were a 3.4-fold increase in the expression of stress response and chaperone proteins and increase of caseinolytic peptidase family proteins. Characterization of these host expression changes witnessed during overexpression of GFP was might suggested the metabolic requirements and networks that may limit protein expression, and will aid in the future development of lactococcal hosts to produce more heterologous protein.

Trap Culture Technique for Propagation of Arbuscular Mycorrhizal Fungi using Different Host Plants

  • Arbuscular mycorrhizal fungi (AMF) spore propagation and long term maintenance is still a complicated technique for farmers. The use of AMF for their ability to promote plant growth and protect plants against pathogen attack and environmental stresses demands AMF propagation for large scale application. This study aimed to propagate AMF spores by trap culture technique and assess their ability to propagate with different host plants in a continuous plant cycle. Mycorrhizal inoculation by trap culture in maize resulted in longer shoots and roots than sudangrass plants. Increase in dry weight with higher percentage also was observed for maize plants. After first and second plant cycle, maize plants had the higher percentage of mycorrhizal response in terms of colonization and arbuscules than sudangrass. Maximum in spore count also achieved in the pots of maize plants. The results show that maize plant is more suitable host plant for AMF spore propagation and trap culture technique can be used effectively to maintain the AMF culture for long time.

A Study ou Iuternet Traffic Coutrol: Blockiug of harmful information based on IP spoofing (인터넷 트래픽 제어에 관한 연구: IP 주소 위조 기법을 사용한 유해 정보 차단 시스템)

  • In this paper, we propose a new system to block harmful Internet information based on IP spoofing. The proposed system is located on a organization's internal network and monitors all outgoing traffic and lets all this traffic go outside. Once the proposed system detects a host's access to a harmful site, it sends the host a pseudo RST packet that pretends to be the response from the harmful site, and prevents the connection between the host and the harmful site. The proposed software system is installed on only a server, and need not be installed on user hosts at all. Thus we can maintain and upgrade the blocking system easily. The performance evaluation of the proposed system shows that it effectively blocks the access to the harmful sites. Since the proposed system is based on IP spoofing, it can be used badly as a hacking tool. Finally we propose some methods to eliminate this possibility.
