Non-disturbing of Decidual Response by Steroid Hormonal Complexes of Pig Testis

  • Sex steroid hormones are key molecules to prepare the decidual response and their levels are important in this process. Imbalances of the levels of steroid hormones are cause of implantation failure and other diseases including physical weakness. Androgen replacement therapy or selective androgen receptor modulator are used to overcome various diseases but long-term use may cause of side effects. In previous report, it is suggested that the steroid hormonal complexes derived from pig enhance the proliferation of satellite cell. Therefore, to evaluate the possible usage of steroid hormonal complex derived from pig testis (tS-C), the effects of tS-C on uterine response were studied using the model of artificial decidua. tS-C did not disturb the rhythmical estrus cycle. Artificial-induced decidual response was normally induced in tS-C administered mice. The histological characters of the decidua of tS-C administered mice were not different from the vehicle. The expression patterns of molecular markers of decidua were not different between vehicle and tS-C group. Collectively these results suggested that tS-C does not disturb the uterine responsibility to the embryo. In addition, our results suggested that tS-C can be applied to overcome the various problems such as loss of muscle mass and anemia.

Hormonal Regulation of the Caprine $\beta$-Lactoglobulin Gene Promoter Activity (염소의 베타-락토글로불린 유전자 프로모터 활성의 호르몬에 의한 조절)

  • Expression of $\beta$-lactoglobulin gene in mammary tissue is strongly induced by lactogenic hormones such as prolactin, glucocorticoid, and insulin. In order to elucidate the regulatory mechanism underlying such hormonal induction, the response of the caprine $\beta$-lactoglobulin gene promoter to lactogenic hormones was analyzed in cultured HC11 mammary cells. Expression with serial deletions of the 5' -regulatory sequence of the $\beta$-lactoglobulin promoter revealed that two regions are responsible for a substantial change in hormonal indudbility. The region upstream of-1692, which exhibited strong repression of the downstream promoter, mediated the induction by insulin. This insulin-response was independent of the other two lactogenic hormones, prolactin and glucocorticoid. The other region from -740 to -470, which showed strong activation of the $\beta$-lactoglobulin promoter in confluent HC11 mammary cells, mediated mainly the response to a glucocorticoid analogue, dexametasone. The induction by the latter region, however, was suppressed by the usptream repression without insulin treatment. These results suggest that the induction of $\beta$-lactoglobulin promoter activity by lactogenic hormones in mammary cells may be achieved by the combined action of derepression by in sulin and activation by glucocorticoid and prolactin. Dexametasone response by the latter region seems to be mediated by the glucocorticoid receptor site around -7OObp.

Third-line Hormonal Therapy to Treat Prostate Cancer Relapse after Initial and Second-line Hormonal Therapy: Report of 52 Cases and Literature Review

  • The aim of this study was to evaluate the efficacy of third-line combined androgen blockade (CAB) therapy for castration-resistant prostate cancer that relapsed after primary and second-line CAB. We retrospectively reviewed the medical records of 52 patients who received first-, second-, and third-line CAB therapy (medical or surgical castration, plus steroidal antiandrogen of chlormadinone acetate, or nonsteroidal antiandrogen of flutamide or bicalutamide). For cumulative analysis, we searched the PubMed database and identified a total of 50 cases published in English. Including our cases, this provided a total of 102 cases for analysis. In our study cohort, 11 cases (21.2%) achieved more than 50% reduction of serum prostate-specific antigen (PSA) on initiation of third-line CAB. We found that third-line CAB with nonsteroidal antiandrogen after second-line CAB with steroidal antiandrogen exhibited favorable results, with a positive response in six of 13 patients (46.2%). Cumulative analysis findings were comparable. Regarding the timing of third-line CAB administration, 15 patients had started at a PSA equal to or less than 4.0 ng/ml, and eight of them (53.3%) showed a positive response to treatment, compared to only three of 37 patients (8.1%) whose PSA at the initiation of third-line therapy was higher than 4.0 ng/ml (p<0.001). We conclude that third-line CAB with nonsteroidal antiandrogen would be particularly useful for patients whose cancer progressed after second-line CAB with steroidal antiandrogen. The timing of treatment seems to be important because the higher the PSA at the start of third-line therapy, the lower the PSA response rate.

Hormonal Regulation of Acetyl-CoA Carboxylase Promoter I Activity in Rat Primary Hepatocytes (흰쥐의 간세포에서 호르몬에 의한 Acetyl-CoA Carboxylase Promoter I Activity 조절에 대한 연구)

  • Acetyl-CoA carboxylase (ACC) is the enzyme that controls no devo fatty acid biogynthesis, and this enzyme catalyzes the carboxylation pathway of acetyl-CoA to malonyl-CoA. Acetyl-CoA carboxylase gene expression was regulated by nutritional and hormonal status. The present study was performed to identify the regulation mechanism of ACC gene promoter I. The fragments of ACC promoter I -1.2-kb region wert recombined to pGL3-Basic vector with luciferase as a reporter gene. The primary hepatocytes from the rat were used to investigate the hormonal regulation of ACC promoter I activity. ACC PI (-1.2)/Luc plasmid was trtransferred into primary hepatocytes using lipofectin. Activity of luciferase was increased two-fold by 10-9M, three-fold by 10-8M, 10-6M, 3.5-fold by 10-6M, and 4.5-fold by 10-7M insulin treatment, respectively. In the presence of dexamethasone (1 $\mu$M), the effects of insulin increased about 1.5-fold, showing the additional effects of dexamethasone. Moreover, the activity of luciferase increased with insulin+dexamethasone, insulin+T3, dexamethasone+T3, and dexamethasone+insulin+T3 treatment approximately 6-, 4-, 6.5-, and 10-fold, respectively. Therefore it can be postulated that 1) these hormones coordinately regulate acetyl-CoA caroxylase gene expression via regulation of promoter activity, 2) the -1.2-kb region of ACC promoter I may have the response element sequences for insulin, dexamethasone, and T3.

  • Ecophysiologically diapause represents a syndrome of physiological and biochemical characteristics, all of which ensure survival during a long period of dormancy. Since, silkworm enters diapause as embryo at the early embryonic stage, the duration of egg life depends on the duration of embryonic diapause. The nature of diapause in silkworm, Bombyx mori, is primarily determined by genetic characters and endocrinologicnl mechanisms, mediated by environmental factors such as temperature and photoperiod. Hibernating potency value besides nucleic acid and carbohydrate metabolism, production and utilization of sorbitol are also equally responsible for induction, initiation, determination, maintenance and termination of diapause. Embryonic diapause in Bombyx moir, induced by active secretion of sub-oesophageal ganglion is attributed to hormonal system and metabolic adjustment, which serves to bring about a new physiological state. Metabolic conversion of trehalose to glycogen at induction, glycogen to sorbitol at initiation and sorbitol to glycogen at termination of diapause is correlated and in each metabolic shift a key enzyme becomes active in response to hormonal and environmental stimulation. An attempt has been made in this review article to discuss briefly the nature of embryonic diapause, influence of various factors on diapause nature, hormonal mechanism of diapause besides biochemical composition of egg, nucleic acid and carbohydrate metabolism, production and utilization of sorbitol in relation to induction, determination, maintenance, initiation and termination of diapause in the silkworm, Bombyx mori.

Thyroid Hormones Receptor/Reporter Gene Transcription Assay for Food Additives and Contaminants

  • Many of thyroid hormones disrupting chemicals induce effects via interaction with thyroid hormone and retinoic acid receptors and responsive elements intrinsic in target cells. We studied thyroid hormones disrupting effects of food additives and contaminants including BHA, BHT, ethoxyquin, propionic acid, sorbic acid, benzoic acid, CPM, aflatoxin B1, cadmium chloride, genistein, TCDD, PCBs and TDBE in recombinant HeLa cells containing plasmid construct for thyroxin responsive elements. The limit of response of the recombinant cells to T3 and T4 was $1\times10^{-12}\;M$. BHA. genistein, cadmium and TBDE were interacted with thyroid receptors with dose-responsive pattern. In addition, BHA, BHT, ethoxyquin, propionic acid, benzoic acid, sorbic acid, and TBDE showed synergism while cadmium chloride antagonism for T3-induced activity. This study elucidates that recombinant HeLa cell is sensitive and high-throughput system for the detection of chemicals that induce thyroid hormonal disruption via thyroid hormone receptors and responsive elements. Also this study raised suspect of BHA. BHT, ethoxyquin, propionic acid, benzoic acid, sorbic acid, TBDE, genisteine and cadmium chloride as thyroid hormonal system disruptors.

The Relationship Between the Expression of Estrogen Receptor ${\beta}$ and Recurrence in Breast Cancer (에스트로겐 수용체 ${\beta}$ 발현과 유방암 재발과의 관련성)

  • Background: It has been reported that estrogen receptor beta ($ER{\beta}$) mRNA expression was down-regulated during carcinogenesis and was inversely related to estrogen receptor alpha ($ER{\alpha}$) expression in breast cancer. The association of $ER{\beta}$ mRNA expression to tamoxifen resistance has also been reported. In this study, the expression of $ER{\alpha}$ and $ER{\beta}$ via immunohistochemistry (IHC) and reverse transcription-polymerase chain reaction (RT-PCR) was prompted, and an attempt was made to find out the relationship between $ER{\beta}$ expression and recurrence in the hormonal therapy group, and between $ER{\beta}$ expression and known prognostic factors. Methods: Tumor specimens were obtained at surgery from 67 female breast cancer patients during the period of September 1995 to December 2000. All the specimens were frozen in liquid nitrogen and kept at $-70^{\circ}C$ until they were used. The medical records were analyzed retrospectively. The expressions of ER were analyzed using IHC and RT-PCR methods. Results: The median follow-up was at 93.0 months (range: 14-157 months). The percentage of $ER{\alpha}+/ER{\beta}+$, $ER{\alpha}+/ER{\beta}-$, $ER{\alpha}-/ER{\beta}+$, and $ER{\alpha}-/ER{\beta}$ group were 35.9% 9.4%, 47.2%, and 7.5%, respectively, in 53 patients with hormonal therapy. $ER{\beta}$ was positive in 42 (82.3%) of 51 ER-positive patients. In the hormonal therapy group, the recurrence rates of each group was 15.8%, 0%, 40.0%, and 0%, respectively. In this group, the $ER{\beta}$ expression tended to recur, but there was no clinical significance (p=0.084). Conclusion: The $ER{\beta}$ expression may be a predictive marker of a poor response to endocrine therapy in breast cancer patients, although this needs to be confirmed in additional studies.

The Effect of Exogenous Hormone Treatment on Spermiation in Rhynchocypris oxycephalus (Sauvage and Dabry)

  • For the evaluation of hormonal control of spermiation in fish, a method to quanify the spermiation response or mature Rhynchocypris oxycephalus (Sauvage and Dabry) to hormonal therapy is described. Spermatocrit was determined after 7 min centrifugation at 18,000 $\times$g and sperm density was estimated by a standard hemocytormer method. Sperm density can be predicted from spermatocrit since their relationship is linear as described by the regression equation, Y=3.68X-27,18 ($R^2$=0.82, N=50), where Y is spermatocrit and X is sperm density. Milt production by mature R. oxycephalus was highest at 24 h after injection of 1,000 IU human chorionic gonadotropin (HCG) and 50 $\mu\textrm{g}$ luteinizing hormone-releasing hormone analogue (LHRHa) per kg body weight. Increased milt production coincided with low spermotocrit and sperm density levels. These results demonstrate that spermition in mature R. oxycephalus can be reliably evaluated by a spermatocrit method and that HCG and LHRHa are effecove in stimulating of spermiation in this species.

Relationship between urinary na, Ca Excretion and Hormonal Factors in Hypertensive and Normotensive Patients (고혈압과 정상혈압 환자에 있어서 Na, Ca 배설 및 관련 요인에 관한 연구)

  • The aim of this study was to investigate the interrelationship among urinary excretion dietary habit of Na, Ca intake and hormonal factors in 22 hypertensive and 30 normotensive hospitalized patients. The results were summarized as follows : 1. Urinary excretion of Na in normotensive patients was not significantly different from that of hypertensive patients, while Ca excretion(as expressed on the basis of mg of creatinine) was significantly higher in hypertensive patients. 2. Habitual dietary intake of Na, Ca in hypertensive patients were not significantly different from those of normotensive patients. 3. There were no significant differences in mean plasma renin activity, aldosterone and parathyroid hormone(PTH) level between two groups. However, systolic pressure significantly correlated with PTH(r=0.2597) and aldosterone level(r=0.24648)(P<0.05). In this study blood pressure did not show any significant relationship between urinary Na excretion and habitual dietary Na intake of Na. It is speculated that individual difference of Na sensitivity might result in heterogenous blood pressure response to dietary Na intake. Higher Ca excretion in hypertensive subjects suggested a future study on the interrelationship between Ca metabolism and aldosterone system in hypertension.

Induction of Lactation and Reproductive Response in Non-producing Buffalo Heifers Following Steroid Hormone Treatment

  • Buffalo heifers weighing from 400 - 500 kg and having a history of reproductive problems like anestrus, irregular estrus or failure to conceive after repeated inseminations were administered subcutaneously with estradiol-$17{\beta}$ and progesterone in two dosage rate 0.1 mg and 0.25 mg, respectively, per kg body weight per day for 7 days in experiment-I and 0.1 mg and 0.125 mg, respectively, per kg body weight per day for 7 days in experiment II. In experiment-I, 9 out of 10 buffaloes responded positively to the hormonal treatment. Milk secretion started between 14-20 days after the start of the treatment. The total milk yield in the successfully induced animals varied from 471.98-625.40 kg. The average daily milk yield varied from 2.08-2.76 kg and peak yield from 3.6-5.3 kg. The time taken to reach peak yield varied from 12-14 weeks. In experiment - II, the established lactation response was absent, although milk secretion process was initiated, the yield could not reach more than 50 - 100 gm at each milking. In experiment - I, the first estrus occurred between days 87 - 231 following the hormonal treatment. Four animals in which lactation was established successfully got pregnant after one or two services. In experiment - II the first estrus occurred between 85 - 173 days following the treatment and only one animal got pregnant.
